3. What we know about volunteer tourism - an approach to motivations and impacts

- Abstract
Generally defined as an activity in which people decide to volunteer in development or conservation projects, voluntourism is one of the most fast-growing alternative tourism markets, arousing interest from several different people and creating an increasing number of volunteers. These developments are being used by many organisations as a way of using volunteers' expertise as important benefits. Our paper analyses and describes the concept of voluntourism, as well as the motivations that drive the volunteers, and the impacts or benefits of their projects' experience in the host community and in themselves. We did a literature review, firstly using a bibliometric analysis, to deeply understand and categorise the generic concept. Secondly, we did ten exploratory online interviews/questionnaires to volunteers with experiences in international volunteering programs. The results show that this topic has become a trend in recent years and that understanding the motivations of volunteers as well as the benefits and impacts of the projects are essential to the development of voluntourism.

4. Impacts of climate changes on risk zoning for cowpea in the Amazonian tropical conditions.

The climate risk zoning was carried out for cowpea cultivated in northeastern Amazonia, Pará state, Brazil. Système d’Analyse Régionale des Risques Agroclimatologiques (SARRA) crop model was used and calibrated from data obtained in field experiments conducted between 2013 and 2016 in Castanhal, state of Pará. Low climate risk areas were defined as those with water requirement satisfaction index (WRSI) greater than or equal to 0.5 during flowering and early grain development and less than 20 mm of total rainfall during grain maturation and harvest for at least 80% of the simulated sowings of the crop. Simulations of the current and future climate conditions were carried out with the regional climate modelling system version 4 (RegCM4) nested with the HadGEM2 global circulation model with RCP 4.5 IPCC-AR4 scenario. The results show that climate changes are likely to reduce the duration of the optimal period for sowing cowpea in the mentioned region. [ABSTRACT FROM AUTHOR]

5. The Dark Side of Leadership.

Previous studies have ignored the leadership role in management and documented complex challenges in this field with respect to effective leadership. Related to the issue, destructive leadership has become an emerging area of research. The purpose of current study is to examine the impact of destructive leadership behaviors on turnover intentions and deviant behavior through the mediation of job stress of leaders. In addition, Self-leadership should be integrated within a nomological network, and its associations with the Dark Triad of personality comprising narcissism, Machiavellianism, and psychopathy, explored. In this study, Narcissism was positively associated with self-leadership scales, whereas Machiavellianism and psychopathy were not. Findings are discussed and future lines of research are proposed. Public Service Obligations are a key mechanism for countries to improve connections among islands and remote regions with the Continent. While Public Service Obligations are allowed by the European Commission, they represent an interference with free market competition, and impact on the entrance of competitors, prices and service frequency. This study employs Canarian air market data for 2002–2015 to econometrically evaluate the effects of two changes in European policy on the number of passengers transported. 13. Marcha Gait Simulation Test Decrease Antioxidative Biomarkers in Four-Beat Gaited Horses.

- Abstract
The aim of the present study was to characterize the modification of anti-oxidative biomarkers, as well as hematological and protein metabolism biomarkers, in four-beat gaited horses after the marcha gait simulation test (MST). Twelve adult Mangalarga Marchador horses were submitted to the MST and blood samples were taken at four time points: pretest/fasting; immediately after the MST; 15 and 120 minutes after the MST. The following measurements were taken: red blood cells count, hemoglobin, hematocrit (HT), white blood cells count, lymphocytes (LYMPH), platelets (PLAT), mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration, the standard deviation of red cell distribution width (RDW-SD), the coefficient of variation of red cell distribution width, glutathione peroxidase (GPX), superoxide dismutase (SOD), total plasma proteins (TPP), uric acid (AcUr), urea (URE), creatinine (CREAT), and albumin (ALB). The results were analyzed using analysis of variance and Tukey test ( P < .05). The results confirmed reductions in GPX and SOD ( P < .05) and increases in AcUr, TPP, ALB, URE, CREAT, red blood cells count, HT, Hb, white blood cells count, LYMPH, PLAT, MCV, and RDW-SD ( P < .05) immediately after the MST. Lymphocytes reduced after the 120-minute recovery period ( P < .05). In conclusion, the MST significantly decreased the antioxidative biomarkers, immediately after the MST, and produce the changes in protein and hematological biomarkers. These results are important for this group of athlete animals, who may participate frequently in competitions with short intervals between gait tests. [ABSTRACT FROM AUTHOR]

