Background Plant organelle genomes are a valuable resource for evolutionary biology research, yet their genome architectures, evolutionary patterns and environmental adaptations are poorly understood in many lineages. Rhodiola species is a type of flora mainly distributed in highland habitats, with high medicinal value. Here, we assembled the organelle genomes of three Rhodiola species (R. wallichiana, R. crenulata and R. sacra) collected from the Qinghai-Tibet plateau (QTP), and compared their genome structure, gene content, structural rearrangements, sequence transfer and sequence evolution rates. Results The results demonstrated the contrasting evolutionary pattern between plastomes and mitogenomes in three Rhodiola species, with the former possessing more conserved genome structure but faster evolutionary rates of sequence, while the latter exhibiting structural diversity but slower rates of sequence evolution. Some lineage-specific features were observed in Rhodiola mitogenomes, including chromosome fission, gene loss and structural rearrangement. Repeat element analysis shows that the repeats occurring between the two chromosomes may mediate the formation of multichromosomal structure in the mitogenomes of Rhodiola, and this multichromosomal structure may have recently formed. The identification of homologous sequences between plastomes and mitogenomes reveals several unidirectional protein-coding gene transfer events from chloroplasts to mitochondria. Moreover, we found that their organelle genomes contained multiple fragments of nuclear transposable elements (TEs) and exhibited different preferences for TEs insertion type. Genome-wide scans of positive selection identified one gene matR from the mitogenome. Since the matR is crucial for plant growth and development, as well as for respiration and stress responses, our findings suggest that matR may participate in the adaptive response of Rhodiola species to environmental stress of QTP. Conclusion The study analyzed the organelle genomes of three Rhodiola species and demonstrated the contrasting evolutionary pattern between plastomes and mitogenomes. Signals of positive selection were detected in the matR gene of Rhodiola mitogenomes, suggesting the potential role of this gene in Rhodiola adaptation to QTP. Together, the study is expected to enrich the genomic resources and provide valuable insights into the structural dynamics and sequence divergences of Rhodiola species.

BackgroundData on drug-coated balloons (DCB) for de novo coronary chronic total occlusion (CTO) are limited. We aimed to investigate the long-term outcomes of substitution of drug-eluting stents (DES) by DCB.MethodsWe compared the outcomes of less DES strategy (DCB alone or combined with DES) and DES-only strategy in treating de novo coronary CTO in this prospective, observational, multicenter study. The primary endpoints were major adverse cardiovascular events (MACE), target vessel revascularization, myocardial infarction, and death during 3-year follow-up. The secondary endpoints were late lumen loss (LLL) and restenosis until 1-year after operation.ResultsOf the 591 eligible patients consecutively enrolled between January 2015 and December 2019, 281 (290 lesions) were treated with DCB (DCB-only or combined with DES) and 310 (319 lesions) with DES only. In the DCB group, 147 (50.7%) lesions were treated using DCB-only, and the bailout stenting rate was relatively low (3.1%). The average stent length per lesion in the DCB group was significantly shorter compared with the DES-only group (21.5 ± 25.5 mm vs. 54.5 ± 26.0 mm, p p p p > 0.999). The Kaplan–Meier estimates of MACE at 3-year (11.8% vs. 12.0%, log-rank p = 0.688) was similar between the groups.ConclusionPercutaneous coronary intervention with DCB is a potential “stent-less” therapy for de novo CTO lesions with satisfactory long-term clinical results compared to the DES-only approach.

