ABSTRACTTo easily edit the genome of naïve human embryonic stem cells (hESC), we introduced a dual cassette encoding an inducible Cas9 into the AAVS1 site of naïve hESC (iCas9). The iCas9 line retained karyotypic stability, expression of pluripotency markers, differentiation potential, and stability in 5iLA and EPS pluripotency conditions. The iCas9 line induced efficient homology–directed repair (HDR) and non-homologous end joining (NHEJ) based mutations through CRISPR-Cas9 system. We utilized the iCas9 line to study the epigenetic regulator, PRC2 in early human pluripotency. The PRC2 requirement distinguishes between early pluripotency stages, however, what regulates PRC2 activity in these stages is not understood. We show reduced H3K27me3 and pluripotency markers in JARID2 2iL-I-F hESC mutants, indicating JARID2 requirement in maintenance of hESC 2iL-I-F state. These data suggest that JARID2 regulates PRC2 in 2iL-I-F state and the lack of PRC2 function in 5iLA state may be due to lack of sufficient JARID2 protein.

For nearly a century developmental biologists have recognized that cells from embryos can differ in their potential to differentiate into distinct cell types. Recently, it has been recognized that embryonic stem cells derived from both mice and humans exhibit two stable yet epigenetically distinct states of pluripotency: naive and primed. We now show that nicotinamide N-methyltransferase (NNMT) and the metabolic state regulate pluripotency in human embryonic stem cells (hESCs).  Specifically, in naive hESCs, NNMT and its enzymatic product 1-methylnicotinamide are highly upregulated, and NNMT is required for low S-adenosyl methionine (SAM) levels and the H3K27me3 repressive state. NNMT consumes SAM in naive cells, making it unavailable for histone methylation that represses Wnt and activates the HIF pathway in primed hESCs. These data support the hypothesis that the metabolome regulates the epigenetic landscape of the earliest steps in human development.

Duchenne muscular dystrophy (DMD) is a lethal muscle-wasting disease. Studies in Drosophila showed that genetic increase of the levels of the bioactive sphingolipid sphingosine-1-phosphate (S1P) or delivery of 2-acetyl-5-tetrahydroxybutyl imidazole (THI), an S1P lyase inhibitor, suppresses dystrophic muscle degeneration. In the dystrophic mouse (mdx), upregulation of S1P by THI increases regeneration and muscle force. S1P can act as a ligand for S1P receptors and as a histone deacetylase (HDAC) inhibitor. Because Drosophila has no identified S1P receptors and DMD correlates with increased HDAC2 levels, we tested whether S1P action in muscle involves HDAC inhibition. Here we show that beneficial effects of THI treatment in mdx mice correlate with significantly increased nuclear S1P, decreased HDAC activity and increased acetylation of specific histone residues. Importantly, the HDAC2 target microRNA genes miR-29 and miR-1 are significantly upregulated, correlating with the downregulation of the miR-29 target Col1a1 in the diaphragm of THI-treated mdx mice. Further gene expression analysis revealed a significant THI-dependent decrease in inflammatory genes and increase in metabolic genes. Accordingly, S1P levels and functional mitochondrial activity are increased after THI treatment of differentiating C2C12 cells. S1P increases the capacity of the muscle cell to use fatty acids as an energy source, suggesting that THI treatment could be beneficial for the maintenance of energy metabolism in mdx muscles.

In this article the author reflects on his editorial affiliation with the "Journal für Entwicklungspolitik" (JEP) in 2002 and his work dealing with big business, the military, and economists and lawyers who contributed to the putsch against Chilean President Salvador Allende. Democratic changes in Chile since 1990 are seen as bringing the nation further into a globalized economy but with the persistence of a transnational corporate and managerial class. The author describes his interview-based research project investigating the development of an elite global ruling class.

An introduction is presented to this 25th anniversary issue of the periodical, whose articles and essays address development research from theoretical and methodological perspectives, international and multidisciplinary approaches, and regional differences in development research.

