Our previous study demonstrated that the total saponins from Paris forristii (PCT3) had obvious inhibitory effect on the proliferation of adriamycin-resistant human breast adenocarcinoma cells (MCF-7/ADM), and this effect was significantly stronger than that in parental cells (MCF-7). This study was designed to test the reversal effect of PCT3 on MCF-7/ADM cells and to understand its mechanism of action. Results demonstrated that low cytotoxic concentrations of PCT3 (0.3, 1 and 3 µg/mL) reversed resistance of MCF-7/ADM cells to ADM, cisplatin (DDP) and 5-fluorouracil (5-FU), with reversal fold of 16.4, 19.5 and 31.7 for ADM, 1.6, 1.4 and 1.4 for DDP, 1.7, 1.8 and 5.6 for 5-FU, respectively. Moreover, PCT3 significantly increased the accumulation of ADM and Rhodamine 123 (Rh123) in MCF-7/ADM cells, suggesting that PCT3 may act by affecting the function of drug efflux pump P-glycoprotein (P-gp), which is encoded by MDR1 gene. Both MDR1 gene and P-gp protein expression was downregulated by PCT3 treatment. Further results demonstrated that p38 mitogen-activated protein kinase (MAPK) and extracellular signal-regulated kinase (ERK) pathway was remarkably activated in MCF-7/ADM cells, inhibition of p38 or ERK attenuated P-gp expression. While, only the phosphorylation level of ERK was downregulated by PCT3, indicating that PCT3 sensitized P-gp overexpressed MCF-7/ADM cells to ADM via inhibition of ERK signaling pathway.

OBJECTIVE: To investigate the effect of corticosteroids therapy on the inflammatory response in a critically ill coronavirus disease 2019 (COVID-19) patient. METHODS: A 55-year old female patient with critical ill COVID-19 was admitted in Taizhou Hospital on January 19, 2020. The patient was treated with methylprednisolone 80 mg on the 2nd day after admission. Thereafter, the dose was adjusted in a timely manner and the therapy lasted for 13 days. The peripheral lymphocyte subsets (CD3 (+)T, CD4 (+) T, CD8 (+) T, NK cells, B cells), as well as serum levels of lymphocyte factors (IL-2, IL-4, IL-6, IL-10, TNF-α, IFN-γ) were dynamically monitored. RESULTS: On D1 of admission, the numbers of peripheral blood CD3 (+) T, CD4 (+) T, CD8 (+) T, and NK cells were significantly lower than the normal range. With the improvement of the disease, the numbers of CD3 (+) T, CD8 (+) T and CD4 (+) T cells gradually recovered and showed a linear growth trend (linear fitting equation: Y=18.59 X+109.4, P < 0.05). On D2 of admission, the patient's IL-6 and IL-10 levels were significantly higher than normal values, IFN-γ was at a normal high value, and then rapidly decreased; IL-2, IL-4, and TNF-α were all in the normal range. On the D6 and D7, the IL-6 and IL-10 decreased to the normal range for the first time. On the D18, the sputum virus nucleic acid test was negative for the first time, and the fecal virus nucleic acid test was still positive; on the D20 the sputum and fecal virus nucleic acid test were both negative. On D34, the patient recovered and was discharged. At the discharge the muscle strength score of the patient was 44 and the daily life ability evaluation was 90. CONCLUSION: In the absence of effective antiviral drugs, early use of appropriate doses of corticosteroids in critically ill patient with COVID-19 can quickly alleviate inflammatory response and improve clinical symptoms, however, it may reduce the number of T cells, and to adjust the dose in time is necessary.