Solution equilibrium calculations were performed in this study to understand the impact of contaminant metal ions on the precipitation efficiency of selected rare earth elements (Ce3+, Nd3+, and Y3+) using oxalic acid as a precipitant. Trivalent metal ions, Al3+ and Fe3+, were found to considerably affect the precipitation efficiency of REEs. When Al3+ and Fe3+ concentrations were increased by 1 × 10−4 mol/L, in order to achieve an acceptable cerium recovery of 93% from solutions containing 1 × 10−4 mol/L Ce3+, oxalate dosage needed to increase by 1.2 × 10−4 and 1.68 × 10−4 mol/L, respectively. Such great impacts on the required oxalate dosage were also observed for Nd3+ and Y3+, which indicates that oxalic acid consumption and cost will be largely increased when the trivalent metal ions exist in REE-concentrated solutions. Effects of the divalent metal ions on the oxalate dosage is minimal. Furthermore, solution equilibrium calculation results showed that the precipitation of Fe3+ and Ca2+ (e.g., hematite and Ca(C2O4)∙H2O(s)) likely occurs during the oxalate precipitation of REEs at relatively high pH (e.g., pH 2.5), which will reduce rare earth oxalate product purity. In addition to the metal ions, anionic species, especially SO42−, were also found to negatively affect the precipitation recovery of REEs. For example, when 0.1 mol/L SO42− occurs in a solution containing 1 × 10−4 mol/L Ce3+ and 4 × 10−4 mol/L oxalate, the pH needed to be elevated from 2.0 to 3.3 to achieve the acceptable recovery. Overall, findings from this study provide guidance for the obtainment of high-purity rare earth products from solutions containing a considerable amount of contaminant metal ions by means of oxalic acid precipitation. Background Tendon is a major component of musculoskeletal system connecting the muscles to the bone. Tendon injuries are very common orthopedics problems leading to impeded motion. Up to now, there still lacks effective treatments for tendon diseases. Methods Tendon stem/progenitor cells (TSPCs) were isolated from the patellar tendons of SD rats. The expression levels of genes were evaluated by quantitative RT-PCR. Immunohistochemistry staining was performed to confirm the presence of tendon markers in tendon tissues. Bioinformatics analysis of data acquired by RNA-seq was used to find out the differentially expressed genes. Rat patellar tendon injury model was used to evaluate the effect of U0126 on tendon injury healing. Biomechanical testing was applied to evaluate the mechanical properties of newly formed tendon tissues. Results In this study, we have shown that ERK inhibitor U0126 rather PD98059 could effectively increase the expression of tendon-related genes and promote the tenogenesis of TSPCs in vitro. To explore the underlying mechanisms, RNA sequencing was performed to identify the molecular difference between U0126-treated and control TSPCs. The result showed that GDF6 was significantly increased by U0126, which is an important factor of the TGFβ superfamily regulating tendon development and tenogenesis. In addition, NBM (nonwoven-based gelatin/polycaprolactone membrane) which mimics the native microenvironment of the tendon tissue was used as an acellular scaffold to carry U0126. The results demonstrated that when NBM was used in combination with U0126, tendon healing was significantly promoted with better histological staining outcomes and mechanical properties. Conclusion Taken together, we have found U0126 promoted tenogenesis in TSPCs through activating GDF6, and NBM loaded with U0126 significantly promoted tendon defect healing, which provides a new treatment for tendon injury.

Background. The revascularization of small vessels using drug-eluting stents remains challenging. The use of the drug-coated balloon is an attractive therapeutic strategy in de novo lesions in small coronary vessels, particularly in the diabetic group. This study aimed to assess the outcomes of DCB-only angioplasty in small vessel disease. Methods. A total of 1198 patients with small vessel disease treated with DCB-only strategy were followed. Patients were divided into the diabetic and nondiabetic groups. Clinical and angiographical follow-up were organized at 12 months. The primary endpoints were target lesion failure and secondary major adverse cardiac events. Results. There was a significantly higher rate of target lesion failure among diabetic patients compared to nondiabetic [17 (3.9%) vs. 11 (1.4%), P = 0.006 ], taken separately, the rate of target lesion revascularization significantly differed between groups with a higher rate observed in the diabetic group [9 (2%) vs. 4 (0.5%), P = 0.014 ]. Diabetes mellitus remained an independent predictor for TLF (HR: 2.712, CI: 1.254–5.864, P = 0.011 ) and target lesion revascularization (HR: 3.698, CI: 1.112–12.298, P = 0.033 ) after adjustment. However, no significant differences were observed between groups regarding the target vessel myocardial infarction (0.6% vs. 0.1%, P = 0.110 ) and MACE [19 (4.4%) vs. 21 (2.7%), P = 0.120 ]. Conclusion. Drug-coated balloon-only treatment achieved lower incidence rates of TLF and MACE. Diabetes is an independent predictor for target lesion failure and target lesion revascularization at one year following DCB treatment in small coronary vessels. We observed no significant differences between groups regarding MACE in one year.