The article discusses the the status of development studies in Austria. In the 1980's researchers coalesced in founding the Mattersburger Kreis of academics whose efforts led to the founding of this "Journal für Entwicklungspolitik," and two decades later the Projekt Internationale Entwicklung and department of Development Studies opened at the University of Vienna. The mission and accomplishments to date are described.

A characteristic feature of tumors is high production of lactic acid due to enhanced glycolysis. Here, we show a positive correlation between lactate serum levels and tumor burden in cancer patients and examine the influence of lactic acid on immune functions in vitro. Lactic acid suppressed the proliferation and cytokine production of human cytotoxic T lymphocytes (CTLs) up to 95% and led to a 50% decrease in cytotoxic activity. A 24-hour recovery period in lactic acid–free medium restored CTL function. CTLs infiltrating lactic acid–producing multicellular tumor spheroids showed a reduced cytokine production. Pretreatment of tumor spheroids with an inhibitor of lactic acid production prevented this effect. Activated T cells themselves use glycolysis and rely on the efficient secretion of lactic acid, as its intracellular accumulation disturbs their metabolism. Export by monocarboxylate transporter-1 (MCT-1) depends on a gradient between cytoplasmic and extracellular lactic acid concentrations and consequently, blockade of MCT-1 resulted in impaired CTL function. We conclude that high lactic acid concentrations in the tumor environment block lactic acid export in T cells, thereby disturbing their metabolism and function. These findings suggest that targeting this metabolic pathway in tumors is a promising strategy to enhance tumor immunogenicity.

This lipid asymmetry, which is maintained by the concerted action of phospholipid transport proteins, is mainly lost during apoptosis. Here, we demonstrate that primary human CD8+cytotoxic T lymphocytes (CTLs) expose PS on T-cell receptor (TCR)–mediated antigen (Ag) recognition. In contrast to PS externalization on apoptotic cells, activation-induced PS exposure is less pronounced and reversible. Fluorescence microscopic analysis revealed that PS is distributed nonhomogenously over the plasma membrane and concentrated in membrane lipid raft domains at the immunologic synapse. By studying the activity of PS transport proteins using a fluorescence-labeled PS analogue, we found that activation of CTLs inhibited the flippase-mediated inward-directed PS transport without affecting the outward transport. Shielding of exposed PS by annexin V protein during Ag recognition diminished cytokine secretion, activation, and cell-to-cell clustering of Ag-specific CTLs. In summary, our data demonstrate for the first time that externalized PS on Ag-stimulated CTLs is linked to T-cell activation and probably involved in cell-to-cell contact formation at the immunologic synapse.

Down-regulation of immune responses by regulatory T (Treg) cells is an important mechanism involved in the induction of tolerance to allo-antigens (Ags). Recently, a novel subset of Ag-specific T-cell receptor (TCR)αβ+CD4-CD8-(double-negative [DN]) Treg cells has been found to be able to prevent the rejection of skin and heart allografts by specifically inhibiting the function of antigraft-specific CD8+T cells. Here we demonstrate that peripheral DN Treg cells are present in humans, where they constitute about 1% of total CD3+T cells, and consist of both naïve and Ag-experienced cells. Similar to murine DN Treg cells, human DN Treg cells are able to acquire peptide–HLA-A2 complexes from antigen-presenting cells by cell contact-dependent mechanisms. Furthermore, such acquired peptide-HLA complexes appear to be functionally active, in that CD8+T cells specific for the HLA-A2–restricted self-peptide, Melan-A, became sensitive to apoptosis by neighboring DN T cells after acquisition of Melan-A–HLA-A2 complexes and revealed a reduced proliferative response. These results demonstrate for the first time that a sizable population of peripheral DN Treg cells, which are able to suppress Ag-specific T cells, exists in humans. DN Treg cells may serve to limit clonal expansion of allo-Ag–specific T cells after transplantation.

