The versatile methodology was developed for synthesizing kujigamberol B, a dinorlabdane diterpenoid isolated from the methanol extract of Kuji amber. A highly efficient intramolecular cyclization is followed by a Sonogashira-coupling reaction during the total synthesis. The synthesized compounds were evaluated for the growth-restoring activity against the mutant yeast (zds1Δ erg3Δ pdr1Δ pdr3Δ) and for the degranulation of RBL-2H3 cells. Lactiplantibacillus species has garnered considerable attention due to its multifaceted probiotic properties, particularly its potential in alleviating food allergies. Despite numerous reports supporting this claim, scant research exists concerning the methodology for screening anti-allergic probiotics. Thus study sought to establish an in vitro splenocyte sensitization model for the purpose of screening potential anti-allergic Lactiplantibacillus strains. Through the splenocyte model, different Lactiplantibacillus strains were assessed for their ability to modulate the Th1/Th2 balance in sensitized splenocytes. The results showed that compared with the of positive control (IFN-γ/IL-4 value was 1.12) and LGG (IFN-γ/IL-4 value was 15.88), Lactiplantibacillus XJ 002, TLF 1-2 and TLF 5-3-2 had the greatest effect on improving IFN-γ/IL-4 value, which were 9.89, 15.81 and 9.72 respectively. The further results showed that the IL-10 levels of Lactiplantibacillus XJ 002, TLF 1-2 and TLF 5-3-2 reached 125.23, 147.44 and 113.16 ng/L, which were significantly higher than the positive control (84.95 ng/L, p < 0.05). Moreover, the IgE reduction rates of Lactiplantibacillus XJ 002, TLF 1-2 and TLF 5-3-2 were 82%, 72% and 40% respectively, which indicated that Lactiplantibacillus XJ 002 and TLF 1-2 showed the best regulatory ability. Subsequently, Lactiplantibacillus XJ 002 and TLF 1-2 were considered promising probiotic candidates following rigorous probiotic evaluation. Remarkably, the inactivated forms of these strains, functioning as postbiotics, exhibited similar anti-allergic effects to those of the live bacteria. Allergic asthma is a disease that pathologically characterized by eosinophilia infiltration, airway inflammation and hyper responsiveness. In this study, we evaluated the anti-inflammatory and anti-allergy possibilities of honokiol, a bi-phenolic compound obtained from species of the genus Magnolia, which has long been involved in traditional Chinese prescriptions for asthma-related lung diseases, in an ovalbumin-induced mouse model of allergic asthma. We found honokiol significantly inhibited the eosinophilia infiltration, reduced the airway inflammation and suppressed the production of inflammatory cytokines) as well as the IgE in serum. Moreover, MMP-9 and? (IL-4 and IFN- NF-κB were found to be involved in the honokiol induced biological process. Recent studies have revealed that various food components affect the immune response. These components act on various immune cells, and their effects are mediated through the intestinal immune system and, in some cases, the intestinal microbiota. In this review, we describe the immunomodulating effects of various food components, including probiotics, prebiotics, polysaccharides, vitamins, minerals, fatty acids, peptides, amino acids and polyphenols. Some of these components enhance immune responses, leading to host defense against infection, whereas others inhibit immune responses, thus suppressing allergy and inflammation. The in vitro antioxidant, anti-obesity, anti-allergy and skin-lightening effects of extracts from strawberry fruits (ripe and unripe) and nine other plant parts of Amaou strawberry ( Fragaria   ×   ananassa var. Amaou ) were comparatively investigated by using the antioxidant assay and inhibition assays of lipase, adipocyte differentiation, β-hexosaminidase release, melanogenesis and tyrosinase. Their total phenolic content and the main phenolic compounds were also determined. The ripe strawberry fruits were found to possess potential anti-obesity and skin-lightening effects. The ethanol extracts from unripe strawberry fruits and several other parts like the crown, stolon leaf and flower showed more strong activities in the bioactivity assays conducted. The phytochemical profile varied among extracts from various parts of the plant. Ellagic acid was found in every part of the strawberry plant except the root. Our study provides valuable evidence that strawberry fruit is a food with potential anti-obesity and skin-lightening functions. The findings also support that several inedible parts of the strawberry plant have great potential to be used as the ingredients of functional food products. [ABSTRACT FROM AUTHOR]

Abstract: Functional food is considered to be any food or food component that provides health benefits beyond basic nutrition. Recently, a great deal of interest has been paid by the consumers towards natural bioactive compounds as functional ingredients in the diets due to their various health beneficial effects. Notably, marine resources have been recognized as rich sources of structurally diverse biologically active compounds with great application potential in marine functional foods. Among them, fucoidans have been found to possess various bioactivities including antioxidant, anti-inflammatory, anti-allergic, anti-tumor, anti-obesity, anti-coagulant, anti-viral, anti-hepatopathy, anti-uropathy, and anti-renalpathy effects. Abstract: The anti-allergic immunomodulatory effects of Quillaja saponin in antigen-stimulated rat basophilic leukemia cell line RBL-2H3 were investigated. Quillaja saponin exhibited a significant inhibitory activity using β-hexosaminidase and histamine release assays on anti-DNP-IgE-sensitized rat basophilic leukemia (RBL)-2H3 cells in a dose-dependent manner when tested in the range of 0.5–8.0μg/mL. In addition, Quillaja saponin also suppressed mRNA expression of interleukins (IL)-4 and IL-6. In order to explore the antigen-allergy mechanisms of Quillaja saponin in mast cells, intracellular Ca2+ level and IgE receptor FcεRI mRNA expression were also determined. It was found that the intracellular Ca2+ level was also decreased with Quillaja saponin tested at various concentrations. Furthermore, real time-PCR results showed that mRNA expression of FcεRI α chain was significantly suppressed by Quillaja saponin, and not of the β and γ chains. [Copyright &y& Elsevier]