REE recovery from a kaolinite sample was conducted in the present study. The results of mineralogical analyses, salt leaching, and acid leaching tests confirmed that REEs predominantly existed as florencite (REEAl3(PO4)2(OH)6) in the material, and the proportion of ion-exchangeable and ion-substituted REEs was negligible. Mechanical grinding for 4 h completely destroyed the crystal structure of the material, but the recovery of total REEs was less than 20%, indicating that insufficient liberation of the REE-bearing particles is not the only factor leading to the low recovery. Thermal treatment of the raw material at 600 °C for 2 h largely enhanced the total REE recovery to as high as 92%. The improved recovery was ascribed to the thermal decomposition of crystallized florencite into an amorphous phase, which was proved through transmission electron microscopy - energy dispersive X-ray analysis. Findings from the study will contribute to REE recovery and comprehensive utilization of kaolinite.

Brassinosteroids (BRs) play essential roles in regulating plant growth and development, however, gaps still remain in our understanding of the BR signaling network. We previously cloned a grain length quantitative trait locus qGL3, encoding a rice (Oryza sativa L.) protein phosphatase with Kelch-like repeat domain (OsPPKL1), that negatively regulates grain length and BR signaling. To further explore the BR signaling network, we performed phosphoproteomic analysis to screen qGL3-regulated downstream components. We selected a 14-3-3 protein OsGF14b from the phosphoproteomic data for further analysis. qGL3 promoted the phosphorylation of OsGF14b and induced the interaction intensity between OsGF14b and OsBZR1. In addition, phosphorylation of OsGF14b played an important role in regulating nucleocytoplasmic shuttling of OsBZR1. The serine acids (Ser258Ser259) residues of OsGF14b play an essential role in BR-mediated responses and plant development. Genetic and molecular analyses indicated that OsGF14b functions as a negative regulator in BR signaling and represses the transcriptional activation activity of OsBZR1. Collectively, these results demonstrate that qGL3 induces the phosphorylation of OsGF14b, which modulates nucleocytoplasmic shuttling and transcriptional activation activity of OsBZR1, to eventually negatively regulate BR signaling and grain length in rice.

A kaolinite containing 245 ppm of REEs was used as a feedstock for REE recovery. Acid leaching test results showed that less than 20% of the REEs were leached using 1.2 mol/L HCl. However, after thermal treatment at 600 °C, a recovery of over 80% was obtained with a much lower acid concentration (0.01 mol/L). Meanwhile, only 3.91% Al, 2.34% Fe, 8.63% K, 9.32% Mg, 0.08% Ti, and 5.34% Ca were simultaneously extracted. Therefore, high-purity rare earth products are likely produced from the kaolinite. To achieve a more comprehensive utilization of the kaolinite, the residual solid after REE recovery was subjected to a selective leaching process to prepare porous silica. Comparisons among three different types of mineral acids showed that H2SO4 provided the best performance. The effects of different factors were investigated using the response surface methodology. A porous silica with as high as 336.95 m2/g specific surface area was ultimately produced.

Although drug-eluting stents are the most common interventional devices for patients with coronary disease, drug-coated balloons (DCBs) represent a novel therapeutic alternative in certain scenarios. This prospective, observational all-comers study explored the clinical outcomes of DCB use in patients with coronary lesions. All patients treated with DCBs were enrolled in this study, including patients with in-stent restenosis (ISR) or de novo lesions. The primary outcome was the target lesion revascularization (TLR) rate at one year. We enrolled 2306 patients with 2660 lesions and performed DCB angioplasty in 399 patients (17.3%) with ISR and 1907 patients (82.7%) with de novo lesions. Saxifraga species are widely distributed in alpine and arctic regions in the Northern hemisphere. Highly morphological diversity within this genus brings great difficulties for species identification, and their typical highland living properties make it interesting how they adapt to the extreme environment. Here, we newly generated the chloroplast (cp) genomes of two Saxifraga species and compared them with another five Saxifraga cp genomes to understand the characteristics of cp genomes and their potential roles in highland adaptation. The genome size, structure, gene content, GC content, and codon usage pattern were found to be highly similar. Cp genomes ranged from 146,549 bp to 151,066 bp in length, most of which comprised 130 predicted genes. Yet, due to the expansion of IR regions, the second copy of rps19 in Saxifraga stolonifera was uniquely kept. Through sequence divergence analysis, we identified seven hypervariable regions and detected some signatures of regularity associated with genetic distance. We also identified 52 to 89 SSRs and some long repeats among seven Saxifraga species. Both ML and BI phylogenetic analyses confirmed that seven Saxifraga species formed a monophyletic clade in the Saxifragaceae family, and their intragenus relationship was also well supported. Additionally, the ndhI and ycf1 genes were considered under positive selection in species inhabiting relatively high altitudes. Given the conditions of intense light and low CO2 concentration in the highland, the products of these two genes might participate in the adaptation to the extreme environment.

