Injectable bone substitutes (IBS) are increasingly being used in the fields of orthopedics and maxillofacial/oral surgery. The rheological properties of IBS allow for proper and less invasive filling of bony defects. Vaterite is the most unstable crystalline polymorph of calcium carbonate and is known to be able to transform into hydroxyapatite upon contact with an organic fluid (e.g., interstitial body fluid). Two different concentrations of hydrogels based on poly(ethylene glycol)-acetal-dimethacrylat (PEG-a-DMA), i.e., 8% (w/v) (VH-A) or 10% (w/v) (VH-B), were combined with vaterite nanoparticles and implanted in subcutaneous pockets of BALB/c mice for 15 and 30 days. Explants were prepared for histochemical staining and immunohistochemical detection methods to determine macrophage polarization, and energy-dispersive X-ray analysis (EDX) to analyze elemental composition was used for the analysis. The histopathological analysis revealed a comparable moderate tissue reaction to the hydrogels mainly involving macrophages. Moreover, the hydrogels underwent a slow cellular infiltration, revealing a different degradation behavior compared to other IBS. The immunohistochemical detection showed that M1 macrophages were mainly found at the material surfaces being involved in the cell-mediated degradation and tissue integration, while M2 macrophages were predominantly found within the reactive connective tissue. Furthermore, the histomorphometrical analysis revealed balanced numbers of pro- and anti-inflammatory macrophages, demonstrating that both hydrogels are favorable materials for bone tissue regeneration. Finally, the EDX analysis showed a stepwise transformation of the vaterite particle into hydroxyapatite. Overall, the results of the present study demonstrate that hydrogels including nano-vaterite particles are biocompatible and suitable for bone tissue regeneration applications.

Die Bestrahlung von Kopf-Hals-Tumore ist ein zentrales Element der Therapie. Für Zinkoxid-Nanopartikel konnte bereits in zahlreichen Studien eine tumorzellschädigende Wirkung nachgewiesen werden. Ziel unserer Untersuchungen war es daher zu evaluieren, ob Zinkoxid-Nanopartikel als Strahlensensitizer in der Therapie von Kopf-Hals-Tumoren nutzbar gemacht werden können. Dazu wurden Tumorzellen mit 0, 2, 4 oder 8 Gray bestrahlt, nachdem sie Zinkoxid-Nanopartikeln ausgesetzt waren (4h, 100 µg/ml). Im Anschluss wurde ein MTT Assay, das die metabolische Aktivität der Zellen nach Behandlung misst, durchgeführt und zusätzlich ein Koloniebildungstest, der eine Quantifizierung des klonogenen Überlebens der Tumorzellen erlaubt. Die Kombination aus einer Bestrahlung mit zwei bzw. vier Gray, entsprechend typischerweise in der Klinik applizierten Einzeldosen, und einer Behandlung von Tumorzellen mit 100 µg/ml Zinkoxid-Nanopartikeln führte zu einer deutlichen Reduktion der Zellviabilität verglichen mit Zellen, die nur bestrahlt wurden. Auch eine Reduktion der Koloniebildung nach Nanopartikelbehandlung zusätzlich zur Bestrahlung mit zwei oder vier Gray konnte nachgewiesen werden. Damit konnte gezeigt werden, dass eine Inkubation mit Zinkoxid-Nanopartikeln, den Bestrahlungserfolg in vitro - gemessen an klonogenem Überleben und Zellviabilität der Tumorzellen - verbessert. Recently, copper selenides have shown to be promising thermoelectric materials due to their possible superionic character resulting from mobile copper cations. Inspired by this recent development in the class of quaternary copper selenides we have focused on the structure-to-property relationships in the solid solution series Cu_2ZnGeSe_(4-x)S_x. The material exhibits an insulator-to-metal transition at higher temperatures, with a transition temperature dependent on the sulfur content. However, the lattice parameters show linear thermal expansion at elevated temperatures only and therefore no indication of a structural phase transformation. ^(63)Cu nuclear magnetic resonance shows clear indications of Cu located on at least two distinct sites, which eventually merge into one (apparent) site above the phase transformation. In this manuscript the temperature dependent lattice parameters and electronic properties of the solid solution Cu_2ZnGeSe_(4-x)S_x are reported in combination with ^(63)Cu NMR, and an attempt will be made to relate the nature of the electronic phase transformation to a superionic phase transformation and a changing covalent character of the lattice upon anion substitution in this class of materials.

