In this work, 1200 V SiC JMOS devices with different Wsch (2 μm, 2.5 μm and 3 μm) are fabricated. The single unclamped inductive switching (UIS) tests under different Vgs_off (-5 V and 0 V) are carried out to investigate the avalanche capability. The avalanche robustness among various Wsch under same Vgs_off is also compared and analyzed by simulation. The different failure mechanisms between different Vgs_off are studied by failure analysis and simulation. The method of improving avalanche ruggedness of JMOS is proposed.

With the development of decentralized networks, smart contracts, especially those for ERC tokens, are attracting more and more Dapp users to implement their applications. There are some functions in ERC token contracts that only a specific group of accounts could invoke. Among those functions, some even can influence other accounts or the whole system without prior notice or permission. These functions are referred to as contract backdoors. Once exploited by an attacker, they can cause property losses and harm users’ privacy. In this work, we propose Pied-Piper, a hybrid analysis method that integrates datalog analysis and directed fuzzing to detect backdoor threats in Ethereum ERC token contracts. First, datalog analysis is applied to abstract the data structures and identification rules related to the threats for preliminary static detection. Then, directed fuzzing is applied to eliminate false positives caused by the static analysis. We first evaluated Pied-Piper on 200 smart contracts, which are injected with different types of backdoors. It reported all problems without false positives, and none of the injected problems was missed. Then, we applied Pied-Piper on 13,484 real token contracts deployed on Ethereum. Pied-Piper reported 189 confirmed problems, four of which have been assigned unique CVE ids while others are still in the review process. Each contract takes 8.03 seconds for datalog analysis on average, and the fuzzing engine can eliminate the false positives within one minute.

Program reduction has demonstrated its usefulness in facilitating debugging language implementations in practice, by minimizing bug-triggering programs. There are two categories of program reducers: language-agnostic program reducers (AGRs) and language-specific program reducers (SPRs). AGRs, such as HDD and Perses, are generally applicable to various languages; SPRs are specifically designed for one language with meticulous thoughts and significant engineering efforts, e.g., C-Reduce for reducing C/C++ programs. Program reduction is an NP-complete problem: finding the globally minimal program is usually infeasible. Thus all existing program reducers resort to producing 1-minimal results, a special type of local minima. However, 1-minimality can still be large and contain excessive bug-irrelevant program elements. This is especially the case for AGR-produced results because of the generic reduction algorithms used in AGRs. An SPR often yields smaller results than AGRs for the language for which the SPR has customized reduction algorithms. But SPRs are not language-agnostic, and implementing a new SPR for a different language requires significant engineering efforts. This paper proposes Vulcan, a language-agnostic framework to further minimize AGRs-produced results by exploiting the formal syntax of the language to perform aggressive program transformations, in hope of creating reduction opportunities for other reduction algorithms to progress or even directly deleting bugirrelevant elements from the results. Our key insights are two-fold. First, the program transformations in all existing program reducers including SPRs are not diverse enough, which traps these program reducers early in 1-minimality. Second, compared with the original program, the results of AGRs are much smaller, and time-wise it is affordable to perform diverse program transformations that change programs but do not necessarily reduce the sizes of the programs directly. Within the Vulcan framework, we proposed three simple examples of fine-grained program transformations to demonstrate that Vulcan can indeed further push the 1-minimality of AGRs. By performing these program transformations, a 1-minimal program might become a non-1-minimal one that can be further reduced later. Our extensive evaluations on multilingual benchmarks including C, Rust and SMT-LIBv2 programs strongly demonstrate the effectiveness and generality of Vulcan. Vulcan outperforms the state-of-the-art language-agnostic program reducer Perses in size in all benchmarks: On average, the result of Vulcan contains 33.55%, 21.61%, and 31.34% fewer tokens than that of Perses on C, Rust, and SMT-LIBv2 subjects respectively. Vulcan can produce even smaller results if more reduction time is allocated. Moreover, for the C programs that are reduced by C-Reduce, Vulcan is even able to further minimize them by 10.07%.