New strategies that enhance both the targetability of chemotherapy drugs and the synergistic effects of chemotherapy and immunotherapy are urgently needed for efficacious solid tumor therapy. In this study, a novel biomimetic nanoparticle system possessing the properties of tumor targeting and immune synergy was designed to meet these requirements. Mesoporous silica nanoparticles loaded with the chemotherapeutic drug doxorubicin (DOX) were coated with cell membranes modified by glycosylphosphatidylinositol (GPI)-anchored anti-HER2 single chain variable fragment (scFv) and the GPI-anchored co-stimulatory molecule CD80 (to promote solid tumor-targeted chemotherapy and cooperated immunotherapy, respectively). The impact of the nanotherapeutic system on both tumor-targeted chemotherapy and cellular immune response was investigated through in vitro and in vivo experiments. The results show that the novel biomimetic therapeutic system effectively promoted antitumor efficiency in vitro and in vivo. Osteoporotic fractures, also known as fragility fractures, are prevalent in the elderly and bring tremendous social burdens. Poor bone quality, weak repair capacity, instability, and high failure rate of internal fixation are main characteristics of osteoporotic fractures. Osteoporotic bone defects are common and need to be repaired by appropriate materials. Proximal humerus, distal radius, tibia plateau, calcaneus, and spine are common osteoporotic fractures with bone defect. Here, the consensus from the Osteoporosis Group of Chinese Orthopaedic Association concentrates on the epidemiology, characters, and management strategies of common osteoporotic fractures with bone defect to standardize clinical practice in bone repair of osteoporotic fractures.

Structural members and kinematic pairs made of rigid materials can be flexibly used in practical applications and obtain shape-changing mechanisms. However, it also leads to a relatively complex mechanical structure and overmuch kinematic pairs, which increases the difficulty of controlling the mechanism and thus limits its application. The emergence of 4D printing technology and smart materials provides new ideas for simplifying shape-changing mechanism structure and innovation. Although smart materials are already being used in many types of mechanisms, they are almost always unique cases. How to organically combine with a specific mechanism, innovate the type of mechanism, and systematically research from structural design to deformation control is still a blank area. Therefore, this paper designs a type of novel mechanism consisting of an angulated scissor unit and a form-controlled unit, the form-controlled planar scaling mechanism, which can achieve drivable self-adjustment of size ratio under predetermined thermal stimulation. The shape-changing mechanism is thus extended to a form-controlled mechanism. This mechanism's construction method and structural principles are analyzed, and the critical structural design parameters required for deformation control are derived. Based on the above research, the core form-controlled unit that controls the deformation of this type of mechanism is designed structurally and extended to deformation control research. Finally, two sets of prototype machines are manufactured and verified the feasibility of the construction method and deformation control scheme and the correctness of the theoretical analysis. This type of mechanism promotes the innovation of mechanism design methods. It shows many application prospects in satellite deployment structures and space probe walking systems.

