ObjectiveThe purpose of this study was to evaluate the safety, efficiency, and cost expenditure of remote programming in patients with Parkinson's disease (PD) after deep brain stimulation (DBS).MethodsA total of 74 patients who underwent DBS at the Department of Neurosurgery, Zhongnan Hospital of Wuhan University between June 2018 and June 2020 were enrolled in this study. There were 27 patients in the remote programming group and 47 patients in the outpatient programming group. Clinical data, programming efficiency, adverse events, expenditure, and satisfaction were compared between the two groups.ResultsA total of 36 times of remote programming were performed on the 27 patients in the remote programming group, and four had mild adverse events during programming, and the adverse events disappeared within 1 week. The satisfaction questionnaire showed that 97.3% of the patients were satisfied with the surgical effect. The patients in the remote programming group (88.9%) were more likely to receive long-term programming after DBS than the patients in the outpatient programming group (74.5%). The Parkinsonism symptoms improved in both programming groups. The majority (18/27) of patients in the remote programming group lived away from the programming center, while the majority (27/47) of patients in the outpatient programming group lived in Wuhan, where the programming center was located (P = 0.046). The cost per patient per programming was US$ 43.5 in the remote programming group and $59.5 (56–82.7) in the outpatient programming group (P < 0.001). The median time cost for each visit was 30 min (25–30) in the remote programming group and 150 min (135–270.0) in the outpatient programming group (P < 0.001).ConclusionRemote programming is safe and effective after DBS in patients with Parkinson's disease. Moreover, it reduces expenditure and time costs for patients and achieves high satisfaction, particularly for patients living far from programming centers.

DNA is the fundamental basis of our genetic information. Throughout evolution, cells have adapted mechanisms that not only allow compaction of DNA into the nucleus, but also maintain easy access to DNA for precise replication, transcription, and damage repair. DNA is wrapped around histone octamers to form nucleosomes, and then a higher-order structure called chromatin. It was revealed that chromatin has regulatory functions for transcription and other biological processes due to its dynamic quality. In eukaryotic organisms, gene regulation is about the balance between compacted DNA and gaining access for RNA polymerase and other regulatory proteins. As the most basic unit of chromatin, a nucleosome has two copies of canonical histones H2A, H2B, H3 and H4 with a segment of about 147 base pairs of DNA. Canonical histones are only expressed in certain times of the cell cycle and they are incorporated into the newly replicated genome. In contrast, histone variants are expressed throughout the cell cycle and are deposited when canonical histones are removed. Histone variants, such as H2A.Z, affect nucleosome properties with distinct modifiable amino acid sequences. Chromatin function is influenced by histone post-translational modifications (PTMs), which can act in a sequential or combinatorial fashion to influence downstream functions. It has been widely acknowledged that histone PTMs such as histone acetylation, crotonylation, and methylation play crucial roles in various biological processes including gene transcription, DNA repair, gene replication, and apoptosis. Additionally, the precursors of histone acetylation and crotonylation, acetyl-CoA and crotonyl-CoA, are regulated by cellular metabolism. This argues that the transcriptional regulatory functions from both PTMs are influenced by cellular metabolism. In recent years, YEATS-domain proteins have gained more appreciation due to their ability to bind both acetylation and crotonylation. As acyllysine readers, YEATS domain proteins amplify the impact of histone acylations on transcriptional regulation through communicating with various transcriptional factors. In budding yeast, there are three YEATS domain proteins: Yaf9, Taf14, and Sas5. Each of them is involved in various essential processes and shows binding preference for specific acylations. Previous work has demonstrated that the YEATS domain of Taf14 binding to crotonylated histone lysine down-regulates pro-growth gene expression during the low oxygen consumption phase, suggesting that transcription is regulated by cellular metabolism through the interactions between YEATS domain and acylation. The work in this dissertation focuses on another YEATS domain protein, Yaf9, and discusses the role it plays in connecting gene expression and cellular metabolism.

Objective To analyze the cardiac safety of trastuzumab on HER2 positive breast cancer patients. Methods Trastuzumab was administrated every three weeks in 185 patients with HER2 positive breast cancer. The initial dose was 8 mg/kg, and the subsequent dose was 6 mg/kg, for 4-36 cycles. The cardiotoxicity of these patients was analyzed. Results The median treatment was 17 cycles in Her-2 positive breast cancer patients who received trastuzumab. Left ventricular ejection fraction (LVEF) level dropped in 88 cases (47.6%) after trastuzumab treatment; 74 patients (40.0%) occurred valvular regurgitation (new+ aggravation); 16 patients (8.0%) occurred the reduction of diastolic function of left ventricle; 88 patients (47.6%) experienced cardiotoxicity, among which 83 patients experienced gradeⅠcardiotoxicity and five patients experienced grade Ⅱ cardiotoxicity, no patient experienced grade Ⅲ cardiotoxicity. Conclusion The treatment of trastuzumab on Chinese patients with HER2 positive breast cancer shows a satisfactory cardiac safety. However, the potential cardiotoxicity of trastuzumab should be carefully monitored during therapy.

