Cryopreservation of sheep embryos with ethylene glycol as a protectant appears to be more effective than glycerol, particularly at the morula stage, as has been demonstrated on the basis of in vitro and in vivo development rates after thawing. In this study we compare the ultrastructure of fresh morulae, thawed morulae, and blastocysts cryopreserved with either ethylene glycol or glycerol at the electron microscopic level, to look for cellular damage that could be responsible for proven differences in embryo survival after transfer. Embryos cryopreserved with glycerol showed unequal degrees of conservation even among blastomeres within a single embryo. In morulae, inner blastomeres were completely damaged, whereas external ones appeared to be intact. Both morulae and blastocysts cryopreserved with ethylene glycol showed a higher uniformity in blastomere conservation than embryos with glycerol. The most remarkable features in this experimental group were the presence of desmosomes following tight junctions between blastomeres and the presence of many microvilli on the outer surface of external blastomeres. These characteristics are similar in fresh embryos of the control group. Our results show that ethylene glycol protects membrane and cytoplasmic structures of embryonic cells from cryoinjury much better than glycerol. In vivo survival of embryos confirmed the ultrastructural observations. A limited permeability of glycerol would explain the observed ultrastructural differences in blastomere integrity, which depends on blastomere location and the differences between morulae and blastocysts. We conclude that the low reproductive yield after cryopreservation using glycerol can be attributed to the lack of protection of inner cells.

We studied the fine structural organization of the meristematic nucleus in roots of Lycopesicon esculentum (tomato) using ultracytochemical and immunocytochemical approaches. The nucleus has a non-reticulate (i.e. low DNA content) structure whose supramolecular organization differs in some respects from that in reticulate nuclei, principally in the organization of the chromocentres associated with the nuclear envelope, with which centromeric structures appear to be associated. The main difference at the nucleolar level is found in the fibrillar centres, which have a low amount of DNA labelling and in which inclusions of condensed chromatin are present only very rarely. The distribution of nucleolar DNA amongst the nucleolar compartments is similar to that in reticulate nucleoli as demonstrated using an anti-DNA monoclonal antibody. Tomato nuclei have nucleolus-associated bodies or karyosomes, like other plant species with a low DNA content and non-reticulate nuclear organization. The nuclear ribonucleoprotein structures in the inter- and perichromatin regions, namely inter- and perichromatin fibrils and granules, show similar ultrastructural and cytochemical characteristics in both types of nuclei.

We isolated the nuclear matrix of proliferative meristematic root cells of Allium cepa bulbs and characterized it ultrastructurally and by one-dimensional SDS-polyacrylamide gel electrophoresis. The general organization of the nuclear matrix in onion root cells is similar to that found in other eukaryotes extracted under the same conditions. There are three main morphological components: the residual pore-lamina complex, the internal matrix and the residual nucleolus. The matrix has a fibrillo-granular structure composed of 5–10 nm fibrils and 20–25 nm granules. The residual protein matrix has a complex pattern, with several polypeptide bands of 12000 to 70000 Mr. The major bands have Mrs of 62000, 56000, 35000 and 27000.

Unicellular Dinoflagellates represent the only eukaryotic Phylum lacking histones and nucleosomes. To investigate whether Dinoflagellates do have a nuclear matrix that would modulate the supramolecular organization of their non-nucleosomal DNA and chromosomes, cells of the free-living unarmored Dinoflagellate Amphidinium carterae were encapsulated in agarose microbeads and submitted to sequential extraction with non-ionic detergents, nucleases and 2 M NaCl. Our results demonstrate that this species has a residual nuclear matrix similar to that of vertebrates and higher plants. The cytoskeleton-nuclear matrix complex of A. carterae shows a relatively intricate polypeptide pattern. Immunoblots with different antibodies reveal several intermediate filament types of proteins, one of which is immunologically related to vertebrate lamins, confirming that these proteins are ancestral members of the IF family, which is highly conserved in eukaryotes. A topoisomerase II homologue has also been identified in the nuclear matrix, suggesting that these structures could play a role in organizing the Dinoflagellate DNA in loop domains. Taken together our results demonstrate that the nuclear matrix is an early acquisition of the eukaryotic nucleus, independent of histones and nucleosomes in such a way that the mechanisms controlling the two levels of organization in eukaryotic chromatin would be molecularly and evolutionarily independent.

The analysis of isolated nucleolar matrices from onion cells by light and electron microscopy, 2-D separation of proteins, and confocal microscopy has confirmed the existence of an organized nucleolar matrix with a complex protein composition to which are attached the insoluble processing complexes. In the present work, we present evidence from immunoblotting, immunofluorescence, immunogold labeling, and preferential cytochemical staining with bismuth salts that an insoluble fraction of the multifunctional protein nucleolin, is a component of the onion nucleolar matrix, and analyse its ultrastructural distribution in the described domains of the matrix.