In this study, a ternary blend modifier (TBM) was prepared with RPE, RPP, and various dosages of PP-g-MAH (i.e., 0%, 1%, 3%, 5%, 7%, and 9% by weight of RPE and RPP) by a twin-screw extruder. The performance of TBM (4% by weight of asphalt) modified asphalt was studied, with the increase of PP-g-MAH content, the penetration decreases gradually, but softening point and viscosity increases and ductility exists minimum value at 7% content, with the increasing of PP-g-MAH percentage, the dynamic rheological properties show that |G*| increases, while the δ and |G*|cosδ decrease, and the fatigue performance (|G*|sinδ) of TBM modified asphalt is worse than virgin asphalt. Dynamic stability test shows that TBM shows the prominent high-temperature properties of a modified (asphalt mixture. BBR test indicates that the m-value of asphalt modified with TBM does not fulfill the requirements (m-value>0.3) except at 5% and 7%, but the S meets the standard requirements (S < 300 MPa), and increases from 0% to 7%, then decreases slightly. In order to illuminate possible reasons, a chemical reaction (ester compounds generated) and physical twines of concerned polymer molecules were put forward, and the results were confirmed by infrared analysis.

Lactic acid production from food waste via fermentation is environmentally sustainable. However, the characteristics of food waste fermentation to produce lactic acid are not well understood due to the complexity of food waste. This study aims to understand the effects of key variables on the characteristics of food waste fermentation to maximize lactic acid production. Food waste was enzymatically hydrolyzed and fermented by Lactobacillus pentosus. Key fermentation variables, including nitrogenous nutrient supplementation, initial sugar concentration, and pH, were investigated in batch fermentation to unveil their effects on fermentation titer, yield, and productivity. The results showed that supplementation of 0.25% (w/v%) yeast extract and peptone to the food waste fermentation media significantly improved fermentation titer and productivity, but further increase in the supplementation level did not improve fermentation. Increasing the initial sugar concentration from 40 g/L to 100 g/L increased the fermentation titer from 41.0 g/L to 93.0 g/L and productivity from 0.34 g/L/h to 0.76 g/L/h. pH 6.0 was the optimal pH for the fermentation. At the optimal conditions, food waste fermentation resulted in the highest fermentation titer, yield, and productivity of 106.7 g/L, 1.12 g/g, and 3.09 g/L/h, respectively. The high fermentation yield of 1.12 g/g might be explained by the extra lactic acid production from unidentified compounds in food waste hydrolysates. By applying fed-batch fermentation, the lactic acid concentration reached 157.0 g/L with a yield and overall productivity of 0.92 g/g and 2.0 g/L/h, respectively. Based on the mass balance, a total of 251 kg lactic acid was produced from 1000 kg food waste. PRACTICAL APPLICATION: Food waste is one of the largest municipal solid wastes in the US, and most food waste ends up in landfills, causing significant economic losses and environmental concerns. In this study, we developed a fermentation process to convert food waste into biorenewable lactic acid and demonstrated that food waste is a superior feedstock for fermentation due to its embedded nutrients. Moreover, due to the embedded nutrients in food waste, the supplementation of yeast extract and peptone to fermentation can be reduced by over 50%, which can reduce the operating cost of lactic acid fermentation on an industrial scale.

MotivationAccurate identification of microRNA (miRNA) targets at base-pair resolution has been an open problem for over a decade. The recent discovery of miRNA isoforms (isomiRs) adds more complexity to this problem. Despite the existence of many methods, none considers isomiRs, and their performance is still suboptimal. We hypothesize that by taking the isomiR-mRNA interaction into account and applying a deep learning model to study miRNA-mRNA interaction features, we may improve the accuracy of miRNA target predictions.ResultsWe developed a deep learning tool called DMISO to capture the intricate features of miRNA/isomiR-mRNA interactions. Based on 10-fold cross-validation, DMISO showed high precision (95%) and recall (90%). Evaluated on three independent datasets, DMISO had superior performance to five tools, including three popular conventional tools and two recently developed deep learning-based tools. Exploring the transformation of spatial mechanisms from their unfolded to controlled folding states to meet the requirements of various application scenarios has long been a hot topic in mechanical structure research. Although conventional spatial mechanisms can be designed to meet almost any application scenario, the design’s complex and excessive combinations of structural components, kinematic pairs, and drive units are unavoidable. It introduces many problems, such as poor reliability, drive complexity, and control difficulties. Based on 4D printing technology, the design of self-folding spatial mechanisms that use pre-stressed response properties under predetermined thermal excitation to achieve different shrinkage ratios integrates the control and drive system and the structural components and kinematic pairs. It brings novel features of self-folding while effectively avoiding many problems associated with conventional mechanical design. Further, the pre-stressed response model introduces the self-folding spatial mechanisms’ excitation, morphing, and driving investigation. Self-folding spatial mechanisms with different shrinkage ratios were prepared via fused deposition modeling, which verified the theoretical analysis and pre-stress response model and the design’s correctness and feasibility by experiments. The existing 4D printing technology lacks a paradigmatic design method in the application field. Contrarily, this work organically combined the conventional mechanical structure design with materials and fabrication via fused deposition modeling. A systematic study of self-folding spatial mechanisms from structural design to morphing control was carried out. This design is expected to introduce a novel paradigm of 4D printing technology in conventional mechanical design and has considerable application prospects in spherical radar calibration mechanisms.