Defects in the epidermal growth factor receptor (EGFR) pathway can lead to aggressive tumor formation. Activation of this pathway during normal development produces multiple outcomes at the cellular level, leading to cellular differentiation and cell cycle activation. To elucidate the downstream events induced by this pathway, we used genome‐wide cDNA microarray technology to identify potential EGFR targets in Drosophila oogenesis. We focused on genes for which the transcriptional responses due to EGFR pathway activation and inactivation were in opposite directions, as this is expected for genes that are directly regulated by the pathway in this tissue type. We perturbed the EGFR pathway in epithelial follicle cells using seven different genetic backgrounds. To activate the pathway, we overexpressed an activated form of the EGFR (UAS‐caEGFR), and an activated form of the signal transducer Raf (UAS‐caRaf); we also over‐ or ectopically expressed the downstream homeobox transcription factor Mirror (UAS‐mirr) and the ligand‐activating serine protease Rhomboid (UAS‐rho). To reduce pathway activity we used loss‐of‐function mutations in the ligand (gurken) and receptor (torpedo). From microarrays containing 6,255 genes, we found 454 genes that responded in an opposite manner in gain‐of‐function and loss‐of‐function conditions among which are many Wingless signaling pathway components. Further analysis of two such components, sugarless and pangolin, revealed a function for these genes in late follicle cell patterning. Of interest, components of other signaling pathways were also enriched in the EGFR target group, suggesting that one reason for the pleiotropic effects seen with EGFR activity in cancer progression and development may be its ability to regulate many other signaling pathways. It has been shown that after allogeneic peripheral blood stem cell transplantation (PBSCT), donor T cells can induce potent graft‐versus‐tumor (GVT) effects in hematologic malignancies and possibly solid tumors such as renal cell carcinoma. Two patients (27 and 30 years old) with metastatic melanoma received allogeneic PBSCT from an HLA‐identical sibling donor after reduced conditioning with fludarabine, carmustine and melphalan. One patient showed a delayed mixed response with complete regression of lymph node metastases but persistent liver metastasis at day +60 and +120, consistent with a GVT response. In order to generate donor‐derived tumor‐reactive cytotoxic T lymphocytes (CTLs), peripheral blood mononuclear cells were stimulated with donor dendritic cells (DCs) loaded with host tumor lysate. Using these culture conditions, a marked increase in CD8+CTLs was observed in both donors exhibiting a strong MHC class I‐restricted cytotoxic activity against the host tumor without cross‐reactivity against nonmalignant host cells. CDR3 spectratyping was used to analyze the complexity of T‐cell subpopulations in both CTL lines. Results demonstrate that oligoclonal T cells are expanded in vitro, exhibiting a marked overexpression of TCRVβ3 (donor 1) and TCRVβ4/Vβ11 (donor 2) subfamilies. Functional (ELISPOT assay) and phenotypic (CDR3 spectratyping, sequencing Vβ transcripts) analysis of patients' T cells at different time points after transplantation demonstrated an expansion of alloreactive T cells with a limited TCR Vβ pattern. The Vβ3 cDNA clone, being predominant in the CTL line from donor 1, could not be identified in patient 1 peripheral blood lymphocytes after transplant. Altogether, our results provide the first evidence that GVT effects against melanoma can induce tumor regression and that oligoclonal donor‐derived CTLs specific against host tumor cells can be generated in vitrothat may be used for adoptive T‐cell transfer after allogeneic transplantation. © 2002 Wiley‐Liss, Inc.

Purpose: To investigate whether amifostine can reduce radiation hematotoxicity.