In this work, we present the fundamentals of the estimation of the energy efficiency in an industrial coal boiler based in novel optical combustion diagnostics variables and several machine learning regression methods. The total radiation Radtand flame temperature Tfwere considered. The inclusion of those variables allows to increase the overall performance in the estimation of the energy efficiency. The comparison in the performance of the tested methods for regression, suggest that Extreme Learning Machines in combination with Partial Least Squares for regression, lead to the best performance with a Pearson correlation coefficient R≈ 0.7 in the test data set.

Objective: The aim of the study was to assess the initial psychometric properties of the Cognitive Distortions Questionnaire (CDQuest) in its Brazilian Portuguese version tested in adult undergraduate students. Methods: Brazilian undergraduate medical and psychology students comprising the sample (n=184) completed the following measures: Cognitive Distortions Questionnaire, Beck Depression Inventory and Beck Anxiety Inventory. These self-report instruments were administered collectively in classrooms. Results: The Cognitive Distortions Questionnaire showed adequate internal consistency (Cronbach's alpha=0.85) and concurrent validity with Beck Depression Inventory (r=0.65, p<0.001) and Beck Anxiety Inventory (r=0.52, p<0.001). Furthermore, it was able to discriminate between groups possessing depressive (Beck Depression Inventory composite score ≥12) and anxious (Beck Anxiety Inventory composite score ≥11) indicators from those not possessing them (p<0.001). Principal components showed the measure was unidimensional, and it explained about 29 percent of the data variance. A confirmatory factor analysis showed that all the regression coefficients are greater than or equal to 0.40 Conclusion: The original Brazilian version of the Cognitive Distortions Questionnaire is adequate for use in the context of national undergraduate students and is able to separate different cognitive distortions. 21. On measuring tele

The authors developed a method for measuring the telic sensitivityof individual members in a group, derived from the sociometric test, which has to be previously performed. Such a measure is subdivided into 4 indices: composite indices of Positiveand Negative Perception, and composite indices of Positiveand Negative Mutuality. Each index scores from 0 to 100. The indices have a normal distribution and, whenever the choices are randomized, an expected mean of 50 (SD= 11.3), irrespective of the size of the groups and the number of the received preferences and rejections. Young girls and boys (mean age 12 years) in actual school groups tend to have higher scores, especially for Positive Perceptionand Positive Mutuality, as opposed to randomized virtual groups. The authors identify other results in order to exemplify future research in clinical and organizational psychology settings Die AutorInnen entwickelten eine Messmethode für das Tele von einzelnen Mitgliedern einer Gruppe, das sich in einem soziometrischen Test zeigt. Eine solche Messung ist unterteilt in vier Indikatoren: Verbundene Indikatoren für positive und negative Wahrnehmung und verbundene Indikatoren für die positive und negative Gegenseitigkeit. Jeder Index hat Werte von 0 bis 100. Die Indikatoren sind normalverteilt und wenn die Wahlen zufallsbestimmt sind, haben sie einen erwarteten Mittelwert von 50 (SD = 11.3), das ist unabhängig von der Gruppengröße und der Anzahl an erhaltenen Wahlen und Ablehnungen. Junge Mädchen und Buben (Durchschnittsalter 12) in Schulgruppen haben höhere Werte besonders in positiver Wahrnehmung und positiver Gegenseitigkeit im Gegensatz zu zufällig zusammengestellten virtuellen Gruppen. 23. A Proton-Pump Inhibitor Modifies the Concentration of Digestion Biomarkers in Healthy Horses.