REE recovery tests were performed on a kaolinite and the corresponding metakaolinite using pH static leaching method. Test results show that over 90% of REEs were leached from the metakaolinite sample at pH 0.00 and 25 °C, while less than 2% of the major constituents were simultaneously extracted. Therefore, selective leaching of REEs from the metakaolinite was achieved through pH static leaching. The leaching residue was then subjected to alkaline activation for zeolites synthesis. The effects of hydrothermal temperature and incubation period under various alkaline conditions on the synthesis of zeolites were systematically investigated. The phase compositions, textual properties, and morphology of the synthesized products were characterized. Pure zeolite A with 100% relative crystallinity was successfully synthesized at 80 °C for 6 h when using 3 M NaOH as the alkaline activator. While as the synthesis conditions became increasingly harsh, the metastable zeolite A gradually transformed into more stable sodalite, and three types of zeolites, including zeolite A, sodalite, and their mixtures, were obtained. Due to the increasing criticality of rare earth elements (REEs), it has become essential to recover REEs from alternative resources. In this study, systematic REEs leaching tests were performed on the calcination product of a coal coarse refuse using hydrochloric acid and different types of organic acid as lixiviants. Experimental results show that the recovery of REEs, especially heavy REEs (HREEs) and scandium (Sc), is improved by using selected organic acids. Citric acid and dl -malic acid afford the best leaching performances; whereas, malonic acid, oxalic acid, and dl -tartaric acid are inferior to hydrochloric acid. Results of zeta potential measurements and solution chemical equilibrium calculations show that malonic acid is more likely adsorbed on the surface of the calcined material compared with citric acid and dl -malic acid. The adsorption may reduce the effective concentration of malonic species in solution and/or increase the amount of REEs adsorbed on the surface, thereby impairing the leaching recovery. Compared with light REEs (LREEs), a stronger adsorption of the HREEs on the surface is observed from electro-kinetic test results. This finding explains why organic acids impose a more positive impact on the leaching recovery of HREEs. By complexing with the HREEs, organic acids can keep the metal ions in solution and improve the leaching recovery. The adsorption of Sc3+ on the surface is the lowest compared with other REEs. Therefore, rather than complexing, the organic anionic species likely play a function of solubilizing Sc from the solid, which is similar to that of hydrogen ions.

Aiming at the problem of unmanned reconfiguration and docking of ground vehicles under complex working conditions, we designed a piece of docking equipment composed of an active mechanism based on a six-degree-of-freedom platform and a locking mechanism with multi-sensors. Through the proposed control method based on laser and image sensor information fusion calculation, the six-dimensional posture information of the mechanism during the docking process is captured in real time so as to achieve high-precision docking. Finally, the effectiveness of the method and the feasibility of the 6-DOF platform are verified by the established model. The results show that the mechanism can meet the requirements of smooth docking of ground unmanned vehicles.