In transgenic strains of Dictyostelium discoideum that express β-galactosidase under the control of a prespore-specific promoter, only early slugs show reporter confined to the prespore zone. As slugs migrate β-galactosidase-positive cells accumulate in the prestalk zone; ultimately, there may be so many that the prestalk-prespore boundary is no longer distinguishable (Harwood, A., Early, A., Jermyn, K. and Williams, J. (1991)Differentiation 46, 7-13). It is not clear whether these ‘anomalous’ reporter-positive cells currently express prespore genes; another possibility is that they are ex-prespore cells that have transformed to prestalk and sorted to the prestalk zone (Sternfeld, J. (1993) Roux Archiv. Dev. Biol. 201, 354-363), while retaining their previously produced reporter. To test the activity of the prespore genes in these cells, we have made prespore reporter constructs whose products decay quickly; these are based on constructs used to investigate protein turnover in yeast (Bachmair, A., Finley, D. and Varshavsky, A. (1986) Science 234, 179-186). In strains bearing such constructs, β-galactosidase-positive cells do not appear in the prestalk zone. The apparent deterioration of the prestalk/prespore pattern in older slugs is thus an artefact of reporter stability.

Dictyostelium discoideumforms elongate cell aggregates called "slugs" which migrate over the substrate before completing their conversion into fruiting bodies. Prespore cellsare found in a zone which occupies the rear four-fifths of the slug. Both front- and rear-prespore cells, marked by a bacterial β-galactosidase gene, sort out to their original positions in experiments in which slugs are reconstituted from disaggregated tissue. When cells from the rear of the prespore zone are transplanted to the middle or front, sorting is also observed: the transplanted cells return rapidly to the rear. Cells from the front of the prespore zone, however, were not observed to "home" to the front after transplantation to the rear. Since front-prespore cells sort out in disaggregation/reaggregation experiments, but fail to do so after transplantation to the rear, it is possible that the transplanted cells are converted to rear-prespore cells by extracellular signals present in the rear of the slug. In an experiment designed to test this hypothesis, front cells were transplanted to the rear and the host and transplant together then subjected to disaggregation/reaggregation. The results showed that front-prespore cells had not been converted to rear-prespore cells. Instead, there was an unanticipated effect: cells placed in the rear of the prespore zone underwent an anteriorshift in positional preference, while cells placed in the frontof the prespore zone showed a posteriorshift. The specific sorting properties of front- and rear-prespore cells thus do not appear to result from the action of positional signals; positional signals destabilize rather than reinforce sorting preferences. Our observations are consistent with a model in which innate differences among cells bias them to differentiate as front-prespore or rear-prespore types, but the proportions of these types are also modulated by a negative-feedback mechanism.

Untersuchungen der Subependymschicht im Bereich der Seitenventrikel bei 185 Hunden im Alter von wenigen Stunden bis zu 17 Jahren ergeben, daß die mitotische Aktivität der Schicht bis ins hohe Alter nachweisbar bleibt. Die neugebildeten Zellen können die Schicht verlassen und sich im Nucleus caudatus, Septum pellucidum, um den ventralen Ventrikelwinkel und vermutlich auch in der Ependymschicht lokalisieren. Es wird versucht, darzustellen, unter welchen Bedingungen — abhängig von Alter, Todesursachen und Befunden am ZNS — die Proliferation der Subependymzellen erfolgt.

The histologic changes in the CNS of 10 dogs with epileptiform cramps and psychic alterations are described. Most of the animals had gastroenteritis of apparently diverse aetiology prior to the onset of the nervous signs.The lesions are characterized by bilaterally symmetrical focal necroses in the grey matter. Oedema and proliferation of blood vessels were present to varying extents in individual cases. What is conspicuous is not only the symmetrical localization of the lesions but also the invatiable damage of the rhinencephalon in the broad sense. There was no significant increase of oligo- or astroglia. In almost all cases Ammon's horn and the piriform lobe were simultaneously affected. In addition the olfactory bulbs, septum pellucidum, dorsomedian cortex, and now and then also the lateral cortical layers and basal ganglia might show accompanying lesions. The remaining brain stem and the cerebellum remained almost entirely spared.The aetiology of the lesions described is undetermined. Exogenous intoxication does not appear likely although it cannot be excluded with certainty. The clinical and histologic findings rather suggest the action of diverse pathogenetic factors. These lead to morphologically similar alterations, which may be a consequence of a deficient provision of the brain tissue with oxygen or with nutrient substances.