In this study, we evaluated the biomechanical characteristics of different locations of medial fixation strategies in double-plate osteosynthesis for fixing AO/ASIF type 33-C2 fractures by means of finite element analysis.We used 3-matic software and UG-NX software to construct AO/ASIF type 33-C2 fractures and lateral less invasive stabilization system (LISS) plates, medial plates (MPs), and medial support pads (MSPs), respectively. Then, the LISS, MP and MSP were assembled into the fracture model separately to form three fixation models: MSP+LISS, anteromedial plate (AMP+LISS), and MP+LISS. In the next procedure, we performed finite element analysis using ANSYS software after meshing the elements of the models in HyperMesh 11.0 software. Loading conditions including lateral-medial four-point bending, anterior-posterior four-point bending, axial loading, and torsional loading were applied to evaluate the biomechanical advantages among the three fixation types. We observed the peak Von Mises Stress (VMS) value, maximum displacement, bending angle in the coronal plane of the fracture, and torsional angle of the fracture to assess the degree of plate deformation and fixation stability.Our results showed that in both lateral-medial four-point bending and anterior-posterior four-point bending, the calculations of MP+LISS were marginally better than those of AMP+LISS. However, with the action of axial loading and torsional loading, the deformation of MP+LISS was distinctly smaller than that of AMP+LISS, and the fixation stability of MP+LISS was also prominently better. Under lateral-medial four-point bending, the VMS on the lateral plate of MSP+LISS (59.977 MPa) was approximately half of the two double-plate models. Under anterior and posterior four-point bending, the 38.209 MPa peak VMS of MSP+LISS was still superior to the other two double-plate models. Under torsional loading, the peak VMS (347.75 MPa), the maximum torsional angle of the femoral head (7.852 °), and the torsional angle of fracture (0.036 °) of MSP+LISS preceded those of the other two models. However, under axial loading, the peak VMS (76.376 MPa) and the maximum displacement (3.1798 mm) of MSP+LISS were slightly higher than those of MP+LISS.The MSP+LISS model showed better biomechanical performance than the double-plate models, which might be an effective solution for the treatment of comminuted distal femur fractures.

Objective To investigate the analgesic mechanism of Tuina (Chinese therapeutic massage) by observing the effect of the N-methyl-D-aspartate receptor subunit 2B (NR2B)/postsynaptic density-95 (PSD-95) pathway on the dendritic structure of spinal cord dorsal horn in rats with lumbar disc herniation. Methods Fifty Sprague-Dawley rats were randomly divided into a blank group, a model group, a Tuina group, a blocker agent group, and a blocker agent + Tuina group. The sciatic nerve chronic constriction injury (CCI) model was prepared by the sciatic nerve ligation method. From the 4th day after modeling, rats in the Tuina group and the blocker agent + Tuina group were subject to daily Tuina intervention, and those in the blocker agent group and the blocker agent + Tuina group were daily intrathecally injected with NR2B blocker agent (MK-801). The spontaneous pain score was used to observe the pain behavior of all rats. The expression levels of NR2B and downstream PSD-95 were measured by immunohistochemistry, and the dendritic structure changes were observed by Golgi staining for rat spinal cord dorsal horn after 14 d of continuous intervention. Results Compared with the blank group, the degree of rat spontaneous pain after CCI was elevated in both the model and the Tuina groups (PPPPPPPPPPPP Conclusion Tuina may exert an analgesic effect by remodeling the dendritic structure in the spinal cord dorsal horn in rats with lumbar disc herniation, and its mechanism may be related to the inhibition of NR2B/PSD-95 signaling pathway.

