A series of Schiff bases of 4-amino-1,2,4-triazole derivatives containing pyrazole (5a–h) were synthesized from condensation of 4-amino-5-(5-methyl-1H-pyrazol-3-yl)-4H-1,2,4-triazole-3-thiol (3) derivative with various aromatic aldehydes (4a–h). The structures of the synthesized compounds were elucidated by IR, 1H NMR, 13C NMR, and mass spectrometry. All the synthesized compounds (5a–h) were screened for their in vivo analgesic and in vitro antioxidant activities revealing significant analgesic and antioxidant properties.

Background:Severe stroke carries high rates of mortality and morbidity. The aims of this study were to determine the characteristics of patients who initially presented with severe ischemic stroke, and to identify acute and subacute predictors of favorable clinical outcome in these patients. Methods:An observational cohort study, Acute Stroke Registry and Analysis of Lausanne (ASTRAL), was analyzed, and all patients presenting with severe stroke – defined as a National Institute of Health Stroke Scale score of ≥ 20 on admission – were compared with all other patients. In a multivariate analysis, associations with demographic, clinical, pathophysiologic, metabolic and neuroimaging factors were determined. Furthermore, we analyzed predictors of favorable outcome (modified Rankin scale score of ≤ 3 at 3 months) in the subgroup of severe stroke patients. Results:Of 1915 consecutive patients, 243 (12.7%) presented with severe stroke. This was significantly associated with cardio‐embolic stroke mechanism (odds ratio [OR] 1.74, 95% confidence interval [CI] 1.19–2.54), unknown stroke onset (OR 2.35, 95% CI 1.14–4.83), more neuroimaging signs of early ischemia (mostly computed tomography; OR 2.65, 95% CI 1.79–3.92), arterial occlusions on acute imaging (OR 27.01, 95% CI 11.5–62.9), fewer chronic radiologic infarcts (OR 0.43, 95% CI 0.26–0.72), lower hemoglobin concentration (OR 0.97, 95% CI 0.96–0.99), and higher white cell count (OR 1.05, 95% CI 1.00–1.11). In the 68 (28%) patients with favorable outcomes despite presenting with severe stroke, this was predicted by lower age (OR 0.94, 95% CI 0.92–0.97), preceding cerebrovascular events (OR 3.00, 95% CI 1.01–8.97), hypolipemic pretreatment (OR 3.82, 95% CI 1.34–10.90), lower acute temperature (OR 0.43, 95% CI 0.23–0.78), lower subacute glucose concentration (OR 0.74, 95% CI 0.56–0.97), and spontaneous or treatment‐induced recanalization (OR 4.51, 95% CI 1.96–10.41). Conclusions:Severe stroke presentation is predicted by multiple clinical, radiologic and metabolic variables, several of which are modifiable. Predictors in the 28% of patients with favorable outcome despite presenting with severe stroke include hypolipemic pretreatment, lower acute temperature, lower glucose levels at 24 h, and arterial recanalization.

To develop and validate a simple, integer-based score to predict functional outcome in acute ischemic stroke (AIS) using variables readily available after emergency room admission.

Le présent travail a pour objectif le traitement des lixiviats de la décharge publique contrôlée de la ville de Fès par des procédés biologiques et chimiques. Une accumulation des métaux lourds par des micro-organismes aérobies spécifiques présents dans le lixiviat réduit davantage les métaux lourds, principalement les ions Mn2+, avec un taux d’abattement compris entre 90,8% et 93,3%, et permet aussi une élimination totale des germes de contamination fécale (coliformes fécaux, streptocoques fécaux), des levures et des champignons.Ce traitement a entraîné également une réduction de la demande chimique totale en oxygène (DCOT) et la demande chimique en oxygène soluble (DCOS), respectivement de 60% et de 88%. Le traitement anaérobie du lixiviat par fermentation pendant 48 heures a montré une réduction de la DCOTde 74%. Le traitement biologique par Sequencing Batch Reactor(SBR) a montré également des résultats permettant une réduction de 87% pour la DCOTet 96,77% pour la demande biologique en oxygène durant 5 jours (DBO5). Le traitement chimique par coagulation-floculation, utilisant la chaux, le chlorure ferrique (FeCl36H2O) et le sulfate d’alumine (Al2(SO4)318H2O), réduit respectivement la DCOTde 36,6%, 81,67% et 85%. La coagulation-floculation avec 200 mg/L de FeCl36H2O a montré une réduction des germes fécaux de plus de 85%. Le couplage du traitement chimique par coagulation-floculation au traitement biologique par SBR permet d’obtenir des résultats satisfaisants en matière d’élimination des éléments minéraux contenus dans les lixiviats et une réduction de la DCOT, de la DBO5et des polyphénols, respectivement de 98%, 99,16% et 94,53%.

Blood lines of polyvinyl chloride (PVC) for hemodialysis usually contain di(2-ethylhexyl) phthalate (DEHP) as a plasticizer. Previous studies show that 1 mg/kg of this plasticizer can leach into the blood during one dialysis session. It is rapidly metabolized in the liver. Mono(2-ehtylhexyl) phthalate (MEHP), its main metabolite can be detected as well. After oral administration to rodents, both compounds caused a variety of adverse biological effects such as testicular atrophy, peroxisome proliferation and hepatic peroxisomal enzyme induction. Male wistar rats were treated intraperitoneally by DEHP and MEHP using twice the dose of that involved in human exposure during a dialysis session. Propranolol metabolism by hepatocytes was investigated after fresh isolation from treated and untreated rats by means of reverse phase HPLC. The choice of propranolol as a substrate was made because of its rather quick liver metabolisation. Phenobarbital was chosen in the study as a reference of enzymatic inducer to evaluate the inducing effect of DEHP and MEHP. Propranolol was metabolized by the hepatocytes of both treated and untreated rats. Hepatocytes isolated from rats treated by phenobarbital, MEHP and DEHP were shown to have a higher speed constant of metabolism indicating a rapid metabolism of propranolol. Under these conditions, in fact, propranolol metabolisation was found to be respectively 6, 2.7, 2 times faster than the propranolol metabolisation of untreated rats. The hypothesis that DEHP and MEHP are enzymatic inducers, particularly cytochrome P450 (CYP) inducers of the xenobiotics metabolism on the intact liver after IP administration has become been found to be valid. The results obtained in this study confirm the value of isolated hepatocytes as anin vivodrug metabolism predictive model.
