Ocean surveying is of great significance to mankind’s development and utilization of the ocean. Island and reef area surveying is an important part of ocean surveying and mapping. The Ice, Cloud, and Land Elevation Satellite-2 (ICESat-2) has been proven to have a certain bathymetric capability. However, the precise extraction of seafloor signal photons in these regions remains a challenge. This study introduces a method for extracting seafloor photons that is water-depth adaptive and works at various depths. In addition, we propose a method to evaluate ICESat-2 seafloor signal photons by underwater light-beam propagation and noise modeling, using the decision tree method to classify signal photons into high-, medium-, and low-confidence levels. The results indicate that the method exhibits better signal continuity, better slope adaptability, and better signal-to-noise ratio (SNR) adaptability in seafloor signal photon detection, and remains more surface object signal photons in island signal photon detection than adaptive variable ellipse filtering bathymetric approach (AVEBM) method. The high-, medium-, and low-confidence seafloor signals exhibit consistencies ( ${R}^{2}$ ) of 0.9954, 0.9926, and 0.9874, respectively. The root-mean-square errors (RMSEs) are 0.49, 0.66, and 0.93 m, and the mean absolute errors (MAEs) are 0.24, 0.44, and 0.86 m, correspondingly. Higher-confidence photons perform significantly better than lower-confidence photons. The confidence evaluation of seafloor photons will provide an important reference for users and will lay the foundation for further research into the use of ICESat-2 for offshore bathymetry.

Due to factors such as overfishing, hydropower development, and water pollution, their wild populations have drastically declined, with many species being classified as endangered or vulnerable. Understanding the driving factors behind the gut microbiota of cold-water fish holds significant implications for conservation. Therefore, to investigate the influence of different factors on the gut microbes of cold-water fish, 112 gut samples from farmed and wild habitats were collected from Schizothorax wangchiachii(herbivorous), Schizothorax kozlovi(omnivorous) and Percocypris pingi(carnivorous), in the Jinping Dahewan range of lower reaches of the Yalong River. The results of 16 S rRNA gene sequencing of the gut contents showed that habitat was the major factor influencing the gut microbes of the three cold-water fishes. In terms of microbial composition, farmed cold-water fish exhibited a dominant bacterial phylum, Proteobacteria (82 %), while those in wild environment presented multiple dominant taxa, such as Proteobacteria (38 %), Fusobacteria (30 %), Firmicutes (11 %), and Cyanobacteria (11 %). Gut microbial diversity, both α diversity and β diversity, in wild cold-water fish was significantly higher than that in farmed individuals. Network complexity of gut microbiota was notably higher in wild cold-water fish than in farmed individuals. From farmed to wild environments, the relative influence of deterministic processes in the gut microbiota assembly mechanism of cold-water fish gradually diminishes, while the contribution of stochastic processes steadily increases. The gut microbiota construction mechanism in farmed cold-water fish was primarily influenced by deterministic processes of homogeneous selection, while in wild cold-water fish, it was mainly driven by stochastic processes of homogeneous dispersal. Overall, this study is an important reference for the study of the gut microbiota of cold-water fish and the conservation of their wild populations.

As the first line of host defense against invading pathogens, neutrophils have an inherent phagocytosis capability for the elimination of foreign agents and target loading upon activation, as well as the ability to transmigrate across blood vessels to the infected tissue, making them natural candidates to execute various medical tasks in vivo. However, most of the existing neutrophil-based strategies rely on their spontaneous chemotactic motion, lacking in effective activation, rapid migration, and high navigation precision. Here, we report an optically manipulated neutrophil microcraft in vivothrough the organic integration of endogenous neutrophils and scanning optical tweezers, functioning as a native biological material and wireless remote controller, respectively. The neutrophil microcrafts can be remotely activated by light and then navigated to the target position along a designated route, followed by the fulfillment of its task in vivo, such as active intercellular connection, targeted delivery of nanomedicine, and precise elimination of cell debris, free from the extra construction or modification of the native neutrophils. On the basis of the innate immunologic function of neutrophils and intelligent optical manipulation, the proposed neutrophil microcraft might provide new insight for the construction of native medical microdevices for drug delivery and precise treatment of inflammatory diseases.

Dynamic rotation and deformation of human red blood cells (RBCs) are extremely important to investigate the survival and mechanical features of cells, which will be of great physiological and pathological significance. Here, we report an optical approach that is capable of both rotating and deforming RBCs with light from two tapered fiber probes (TFPs). With laser beams at the wavelength of 980 nm injected into the TFPs, a single RBC was rotated around different axes while single or multiple RBCs were stretched by adjusting the points of action and magnitude of the optical forces from the TFPs. The biological safety of the approach was also discussed by taking the laser power required into account.