A histologic study was made of the central nervous system of 108 dogs with neurological illness. One group comprised 71 cases of distemper encephalitis. The second group of 37 cases encompassed toxoplasma encephalitis (14), symmetrical cerebral necrosis (10), reticulosis (6), lymphocytic meningoencephalitis (3), leukoencephalitis (2) and canine viral hepatitis (2).Inclusion bodies can be found in all dogs with distemper encephalitis. In most cases this diagnosis is easy, even without the inclusion bodies. However, detection of inclusions becomes important in cases showing a form of distemper encephalitis atypical with respect to histologic type or localization. In one case each, focal symmetrical necroses and toxoplasmosis-induced lesions were observed in association with distemper infection of the CNS.The regularity with which inclusion bodies can be found in all cases of distemper encephalitis means that in their absence, lesions induced by the distemper virus can be excluded with considerable assurance.The dependence of the density of the inclusion bodies on the type of tissue changes, the histochemical and tinctorial features of the inclusion bodies, the occurrence of lesions in the cerebellar granule cells, and the formation of cell syncytia by glial cells with inclusion bodies are discussed with respect to their specificity for distemper viral infection of the CNS.

THE complexity of the central nervous system of vertebrates makes it difficult to identify the cell type responsible for the biochemical or pharmacological effects of drugs observed in the whole tissue. This difficulty does not exist for clonal cell lines derived from tumours of the nervous system. Such lines derived from mouse neuroblastoma C1300 show a number of differentiated functions of neurones1. Many of these properties are more strongly expressed in clonal lines of hybrid cells obtained by fusion of mouse neuroblastoma and rat glioma cells (refs 2–4 and B.H., unpublished). In suitable conditions, these undifferentiated cells differentiate by extending long processes3–5, which contain structures resembling synaptic vesicles6. In addition, their membranes become excitable by an electric current (refs 3 and 4 and B.H., unpublished) or by neurotransmitters (refs 3, 4 and 7, and B.H., unpublished). On exposure to prostaglandin E1(PGE1) hybrid cells strongly increase their intracellular levels of cyclic AMP8. This effect of PGE1is antagonised by α adrenergic agonists7and morphine7,9.

WORK on cell lines derived from tumours of the nervous system has been carried out to establish models for neurones and glial cells. Clonal cell lines derived from mouse neuroblastoma C 1300 possess many properties characteristic of neurones1–11. While the neuroblastoma cells increase in their intracellular levels of adenosine 3′,5′-cyclic monophosphate (cyclic AMP) in the presence of prostaglandin E1(PGE1) (refs 12 and 13), acetylcholine14and adenosine15, rat glioma cells16do so when exposed to noradrenaline17,18or PGE1(ref. 19).

NARCOTICS do not normally occur in vivo. “Opiate receptors” must therefore have evolved to recognise endogenous ligands, which are mimicked by the opiates1. Indeed such compounds have been isolated2–5and their structure elucidated as that of the two pentapeptides leucine and methionine-enkephalin6. Neuroblastoma × glioma hybrid cells 108CC15 display many properties characteristic of neurones7. In these cells morphine and other narcotics8–10prevent the increase in the level of cyclic AMP that is caused by prostaglandin E1(PGE1)11. The opiates also increase the level of cyclic GMP in the hybrid cells12. On long term incubation with morphine, the hybrid cells show phenomena which can be interpreted as biochemical correlates of morphine tolerance, dependenceand withdrawal9,13. Here we report that leucine-enkephalin (leu-enk) has effects like those of opiates on the hybrid cells.

