Objective Our study aimed to elucidate the effect of PAI-1 (Plasminogen Activator Inhibitor-1) and t-PA (Tissue-type Plasminogen Activator) in tissue remodeling in nasal polyps patients. Methods Samples were streamed as early Nasal Polyps (eNP, n = 10) and inferior tissue from the same patient, mature Nasal Polyps (mNP, n = 14), and Control group (n = 15), respectively. Immunohistochemistry and immunofluorescence were applied to detect localization. Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and Western blot were used to measure different levels among three groups. The mNP tissue was cultured in vitro and treated with TGF-β1 (Transforming Growth Factor-beta 1) activator, TGF-β1 inhibitor (SB431542), and PAI-1 inhibitor (TM5275); then Western blot, qRT-PCR, and ELISA were used to assess changes. Results The immunohistochemistry and immunofluorescence showed that PAI-1 expression decreased in eNP and mNP, mainly in epithelium and glands. The transcriptional expression and protein level of TGF-β1/t-PA/PAI-1/Collagen1 were lower in eNP than IT while mNP group demonstrated lower mRNA expression and protein level of TGF-β1/t-PA/PAI-1/Collagen1 than Control group. In mNP tissue culture in vitro, TGF-β1 activator elevated t-PA, PAI-1, and Collagen1 with higher release of PAI-1 and Collagen1 in supernatant, whereas SB431542 suppressed above reactions; TM5275 lowered transcriptional and protein level of Collagen1 in supernatant. Conclusion Early Nasal polyps’ formation in middle meatus mucous is related with fibrillation system PAI-1/t-PA and tissue remodeling; moreover, nasal polyps’ development is regulated by TGF-β1-mediated PAI-1 reduction. Currently, cryptocurrency has become a research and investment topic of great concern and attracted considerable attentions in a wide range of fields. Stocks are well known as a form of investment, and there are countless studies on how its price trends. Cryptocurrencies, however, are not easily predicted due to their extremely high volatility. This paper implements the forecasting results by taking three major cryptocurrencies (i.e., Bitcoin, Ethereum, Dogecoin) as examples for the period starting from January 1st, 2020 to May 31st 2022. Four popular prediction models (XGBoost, LightGBM, GARCH, ARIMA) are applied in Python by training models and testing the prediction results. According to the analysis, XGBoost and LightGBM can forecast the future prices for all three cryptocurrencies exactly apart from the turning points when prices rise and drop suddenly. Although the predicting trends of GARCH, ARIMA have differences from the real price, they can forecast well except the unexpected situations such as COVID-19. Overall, relatively reliable and accurate prediction models can be provided for investors to apply and make wise decisions in investments based on the results of this study.

Mechanical fault detection has an important influence on production schedule and efficiency. With the development of intelligent technology, more and more intelligent detection technologies are applied to mechanical fault detection. In order to detect mechanical faults more efficiently and accurately, this experiment proposes a production knowledge base model based on genetic algorithm (GA algorithm). The model uses the unique biological genetics principle of genetic algorithm to evolve the interested population, and can conduct spatial search to find the global optimal solution. By comparing the performance of GA algorithm model with other similar detection models, it is found that the model proposed in the experiment has obvious advantages in mechanical fault detection performance. The experimental results show that the maximum accuracy of the GA algorithm is 0.935, 0.074 higher than the support vector machine (SVM) model, 0.118 higher than the linear discriminant analysis (LDA) model, 0.032 higher than the random forest (RF) model, and 0.166 higher than the K nearest neighbor (KNN) model. In addition, the error value of GA algorithm is the lowest among these models, which is 0.028. This proves that the genetic algorithm model has higher diagnostic accuracy and can play an important role in mechanical fault detection.