We defined preoperative anemia as the last hemoglobin measurement level before surgery < 130 g/L for men and < 120 g/L for women. The primary outcome was a composite of in-hospital major complications, including pneumonia, respiratory failure, gastrointestinal bleeding, urinary tract infection, incision infection, deep venous thrombosis, pulmonary embolism, angina pectoris, arrhythmia, myocardial infarction, heart failure, stroke, and death. Secondary outcomes were cardiovascular events, infection, pneumonia, and death. We used multivariate negative binomial or logistic regression to evaluate the impact of anemia and its severity, categorized as mild (90– 130 g/L for men, 90– 120 g/L for women) or moderate-to-severe (< 90 g/L for both) anemia on outcomes.Results: Of the 3540 included patients, 1960 had preoperative anemia. 188 anemic patients experienced 324 major complications, while 63 non-anemic patients had 94 major complications. The risk of major complications was 165.3 (95% CI, 149.5– 182.4) and 59.5 (95% CI, 48.9– 72.3) per 1000 persons in anemic and non-anemic patients, respectively. Anemic patients were more likely to have major complications than non-anemic patients (adjusted incidence rate ratio (aIRR), 1.87; 95% CI, 1.30– 2.72), which was consistent in mild (aIRR, 1.77; 95% CI, 1.22– 2.59) and moderate-to-severe (aIRR, 2.97; 95% CI, 1.65– 5.38) anemia. Preoperative anemia also increased the risk of cardiovascular events (aIRR, 1.96; 95% CI, 1.29– 3.01), infection (aIRR, 1.68; 95% CI, 1.01– 2.86), pneumonia (adjusted odds ratio (aOR), 1.91; 95% CI, 1.06– 3.57), and death (aOR, 3.17; 95% CI, 1.06– 11.89).Conclusion: Our findings suggest that even mild preoperative anemia is associated with major postoperative complications in hip fracture patients. This finding highlights considering preoperative anemia as a risk factor in surgical decision-making for high-risk patients.Keywords: anemia, hip fracture, postoperative complications, risk factor

Fetal cystic hygroma (CH) is associated with poor prognosis and chromosomal anomalies. Recent studies have suggested that the genetic background of affected fetuses is essential for predicting pregnancy outcomes. However, the detection performance of different genetic approaches for the etiological diagnosis of fetal CH remains unclear. In this study, we aimed to compare the diagnostic efficiency of karyotyping and chromosomal microarray analysis (CMA) in a local fetal CH cohort, and tried to propose an optimized testing strategy that may help improve the cost-effectiveness of disease management. We reviewed all pregnancies that underwent invasive prenatal diagnosis between January 2017 and September 2021 at one of the largest prenatal diagnostic centers in Southeast China. We collected cases identified by the presence of fetal CH. Prenatal phenotypes and laboratory records of these patients were audited, collated, and analyzed. The detection rates of karyotyping and CMA were compared, and the concordance rate of these two methods was calculated. A total of 157 fetal CH cases were screened from 6,059 patients who underwent prenatal diagnosis. Diagnostic genetic variants were identified in 44.6% (70/157) of the cases. Karyotyping, CMA, and whole-exome sequencing (WES) identified pathogenic genetic variants in 63, 68, and 1 case, respectively. The Cohen’s κ coefficient between karyotyping and CMA was 0.96, with a concordance of 98.0%. Of the 18 cases in which cryptic copy number variants PIGN gene in a case undiagnosed by CMA and karyotyping. Our study demonstrated that chromosomal aneuploidy abnormalities are the main genetic cause of fetal CH. Based on this, we recommend karyotyping combined with rapid aneuploidy detection as a first-tier approach for the genetic diagnosis of fetal CH. WES and CMA could improve the diagnostic yield when routine genetic tests fail to determine the cause of fetal CH.