The immune attack against malignant tumors requires the concerted action of CD8+cytotoxic T lymphocytes (CTL) as well as CD4+T helper cells. The contribution of T cell receptor (TCR)αβ+CD4−CD8−double-negative (DN) T cells to anti-tumor immune responses is widely unknown. In previous studies, we have demonstrated that DN T cells with a broad TCR repertoire are present in humans in the peripheral blood and the lymph nodes of healthy individuals. Here we characterize a human DN T cell clone (T4H2) recognizing an HLA-A2-restricted melanoma-associated antigenic gp100-peptide isolated from the peripheral blood of a melanoma patient. Antigen recognition by the T4H2 DN clone resulted in specific secretion of IFN-γ and TNF. Although lacking the CD8 molecule the gp100-specifc DN T cell clone was able to confer antigen-specific cytotoxicity against gp100-loaded target cells as well as HLA-A2+gp100 expressing melanoma cells. The cytotoxic capacity was found to be perforin/granzymeB-dependent. Together, these data indicate that functionally active antigen-specific DN T cells recognizing MHC class I-restricted tumor-associated antigen (TAA) may contribute to anti-tumor immunity in vivo.

The immune attack against malignant tumors requires the concerted action of CD8+ cytotoxic T lymphocytes (CTL) as well as CD4+ T helper cells. The contribution of T cell receptor (TCR)αβ+ CD4− CD8− double-negative (DN) T cells to anti-tumor immune responses is widely unknown. In previous studies, we have demonstrated that DN T cells with a broad TCR repertoire are present in humans in the peripheral blood and the lymph nodes of healthy individuals. Here we characterize a human DN T cell clone (T4H2) recognizing an HLA-A2-restricted melanoma-associated antigenic gp100-peptide isolated from the peripheral blood of a melanoma patient. Antigen recognition by the T4H2 DN clone resulted in specific secretion of IFN-γ and TNF. Although lacking the CD8 molecule the gp100-specifc DN T cell clone was able to confer antigen-specific cytotoxicity against gp100-loaded target cells as well as HLA-A2+ gp100 expressing melanoma cells. The cytotoxic capacity was found to be perforin/granzymeB-dependent. Together, these data indicate that functionally active antigen-specific DN T cells recognizing MHC class I-restricted tumor-associated antigen (TAA) may contribute to anti-tumor immunity in vivo.

In eukaryotic cells the phospholipid phosphatidylserine (PS) is restricted to the inner plasma membrane leaflet. This lipid asymmetry which is maintained by the concerted action of phospholipid transport proteins is mainly lost during apoptosis. Here, we demonstrate that primary human CD8+ cytotoxic T lymphocytes (CTL) expose PS upon T-cell receptor (TCR)-mediated antigen recognition: antigen-specific CTL, recognizing the HLA-A2 binding Melan-A peptide, demonstrated a marked exposure of PS as determined by annV-FITC staining, a highly specific PS-binding protein, after 4 h of stimulation with Melan-A-loaded antigen presenting cells (APC). PS exposure was Ag-specific (>80% annV-positive T cells) but control peptide (gp100)-pulsed and unpulsed APC also induced a slight background PS exposure on the CTL. To follow more precisely the fate of annV-positive CTL after antigen-specific stimulation, we labeled Melan-A-specific CTL with the membrane dye PKH-26 and isolated all annV+ PKH-26+ CTL 4 h after stimulation with peptide-pulsed APC cells by cell sorting. Results demonstrate that the annV-positive T-cell population is heterogenous: while the annV-high T-cell population retained the annV-high phenotype and consisted of apoptotic T cells, the annV-intermediate(int) T-cell population revealed a constant decrease of annV binding and became annV-negative at 54 to 72 h after stimulation. Using three independent assays for apoptosis we found that annV-int T cells are propidium iodide negative, do not exhibit DNA strand brakes and contain no active caspase 3. In contrast, annV-int CTL revealed a strongly activated phenotype indicated by an upregulation of CD69 expression and downregulation of the TCRaß chain expression. Fluorescence microscopic analysis demonstrated that PS is distributed inhomogenously over the plasma membrane and concentrated in membrane lipid raft domains at the immunological synapse. By studying the activity of PS transport proteins using a fluorescence-labeled PS analogue, we determined a constitutive outward transfer of PS molecules in Melan-A-specific CTL. The constitutive PS outward transport was not further accelerated after antigen re-stimulation. In sharp contrast, the inward transporting flippase was strongly inhibited in stimulated CTL resulting in an accumulation of PS molecules on the cell surface. Shielding of exposed PS by annexinV protein during antigen recognition diminished cytokine secretion, activation and cell-cell clustering of antigen-specific CTL. In summary, our data demonstrate for the first time that externalized PS on antigen-stimulated CTL is linked to T-cell activation and probably involved in cell-cell contact formation at the immunological synapse.