This study performed a comprehensive investigation of Hyperici Perforati Herba polysaccharide (HPHP) regarding the development and optimization of extraction methods, elucidation of structure and characteristics, and determination of antioxidant activities. An ultrasonic-assisted extraction method, which offered advantages in terms of the extraction yield and energy efficiency, was developed by response surface analysis. The following optimum conditions were determined: a crushing degree at 65 mesh, ultrasonic time at 50 min and temperature of 43 °C. Through enzyme-mediated deproteination via the Sevag method, activated carbon depigmentation, and DEAE-52 and Sephadex G-100 column elution, three HPHPs were obtained, and their monosaccharides mainly included mannose, galactose, glucose and arabinose. The molar weights were 8.347, 1.199 and 22.426 kDa, respectively. The HPHP structures were an amorphous aggregate of spherical-like shapes with a rough surface of pores and crevices, which presented characteristic Fourier transform infrared (FT-IR) and nuclear magnetic resonance (NMR) spectra of polysaccharides. Their main glucosidic linkage is the α-type configuration. Moreover, HPHPs exhibited strong scavenging activity for DPPH·, ABTS·

To present a case series of the exstrophy-epispadias complex (EEC) with isolated ectopic bowel segment (IEBS) with the literature review, highlighting the clinical findings and treatments.We present 3 cases of bladder exstrophy (BE) with IEBS in our institute and reviewed the literature in PubMed with the terms "("bladder exstrophy" OR "epispadias") AND ("visceral sequestration" OR "sequestered" OR "ectopic bowel")."There were 2 males and 1 female. The IEBS was detected by physical examination in 2 cases and by ultrasonography in another one. All cases were BE accompanying with lower abdominal mass which adhered to the bladder wall but was separated from the digestive system. All cases underwent the IEBS excision and BE repair simultaneously. Pathological result of IEBS suggested the histological structures of colon. There were totally 13 cases of EEC with IEBS reported in the literature, including 2 (15%) epispadias, 9 (69%) covered BE, 1 (8%) duplicate BE and 1 (8%) classic bladder exstrophy. Although their clinical manifestations were various, IEBS excision were safely conducted in all cases.EEC with IEBS is an extremely rare congenital malformation. Physical and imaging examinations are important for diagnoses. Surgical excision is safe and effective for managing IEBS.

The cause of quicksand in soil depends on the nature of soil. Under the action of permeability, quicksand is easy to occur when the pore ratio of soil is large, the water content is large, the clay content is small, the silt content is large, the permeability coefficient is small and the drainage performance is poor. Therefore, quicksand phenomenon is easy to occur in fine sand, silt and loam, but whether quicksand phenomenon occurs or not also depends on certain external conditions, that is, the hydrodynamic pressure (permeability) generated by the seepage of groundwater in the soil. When the upward seepage force of unit granular soil is greater than or equal to its own gravity, the soil will suspend and move. In the process of underground engineering construction, if the problem of quicksand is not solved, the structure will displace with the flow of sand layer, which will change the bearing layer of the foundation and do great harm to the structure. This phenomenon should be eliminated. Practice shows that many accidents of underground structures are related to quicksand caused by the change of seepage conditions during construction. The disturbance of soil in pipe jacking construction is the fundamental cause of soil deformation. It destroys the original natural equilibrium state of soil, changes the stress-strain state of soil, and makes the undisturbed soil experience complex stress paths such as extrusion, shear and deformation. In this paper, a numerical model is established to analyze the surface settlement trend of pipe jacking in quicksand covered stratum, and the methods of grouting behind pipe jacking wall and excavation surface stability measures are verified to control the settlement measures in the construction process.

To characterize quick contrast sensitivity function (qCSF) in keratoconus and its correlation with corneal topographic parameters.Patients with keratoconus (n = 120) who visited the Fudan Eye and ENT Hospital between April and June 2021 were enrolled in our study. A total of 215 eyes were subdivided into three groups according to maximum keratometry (KSignificant differences in the area under the log CSF (AULCSF) and CSF Acuity among the three groups were found, which decreased with an increase in KThe use of qCSF test can serve as a feasible tool to evaluate visual quality and severity of keratoconus, since changes in CS significantly correlated with keratoconus severity.