To determine the concentration of various blood biomarkers associated with digestion in healthy horses treated with different doses of omeprazole (OMPZ), four Arabian horses without gastric ulcers were selected and distributed in one factorial with four oral treatments (Control; OMPZ BOLUS ; OMPZ 4mg/kg ; OMPZ 1mg/kg ). Control did not receive any treatment. OMPZ BOLUS were given 4 mg/kg of omeprazole in a single dose on the day before blood sampling. In the OMPZ 4mg/kg and OMPZ 1mg/kg treatments, horses were treated over 11 days. All treatments were performed 16 hours before morning feeding. The “washout” period was 21 days between rounds. After an overnight fasting period, blood samples were collected from all animals (T1), after which the animals received supplementation, and blood samples were collected after 30 minutes (T2), 1.0 hours (T3), 1.5 hours (T4), 2.0 hours (T5), 3.0 hours (T6), 4.0 hours (T7), 5.0 hours (T8), 6.0 hours (T9), and 7.0 hours (T10). Blood samples were analyzed for total plasma protein (TPP), glucose, urea, creatinine, uric acid, cholesterol, triglycerides, calcium, phosphorus, and magnesium. The results showed differences between treatments for urea, cholesterol, uric acid; between phases for glucose; and between phase and treatment for creatinine, triglycerides, phosphorus, and magnesium; however, there was no difference for TPP and calcium between treatment and between phases. Oral administration of OMPZ in healthy equines interfered with the metabolism of digestion biomarkers of lipid, mineral, and protein metabolism, although the animals were treated for a maximum of 11 days. Horses treated with a proton-pump inhibitor need to be evaluated regularly to avoid significant modification in their metabolic parameters. [ABSTRACT FROM AUTHOR]

- Abstract
In order to verify the chemical qualities of polar extract of Green Propolis produced in the State of Minas Gerais, Brazil, was analyzed by means of RP-HPLC, the concentration of eleven flavonoids, Artepillin C and p-coumaric acid were analyzed by means of RP-HPLC. Samples were collected in the months of February, March, May, July, September, and October, during a period of six years (2008 to 2013) and the results compared with purchased standards. Artepillin C was the main constituent (80-90%) found in all samples. Lower concentrations of p-coumaric acid, chrysin and kaempferide were observed in March, and of ricin and galangin in September. Respectively, these months correspond to the end of Summer and Spring period in Brazil. The variation in concentration of the analyzed constituents always occurred in the same month, during the entire six years of evaluation period. All thirteen constituents of Green Propolis were detected throughout the study period, and the average concentration of each one was similar in relation to the respective monthly collection period of each year. Due to similarities among chemical constituents of Green Propolis with those present in B. dracuncufolia, this plant was identified as being the principal source of Green Propolis.

42. Influence of Liquid Length Variation in Hydraulic Transients

- Abstract
When a problem of hydraulic transients with entrapped air in pipelines andor with watercolumn separation is tackled, the variation of the length of the liquid column is not usually taken into account. In this paper due largely to the assumption of a system with a cross section at the airwater interface in a horizontal pipeline, it is shown that in a system with entrapped air and without changes of potential energy at the airwater interface horizontal pipe the final pressure obtained in a mass oscillation in which the kinetic energy of the fluid and the liquid column length variation are neglected is the same as the final pressure obtained from the consideration of both effects. This peak pressure is also independent of the assumed polytropic exponent for the entrappedair compression. Nevertheless, the instant when the maximum is achieved is different.

Myogenic differentiation is a multistep process that begins with the commitment of mononucleated precursors that withdraw from cell cycle. These myoblasts elongate while aligning to each other, guided by the recognition between their membranes. This step is followed by cell fusion and the formation of long and striated multinucleated myotubes. We have recently shown that cholesterol depletion by methyl-β-cyclodextrin (MβCD) induces myogenic differentiation by enhancing myoblast recognition and fusion. Here, we further studied the signaling pathways responsible for early steps of myogenesis. As it is known that Wnt plays a role in muscle differentiation, we used the chemical MβCD to deplete membrane cholesterol and investigate the involvement of the Wnt/β-catenin pathway during myogenesis. We show that cholesterol depletion promoted a significant increase in expression of β-catenin, its nuclear translocation and activation of the Wnt pathway. Moreover, we show that the activation of the Wnt pathway after cholesterol depletion can be inhibited by the soluble protein Frzb-1. Our data suggest that membrane cholesterol is involved in Wnt/β-catenin signaling in the early steps of myogenic differentiation. [ABSTRACT FROM AUTHOR]

