In the athletes the wide use of Anabolic Androgenic Steroids (AAS) cause series damage in various organs, in particular, analyzing the liver, elevation on the levels of liver enzymes, cholestatic jaundice, liver tumors, both benign and malignant, and peliosis hepatis are described. A prolonged AAS administration provokes an increase in the activities of liver lysosomal hydrolases and a decrease in some components of the microsomal drug-metabolizing system and in the activity of the mitochondrial respiratory chain complexes without modifying classical serum indicators of hepatic function. Liver is a key organ actively involved in numerous metabolic and detoxifying functions. As a consequence, it is continuously exposed to high levels of endogenous and exogenous oxidants that are by-products of many biochemical pathways and, in fact, it has been demonstrated that intracellular oxidant production is more active in liver than in tissues, like the increase of inflammatory cytokines, apoptosis and the inhibitors of apoptosis NF-B and Heat Shock Proteins.

This study aims to evaluate the use of a placenta histoculture system based on placental villi culture on collagen sponge gels at the liquid/air interface, for the detection of zidovudine-induced apoptosis. Explant cultures from full term placentas were exposed to different concentrations of zidovudine (1, 10, 100 uM) up to 7 days and samples were analysed for apoptosis with different methods after 3, 24, 48 hours and 7 days of exposure. Apoptotic phenomena were demonstrated at 10 and 100 uM zidovudine, more delayed at the lowest concentration: this demonstrates that zidovudine-induced placenta apoptosis in vitrois both time- and dose-dependent and that also concentrations near to those achievable in vivomight cause placenta apoptosis. This has to be taken into account when considering possible consequences of antiretroviral treatments during pregnancy. Placental histocultures on collagen sponge cells are a reliable tool for the study of antiretroviral-induced toxicity in placenta: in fact, explants cultured for more prolonged periods (up to 7–14 days) in a “physiological milieu” allow the detection of biological effects otherwise not evident in the commonly used, short-term, placental cultures.

In this study the authors investigated the presence of serum antibodies to Human Herpesvirus 8 (HHV-8) in a group of women of child-bearing age in the Apulia region (South Eastern Italy). A seroprevalence of 16.8% was observed, increasing, although non significantly, with the age of the women (10.6% in women between 19 and 25 years, 25.3% in women aging more than 35 years). The presence of antibodies to Hepatitis C Virus (HCV) was significantly associated to the detection of antibodies to HHV-8. Possible mother-to-child transmission of HHV-8 as well as the outcome of fetuses or children born to HHV-8 positive mothers are still a matter of debate. This study, showing the wide diffusion of HHV-8 infection in healthy women of child-bearing age in our geographical area, highlights the urgency of studies aimed to better clarify these relevant topics.