Charge transfer dynamics is decisive for the performance of perovskite light emitting diodes (PeLEDs), and deep insight into the charge transfer process inside the working device is indispensable. Here, the influence of the hole transport layer on charge transport and recombination processes in PeLEDs is investigated via impedance spectroscopy. The results demonstrate that the rational interfacial energy level alignment can improve the radiative recombination by reducing the leakage current and carrier transport resistance. Shockley-Read-Hall recombination and Auger recombination enlarge the lifetime of carrier transfer in the working devices as determined from the electroluminescence spectrum. Our work provides a distinctive and reliable method to explore the charge transfer property and highlights the importance of interfaces to boost the performance of PeLEDs.

The ecological validity of emotion-inducing scenarios is essential for emotion research. In contrast to the classical passive induction paradigm, immersive VR fully engages the psychological and physiological components of the subject, which is considered an ecologically valid paradigm for studying emotion. Several studies investigate the emotional responses to different VR tasks or games using subjective scales. However, little research regards VR as an eliciting material, especially when systematically analyzing emotional processes in VR from a neurophysiological perspective. To fill this gap and scientifically evaluate VR's ability to be used as an active method for emotion elicitation, we investigate the dynamic relationship between explicit information (subjective evaluations) and implicit information (objective neurophysiological data). A total of 28 participants are enlisted to watch eight VR videos while their SAM/IPQ scores and EEG data are recorded simultaneously. In ecologically valid scenarios, the subjective results demonstrate that VR has significant advantages for evoking emotion in arousal-valence. This conclusion is backed by our examination of objective neurophysiological evidence that VR videos effectively induce high-arousal emotions. In addition, we obtain features of critical channels and frequency oscillations associated with emotional valence, thereby validating previous research in more lifelike circumstances. In particular, we discover hemispheric asymmetry in the occipital region under high and low emotional arousal, which adds to our understanding of neural features and the dynamics of emotional arousal. As a result, we successfully integrate EEG and VR to demonstrate that VR is more pragmatic for evoking natural feelings and is beneficial for emotional research. Our research has set a precedent for new methodologies of using VR induction paradigms to acquire a more reliable explanation of affective computing.

Markers are needed to increase the diagnostic accuracy of prostate-specific antigen (PSA) in prostate cancer (PCa) screening. Mounting evidence has shown that plasma proteins can be hopeful biomarkers for cancer diagnosis.Tandem mass tag (TMT)-based proteomics and parallel reaction monitoring (PRM) analysis were used to screen the differential proteins and further validated in other independent studies (n = 539). Receiver-operating characteristic (ROC), decision curves and nomograms were applied to assess the diagnostic accuracy of biomarkers.Three candidate proteins (DBP, LCAT and ORM2) were preliminarily screened. Subsequent validation studies revealed significant upregulation of ORM2 in PCa patients across other independent cohorts. ORM2 yielded excellent discriminative power for PCa from benign prostatic hyperplasia (BPH) patients (AUC = 0.861 and 0.814 in validation phases 2a and 2b, respectively). Importantly, the combination of ORM2 and PSA gave better predictive accuracy than PSA alone. We incorporated age, PSA and ORM2 into a nomogram, which yielded C-index of 0.883 in validation phase 2a. A similar C-index of 0.879 was obtained in external validation phase 2b.In summary, our study suggests that ORM2 could be treated as a complementary biomarker for PSA in distinguishing PCa from BPH.

This paper studies the simplest system that can possess left-right symmetrical and asymmetrical surroundings, three bubbles in a line. Assuming that the deformations are small, the surfaces of bubbles are described by a combination of the first three Legendre polynomials, that is, spherical symmetrical mode P0, L-R antisymmetrical mode P1, and symmetrical mode P2. A dynamical model is built to describe aspherical oscillations of central and two side bubbles. It is found that when three identical bubbles are separated uniformly, the central bubble only has a P2 component and P1 component tends to zero, while two side bubbles have both P1 and P2 components. When three identical bubbles are separated by different distances, they can be degenerated into a two-bubble system and a free bubble. The bubble deformations contain both P1 and P2 components in the two-bubble system, while both aspherical components P1 and P2 of the free bubble tend to zero. If side bubbles are different in ambient radii but located symmetrically on the left and right of the central bubble, the side bubble pulsated more strongly plays an important role on the deformation of the central one.