Background: The molecular mechanism underlying the progression of gastric cancer (GC) remains unclear, the overexpression level of SALL4 has been demonstrated in many cancers and participated in tumorigenesis, however, it is still ambiguous for the expression and function of SALL4 in GC, especially its upstream mechanistic modulators.Method: Analysis of discrepant gene expression in GC and normal gastric tissues from The Cancer Genome Atlas (TCGA) dataset. Cultured GC cell lines were treated with siEZH2 and siKDM6A and checked for their influences on SALL4, the transduction molecules of KDM6A/EZH2-SALL4-β-catenin signaling were quantified in the GC cells.Results: Here, we showed that only SALL4 levels of SALL family members were upregulated in nonpaired and paired GC tissues than those in corresponding normal tissues and were associated with its histological types, pathological stages, TNM stages, and overall survival from the TCGA dataset. SALL4 level was elevated in GC cells compared to normal gastric epithelial cell line (GES-1) and was correlated to cancer cell progression and invasion through the Wnt/β-catenin pathway in GC, which levels would be separately upregulated or downregulated by KDM6A or EZH2.Conclusion: We first proposed and demonstrated that SALL4 promoted GC cell progression via the Wnt/β-catenin pathway, which was mediated by the dual regulation of EZH2 and KDM6A on SALL4. This mechanistic pathway in gastric cancer represents a novel targetable pathway.

Structural variations (SVs) are a major contributor to genetic diversity and phenotypic variations, but their prevalence and functions in domestic animals are largely unexplored. Here we generated high-quality genome assemblies for 15 individuals from genetically diverse sheep breeds using Pacific Biosciences (PacBio) high-fidelity sequencing, discovering 130.3 Mb nonreference sequences, from which 588 genes were annotated. A total of 149,158 biallelic insertions/deletions, 6531 divergent alleles, and 14,707 multiallelic variations with precise breakpoints were discovered. The SV spectrum is characterized by an excess of derived insertions compared to deletions (94,422 vs. 33,571), suggesting recent active LINE expansions in sheep. Nearly half of the SVs display low to moderate linkage disequilibrium with surrounding single-nucleotide polymorphisms (SNPs) and most SVs cannot be tagged by SNP probes from the widely used ovine 50K SNP chip. We identified 865 population-stratified SVs including 122 SVs possibly derived in the domestication process among 690 individuals from sheep breeds worldwide. A novel 168-bp insertion in the 5′ untranslated region (5′ UTR) ofHOXB13is found at high frequency in long-tailed sheep. Further genome-wide association study and gene expression analyses suggest that this mutation is causative for the long-tail trait. In summary, we have developed a panel of high-quality de novo assemblies and present a catalog of structural variations in sheep. Our data capture abundant candidate functional variations that were previously unexplored and provide a fundamental resource for understanding trait biology in sheep.

To evaluate longitudinal changes in macular choroidal thickness (mCT) in myopic children treated for 1 year with repeated low-level red-light (RLRL) therapy and their predictive value for treatment efficacy on myopia control.A secondary analysis of data from a multicenter, randomized controlled trial (RCT; NCT04073238).Myopic children aged 8-13 years who participated in the RCT at 2 of 5 sites where mCT measurements were available.Repeated low-level red-light therapy was delivered using a home-use desktop light device that emitted red-light at 650 nm. Choroidal thickness was measured by SS-OCT at baseline and 1-, 3-, 6-, and 12-month follow-ups. Visual acuity, axial length (AL), cycloplegic spherical equivalent refraction (SER), and treatment compliance were measured.Changes in mCT at 1, 3, 6, and 12 months relative to baseline, and their associations with myopia control.A total of 120 children were included in the analysis (RLRL group: n = 60; single-vision spectacle [SVS] group: n = 60). Baseline characteristics were well balanced between the 2 groups. In the RLRL group, changes in mCT from baseline remained positive over 1 year, with a maximal increase of 14.755 μm at 1 month and gradually decreasing from 5.286 μm at 3 months to 1.543 μm at 6 months, finally reaching 9.089 μm at 12 months. In the SVS group, mCT thinning was observed, with changes from baseline of -1.111, -8.212, -10.190, and -10.407 μm at 1, 3, 6, and 12 months, respectively. Satisfactory myopia control was defined as annual progression rates of less than 0, 0.05, or 0.10 mm for AL and less than 0, 0.25, or 0.50 diopters for SER. Models that included mCT changes at 3 months alone had acceptable predictive discrimination of satisfactory myopia control over 12 months, with areas under the curve of 0.710-0.786. The predictive performance of the models did not significantly improve after adding age, gender, and baseline AL or SER.This analysis from a multicenter RCT found RLRL induced sustained choroidal thickening over the full course of treatment. Macular choroidal thickness changes at 3 months alone can predict 12-month myopia control efficacy with reasonable accuracy.Proprietary or commercial disclosure may be found after the references.