The paper discusses the internal correlation among core curriculums in the chemical engineering and technology. By integrating courses, joint construction and teaching, a main line is found to enhance the engineering abilities of students. And the establishment of curriculum group has strengthened the correlation among courses, avoiding course overlapping as well as the waste of class hour. It adopts a teaching model characterized by joint teaching in the group, and analysis of typical cases, which integrates all knowledge, enabling students to realize the significance and practicality of expertise in the practical projects, arouse their enthusiasm, and raise their consciousness of engineering and innovation.

A vertical viewing 0.65 THz Solid-state Source Interferometer (SSI) has been installed for line-integrated density measurements on Experimental Advanced Superconducting Tokamak (EAST) . The SSI utilizes two independent solid-state diode sources with 2 mW output power and a base frequency multiplier, which provides a wide range of Intermediate Frequencies (IF) . The high time resolution of 0.1 μs at a 10 MHz IF has been successfully achieved. The IF signal remains almost constant for several hours without any active frequency stabilization. A sensitivity in the integrated density of 0.77× 1016 m−2 has been obtained. The density measurement has been implemented with IF ~ 0.85 MHz, in a plasma with Ip=600 kA and PLHW=2.1 MW on EAST . The line-averaged density, 3.5× 1019 m−3, measured by the SSI shows good agreement with the results measured by the HCN interferometer system and the POlarimeter-INTerferometer (POINT) system in the same discharge.

Chrysanthemum is one of the four most important cut flowers in the world and is also China′s main export flower.To study the effects of the red to far red ratio of light on the morphological index,leaf area,and dry matter partitioning of chrysanthemum,experiments were conducted in the experimental greenhouse of Nanjing University of Information Science and Technology from October 2010 to February 2011.The greenhouse was 9.6 m wide and 30.0 m long,with top and shoulder heights of 5.0 m and 4.5 m,respectively.The experimental material was Chrysanthemum morifolium Ramat.cv.'Jingba').The plants were transplanted on 6 October,when the seedlings were approximately 20 cm high with 6—10 leaves.A mixture of vermiculite and perlite with a volume ratio of 2∶1 was used as the culture substrate.The planting space was 20 cm × 20 cm in size.During the vegetative growth phase,fluorescent lamps(photosynthetically active radiation(PAR) =200 μmol·m-2·s-1) were used to supplement the light for 5 h(18:00—23:00) to extend the illumination time.When the plant height reached 50 cm,natural light was shaded using black plastic film,and LED light sources with four different R:FR ratios were used to produce short-day treatments.Each LED light source consisted of 360 evenly and proportionally arranged red and far-red LED lamps.The red(R:(660±30) nm): far-red(FR:(730±30) nm) energy ratios of the four experimental treatments were 0.5,2.5,4.5,and 6.5.The LED light sources(50 cm long × 50 cm wide) were fixed 15 cm above the canopy.Each treatment had 30 chrysanthemum plants.All plants were irrigated with a nutrient solution with a conductivity of 1.5 ms/cm(200 μg/g N;80 μg/g P;170 μg/g K).The morphological index was calculated for each plant based on leaf number,plant height,internode length,stem diameter,flower diameter,whole-plant diameter,total leaf area,and dry matter partitioning.The maximum plant leaf number,plant height,stem diameter,flower diameter,leaf area and total dry weight of chrysanthemum occurred at R:FR = 2.5,and the longest internodes were at R/FR=0.5.The relationship between the shoot dry-matter weight per plant and the value of R:FR in all treatments was best described with a exponential-linear model.As the duration of R:FR treatment increased,the shoot and root dry-matter partitioning indices did not significantly differ overall,while the leaf dry-matter partitioning index decreased,the stem dry-matter partitioning index initially increased and then decreased,and the flower dry-matter partitioning index increased.At R:FR=2.5,the chrysanthemum leaf and flower dry-matter partitioning indices were the highest,while the stem dry-matter partitioning index was the lowest;however,the stem dry-matter partitioning index was the highest and the leaf dry-matter partitioning index was the lowest at R/FR=6.5,and the flower dry-matter partitioning index was the lowest at R/FR=0.5.We conclude that increasing the far-infrared component of light was not conducive to increasing flower dry matter,but increasing the proportion of red light can promote stem dry-matter accumulation and a red: far red ratio of 2.5 can improve dry matter transfer into flowers.