Recovery of rare earth elements (REE) from coal-related resources has recently received significant interest due to supply concerns and their key roles in critical industries and defense-related technologies. An integrated flowsheet consisting of sorting, crushing/grinding, physical separation, acid leaching, solvent extraction, and selective precipitation was designed to achieve continuous production of rare earth products from various coal-related resources such as coarse refuse and acid mine drainage. A pilot plant was constructed that enabled continuous testing of the circuitry on a range of different coal-based feedstocks. Despite achieving significant success in producing high-grade rare earth oxide products, low REE recovery values and high operational costs necessitated a re-evaluation of the process flowsheet. This article describes the circuitry improvements that were based on both laboratory and pilot plant test results. Roasting as a pre-leach treatment was found to significantly improve the leaching rate and overall REE recovery values. After roasting West Kentucky No. 13 and Fire Clay coal refuse materials at 600 °C, REE recovery increased by 60 and 40 absolute percentage points, respectively. Rare earth concentration in the pregnant leachate solution (PLS) was increased to around 700 ppm prior to solvent extraction using a staged precipitation method. REEs in the precipitate were re-dissolved, purified, and extracted from the PLS using solvent extraction and selective precipitation to values of around 80%. As a result of the improvements, chemical consumption in the acid leaching and solvent extraction processes was significantly reduced. A techno-economical assessment of the improved flowsheet indicated significant commercialization potential.

It is critical to separate carbon from fly ash due to its increasing amount being produced per year and component requirements for secondary utilizations. Flotation is an efficient technique to fulfill this task. Effects of diesel emulsification using sodium petroleum sulfonate on unburned carbon flotation from a fly ash sample were studied. Flotation tests showed that carbon recovery was increased from 41% to 86% using the emulsified collector at the same dosage level (0.5 kg/ton) as diesel. Zeta potential measurements indicated that both the carbon and emulsified collector droplets carried negative charges within the studied pH range. For fly ash particles with high carbon content, adsorption bands of carbonyl bond (C O) were detected in the FTIR spectra, which agreed with the XPS characterization findings. XPS analysis indicated that the emulsified diesel (ED) collector interacted more intensively with the surface carbon-oxygen groups relative to the normal diesel collector. Finally, the improved efficiency of ED is proved to be related to its polar groups and reduced oil droplet size. The polar groups of ED (mainly sulfonate) interacted with the surface oxygen containing groups (e.g., C O and COOH) through hydrogen bonding and Lewis acid base interaction.

Coronavirus disease 2019 (COVID-19) has been spreading rapidly and is still threatening human health currently. A series of measures for restraining epidemic spreading has been adopted throughout the world, which seriously impacted the gross domestic product (GDP) globally. However, details of the changes in the GDP and its spatial heterogeneity characteristics on a fine scale worldwide during the pandemic are still uncertain. We designed a novel scheme to simulate a 0.1° × 0.1° resolution grid global GDP map during the COVID-19 pandemic. Simulated nighttime-light remotely sensed data (SNTL) was forecasted via a GM(1, 1) model under the assumption that there was no COVID-19 epidemic in 2020. We constructed a geographically weighted regression (GWR) model to determine the quantitative relationship between the variation of nighttime light (ΔNTL) and the variation of GDP (ΔGDP). The scheme can detect and explain the spatial heterogeneity of ΔGDP at the grid scale. It is found that a series of policies played an obvious role in affecting GDP. This work demonstrated that the global GDP, except for in a few countries, represented a remarkably decreasing trend, whereas the ΔGDP exhibited significant differences.

Deregulation of gene expression is associated with the pathogenesis of numerous human diseases including cancer. Current data analyses on gene expression are mostly focused on differential gene/transcript expression in big data-driven studies. However, a poor connection to the proteome changes is a widespread problem in current data analyses. This is partly due to the complexity of gene regulatory pathways at the post-transcriptional level. In this study, we overcome these limitations and introduce a graph-based learning model, PTNet, which simulates the microRNAs (miRNAs) that regulate gene expression post-transcriptionally in silico. Our model does not require large-scale proteomics studies to measure the protein expression and can successfully predict the protein levels by considering the miRNA–mRNA interaction network, the mRNA expression, and the miRNA expression. Large-scale experiments on simulations and real cancer high-throughput datasets using PTNet validated that (i) the miRNA-mediated interaction network affects the abundance of corresponding proteins and (ii) the predicted protein expression has a higher correlation with the proteomics data (ground-truth) than the mRNA expression data. The classification performance also shows that the predicted protein expression has an improved prediction power on cancer outcomes compared to the prediction done by the mRNA expression data only or considering both mRNA and miRNA. Although the neural correlates of novelty and appropriateness of creative insight during cognitive tasks have been investigated in several studies, they have not been examined during mental distress in a psychotherapeutic setting. This study aimed to reveal the promoting effects of novelty and appropriateness processing on therapeutic insight in a micro-psychotherapeutic setting. We examined the effects of appropriateness (between-subject factor: appropriateness group, 20 participants; inappropriateness group, 21 participants) by manipulating the preceding negative scenarios that either fit or did not fit the subsequent solutions, and those of novelty (within-subject factor) by varying the linguistic expressions for describing solutions (metaphorical, literal, or problem-restatement). Event-related functional magnetic resonance images were collected. We found the following effects: an interactive effect of the two factors on insightfulness and activation in the bilateral hippocampus and amygdala, right superior frontal gyrus, and left superior/middle temporal gyrus; a simple effect of novelty on activation in the bilateral inferior frontal gyrus, fusiform gyrus, and inferior/middle occipital gyrus; and a simple effect of appropriateness on activation in the left inferior parietal lobule. Our findings indicate that solutions with high novelty and appropriateness generate the highest levels of therapeutic insightfulness as well as the strongest activation in the hippocampus and amygdala, which may be involved in episodic memory encoding.

