The pathophysiological events of secondary brain injury contribute to poor outcome after traumatic brain injury (TBI). The neuroprotective effects of mesenchymal cells have been extensively studied and evidence suggests that their effects are mostly mediated through paracrine effects. Human umbilical cord perivascular cells (HUCPVCs) are mesenchymal stem cells with potential therapeutic value in TBI. In this study, we assessed the effect of HUCPVC-conditioned media (CM) in an established adult zebrafish model of TBI induced by pulsed high-intensity focused ultrasound (pHIFU). This model demonstrates similarities to mammalian outcome after TBI. Administration of HUCPVC-CM 1 h postinjury (hpi) resulted in improved outcome after pHIFU-induced TBI. Western blot and immunohistochemistry results demonstrated that the HUCPVC-CM reduced (p< 0.05) reactive astrogliosis at 24 hpi. Moreover, at 24 hpi, the HUCPVC-CM treatment resulted in reduced apoptosis in HUCPVC-CM-treated zebrafish. Behavioral analysis demonstrated improvement in locomotor activity (p< 0.05) and anxiety (p< 0.05) at 6 and 24 hpi following HUCPVC-CM treatment. Overall, HUCPVC-CM treatment improved acute outcome measures in pHIFU-injured zebrafish. Collectively, the data demonstrate a cell-free treatment approach for traumatic brain injuries.

Controlled, localized, and timely activation of nanosized drug delivery systems (NSDDSs), using an external stimulus such as therapeutic ultrasound (TUS), can improve the efficacy of cancer treatments compared to either conventional chemotherapy methods or passive NSDDSs alone. Specifically, TUS induces thermal and mechanical effects that trigger drug release from NSDDSs and overcomes drug delivery barriers in tumor microenvironments to allow nanoparticle drug carriers to penetrate more deeply into tumor tissue while minimizing side effects. This review highlights recent advancements, contemplates future prospects, and addresses challenges in using TUS-mediated NSDDSs for cancer treatment, encompassing preclinical and clinical applications.

A distributed Brillouin scattering sensor has been employed to identify several inner wall cutouts in an end-capped steel pipe by measuring the axial and hoop strain distributions along the outer surface of the pipe. The locations of structural indentations that constitute 50–60% of the inner pipe wall are found and distinguished by use of their corresponding strain—pressure data. These results are quantified in terms of the fiber orientation, defect size and depth, and behavior relative to those of unperturbed pipe sections.

The strain measurement of a 1.65-m reinforced concrete beam by use of a distributed fiber strain sensor with a 50-cm spatial resolution and 5-cm readout resolution is reported. The strain-measurement accuracy is ±15 µε (µm/m) according to the system calibration in the laboratory environment with non-uniform-distributed strain and ±5 µε with uniform strain distribution. The strain distribution has been measured for one-point and two-point loading patterns for optical fibers embedded in pultruded glass fiber reinforced polymer (GFRP) rods and those bonded to steel reinforcing bars. In the one-point loading case, the strain deviations are ±7 and ±15 µε for fibers embedded in the GFRP rods and fibers bonded to steel reinforcing bars, respectively, whereas the strain deviation is ±20 µε for the two-point loading case.

The effect of the finite extinction ratio of an electro-optic modulator (EOM) on the Brillouin frequency measurement of a distributed Brillouin-based fiber optic sensor is studied. An EOM with a finite extinction ratio limits the application of Brillouin optical time domain analysis in a distributed Brillouin-based fiber optic sensor. This results in confusion in specifying the location of the strained region and in error in detecting the Brillouin frequency and hence in strain and temperature measurement.

During normal operation in Canada deuterium uranium (CANDU®) reactors, the stress corrosion cracking (SCC) of fuel sheathing is mitigated effectively, in part, using a thin graphite-based coating known as CANDU lubricant (CANLUB). Mechanisms typically proposed for the demonstrated SCC mitigation offered by CANLUB include lubrication and/or chemical interactions. An additional possibility, that was recently suggested, involves the sequestering of iodine through its interaction with alkali metal and/or alkaline earth metal impurities in the CANLUB coating. This possibility is supported by the systematic analysis and testing in this paper, wherein three prevalent impurities (Na, Ca, and Mg) found in CANLUB were incorporated into SCC slotted ring experiments as metal oxides. When the amount of metal oxide (Na2O, CaO, or MgO) matched or exceeded the amount of iodine (6 mmol = 16 mg/cm3), Na2O and CaO protected the rings from corrosion whereas MgO enhanced their corrosion. When Zircaloy-4 sheathing is subjected to mechanical stress, high temperature, and high concentrations of iodine vapor, it is better protected by siloxane coatings than by graphite-CANLUB coatings. Consequently, since metal impurities (Na, Ca, and Mg) are found more abundantly in siloxane coatings than in graphite-CANLUB coatings, Zircaloy-4 slotted rings were coated with graphite-CANLUB containing Na, Ca, and/or Mg at those more abundant concentrations. Since these concentrations remain below 6 mmol, SCC test results suggest that the siloxane's superior adhesion is an essential first step in preventing corrosion induced by 6 mmol of iodine.

For more than 50 years, a thin (3–20 μm) graphite coating has played an important role in limiting the stress corrosion cracking (SCC) of Zircaloy-4 fuel sheathing in CANDU® nuclear reactors. Siloxane coatings, which were examined alongside graphite coatings in the early 1970s, demonstrated even better tolerance against power-ramp-induced SCC and exhibited better wear resistance than graphite coatings. Although siloxane technology developed significantly in the 1980s/1990s, siloxane coatings remain unused in CANDU reactors, because graphite is relatively inexpensive and performs well in-service. However, advanced CANDU designs will accommodate average burnups, exceeding the threshold tolerable by the graphite coating (450  MWh/kgHE). In addition, siloxane coatings may find applicability in pressurized and boiling water reactors, wherein the burnups are inherently larger than those in CANDU reactors. Consequently, a commercially available siloxane coating is evaluated by its present-day chemistry, wear resistance, and performance in hot, stressful, and corrosive environments. After subjecting slotted Zircaloy-4 rings to iodine concentrations exceeding the estimated in-reactor concentration (1  mg/cm3), mechanical deflection tests and scanning electron microscopy (SEM) show that the siloxane coating outperforms the graphite coating in preserving the mechanical integrity of the rings. Furthermore, the baked siloxane coating survived a 50-day exposure to thermal neutron flux ((2.5±0.1)×1011  n/cm2 s) in the SLOWPOKE-2 nuclear reactor at the Royal Military College of Canada.