Eine zentrale Herausforderung des 21. Jahrhunderts ist die Reduktion von Treibhausgasen wie beispielsweise Kohlenstoffdioxid (CO2). Ein vielversprechender Ansatz hierfür ist die katalytische Umsetzung von CO2 zu Synthesegas (CO + H2) über die umgekehrte Wassergas-Shift-Reaktion. Nanopartikel sind ein wichtiges Forschungsgebiet aufgrund ihrer einzigartigen Eigenschaften und vielfältigen Anwendungsmöglichkeiten, einschließlich als Katalysatoren für die umgekehrte Wassergas-Shift-Reaktion. Im Fokus dieser Arbeit steht die Synthese von mono- und bimetallischen Nickel-Nanopartikeln und deren Aktivität für die umgekehrte Wassergas-Shift-Reaktion. Im ersten Teil dieser Arbeit wird das weithin bekannte thermische Reduktionsverfahren zur Herstellung von Nickel-Nanopartikeln vereinfacht und modifiziert. Die monometallischen Nickel-Nanopartikel werden eingehend charakterisiert und der Einfluss der Synthesebedingungen auf die finalen Nanopartikel sowie deren katalytische Aktivität für die Umwandlung von CO2 zu Synthesegas untersucht. In einem zweiten Teil wird das modifizierte Syntheseverfahren auf bimetallische Nickel-Nanopartikel durch Co-Reduktion von Nickel mit entweder Kupfer oder Kobalt übertragen. Die Nanopartikel werden eingehend untersucht und die Kern-Schale-Struktur der Nickel-Kupfer-Nanopartikel wird identifiziert und detailliert charakterisiert. Der Einfluss der Reaktionsparameter auf die finalen Nanopartikel und die Korrelation von Größe, Form und Kupfergehalt der Nanopartikel auf die Aktivität für die umgekehrte Wasser-Gas-Shift-Reaktion werden untersucht., A great challenge of the 21st century is the minimization of greenhouse gases such as CO2. A promising approach is the catalytic conversion of CO2 to synthesis gas (CO + H2) via the reverse water gas shift reaction. Nanoparticles are an important field of research due to their unique properties and variety of applications including catalysts for the reverse water gas shift reaction. This work focusses on the synthesis of mono- and bimetallic nickel nanoparticles and their activity for the reverse water gas shift reaction. In the first part of this work the wellknown thermal reduction procedure for the preparation of nickel nanoparticles is adapted and simplified. The monometallic nickel nanoparticles are characterized thoroughly and the impact of the synthesis conditions on the final nanoparticles and their catalytic activity for the conversion of CO2 to CO are investigated. In a second part the adapted synthesis procedure is transferred to bimetallic nickel nanoparticles by co-reduction of nickel with either copper or cobalt. The nanoparticles are investigated thoroughly and the core-shell structure of nickel copper nanoparticles is identified and characterized in detail. The impact of the reaction parameters on the final nanoparticles as well as the correlation of size, shape and copper content of the nanoparticles on the activity for the reverse water gas shift reaction is investigated.

Die Bildungsmechanismen von Metalloxiden, speziell der Eisen- und Aluminiumoxide, im wässrigen Medium sind aufgrund der Vielzahl kurzlebiger Intermediate und komplexen Reaktionspfaden nur wenig verstanden. Ein Verständnis der Reaktionsmechanismen kann jedoch mit Hilfe von zeitaufgelösten Untersuchungen wesentlich vertieft werden. Der Fokus dieser Arbeit liegt auf der Verwendung von zeitaufgelösten Untersuchungen, die größtenteils auf röntgenbasierenden Analyseverfahren beruhen, um die intermediären Phasen direkt unter realistischen Bedingungen zu identifizieren. Im ersten Teil dieser Arbeit erfolgte die Untersuchung der Reaktionsmechanismen von Eisenoxiden und -oxidhydroxiden direkt in einer stabilisator-unterstützten Synthese. Nach einer Optimierung der instrumentellen Aufbauten, wurden erstmals zeitaufgelöste, simultane Röntgenkleinwinkelstreu- und Röntgenabsorptionsspektroskopie-Experimente an einem Eisenoxid-System durchgeführt, indem ein akustischer Levitator als Probenhalter verwendet wurde. Als Unterstützung wurden weitere analytische Methoden verwendet, um ein Gesamtbild über die intermediären Spezies in den drei vorliegenden Reaktionsmechanismen zu erhalten. Mit Hilfe von Röntgenphotoelektronenspektroskopie in Kombination mit einem Mikrojet konnten Spezies, wie Eisen-oxo-Oligomere, als Intermediate in den frühen Stadien des Hydrolyseprozesses von Akaganeit-Nanopartikeln detektiert werden. Das erste Photoelektronenspektrum für diese Spezies konnte gezeigt werden. Im letzten Teil dieser Arbeit wurden zeitaufgelöste Untersuchungen an einem Aluminiumoxo- System durchgeführt. Es gelang erstmals durch die Kombination von Weitwinkelstreuexperimenten mit dem akustischen Levitator, die Kristallisation des Al13-Clusters in einer sulfathaltigen-Lösung als Aluminiumsulfat-Cluster Al13SO4 in situ zu detektieren., The formation mechanisms of metal oxides, especially iron and aluminium oxides, in aqueous media are poorly understood due to the large number of short-lived intermediates and complex reaction paths. However, an understanding of the reaction mechanisms can be considerably deepened with the help of time-resolved investigations. This work focuses on the use of time-resolved investigations, which rest largely on X-ray