Backgroud: Both alcohol consumption and the proprotein convertase subtilisin/kexin type 9 (PCSK9) gene polymorphism modulate serum lipid levels, but their interactions on serum lipid profiles are still unknown. The present study was undertaken to detect the interactions of PCSK9 E670G polymorphism and alcohol consumption on serum lipid levels. Methods: Genotypes of the PCSK9 E670G in 1352 unrelated subjects (785 non-drinkers and 567 drinkers) were determined by polymerase chain reaction and restriction fragment length polymorphism combined with gel electrophoresis, and then confirmed by direct sequencing. The interactions between PCSK9 E670G genotypes and alcohol consumption on serum lipid parameters were detected by using a factorial design covariance analysis after controlling for potential confounders. Results: The levels of serum triglyceride, high-density lipoprotein cholesterol, apolipoprotein (Apo) A1, and the ratio of ApoA1 to ApoB were higher in drinkers than in non-drinkers (P < 0.01 for all), whereas the levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and ApoB were lower in drinkers than in non-drinkers (P < 0.001 for all). The genotypic and allelic frequencies of PCSK9 E670G were not different between non-drinkers and drinkers (P > 0.05 for each). The subjects with AA genotype in non-drinkers had higher serum LDL-C levels than the subjects with AG genotype, whereas the subjects with AG genotype in drinkers had higher serum TC levels than the subjects with AA genotypes (P < 0.05 for each). The effects of alcohol consumption on TC and LDL-C levels depended upon genotypes, the subjects with AA genotype had lower serum TC and LDL-C levels in drinkers than in non-drinkers.Conclusions: Alcohol consumption can modify the effects of the PCSK9 E670G polymorphism on serum TC and LDL-C levels. The subjects with AA genotype of the PCSK9 E670G benefit more from alcohol consumption than the subjects with AG genotype in decreasing serum TC and LDL-C levels.

The interactions of single nucleotide polymorphisms (SNPs) and cigarette smoking on blood pressure levels are limited. The present study was undertaken to detect nine lipid-related SNPs and their interactions with cigarette smoking on blood pressure levels. Genotyping of ATP-binding cassette transporter A1 (ABCA-1) V825I, acyl-CoA:cholesterol acyltransferase-1 (ACAT-1) rs1044925, low density lipoprotein receptor (LDL-R) AvaⅡ, hepatic lipase gene (LIPC) -250G>A, endothelial lipase gene (LIPG) 584C>T, methylenetetrahydrofolate reductase (MTHFR) 677C>T, proprotein convertase subtilisin-like kexin type 9 (PCSK9) E670G, peroxisome proliferator-activated receptor delta (PPARD) +294T>C, and Scavenger receptor class B type 1 (SCARB1) rs5888 was performed in 935 nonsmokers and 845 smokers. The interactions were detected by factorial regression analysis. The frequencies of genotypes (ACAT-1 and LIPG), alleles (ABCA-1), and both genotypes and alleles (LDL-R, LIPC, PPARD and SCARB1) were different between nonsmokers and smokers (P < 0.05-0.001). The levels of pulse pressure (PP, ABCA-1), and systolic, diastolic blood pressure (SBP, DBP) and PP (LIPC) in nonsmokers were different among the genotypes (P < 0.01-0.001). The levels of SBP (ABCA-1, ACAT-1, LIPG and PCSK9), DBP (ACAT-1, LDL-R, LIPC, PCSK9 and PPARD), and PP (LIPC, LIPG, MTHFR and PCSK9) in smokers were different among the genotypes (P < 0.01-0.001). The SNPs of ABCA-1, ACAT-1 and PCSK9; ACAT-1, LDL-R, MTHFR and PCSK9; and ABCA-1, LIPC, PCSK9 and PPARD were shown interactions with cigarette smoking to influence SBP, DBP and PP levels (P < 0.05-0.001); respectively. The differences in blood pressure levels between the nonsmokers and smokers might partly result from different interactions of several SNPs and cigarette smoking.

Oxidative stress and inflammation are the important pathological basis of atherogenesis. So, attenuating oxidative stress and inflammation has a very important significance in the prevention and treatment of atherosclerosis. The aim of present study was to investigate whether anti-atherosclerotic effect of Tongxinluo (TXL), a compound traditional Chinese medicine, is related to its anti-oxidation and anti-inflammation in human cardiac microvascular endothelial cells (HCMEC). We found that TXL treatment significantly reduced serum lipid levels and atherosclerotic plaque formation of apoE-deficient mice, and improved endothelial cell function as evidenced by increased expression of CD31 and eNOS. TXL pretreatment could abrogate the up-regulation of ROS and MDA induced by C16. Further experiments showed that the anti-oxidative effect of TXL may be related to inhibiting the expression of p22(phox), p47(phox) and HO-1 in HCMECs. We also found that TXL could inhibit the release of IL-1β and TNFα induced by C16, which is mediated by inhibiting the expression and activation of NF-κB. In conclusion, TXL decreases atherosclerotic plaque formation and improves endothelial cell function by inhibiting oxidative stress and inflammation in HCMECs. This finding provides a new molecular mechanism for the anti-atherosclerotic effect of TXL.