Summary “Micropuffs” and coiled bodies are two nuclear bodies which have previously been described in a wide range of plant species. In this work, we have demonstrated, by means of ultrastructural cytochemical, autoradiographic and immunocytochemical methods, that the so-called “micropuffs” contain chromatin, which is organized in a supranucleosomal structure, is inactive in transcription, and has no apparent relationship with the nucleolus. These features, as well as the distribution of these structures during the interphase periods, allowed us to conclude that they represent centromeric structures, so, we propose that they be denoted by this name. Coiled bodies show the same silver-staining pattern as the nucleolus organizer (NOR), but do not appear to contain NOR chromatin nor does the early processing of rRNA precursor seem to take place in them. We postulate they are involved in later nuclear RNA processing and storage. This work clarifies some previous misunderstandings with regard to these bodies.

We have used polyclonal and monoclonal antibodies against different lamins from vertebrates, and the IFA antibody recognizing all kinds of intermediate filament proteins, to investigate the lamins of the nuclear matrix of Allium cepa meristematic root cells. All the antibodies react in the onion nuclear matrix with bands in the range of 60-65 kDa, which are enriched in the nuclear matrix after urea extraction, and do not crossreact with other antibodies recognizing intermediate filaments in plants (AFB, anti-vimentin and MAC 322), ruling out crossreaction with contaminating intermediate filaments of cytoplasmic bundles. In 2-D blots the chicken anti-lamin serum reacts with one spot at 65 kDa and pI 6.8 and the anti B-type lamin antibodies with another one at 64 kDa and pI 5.75. Both crossreact with IFA. The lamin is localized at the nuclear periphery and the lamina by indirect immunofluorescence. Immunogold labelling of nuclear matrix sections reveals that the protein is not only associated with the lamina, but also with the internal matrix. Taken together these results reveal that higher plants, which do not possess an organized network of cytoplasmic intermediate filaments, nevertheless present a well-organized lamina containing lamins in which at least one of them is immunologically related to vertebrate lamin B. Our data confirm that lamins are very old members of the intermediate filament proteins that have been better conserved in plants during evolution than their cytoplasmic counterparts.

In plant cells nucleolar fibrillar centres (FCs) undergo ultrastructural changes, depending on the nucleolar activity. We have found two types of FC structure in nucleoli with either high or low activity, to which we have given the conventional names of homogeneous and heterogeneous, respectively. The first type is characterized by the presence of fibres that we describe structurally and cytochemically as decondensed chromatin; the second type, in addition to these fibres, contains a variable number of dense cores made up of condensed chromatin. Moreover, RNA does not appear to be present in FCs, while proteins are a major component. Autoradiography after tritiated uridine incorporation shows that FCs are not the site of transcription, but that this takes place in the fibrillar component; the same result is obtained using the lead acetate fixation technique for detecting orthophosphate ions. This fact leads us to think that FCs are not the whole interphasic counterpart of the mitotic nucleolar organizing region (NOR), as stated by other authors, but only the portion of the NOR that is temporarily inactive in transcription; the transcriptionally active part of the NOR is in the fibrillar component, bound to its earliest product of transcription. Thus, FCs and the fibrillar component constitute a functional unit.

This paper deals with the fine structure of the fibrillar centres of the nucleolus inAllium cepacells in ultrathin, sections of in vivo fixed roots. The ultrastructural observations have allowed us to consider each nucleolar fibrillar centre as an active zone in the nucleolar chromatin loop, and to propose a possible model for the organization of the different components of the nucleolus within it.

The human ets-2 gene is a homolog of the v-ets oncogene of the E26 virus and codes for a 56-kilodalton nuclear protein. The ets-2 protein is phosphorylated and has a rapid turnover, with a half-life of 20 min. When human lymphocytic CEM cells were treated with the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA), the level of the ets-2 protein was quickly elevated 5- to 20-fold. This effect of TPA was mimicked by a synthetic diacylglycerol, 1-oleoyl-2-acetyl glycerol, and was blocked by the protein kinase C inhibitor H7, indicating that protein kinase C is involved in the induction. The increase in the ets-2 protein was due to stabilization of the protein, because the protein had a half-life of more than 2 h in the presence of TPA and the ets-2 mRNA level did not increase significantly upon TPA treatment. The protein synthesis inhibitor cycloheximide enhanced the effect of TPA on the ets-2 protein and could itself slow turnover of the protein. Properties of the ets-2 protein, such as nuclear localization, phosphorylation, rapid turnover, and response to protein kinase C, indicate that this protein belongs to a group of oncogene proteins which are generally thought to have regulatory functions in the nucleus (e.g., myc, fos, myb, and p53). Our results suggest that protein kinase C, either directly or indirectly, regulates the level of the ets-2 protein by posttranslational mechanisms.

This work is an ultrastructural and cytochemical study of a structure observed in the nucleolus of Allium cepa meristematic cells after nucleolar disgregation, by a continuous treatment of 12 h with ethidium bromide. The ultrastructural and cytochemical data allow us to consider this structure as the intranucleolar chromatin collapsed by the effect of the drug.
