The separate (+) and (−) enantiomers of the acyclic guanosine analogue 9-[4-hydroxy-2-(hydroxymethyl)-butyl]guanine (2HM-HBG) were tested for inhibition of varicella-zoster virus (VZV), herpes simplex virus type 1 (HSV-1) and herpes simplex virus type 2 (HSV-2). In all cases the (−) enantiomer was the most active enantiomer. The (+) enantiomer was 10 times less active than the racemate against VZV and inactive against HSV-1 and -2.The parent compound, 9-(4-hydroxybutyl)guanine, containing an unbranched side chain, was inactive against VZV, whereas substitution with a hydroxymethyl group at the 2 or 3 position of the side chain conferred anti-VZV activity. The effect of hydroxymethyl substitution may increase the recognition of the compound by the VZV thymidine kinase by increasing its similarity to the natural substrate thymidine. Further substitution of the side chain of the parent compound with oxygen, fluorine or hydroxyl groups did not confer antiviral activity against VZV.Two VZV strains were isolated which could be grown in the presence of high concentrations of 2HM-HBG and which were also cross-resistant to other nucleoside analogues. These strains induced very little viral thymidine kinase activity in infected cells and thus were probably deficient for a functional thymidine kinase.2HM-HBG exhibited a persistent antiviral effect even after the nucleoside was removed from the medium of VZV-infected cells, whereas acyclovir did not show this effect.

Traffic tests conducted near Barrow, Alaska with a 7-ton SK-5 Air Cushion Vehicle have shown that these types of vehicles can provide year-round high-speed transport capability over a variety of relatively level, low strength terrains. The ecological impact of ACV traffic over easily degradable tundra terrains is not nearly as significant as that of wheeled or tracked vehicle traffic.

The acyclic guanosine analog (R,S)-9-[4-hydroxy-2-(hydroxymethyl)butyl]guanine, (+/-)2HM-HBG, is an effective inhibitor of herpes simplex virus and varicella-zoster virus infections in vitro. This report is concerned with the pharmacokinetic evaluation of the drug in rats and monkeys and its antiviral activity in African green monkeys infected with simian varicella virus (SVV), a virus closely related to varicella-zoster virus that is also susceptible to inhibition by (+/-)2HM-HBG. Elimination half-lives in plasma following intravenous administration to monkeys (100 mumol/kg of body weight) ranged from 1.8 to 2.2 h, and total body clearance was 9.0 +/- 0.4 ml/min per kg (mean +/- standard error). After oral administration, levels in plasma were low, with a maximum concentration of the drug of only 3.1 +/- 0.8 microM, a time to reach maximum concentration of drug of 2.7 +/- 0.4 h, and an oral bioavailability of 10.6 +/- 1.4%. Because of the low oral bioavailability, SVV-infected monkeys were treated intramuscularly with (+/-)2HM-HBG. (+/-)2HM-HBG at a dosage of 10 mg/kg of body weight per day allowed moderate viremia, whereas a dosage of 30 mg/kg of body weight per day strongly suppressed viremia with minimal numbers of virus plaques from blood specimens collected at days 3, 5, and 7 postinfection and complete clearance at day 9 postinfection. Titers of antibody to SVV were also low. Treatment three times daily was somewhat more efficacious than treatment twice daily. Thus, (+/-)2HM-HBG is an effective inhibitor of SVV replication in vivo, despite the fact that leves of (+/-)2HM-HBG in plasma were low at extended periods of time and below the concentration of drug giving 50% inhibition of plaque formation obtained in vitro.