based analysis techniques, to identify the intermediate phases directly under realistic conditions. In the first part of this work, the reaction mechanisms of iron oxides and iron oxide hydroxides were investigated directly in a stabilizer-assisted synthesis. Time-resolved, simultaneous small-angle X-ray scattering and X-ray absorption spectroscopy experiments were performed on an iron oxide system for the first time using an acoustic levitator as sample holder after the optimisation of the instrumental set-ups. Further analytical methods were used to obtain detailed insights of the intermediate species in the three reaction mechanisms. Using X-ray photoelectron spectroscopy in combination with a microjet, species such as iron oxo oligomers could be detected as intermediates in the early stages of the hydrolysis process of Akaganeit nanoparticles. The first photoelectron spectrum for this species could be shown. In the last part of this work, time-resolved investigations were carried out on an aluminium oxo system. In situ-investigations were possible by combining wide-angle X-ray scattering experiments with the acoustic levitator. For the first time it was possible to detect the early stage crystallization of the Al13 cluster in a sulfate-containing solution as an aluminum sulfate cluster Al13SO4 directly in solution.

Fluorhaltige-Koordinationspolymere (FCPs) wurden durch mechanochemische Reaktionen synthetisiert. Die Erdalkalimetallhydroxide, die in ihrem Wassergehalt variieren, wurden als anorganische Quellen verwendet. Die perfluorierten Benzol Dicarboxylsäuren und ihre fluorfreien Analoga wurden als organische Linker verwendet. Die erhaltenen FCPs werden mit den fluorfreien Verbindungen (CPs) verglichen, die unter den gleichen Mahlbedingungen synthetisiert wurden. Der Austausch von Wasserstoff- durch Fluoratome beeinflusst sowohl die thermischen als auch die strukturellen Eigenschaften der FCPs. Letztere werden auch von den Unterschieden der Geometrien und von der Natur der organischen Linker beeinflusst. Während des Mahlprozesses dient Wasser dabei hauptsächlich als Vermittlermedium und zur Stabilisierung der neuen Strukturen. Die Unterschiede in den Kationengrößen zwischen Ca2+- und Sr2+-Ionen wirken sich geringfügig auf ihre Koordination durch die Dicarboxylatanionen aus. Hier kristallisieren Ca- und Sr-Verbindungen zumeist isomorph. Im Gegensatz dazu beeinflusst der große Ionenradius der Ba2+-Kationen die lokalen Koordinationsumgebungen. Die durch Mahlen erhaltenen Verbindungen sind hydratisiert und weisen eine kleine spezifische Oberfläche auf, die nach der thermischen Behandlung zunehmen kann. Die FCPs sind bis zu 300 °C stabil, während die CPs sich erst oberhalb von 400 °C zersetzen. Die hydratisierten Proben wandeln sich beim Tempern in neue dehydratisierte Phasen um. Die Phasenumwandlung kann reversible ablaufen. Zusätzlich wurde der Effekt des Austausches einer Carboxylgruppe durch eine Aminogruppe in einem organischen Liganden untersucht. In den CPs auf der Basis von Anthranilsäure beeinflussen die unterschiedlichen Kationengrößen der Ca2+-, Sr2+-, und Ba2+-Ionen sowohl die Koordinationsumgebung als auch die Dimensionalität der CPs.Die physikalisch-chemischen Eigenschaften der neuen Materialien wurden systematisch durch verschiedene analytische Techniken. A series of fluorinated coordination polymers (FCPs) were mechanochemically synthesized using alkaline earth metal hydroxides (M = Ca, Sr, Ba) that vary in their water content as inorganic sources. The perfluorinated benzene-dicarboxylic acids and their fluorine-free analogs were used as organic linkers. The obtained FCPs are compared to their synthesized fluorine-free counterparts (CPs) under the same conditions. The presence of fluorine influences both thermal and structural properties of the resulting FCPs. The latter are also strongly affected by the difference in geometries and nature of organic linkers. Water introduced to grinding acts as a mediator for the milling process and as a reactant for stabilizing the resulting structures. The difference in cation size between Ca2+- and Sr2+-ions has a minor effect on their coordination with perfluorinated or fluorine free benzene-dicarboxylate anions. Here, Ca- and Sr-compounds crystallize isomorphously (an exception was recorded for ortho-phthalate systems). In contrast, the relatively larger size of Ba2+-cation strongly influences the coordination environment. The obtained compounds by milling are hydrated and exhibit small surface areas that can increase after thermal post-treatment. The FCPs are stable up to 300 ˚C. On the other hand, the nonfluorinated CPs begin to decompose above 400 ˚C. The hydrated samples transform into new dehydrated phases upon thermal annealing. The hydrated-dehydrated phase transformation can be reversible. Moreover, the effect of replacement of one carboxylic group by an amino group in an organic ligand was explored. In the CPs based on anthranilic acid, the variations in cation size between Ca2+-, Sr2+-, and Ba2+-ions affect both coordination environment and dimensionality of the resulting CPs. The physicochemical properties of the new materials were systematically investigated applying different analytical techniques.