China is the country with the largest number of disabled people in the world. By 2022, according to the statistics of the China Disabled Persons Federation, the total number of disabled people in China has reached 85 million. In addition, most disabled people have special needs for the structure, materials and auxiliary accessories of clothing due to physical or psychological reasons, so we call this kind of clothing "adaptive clothing". In recent years, developed countries have made some achievements in the theoretical research and development of adaptive clothing. Clothing companies in some countries, including the United States, Britain, Australia, Russia and so on, have developed and formed, with a complete range of clothing, and normal production and sales. However, at present, domestic research in this field focuses more on the theoretical level and lacks practical data and experience. The adaptive clothing market is almost a blank, which requires further follow-up research. Based on the current situation of domestic adaptive clothing, this paper analyzes the theoretical research and development of domestic adaptive clothing, combines with the practical level to summarize the reasons for the lagging research of adaptive clothing and puts forward countermeasures, so as to provide reference for the systematic development and research of adaptive clothing. It is of great significance to care for disabled groups, promote social harmony, open the potential clothing market, create economic benefits and improve people's happiness index.

Background. Spontaneous preterm birth is one of the most common pregnancy complications in obstetric clinical practice, and its etiology is complex. The problems of low survival and high morbidity rates of premature infants need to be solved urgently. The platelet-to-lymphocyte ratio (PLR) and lymphocyte-to-monocyte ratio (LMR) are two novel biomarkers of inflammation, and several studies have linked PLR and LMR to spontaneous preterm birth. These systematic review and meta-analysis are aimed at analyzing the relationship between PLR and LMR in patients with spontaneous preterm birth to provide new ideas for the early prevention and treatment of spontaneous preterm births. Methods. Cochrane Library, EMBASE, PubMed, and China National Knowledge Infrastructure databases were inspected to gather PLR and LMR in patients with spontaneous preterm birth, all from the database to February 2022. Interstudy heterogeneity was evaluated using Cochran’s Q test and I 2 statistic. Differences in PLR and LMR between patients with spontaneous preterm birth and full-term controls were evaluated by computing standardized mean differences and 95% confidence intervals. Publication bias and sensitivity analyses were also performed. Results. Nine studies were included in the meta-analysis based on the inclusion and exclusion criteria. The meta-analysis showed that serum PLR values were remarkably larger for patients with spontaneous preterm birth than for full-term controls ( SMD = 0.49 , 95% CI: 0.13 to 0.84, P = 0.007 ), whereas the difference between serum LMR in patients with spontaneous preterm birth and full-term controls was not statistically significant (SMD: 0.35, 95% CI: -0.18, 0.88, P = 0.199 ). The results of Begg’s and Egger’s tests revealed that the publication bias of the meta-analysis was not significant. The outcomes of the sensitivity analysis showed that the individual studies did not influence the meta-analysis results. Conclusions. Current evidence shows that PLR is strongly associated with spontaneous preterm birth, whereas LMR is not. PLR has a certain clinical value in diagnosing and treating spontaneous preterm births, and our research will provide strong theoretical support for clinical work. In the future, it will be necessary to further explore the reasons for the increased PLR in the serum of patients with spontaneous preterm birth and other mechanisms inducing spontaneous preterm birth.