High expression of long intergenic non-protein coding RNA 987 (LINC00987) is strongly associated with low overall survival of osteosarcoma; however, its role in osteosarcoma remains unclear. This study explored the biological function and underlying mechanism of LINC00987 in osteosarcoma. In this study, LINC00987 expression in osteosarcoma cells was analyzed using Cancer Cell Line Encyclopedia and qRT-PCR. The proliferation and migration and invasion in osteosarcoma cells were evaluated using Cell Counting Kit-8 and Transwell assays, respectively. Bioinformatic analysis was used to predict the LINC00987-bound miRNAs and miR-376a-5p-bound mRNAs. Dual-luciferase reporter assays were used to assess the interaction between miR-376a-5p, LINC00987, and forming-binding protein 1 (FNBP1). FNBP1 expression was measured by western blotting. LINC00987 was found to be upregulated in osteosarcoma cells. LINC00987 silencing suppressed proliferation, migration, and invasion of osteosarcoma cells. Additionally, miR-376a-5p expression was downregulated in osteosarcoma cells. miR-376a-5p knockdown reversed the effect of LINC00987 silencing on the biological function of osteosarcoma cell. miR-376a-5p was found to target LINC00987 and FNBP1. FNBP1protein level was increased in osteosarcoma cells; however, it was inhibited by silencing LINC00987 and enhanced by silencing miR-376a-5p. In conclusions, this study suggests LINC00987 silencing inhibits osteosarcoma cell proliferation, migration, and invasion by sponging miR-376a-5p to regulate FNBP1 expression. LINC00987 as a potential therapeutic target for osteosarcoma.

Neonicotinoids (NEOs) are extensively applied in global agricultural production for pest control but have adverse effects on human health. In this study, the concentrations of six NEOs and three characteristic metabolites were investigated by collecting 200 serum samples from an elderly population in China. Results showed that the NEOs and their metabolites were widely detected (89%-98 %) in the serum samples from the osteoporosis (OP) (n = 120) and non-OP (n = 80) population, and their median concentrations ranged from 0.04 ng/mL to 5.99 ng/mL and 0.01 ng/mL to 2.02 ng/mL, respectively. N-desmethyl-acetamiprid (ACE-dm) was the most abundant NEOs in the serum samples. Gender-related differences were found in concentrations of most NEOs and their metabolites in serum, with males having higher target analytes than females. Significantly (p 0.05) positive correlations were observed among most NEO concentrations, suggesting that exposure source of these substances is common or related. However, associations between the concentrations of characteristic metabolites and their corresponding NEOs were insignificant, probably because the exogenous intake are the primary sources of metabolites of NEOs instead of the internal biotransformation. The associations between NEO concentrations (i.e., ACE-dm, dinotefuran, and olefin-imidacloprid) and OP (OR = 2.33-6.92, 95 % CI = 0.37-16.9, p-trend 0.05) indicate that NEO exposure is correlated with increased odds of prevalent OP. This study is the first to document the profiles of NEOs and their metabolites in serum samples collected from an elderly population in South China and examine the relationships between NEO exposure and OP.