The triphosphates of the antiherpesvirus acyclic guanosine analogs 9-[4-hydroxy-2(hydroxymethyl)butyl] guanine (2HM-HBG), 9-(2-hydroxyethoxymethyl)guanine (acyclovir [ACV]), and 9-(3,4-dihydroxybutyl)guanine (buciclovir) were examined for their effects on partially purified varicella-zoster virus (VZV) DNA polymerase as well as cellular DNA polymerase alpha. The triphosphate of 2HM-HBG competitively inhibited the incorporation of dGMP into DNA catalyzed by the VZV DNA polymerase. 2HM-HBG-triphosphate (2HM-HBG-TP) had a higher affinity for the dGTP-binding site on the VZV DNA polymerase than did dGTP; apparent Km and Ki values of dGTP and 2HM-HBG-TP were 0.64 and 0.034 microM, respectively. ACV-triphosphate (ACV-TP) was found to be the most potent inhibitor of VZV DNA polymerase. ACV-TP had a 14 and 464 times better direct inhibitory effect than 2HM-HBG-TP and buciclovir-triphosphate, respectively. The cellular (human embryonic lung fibroblast) DNA polymerase alpha inhibition was related to viral polymerase inhibition as efficacy ratios: 2HM-HBG-TP had a ratio of more than 1,000, which appeared to be similar to that of ACV-TP.

The activity and mode of action of the new nucleoside analog (RS)-9-[4-hydroxy-2-(hydroxymethyl)butyl]guanine (2HM-HBG) against varicella-zoster virus (VZV) were determined. In cell culture, replication of different strains of VZV was inhibited to 50% by 0.4 to 0.7 microM 2HM-HBG, while 685 microM was required to inhibit 50% of the DNA synthesis in uninfected human lung fibroblasts. A thymidine kinase-negative VZV strain was not inhibited by 100 microM 2HM-HBG. Inhibition of VZV replication was not reversible after 7 to 14 days of incubation, depending on the multiplicity of VZV. 2HM-HBG was shown to be selectively phosphorylated by purified VZV thymidine kinase, with an inhibition constant of 32.5 microM. The antiviral activity of 2HM-HBG in cell culture was decreased by the addition of deoxythymidine and deoxycytidine but not by other ribo- or deoxyribonucleosides.

This study was performed to obtain information on the enzymatic background to the antiviral activity of acyclic guanosine analogs. Five acyclic guanosine analogs, the (R)- and (S)-enantiomers of 9-(3,4-dihydroxybutyl)guanine, 9-(4-hydroxybutyl)guanine, 9-[(2-hydroxyethoxy)methyl]guanine, and 9-[[2-hydroxy-1-(hydroxymethyl)ethoxy]methyl]guanine, were compared in enzyme kinetic experiments using purified varicella-zoster virus and human placenta mitochondrial thymidine kinase (TK). All analogs showed competitive patterns of inhibition in the phosphorylation of thymidine by varicella-zoster virus TK, but only low affinities and phosphorylation rates were observed. No affinity for the mitochondrial TK was observed for any of the analogs.

Auf einem 2,4 km langen Straßen- und Wegesystem wurden von 33 sehr guten Berufsfahrern 491 Versuchsfahrten mit Geschwindigkeiten bis zu 45 km/h durchgeführt. Die Fahrweise (Geschwindigkeit, Beschleunigung, Verzögerung, Lenkung) wurde fortlaufend registriert. Jeder Fahrer fuhr nüchtern und unter Alkoholeinfluß.

Bei steigender Blutalkoholkonzentration bis zu 1%. wird die Zahl der kleinen Beschleunigungen und Verzögerungen größer, die mittlere Beschleunigung kleiner. Die Fahrweise ist verlangsamt und unruhig.

Eine Methode zur Erfassung der Fahrweise durch fortlaufende Registrierung der Fahrzeugbewegungen wurde entwickelt. Im normalen Straßenverkehr eines durchschnittlichen Straßensystems mit einer Länge von 117,0 km wurden 105 Fahrten unternommen. Das Fahrerkollektiv war entsprechend der allgemeinen Fahrerfahrung der Kraftfahrer ausgewählt. Die Meßergebnisse wurden mit mathematisch-statistischen Methoden in Leistungsfaktoren umgewandelt, die die Geschwindigkeitsund die Gleichmäßigkeitsleistung messen. Die Geschwindigkeitsleistungen, die weniger als±3% vom Mittelwert abweichen, gelten als Normalleistungen. Die Geschwindigkeitsleistung streut von -8% bis + 9,2% der Durchschnittsleistung.

Versuchsfahrten mit einem Kraftwagen und Geschwindigkeitsaufzeichnungen werden beschrieben. Als Normalleistung wurden die Fahrten bei Tageslicht auf trockener Straße gewertet.