The IGF1 signal pathway is highly activated in some subtype of gastric cancer(GC) that exhibits poor survival and chemotherapy resistance. Although the results of clinical trials of anti-IGF1R monoclonal antibodies and IGF-1R inhibitors have been mostly disappointing in unselected cancer patients, some patients benefit from anti-IGF1R therapy in these failed studies. Therefore, it is necessary to characterize the complex IGF signaling in GC and help refine the strategies targeting the IGF1 pathway. We found that GC cell lines exhibit differential responses to the specific IGF1R inhibitor OSI906. According to the phosphorylation status of Akt upon the OSI906 treatment, we divided the GC cell lines into IGF1R-dependent and IGF1R-independent cells. Both in vitro and in vivo experiments indicate that Dox-induced knockdown of NEDD4 significantly suppresses tumor growth of IGF1R-dependent GC cells and NEDD4 overexpression promotes tumor growth of IGF1R-dependent GC cells. In contrast, the proliferation of IGF1R-independent GC cells is not affected by NEDD4 silencing and overexpression. The rescue experiments show that a PTEN-IRS1 axis is required for NEDD4-mediated regulation of Akt activation and tumor growth in GC cells. Clinically, NEDD4 is expressed higher in IGF1-high GC tissues compared with IGF1-low GC tissues and normal tissues, and the co-high expression of NEDD4 and IGF1 predicts a worse prognosis in GC patients. Taken together, our study demonstrated that NEDD4 specifically promotes proliferation of GC cells dependent on IGF1/IGF1R signaling by antagonizing the protein phosphatase activity of PTEN to IRS1, and targeting NEDD4 may be a promising therapeutic strategy for IGF1 signal pathway-driven gastric cancer.

To investigate the feasibility and clinical efficacy of laparoscopic ureteroplasty with oral mucosal graft for ureteral stricture and describe the initial experience of eighteen operations.A retrospective analysis was performed on the clinical data of 18 patients who underwent laparoscopic ureteroplasty with oral mucosal graft for long segment or complex ureteral stricture admitted to the Second Affiliated Hospital of Anhui Medical University from July 2018 to June 2021. After the stricture ureter segment was freed during the operation, the ureteral stenosis segment was longitudinally cut. The required oral mucosa is removed according to the length of the stenosis. Oral mucosal grafts were harvested and placed in the ureter as an anterior onlay with omental wrapping.Ureteral repair was performed laparoscopically in all cases, with no conversion to open surgery and no serious complications. The median length of ureteral stenosis was 3 cm (range, 2-7 cm), the mean operative time was 205.8 ± 33.4 min, indwelling time of the drainage tube was 4.9 ± 1.6 days, and the median length of postoperative stay was 7 days (range, 4-14 days). The double J tube was removed three to six weeks postoperatively. One of the eighteen patients had restenosis after surgery, and the other patients showed no deterioration of the severity of the obstructive uropathy after follow-up imaging examination.Laparosopic ureteroplasty with oral mucosal graft is a simple, safe and efficient option for the treatment of ureteral stricture. Although the results of the initial experience are encouraging, a large cohort study with longer follow-up period is need to be done.

Scanners are commonly applied for detecting vulnerabilities in web applications. Various scanners with different strategies are widely in use, but their performance is challenged by the increasing diversity of target applications that have more complex attack surfaces (i.e., website paths) and covert vulnerabilities that can only be exploited by more sophisticated attack vectors (i.e., payloads). In this paper, we propose Scanner++, a framework that improves web vulnerability detection of existing scanners through combining their capabilities with attack intent synchronization. We design Scanner++ as a proxy-based architecture while using a package-based intent synchronization approach. Scanner++ first uses a purification mechanism to aggregate and refine attack intents, consisting of attack surfaces and attack vectors extracted from the base scanners’ request packets. Then, Scanner++ uses a runtime intent synchronization mechanism to select relevant attack intents according to the scanners’ detection spots to guide their scanning process. Consequently, base scanners can expand their attack surfaces, generate more diverse attack vectors and achieve better vulnerability detection performance. For evaluation, we implemented and integrated Scanner++ together with four widely used scanners, BurpSuite, AWVS, Arachni, and ZAP, testing it on ten benchmark web applications and three well-tested real-world web applications of a critical financial platform from our industry partner. Working under the Scanner++ framework helps BurpSuite, AWVS, Arachni, and ZAP cover 15.26%, 37.14%, 59.21%, 68.54% more pages, construct 12.95×, 1.13×, 15.03×, 52.66× more attack packets, and discover 77, 55, 77, 176 more bugs, respectively. Furthermore, Scanner++ detected eight serious previously unknown vulnerabilities on real-world applications, while the base scanners only found three of them.