ABSTRACTClostridium difficiletoxins A and B (TcdA and TcdB) are homologous glycosyltransferases that inhibit a group of small GTPases within host cells, but several mechanisms underlying their pathogenic activity remain unclear. In this study, we evaluated the effects of TcdA on the Wnt/ß-catenin pathway, the major driving force behind the proliferation of epithelial cells in colonic crypts. IEC-6 and RKO cells stimulated with Wnt3a-conditioned medium were incubated with 10, 50, and 100 ng/ml of TcdA for 24 h, resulting in a dose-dependent inhibition of the Wnt signaling, as demonstrated by a T-cell factor (TCF) reporter assay. This was further confirmed by immunofluorescence staining for nuclear localization of ß-catenin and Western blotting for ß-catenin and c-Myc (encoded by a Wnt target gene). Moreover, our Western blot analysis showed a decrease in the ß-catenin protein levels, which was reversed by z-VAD-fmk, a pan-caspase inhibitor. Nonetheless, TcdA was still able to inhibit the Wnt/ß-catenin pathway even in the presence of z-VAD-fmk, lithium chloride (a GSK3ß inhibitor), or constitutively active ß-catenin, as determined by a TCF reporter assay. Furthermore, preincubation of RKO cells with TcdA for 12 h also attenuated Wnt3a-mediated activation of Wnt signaling, suggesting that inactivation of Rho GTPases plays a significant role in that inhibition. Taken together, these findings suggest that attenuation of the Wnt signaling by TcdA is important for TcdA antiproliferative effects.

45. Antifungal Activity of Green and Red Brazilian Propolis Extracts

Background: Diseases associated to Candida spp. are recurrent and can be difficult totreat, mainly due to the new strains resistant to the limited number of available antifungals. Objectives: The objective of this study is to evaluate the in vitro and in vivo activity of Braziliangreen propolis (GrProp) and red propolis (RdProp) ethanolic extracts against standard strains ofCandida albicans, C. tropicalis, C. krusei, C. glabrata, C. parapsilosis and 10 clinical isolates ofC. albicans. Methods: Antifungal activity in vitro was tested using the M-27-A3/CLSI protocol. The in vivo antifungalactivity was evaluated using Tenebrio molitor model and, the effect of extracts on adhesionof C. albicans in human buccal epithelial cells (BECs) was also studied. Results: GrProp and RdProp exhibited antifungal activity against at least one of the Candidastrains tested. The adhesion inhibition of C. albicans in BECs was 45% (GrProp28), 60% (Gr-Prop50) and 82% (RdProp), in comparison to amphotericin B (82%). All propolis extracts showedsynergistic activity with fluconazole and amphotericin B. GrProp50 (10 mg/kg) showed better protectionof T. molitor, blocking the progression of C. albicans infection, increasing survival and delayedthe larvae death. Conclusion: Brazilian GrProp and RdProp extracts inhibit the in vitro C. albicans growth and protectT. molitor against infection by this yeast. The physiochemical parameters found for the analyzedsamples were in accordance to the standards established by the Brazilian Legislation forpropolis and derivatives. GrProp and RdProp have potential to be used against Candida spp. infections,mainly in association with fluconazole or amphotericin B.