Im Rahmen dieser Dissertation wurden (I) erstmalig Metallphosphonate mechanochemisch synthetisiert, (II) 21 neue Strukturen mit Hilfe der Röntgenpulverdiffraktometrie gelöst und (III) die Reaktionsverläufe mittels in situ Methoden untersucht. Die Mechanochemie erwies sich als sehr geeignete Methode, um Metallphosphonate mit verschiedenen Metallen, Liganden und Strukturen darzustellen. Durch Variation des Verhältnisses der Edukte können die Reaktionen gesteuert werden. Etliche neue Metallphosphonat-Strukturen konnten mit Hilfe der erhaltenen Röntgenpulverdiffraktometrie-Daten gelöst werden. Dabei war auch die Aufklärung seltener Strukturmotive möglich. Durch die Resultate existiert nun eine schnelle, einfache und umweltfreundliche Alternative für die Herstellung von Metallphosphonaten. Im Rahmen dieser Arbeit wurden drei experimentelle Aufbauten für die in situ Untersuchung von Reaktionen entwickelt: (i) für mechanochemische Reaktionen mittels Synchrotron-Röntgenpulverdiffraktometrie und Raman-Spektroskopie, (ii) für mechanochemische Reaktionen mittels Thermographie und Raman-Spektroskopie sowie (iii) für Fällungsreaktionen bei Raumtemperatur mittels Synchrotron-Röntgenpulverdiffraktometrie. Durch die erhaltenen Ergebnisse konnten tiefgreifende Einblicke in die Bildungsmechanismen von Metallphosphonaten gewonnen werden. Für die mechanochemische Darstellung von Metallphenylphosphonaten wurde ein mehrstufiger Diffusionsmechanismus gefunden. Anhand der thermographischen Messungen wird deutlich, dass die Reaktionstemperatur einen großen Einfluss auf die Gesamttemperatur des Systems hat. Klassische Erklärungsmodelle für mechanochemische Reaktionen können für die hier untersuchten Systeme ausgeschlossen werden. Die in dieser Arbeit gewonnen Erkenntnisse über die Bildungsmechanismen und Strukturen der di- und tetravalenten Metallphosphonate liefern einen wichtigen Beitrag auf dem Weg zur zielgerichteten Darstellung von Metallphosphonaten., In this dissertation (I) metal phosphonates were mechanochemically synthesized for the first time, (II) 21 new structures were solved from powder X-ray diffraction data and (III) the reaction pathways were investigated with in situ methods. Mechanochemistry has shown to be very suitable for synthesizing metal phosphonates with different metals, ligands, and structures. By varying the ratio of the reactants, it is possible to control the reaction pathway. Several new metal phosphonate structures were solved from the powder X-ray diffraction data. It was also possible to elucidate rare structures. The results demonstrate a new fast, facile, and environmental friendly alternative for the preparation of metal phosphonates. During this dissertation three experimental setups for the in situ investigation of reactions were developed: (i) for mechanochemical reactions via synchrotron-powder X-ray diffraction and Raman spectroscopy, (ii) for mechanochemical reactions via thermography and Raman spectroscopy and (iii) for precipitation reactions via synchrotron-powder X-ray diffraction. The gained results provided an insight into the formation of metal phosphonates. For the mechanochemical synthesis of metal phenylphosphonates a multi-step diffusion mechanism was found. From the thermography studies, it is proven that the reaction temperature has a big impact on the temperature of the whole system. Classical theories for mechanochemical reactions can be ruled out for the investigated systems. The results gained in this thesis about the formation mechanisms and structures of di- and tetravalent metal phosphonates provide an important contribution for the targeted synthesis of metal phosphonates.
