Host defense peptides (HDP) are multifunctional effectors of the innate immune system, which has antimicrobial and pleiotropic immunomodulatory functions. Although there is a very sophisticated superposition of adaptive immune systems in vertebrates, this system is still essential. As an important family of HDP, cathelicidins are also known for their broad-spectrum antibacterial activity against bacteria, fungi, and enveloped viruses. It has been found in humans and other species, including cattle, pigs, sheep, goats, chickens, rabbits, and some kind of fish. Among them, cathelicidins in birds were described for the first time in 2005. This review focuses on the structure, biological activities, expression, and regulation of avian cathelicidin, especially main effects of host defense cathelicidin on potential therapeutic applications. According to the results obtained both in vitro and in vivo, good perspectives have been opened for cathelicidin. Nevertheless, further studies are needed to better characterize the mechanisms of action underlying the beneficial effects of cathelicidin as novel therapeutic alternatives to antibiotics.

AbstractCathelicidin host defense peptides (cHDPs) widely exist in animals with the functions of innate immunity and acquired immunity. To investigate the tissue and sequential expression patterns of the dCATHgene, which encodes the only cHDPs found in waterfowls, we analyzed the internal organs of healthy ducks (Shaxoxing Sheldrake) from 1-day-old to 28-day-old raised in the natural environment. The dCATH mRNA expression level across tissues was determined viaquantitative real-time polymerase chain reaction (qRT-PCR). Monoclonal antibody against the mature dCATH peptide used in enzyme-linked immunosorbent assay (ELISA) was prepared by the lymphocyte hybridoma technique for the first time. The concentration of the dCATH peptide in tissues was determined by ELISA. It showed that the dCATH mRNA had a clear tissue-specificity and age-dependent expression pattern, and it was highly expressed in the bone marrow and moderately expressed in the liver, kidney, spleen, pancreas and bursa of healthy duck. The peptide levels of dCATH in liver, kidney, spleen, pancreas and bursa changed with the age of duck, and the peptide concentration in these organs was between 88 and 128 ng/g tissue. For the first time, we characterized the concentration of the cHDP dCATH in vivo. Our study may represent new insights into the crosstalk of innate immunity, tissue development and age.

ABSTRACTAntimicrobial peptides (AMPs) have been paid considerable attention because of their broad-spectrum antimicrobial activity and a reduced possibility of the development of bacterial drug resistance. Fowlicidin-3 (Fow-3) is an identified type of chicken cathelicidin AMP that has exhibited considerable antimicrobial activity and cytotoxicity. To reduce cell toxicity and improve cell selectivity, several truncated peptides of fowlicidin-3, Fow-3(1-15), Fow-3(1-19), Fow-3(1-15-20-27), and Fow-3(20-27), were synthesized. Our results indicated that neither the N- nor C-terminal segment alone [Fow-3(1-15), Fow-3(1-19), Fow-3(20-27)] was sufficient to confer antibacterial activity. However, Fow-3(1-19) with the inclusion of the central hinge link (-AGIN-) retained substantial cell toxicity, which other analogs lost. Fow-3(1-15-20-27) displayed potent antimicrobial activity for a wide range of Gram-negative and Gram-positive bacteria and no obvious hemolytic activity or cytotoxicity. The central link region was shown to be critically important in the function of cell toxicity but was not relevant to antibacterial activity. Fow-3(1-15-20-27) maintained antibacterial activity in the presence of physiological concentrations of salts. The results from fluorescence spectroscopy, scanning electron microcopy, and transmission electron microcopy showed that Fow-3(1-15-20-27) as well as fowlicidin-3 killed bacterial cells by increasing membrane permeability and damaging the membrane envelope integrity. Fow-3(1-15-20-27) could be a promising antimicrobial agent for clinical application.