Ethnopharmacological relevance Chong-lou (Paris polyphylla var. yunnanensis or P. polyphylla var. chinensis) is traditionally used as an anticancer medicine in China. It is also the material basis of some Chinese patent anticancer medicines, such as Gan-Fu-Le capsules, Bo-Er-Ning capsules, Lou-Lian capsules, Ruan-Jian oral liquid, and Qi-Zhen capsules. P. forrestii, a substitute for Chong-lou, is planted at a large scale in the Yunnan Province of China. Aim of the study To clarify the active chemical constituents of P. forrestii and evaluate the in vitro and in vivo anticancer activities of the total saponins from P. forrestii. Materials and methods The total saponins of P. forrestii were extracted and separated to yield pure compounds by chromatographic techniques, and the structures of the isolates were elucidated by spectroscopic methods. The cytotoxicity of the crude extracts, total saponins, and chemical constituents were evaluated using an MTS assay. In vivo antitumor activities of the total saponins from P. forrestii were measured using H22 tumor-bearing mice by intraperitoneal (ip) administration. Results Eight compounds, including polyphyllin D (1), formosanin C (2), dioscin (3), diosgenin-3-O-α- l -rhamnopyranosyl-(1→2)-β- d -glucopyranoside (4), paris saponin H (5), pennogenin-3-O-α- l -rhamnopyranosyl-(1→2)-[α- l -rhamnopyranosyl-(1→4)]-β- d -glucopyranoside (6), pariposide A (7), and crustecdysone (8), were isolated from the total saponins of P. forrestii. The total saponins and compounds 1–6 showed significant inhibitory activity against the growth of the HL-60, SMMC-7721, A-549, MCF-7, and SW480 cell lines. The total saponins from P. forrestii had a tumor-inhibitory effect in H22 tumor-bearing mice upon ip (2.25 mg/kg dose) administration, with an inhibition rate of 42.6% compared with cisplatin (ip, 2 mg/kg dose, 53.9% inhibition rate). Conclusion The results support that P. forrestii could be a substitute for P. polyphylla var. yunnanensis as an anticancer medicine.

Mineralogy and geochemistry of the four main workable coal seams (No.6, No.7, No.8, and No.11) of Late Permian age from the Songzao Coalfield, Chongqing, Southwest China, were examined using inductively coupled plasma-mass spectrometry (ICP-MS), X-ray fluorescence spectrometry (XRF), cold-vapor absorption spectrometry (CV-AAS), ion-selective electrode (ISE), scanning electron microscopy equipped with an energy-dispersive X-ray spectrometer (SEM-EDX), and X-ray diffraction analysis (XRD). The results showed that the main workable No.8 Coal that accounts for about 60% of the total coal reserves in the Songzao Coalfield was not enriched in hazardous trace elements. The No.11 Coal has high concentrations of alkaline elements, Be (9.14 μg/g), Sc (12.9 μg/g), Ti (9508 μg/g), Mn (397 μg/g), Co (23.7 μg/g), Cu (108 μg/g), Zn (123 μg/g), Ga (32 μg/g), Zr (1304 μg/g), Nb (169 μg/g), Hf (32.7 μg/g), Ta (11.4 μg/g), W (24.8 μg/g), Hg (0.28 μg/g), Pb (28.1 μg/g), Th (24.1 μg/g), and rare earth elements (509.62 μg/g). The concentration of Nb and Ta in the No. 11 Coal is higher than the industrial grade, and their potential utilization should be further studied. Besides pyrite, quartz, calcite, and clay minerals, trace minerals including chalcopyrite, marcasite, siderite, albite, mixed-layer clay minerals of illite and smectite, monazite, apatite, anatase, chlorite, and gypsum were found in the No.11 Coal. It should be noted that alabandite of hydrothermal origin and anatase occurring as cement were identified in coal. In addition, the clayey microbands derived from alkaline volcanic ashes were identified in the coal. The dominant compositions of these clayey microbands were mixed-layer clay minerals of illite and smectite, which were interlayered with organic bands. The modes of occurrence of alkaline volcanic ash bands indicate that the volcanic activities were characterized by the multiple eruptions, short time interval and small scale for each eruption during peat accumulation. The alkaline volcanic ashes were the dominant factors for the enrichment of alkaline elements, Nb, Ta, Zr, Hf, and rare earth elements, and the sulfide minerals are the main carriers of Ga, Cu, and Hg in the No. 11 Coal.

Parts of Xuan Wei County, Yunnan Province, China have the highest known lung cancer mortality in nonsmoking women. This high mortality displays a clear spatial relationship to the mines producing coal from the uppermost Permian. Geochemical, petrographic, and grain size analyses were undertaken on a set of coal samples from Xuan Wei.