The aim of this work was to use Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares regression (PLSR) to estimate the oil content of individual Holm oak (Quercussp.) acorn kernels from different trees, sites and years that should be used in the future for molecular marker association studies. Sampling of acorns in two consecutive years (2003 and 2004) and from different sites in Portugal provided independent sample sets. A total of 89 samples (acorn kernels) representative of the natural oil content range were extracted. The results of the analyses performed by three people revealed accuracy of the oil extraction procedure (n-hexane) and the precision (repeatability) of this method, assessed during a four-day period, gave a standard deviation of 0.1%. Careful wavenumber selection and several steps of validation of the PLSR models led to a final robust model that allowed the precise prediction of the oil content of individual acorns. By using the wavenumber ranges from 5995 to 5323 cm−1and from 4478 to 4177 cm−1of the vector normalised spectra, a PLSR model with a coefficient of determination (r2) of 0.992 and a root mean square error of cross-validation (RMSECV) of 0.37% was achieved. The RPDvalue of about 10 and a bias of almost zero showed that the developed models are good for process control, development, and applied research. Oil content estimation of individual Quercussp. acorns by FT-NIR and PLSR was shown to be possible. The varying water content detected in the spectra of the milled kernels after drying in similar conditions, within and especially between years, could be handled.

Development of novel computational approaches for modeling protein properties from their primary structure is a main goal in applied proteomics. In this work, we reported the extension of the autocorrelation vector formalism to amino acid sequences for encoding protein structural information with modeling purposes. Amino Acid Sequence Autocorrelation (AASA) vectors were calculated by measuring the autocorrelations at sequence lags ranging from 1 to 15 on the protein primary structure of 48 amino acid/residue properties selected from the AAindex database. A total of 720 AASA descriptors were tested for building predictive models of the thermal unfolding Gibbs free energy change of human lysozyme mutants. In this sense, ensembles of Bayesian-Regularized Genetic Neural Networks (BRGNNs) were used for obtaining an optimum nonlinear model for the conformational stability. The ensemble predictor described about 88% and 68% variance of the data in training and test sets, respectively. Furthermore, the optimum AASA vector subset was shown not only to successfully model unfolding thermal stability but also to distribute wild-type and mutant lysozymes on a stability Self-organized Map (SOM) when used for unsupervised training of competitive neurons.

Abstract: Background: We evaluated, in a nonrandomised, institutional, prospective study, the efficacy of photodynamic therapy (PDT) with verteporfin in age-related macular degeneration (AMD) eyes with polypoidal choroidal vasculopathy (PCV) and subfoveal exudation. Methods: A prospective clinical and angiographic study was done in 40 consecutive eyes with PCV treated with PDT using masked best-corrected visual acuity (VA) and fluorescein and indocyanine green angiographic features at baseline and over 2 years. Results: Twenty-one eyes completed 1-year follow-up and showed, after a mean 2.9 PDT sessions, VA improvement in 12 eyes, no change in five eyes, and VA decrease in four eyes. Leakage was absent at the retinal and choroidal level in 14 eyes at 1 year. Recurrence occurred in one eye during the first year. Six eyes completed 2 years of follow-up and showed, after a mean 4 PDT sessions, VA improvement in five eyes and VA decrease in one eye. Leakage was absent at the retinal and choroidal level in five eyes. Recurrence occurred in four of these six eyes during the second year of follow-up. No serious adverse events were observed during the 2 years of follow-up. Conclusions: PDT with verteporfin was shown to be safe and effective for treating AMD eyes with PCV with subfoveal involvement. VA improvement and absence of leakage were achieved, respectively, in 57.1% and 66.6% of the eyes at 1 year. Recurrences were more frequent during the second year of follow-up.

50. Antimicrobial Activity of Propolis on Oral Microorganisms

Abstract.: Formation of dental caries is caused by the colonization and accumulation of oral microorganisms and extracellular polysaccharides that are synthesized from sucrose by glucosyltransferase of Streptococcus mutans. The production of glucosyltransferase from oral microorganisms was attempted, and it was found that Streptococcus mutans produced highest activity of the enzyme. Ethanolic extracts of propolis (EEP) were examined whether EEP inhibit the enzyme activity and growth of the bacteria or not. All EEP from various regions in Brazil inhibited both glucosyltransferase activity and growth of S. mutans, but one of the propolis from Rio Grande do Sul (RS2) demonstrated the highest inhibition of the enzyme activity and growth of the bacteria. It was also found that propolis (RS2) contained the highest concentrations of pinocembrin and galangin.