A heterodyne interferometer operating at the frequency f = 890 GHz has been designed for measuring the electron density of atmospheric pressure air plasmas, it's density range is from 1015 to 3×1019 m−3 and the pressure range is from 1 Pa to 20 kPa. The system is configured as a Mach\nobreakdash-Zehnder type interferometer. The light source is hydrogen cyanide (HCN) laser with a wavelength of 337 μm, which is suitable for the high density area, 1018 to 3×1019 m−3, of atmospheric pressure air plasmas. The HCN laser power is up to 100 mW. The intermediate frequency, shifted by 500 kHz, is generated by the Doppler shift with a high speed rotating grating (HSRG), which is installed in a vacuum vessel with gas pressure less than 1 Pa. With the HSRG, the temporal resolution of the HCN interferometry can reach to 2 μs. Beam path analysis based on Gaussian optics is used in the design of the optical system. The VDI planar-diode Integrated Conical Horn Fundamental Mixers optimized for high sensitivity—typical RF responsivity is around 900 V/W —will be used in the HCN interferometry. The detailed optical design and opto-mechanical design for the HCN interferometry are presented.

Offshore wind farms offer many benefits over traditional energy sources and are expected to contribute significantly to a reduction in climate change in coming years. Many countries have made commitments to reduce their carbon emissions and are therefore planning to expand their current renewable energy sectors. The use of this “renewable” resource to generate electricity, through the development of offshore windfarms, is recognized as one of the key elements in meeting the commitment to reducing greenhouse gases, and is supported by a number of global conservation organizations, including Greenpeace, the World Wide Fund for Nature and Friends of the Earth. The seabed is gentle flat, water depth is shallow and wind resources are abundant in the Yellow sea, which are suitable for construction of wind farm. A total of 50 wind turbines with 4.0 MW single unit capacity and 200 MW total installed capacity are designed and installed in this project. The wind turbines are arranged along the boundary of the wind farm, the spacing between East and West is 470-1850m, and the spacing between North and south is 620-1600 m. A 220 kV offshore booster station is set up to the west of the middle of the wind farm. All the power generated by the wind turbines can be boosted by 220 kV offshore booster station and connected to the landing point of the submarine cable by three 1 x 500 220 kV single core XLPE submarine cables in one circuit. The length of the 220 kV submarine cable route is about 31.8 km. Survey contents include Bathymetric and topographic survey, seabed conditions, shallow geology, soil physical and mechanical properties, corrosive environmental parameters, marine hydrographic and meteorological elements and so on. Equipment and sensors, such as side scan sonar, sub-bottom profiler, multi-channel seismic equipment, magnetometer, sampler and CPT were employed in this survey. This survey finished 1500km geophysical survey lines, 50 stations of drilling, 55 stations of in-situ test, and 12 stations of marine hydrographic observation within 120 days. The purpose of construction of submarine scientific observation network in the East China Sea from the seabed to the sea surface, is to deeply understand the material exchange process and mechanism between the Yangtze River estuary and the East China Sea under the influence of human activities and its eco-environmental effects, and study the formation mechanism, biogeochemical process and its impact on the ecological environment of the hypoxia zone in the East China Sea. From May to August, 2018, submarine cable route survey for this project was carried out in the East China Sea. The survey vessel used is the M.V.Heke 2. Survey contents include Bathymetric and topographic survey, seabed conditions, shallow geology, soil physical and mechanical properties, corrosive environmental parameters, marine hydrographic and meteorological elements and so on. Equipment and sensors, such as side scan sonar, sub-bottom profiler, magnetometer, sampler and CPT were employed in this survey. This survey finished 6500km geophysical survey lines, 103 stations of seabed sampling, 55 stations of in-situ test, 48 stations of marine hydrographic observation within 95 days. This work was completed under the guidance of the national standards of the People's Republic of China (GB/T 17502-2009) "Specifications for submarine cable and pipeline route investigation ". The HCN laser on EAST is a continuous wave glow discharge laser with 3.4 m cavity length and 120 mW power output at 337 μ m wavelength. Without a temperature-controlled system, the cavity length of the laser is very sensitive to the environmental temperature. An external power feedback control system is applied on the HCN laser to stabilize the laser output power. The feedback system is composed of a stepping motor, a PLC, a supervisory computer, and the corresponding control program. One step distance of the stepping motor is 1 μ m and the time response is 0.5 s. Based on the power feedback control system, a stable discharge for the HCN laser is obtained more than eight hours, which satisfies the EAST experiment.