Some triply excited states of He- ion are studied using the saddle-point complex-rotation method. The energies and Auger widths of these resonances are calculated with the relativistic corrections and mass polarization taken into account. The total Auger width is obtained by coupling the important open channels and summing over the other channels. These results are in good agreement with other theoretical and experimental data in the literature.

The fretting wear-resistant properties of ion-sulphuration+shot-peening and shot-peening+ion-sulphuration duplex-treated coatings on 1045 steel substrate were investigated. The results show that the fretting wear-resistance of shot-peening+ion-sulphuration coating was better than that of the ion-sulphuration+shot-peening coating. The fretting wear process of the duplex-treated coatings in the gross slip regime could be described as the sulphuration coatings playing an important lubricating role in the incubation period. Then a series of changes took place continuously and alternatively on the contact surfaces, including adhesion, material transfer, oxidation, delamination of the oxide layer and degradation. In the last stage, the trapping, pulverization and elimination of debris restrained the adhesion between the contact surfaces and led to a decrease in the friction coefficient.

By using the nonequilibrium Green's function method, the ballistic thermal rectification in the three-terminal graphene nanojunction is studied. The dynamics of atoms is described by the interatomic fourth-nearest neighbor force-constant model. The nanojunction has a Y-shaped structure, created by a combination of a straight graphene nanoribbon and a leaning branch as the control terminal holding a fixed temperature. No heat flux flows through the control terminal. There exists a temperature bias between the two ends of the graphene nanoribbon serving as the left and right terminals, respectively. The primary goal of this paper is to demonstrate that the ballistic thermal rectification can be introduced by the asymmetric structure with different connection angles between terminals. The control terminal has a smaller connection angle with respect to the left terminal than to the right terminal. The forward direction is defined as being from the left terminal to the right terminal. The results demonstrate that, given the same control temperature and absolute temperature bias, the heat flux in the graphene nanoribbon tends to run preferentially along the forward direction. When the difference between the connection angles increases, the rectification ratio rises. Compared with that of the zigzag graphene nanoribbon, the rectification ratio of the armchair nanoribbon is much sensitive to the direction the control terminal. However, the greatest rectification ratio is found in the zigzag graphene nanoribbon which has a connection angle of 30 degrees with respect to the armchair branch. In addition, the direction of the control terminal can be adjusted to raise more than 50% of the rectification ratio of the graphene thermal rectifier based on the width discrepancy between the left and right terminals. The mechanism of the ballistic thermal rectification is also discussed. In the three-terminal graphene nanojunction, a smaller connection angle with respect to the control terminal leads to more phonon scatterings. The confirmation of this conclusion comes from a comparison of phonon transmission between different couples of terminals, which shows that in most of the frequency spectrum, the phonon transmission between the control terminal and the left terminal is smaller than between the control terminal and the right terminal. Given the same control terminal temperature and temperature bias, the asymmetric connection angles therefore will introduce a higher average temperature of the left and right terminals, and a larger heat flux in the forward process. Moreover, the average temperature difference between in the forward process and in the reverse process is found to be proportional to the temperature bias, and the proportionality coefficient will become bigger if the asymmetry is strengthened.

Highly c-axis oriented Mg0.05Zn0.95O thin films were fabricated on Si(111) substrates at different temperature by pulsed laser deposition. The influence of the substrate temperature on the structure and luminescent properties of Mg0.05Zn0.95O thin films were studied by XRD and photoluminescence (PL) spectra and the relationship between the crystalline quality and PL properties of thin films was discussed. The results indicate that the Mg0.05Zn0.95O thin films grown at substrate temperature of 450℃ show good c-axis orientation and strong PL peaks. Different PL spectra are acquired at room temperature using xenon lamps as excitation light sources, with excitation wavelengths of 240, 300 and 325nm, respectively. The UV peaks and violet peaks were found to originate from free excition radiation recombination and the PL intensity was related to crystalline quality, and the blue-green peaks may be related to the oxygen vacancies. We also discuss the physical mechanism throngh which the substrate temperature affects the red shift and blue shift of UV PL peaks.