Various subsets of T lymphocytes have been isolated in mice and humans that have the ability to down-regulate the proliferation of autoimmune effector cells. Recently, a novel subset of Ag-specific T-cell receptor (TCR)αβ+ CD4−CD8− (double negative, DN) Treg cells has been found to be able to prevent the rejection of skin and heart allografts by specifically inhibiting the function of anti-graft-specific CD8+ T cells. Here we demonstrate that peripheral DN Treg cells are present in humans, where they constitute about 1% of total CD3+ T cells, and consist of both naïve and Ag-experienced cells. Furthermore, analysis of T-cell receptor excision circles (TRECs) indicate that DN T cells are not recent thymic emigrants, but rather an expanded T-cell subset. MHC multimer staining revelaed a distinct population of DN T cells recognizing common MHC class I-restricted CMV and EBV antigens. DN T cells exhibited a strong proliferative response upon stimulation with allogeneic antigen presenting cells (APC) and secreted high amounts of IFN-γ but no IL-2, with some IL-5, and marginal levels of IL-4 and IL-10. Similar to murine DN Treg cells, human DN Treg cells are able to acquire peptide-HLA-A2 complexes from APCs by cell contact-dependent mechanisms. Furthermore, such acquired peptide-HLA complexes appear to be functionally active, in that CD8+ T cells specific for the HLA-A2-restricted self peptide, Melan-A, became sensitive to apoptosis by neighboring DN T cells after acquisition of Melan-A-HLA-A2 complexes and revealed a reduced proliferative response. These results demonstrate for the first time that a sizeable population of peripheral DN Treg cells exists in humans that are able to suppress Ag-specific T cells. DN Treg cells may serve to limit clonal expansion of allo-Ag-specific T cells after transplantation.

A characteristic feature of inflammatory lesions or tumor sites is local acidosis, which is attributed to the local increase in lactic acid production. We studied the effect of such an acidic environment on the immune functions of antigen-specific CD8+ T-cells by incubating the cells in the presence of various concentrations of lactic acid for up to 48h. CD8+ T-cells were isolated from healthy donors and expanded by weekly stimulation with autologous dendritic cells pulsed with a mutated HLA-A2-binding Melan-A (ELAGIGILTV) peptide. The obtained T cell population consisted of at least 90% CD8+ and about 60% Melan-A specific T cells, as determined by Melan-A multimer staining. Incubation of CD8+ T cells with up to 20mM lactic acid for 24h did not cause T-cell apoptosis or cell death as determined by combined annexin/propidium iodide staining. However, the proliferative capacity of CD8+ T cells, as determined by 3H-thymidine uptake, was strongly inhibited. Similar results were obtained when we determined cytokine production and cytotoxic activity of the cells after a 24h culture period in 5-20 mM lactic acid. Production of both, IL-2 and IFN-gamma was strongly diminished in comparison to untreated cells, as determined by intracellular staining after stimulation with PMA/ionomycin for 5h in the presence of monensin. Analysis of the antigen-specific cytolytic capacity revealed that CD8+ T cells pre-cultured with lactic acid were less effective in killing antigen-loaded T2 target cells as compared to untreated CD8+ T cells. In parallel, the intracellular contents of the cytotoxic effector molecules granzyme-B and perforin was diminished. Re-adjusting the pH of the medium to a physiological value of pH7.4 could partially revert the effect of lactic acid. Treatment of CD8+ T cells with sodium lactate instead of lactic acid had no inhibitory effect. We conclude, that lactic acid is an important modulator of CD8+ T-cell function and may contribute, together with other factors, to immune escape mechanisms in the tumor environment.