Der Personenwagen stellt die geringsten Anforderungen an den Fahrer. Das Motorrad besitzt eine rund 15mal empfindlichere Lenkung, ein doppelt so großes Beschleunigungsvermögen und eine nur halb so große Bremsverzögerung.

Nach der Gabe von Vitamin B6 im Rauschzustand zeigte sich bei den Versuchspersonen eine unterschiedliche Reaktionsweise. Teilweise war keinerlei Wirkung, teilweise eine deutliche Zustandsverschlechterung und zum Teil eine fast schlagartige Besserung mit verbleibendem und auch zunehmendem Kopfschmerz zu beobachten.

Ein qualitativer und quantitativer Nachweis des in Winterspritzmitteln enthaltenen 4,6-Dinitro-o-kresols (DNoC) mit Nitron aus Leichenorganen wird beschrieben. Die Unterscheidung gegenüber Pikrinsäure ist somit möglich.

Die Bewertung von Alkoholbefunden aus Leichenblut und Harn wird diskutiert. Ein Anstieg der Herzblut-Alkoholkonzentration um 0,6‰ durch Diffusion wird beschrieben.

Es wurden 91 auswertbare Trinkversuche entsprechend den Trinksitten des einzelnen durchgeführt, um die Anstiegsgeschwindigkeit der Blutalkoholkonzentration nach der Gabe von Weinbrand, Doppelkorn und Steinhäger (39–40Vol.-%) zu untersuchen. Entsprechend der Magenfüllung wurden 3 Gruppen gebildet, die Anstiegszeiten der Blutalkoholkonzentration mit deutlichen Unterschieden ermittelt.

Für die Lenkweise konnten keine Leistungsfaktoren definiert werden. Die Werte geben nur die Änderung der Lenkweise unter Alkoholeinfluß gegenüber der der Nüchternfahrt an. Die Anzahl der Lenkradauscohläge ist im Mittel bei zunehmender Blutalkoholkonzentration bis 1‰ um 18%, zwischen 1,0 und 1,5‰ und im Gipfel um 24%, bei abnehmender Konzentration zwischen 1,5 und 1,1‰ um 20%, zwischen 1,1 und 0,1‰ um 18% gegenüber den Werten der Nüchternfahrten verschieden. Die Größe der Lenkradausschläge ist im Mittel bei zunehmender Blutalkoholkonzentration bis 1,0‰ um 24,5%, zwischen 1,0 und 1,5‰ um 35%, im Gipfel um 38% und bei abnehmender Konzentration um 28% verschieden. Die Lenkweise ist von derjenigen der Nüchternfahrten auf gerader, breiter Fahrbahn stärker verschieden als auf Schwierigkeiten bietender Strecke.

Die Begriffe des Konzentrationsabfalles \ und des Reduktionsfaktors sowie die Variationsbreiten dieser Größen werden diskutiert. Die mittlere Abbaugeschwindigkeit während langer Zeitspannen lag bei Blutalkoholkonzentrationen oberhalb 1,5‰ mindestens bei 0,16‰, wenn der errechnete Höchstwert (r= 0,7) annähernd erreicht wurde. Die statistische Auswertung von den 4118 Einzelentnahmen und den Entnahmen in zeitlichem Abstand bei 922 Personen zeigt die am häufigsten beobachtete Blutalkoholkonzentration bei 1,8‰und ein mittleres \60von 0,184‰. Der Faktor β nimmt mit steigender Blutalkoholkonzentration zu. Die Unterschiede zwischen den Bereichen 0,41—1,2‰ und oberhalb 2,0‰ sind statistisch gesichert, die mittleren \60-Werte von 0,17 bzw. 0,203‰ damit signifikant.

We therefore analysed the dThd concentration in human plasma and skin and in organs of guinea pig, the common experimental animal. High-performance liquid chromatography showed low amounts of dThd in human skin, 0.20–1.15 nmolg, whereas guinea pig skin and spleen had 20–30 nmolgand the concentration in guinea pig plasma was 10-times higher than in human plasma. These animals are therefore in this respect less suitable as accurate models for antiviral nucleoside activity in humans.