To improve hydrophobicity and thermal stability, polydimethysiloxane (PDMS) emulsion and silica sol were used for depositing organic/inorganic hybrid coatings in wood. PDMS emulsion could provide the hydrophobic film to improve the hydrophobicity and dimensional stability owing to its low-surface-energy. Silica sol could significantly enhance the surface hardness and thermal stability due to its penetration in cell walls, indicating the pore-filling effect in wood. Moreover, in the hybrid system, silica incorporation in PDMS emulsion helped to form integrate coatings in wood via Si-O-Si cross-linked networks. The hydrophobicity, surface hardness and thermal stability of treated wood were related to the loadings of silica sol in the PDMS. Stiff silica could compensate the negative effect on thermal stability caused by PDMS, and synergistically improve the surface hydrophobicity and hardness of wood. This work opens a facile method to produce bio-based materials with satisfied hydrophobicity and thermal stability to be used in humid environments.

To assess the outcomes and risk factors for adult patients with acute fulminant myocarditis (AFM) supported with venoarterial extracorporeal membrane oxygenation (VA ECMO) in China mainland.Data were extracted from Chinese Society of ExtraCorporeal Life Support (CSECLS) Registry database. Data from adult patients who were diagnosed with AFM and needed VA ECMO in the database were retrospectively analyzed. The primary outcome was 90-day mortality after ECMO initiation in patients with AFM supported with VA ECMO. Cox proportional hazard regression model was used to examine the risk factors associated with 90-day mortality.Among 221 patients enrolled and followed up to 90 days, 186 (84.2%) patients weaned from ECMO and 159 (71.9%) patients survived and discharged home. The median age was 38 years (IQR 29-49) and males (n = 115) represented 52.0% of the total accounted patients. The median ECMO duration was 134 h (IQR 96-177 h). The main adverse event during ECMO course was bleeding (16.3%), followed by infection (15.4%). In the multivariate Cox model analysis, cardiac arrest prior to ECMO initiation (adjusted HR 2.529; 95%CI: 1.341-4.767, p = 0.004), lower pH value (adjusted HR 0.016; 95%CI: 0.010-0.059, p 0.001) and higher lactate concentration at 24 h after ECMO initiation (adjusted HR 1.146; 95%CI: 1.075-1.221, p 0.001) were associated with 90-day mortality.71.9% patients with AFM (clinical diagnosed) supported with VA ECMO survived. Cardiac arrest prior to ECMO, lower pH and higher lactate concentration at 24 h after ECMO initiation were correlated with 90-day mortality of AFM patients supported with VA ECMO.

Fish possess a powerful IFN system to defend against aquatic virus infections. Nevertheless, spring viremia of carp virus (SVCV) causes large-scale mortality in common carp and significant economic losses to aquaculture. Therefore, it is necessary to investigate the strategies used by SVCV to escape the IFN response. In this study, we show that the SVCV nucleoprotein (N protein) negatively regulates cellular IFN production by degrading stimulator of IFN genes (STING) via the autophagy-lysosome–dependent pathway. First, overexpression of N protein inhibited the IFN promoter activation induced by polyinosinic-polycytidylic acid and STING. Second, the N protein associated with STING and experiments using a dominant-negative STING mutant demonstrated that the N-terminal transmembrane domains of STING were indispensable for this interaction. Then, the N protein degraded STING in a dose-dependent and autophagy-lysosome–dependent manner. Intriguingly, in the absence of STING, individual N proteins could not elicit host autophagic flow. Furthermore, the autophagy factor Beclin1 was found to interact with the N protein to attenuate N protein–mediated STING degradation after beclin1 knockdown. Finally, the N protein remarkably weakened STING-enhanced cellular antiviral responses. These findings reveal that SVCV uses the host autophagic process to achieve immune escape, thus broadening our understanding of aquatic virus pathogenesis.
