Twenty one UV-sensitive rad mutants were tested for their sensitivity towards DEB. All mutants were more sensitive to this treatment than the wild type. Seven mutants were classified as supersensitive to DEB (radl-1, 2, 3, 6, 15 and 18-2), while only rad2 and rad3 can be classified as supersensitive to UV. For all mutants ability for liquid holding recovery (LHR) after UV and DEB was compared. Mutants radl-1, 3, 5, 6, 9 and 11 differ in their response to LH afterr the two treatments. Survival of radl-1 and rad3 increases significantly during LH after DEB but not after UV exposure. In contrast rad5, 6, 11 and 22 show marked LHR after UV but no increase of survival after DEB treatment.

Maximal liquid-holding recovery (LHR) of the DEB-treated rad3 mutant occurs at 30° C in buffer supplemented with glucose. Addition of cycloheximide (CHX) to the buffer, the increase in cell density above 2 × 107/ml as well as lowering of temperature during liquid holding (LH) below 27° C decrease considerably the cell capacity for recovery. LHR does not take place at 5° C. No measurable DNA synthesis or degradation occurs in cells held in buffer alone, while addition of 0.02% glucose results in incorporation of radioactivity into DNA both of DEB-treated and control cells. Similarly, protein synthesis was observed only in cultures held in buffer supplemented with glucose. Cells transfered to growth medium directly after treatment complete one round of DNA replication and at least one division cycle, but further DNA replication and cell division are inhibited. Cells placed in growth medium after 5 days LH show an increased rate of DNA replication and cell division. Completion of the first posttreatment round of DNA replication in growth medium abolishes ishes the cell capacity for LHR. DEB treatment results in abnormal cell division of the rad3 mutant, giving ‘colonies’ consisting of several cells, usually abnormal in shape, held together by common cell walls.

The conditional cdc8 mutant is known to be defective, under restrictive conditions, in the elongation of DNA during synthesis. In yeast the CDC8 gene encodes thymidylate kinase. We show here that UV-induced gene conversion and gene mutation events require the participation of this CDC8 gene. Thus, the same thymidylate kinase is incolved both in DNA replication and in UV-induced gene conversion and gene mutation in yeast.

The method of analysis of DNA in mammalian cells by alkaline elution from filters (Kohn et al. 1974) was adapted for studies on yeast DNA. By this technique spheroplasts obtained from yeast cells are lysed on filters and single-stranded DNA fragments selectively eluted by alkaline solutions. The procedure was applied to monitor the occurrence of replication intermediates and production of DNA single-strand breakage by MMS, and its repair in growth medium.

It has been found that the repair of single strand breaks is defective in the DNA replication mutants cdc8-1 and cdc8-3 of Saccharomyces cerevisiae both in permissive (23°C) and restrictive conditions (36°C). In permissive conditions we observed a significant delay in single strand break repair in a diploid strain HB7 (cdc8-1/cdc8-1), as compared with the wild-type strain. Under restrictive conditions no repair was observed, but rather degradation of MMS-damaged DNA occurred. It has been also found that the repair of single strand breaks in yeast is inhibited by cycloheximide but not by hydroxyurea.

Incubation of cdc8 mutants of the yeast Saccharomyces cerevisiae in YPD under permissive conditions, when DNA replication is taking place, prior to transfer to restrictive conditions, strongly stimulates induction of cdc+ colonies of ethyl methane sulphonate (EMS)- and methyl methane sulphonate (MMS)-treated yeast strains HB23 (cdc8-1/cdc8-3), HB26 (cdc8-3/cdc8-3) and HB7 (cdc8-1/cdc8-1). After diepoxybutane (DEB) treatment, both the induction of cdc+ colonies and their stimulation after incubation in YPD under permissive conditions is low. The results obtained show that stimulation of induction of cdc+ colonies under permissive conditions occurs not only after UV-treatment, but also after treatment with such mutagens as EMS and MMS.

Summary: The induction of mitotic recombination in theCDC8 locus was studied in a diploid strain heteroallelic forcdc8 mutations (cdc8-1/cdc8-3); mitotic reversion was studied in strainscdc8-1/cdc8-1 andcdc8-3/cdc8-3. Conversion and reversion did not occur in those cells blocked at the S stage of the cell cycle by exposure to a nonpermissive temperature. In stationary phase cells irradiated just prior to exposure to temperature stress, the induction of recombinants was rather low and the induction of revertants was minimal. Conversely, a significant induction ofcdc + occurred in logarithmic phase cells subjected to the same treatment. Irradiation of synchronously dividing cultures revealed that intragenic recombination occurs at all three stages of the cell cycle- G1, S and G2. It was also found that UV-induced gene reversion can occur during the S and G2 stages, but not during the G1 stage of the cell cycle.

The Y chromosome heterochromatin in Rumex thyrsiflorus has been analyzed. In natural populations the Y chromosome shows a higher morphological variability than the X chromosome. The total duration of replication of Y chromosomes is about 2 hrs longer than that of euchromatin. Autoradiography with tritiated thymidine showed that chromocentres formed by Y chromosomes in interphase nuclei retain their heterochromatic form during DNA replication. — Y chromosome heterochromatin in interphase nuclei is stained pink, while the rest of the nucleus stains green after fast green-eosin staining for histones. — During the premeiotic stage of PMC development Y chromosomes are no longer visible as compact bodies and become more fuzzy in appearance. A diffuse state of Y coincides with intense RNA synthesis. Therefore genetic activity of Y chromosomes or their parts during premeiotic stage of microsporogenesis is postulated.

The frequency of aberrations induced by DEB depends on the temperature and concentration of the agent. The effect of DEB is delayed. Pretreatment with EDTA increased the frequency of aberrations. The frequency of aberrations was also increased after simultaneous treatment with DEB and chloramphenicol or streptomycin. Pre- and posttreatment with protein synthesis inhibitors had a reverse effect, the frequency of aberrations was decreased.

The crude enzyme powder from Rhizopus nigricans was immobilized by sorption and subsequent cross-linking with glutaraldehyde on collagen and polytetrafluoroethylene (PTFE) membranes. Lipolytic membranes were applied to plant oils hydrolysis. The comparison of the two types of membranes by calculating the time required to obtain 1 mole of free fatty acids (FFA) from 1 m2 of membrane area, indicates that hydrophobic PTFE membrane is a better one in spite of the fact that the amount of protein sorbed on PTFE membrane is about three times smaller than that for collagen membrane. The hydrolysis of sunflower oil was the most efficient at the temperature of 37 °C and a pH of 7. At these conditions the specific activity after immobilization was about four times higher than that of the soluble enzyme.

Abstract: Faraday rotation and linear birefringence of GdCrO3 were measured in the spectral range 0.8 to 2.0 μm at liquid nitrogen temperature. Good agreement is obtained between the experimental data and the theoretical values of rotation, calculated by the use of the formula describing Faraday rotation in the presence of the linear birefringence. The intrinsic Faraday rotation as a function of the wavelength is determined and it is probably due to higher-energy optical transitions.