Abstract: The key feature of food allergy is a T helper-2 cell (Th2)-predominant allergen-specific immune response. Rutin can increase PPAR-γ expression to improve diabetes; however, this effect whether attenuating Th2 signal is still unknown. The 75% ethanol extracts of buckwheat (EEB) and rutin (major active compound) significantly suppress interleukin-4 (IL-4) secretion from splenocytes in ovalbumin (OVA)-induced sensitive Balb/c mice; therefore, the inhibitory effects of EEB and rutin on Th2 signaling pathway in PMA/ionomycin-induced EL4 T cells were subsequently investigated. At the transcriptional level, rutin reduced the expression of Th2 markers such as GATA3 and p-STAT6 in PMA/ionomycin-induced EL4 T cells. Moreover, we used the PPAR-γ antagonist GW9662 to confirm that rutin skewed the immunological function toward Th2 activity through regulation of PPARγ expression. Taken together, EEB and rutin may have the potential to regulate immune responses, thereby deeming them useful in reducing food hypersensitivities and improving anti-cellular allergic responses. The article presents a study which investigates the anti-allergic effects of the ethanol extract of marc (SFEM) to isolate the ethanol extract with anti-inflammatory activity to determine its structure. The study used silica gel and ODS column chromatography to separate the ethyl acetate fraction (EtOAc). Results show that actinidiamide was found to inhibit the production of nitric oxide (NO).

Abstract: The anti-allergic effect of an edible brown alga, Eisenia arborea, was investigated by determining the anti-degranulation activity on RBL cells or KU812 cells and the inhibitory effects on enzymatic activities involved in eicosanoid (leukotoriene and prostaglandin) synthesis. When the stimulated cells were treated with a M/C (methanol:chloroform=1:2, v/v) extract of the alga, the histamine release of the cells was suppressed in a dose-dependent manner, and eicosanoid release from the cells was also strongly inhibited. Furthermore, the M/C extract and six phlorotannins (eckol, 6,6′-bieckol, 6,8′-bieckol, 8,8′-bieckol, phlorofucofuroeckol-A, phlorofucofuroeckol-B) derived from the extract inhibited enzymatic activities responsible for eicosanoid synthesis. This is the first report of the enzymatic activity inhibition by the phlorotannins 6,6′-bieckol, 6,8′-bieckol and PFF-B. Since histamine and eicosanoids are known to mediate the allergic reaction, inhibition of these mediators and enzymatic activities could be responsible for the anti-allergic effect of the alga. Allergies are a global issue. However, medical intervention for allergy treatment is limited. Recent studies have focused on allergy prevention with food components. Both the lactic acid bacteria Enterococcus faecalis IC-1 (IC-1) and the flavonoid luteolin have been shown to have an anti-allergic effect. Following the concept of diet diversity and synergistic effects, the purpose of this study was to explore the anti-allergic activity of a combination of IC-1 and luteolin. A combination of IC-1 and luteolin had anti-allergic effects with the passive cutaneous anaphylaxis reaction and strongly inhibited mast cell degranulation in a Caco-2/RBL-2H3 cells co-culture system. Oral administration with a combination of IC-1 and luteolin significantly decreased blood IgE content in ovalbumin (OVA)-induced allergic mice and alleviated symptoms of an allergy attack. These might have been done through correcting Th2 shifting with the allergy state since a combination of IC-1 and luteolin increased the IFN-γ content in blood. Furthermore, in vitro differentiation of the Th2 subset confirmed that a combination of IC-1 and luteolin inhibited naïve T cell from differentiating into a Th2 subset through inhibition of Th2 transcription factor GATA-3 expression. The combination also promoted the gene expression level of IFN-γ in mice bone marrow-derived dendritic cells. These results suggested a synergistic anti-allergic effect using a combination of IC-1 and luteolin both in reducing IgE content and in inhibiting mast cell degranulation during the allergy state. The article discusses a study which examines the impact of coffee on the antigen-specific immune responses of naïve splenocytes. Findings of the study reveal that coffee contains ingredients which could suppress IL-I2 secretion and produce anti-allergic activities. It also indicates that the effect of coffee on immune system can be associated to multiple factors.

This study was conducted to isolate and identify an active compound possessing anti-allergic property from the Jeju Island inhabiting Sargassum horneri (S. horneri). The ethanol extract of S. horneri (SHE) exhibited an anti-allergic effect via the reduction of β-hexosaminidase release in immunoglobulin E (IgE)/bovine serum albumin (BSA)-stimulated bone marrow-derived cultured mast cells (BMCMCs). SHE was subjected to liquid phase fractionation obtaining the hexane fraction, and this was further purified via ODS column chromatography and prep-C18 HPLC. The isolated compound was identified as mojabanchromanol (MC) by 1H, 13C NMR, and LCMS analysis. Isolated MC showed a higher inhibitory effect on the β-hexosaminidase release than those of SHE without cytotoxicity. Moreover, MC inhibited the mRNA expression levels of allergic cytokines in BMCMCs. This is the first report to inform on the anti-allergic property of MC isolated from S. horneri. In conclusion, SHE, as well as MC, is applicable in the development of functional health food, cosmetics, and pharmaceutical products exhibiting anti-allergic effects.
