Die Stadt- und Landesbibliothek (SLB) Potsdam ist im Land Brandenburg für die Erfüllung der Pflichtexemplaraufgabe verantwortlich, die auf gesetzlichen Grundlagen wie dem Brandenburgischen Pressegesetz (BbgPG) von 1993 und der nachgeordneten Pflichtexemplarverordnung (PflEV) von 1994 basiert. Seit 2012 umfasst die Sammlungspflicht neben gedruckten auch Netzpublikationen. Die wachsenden Anforderungen an die E-Pflicht stellen neue Herausforderungen dar, denen durch organisatorische Anpassungen und technologische Weiterentwicklungen in Kooperationen, wie die Nutzung des OPUS-Repositoriums „BrandenburgDOK“, Rechnung getragen wird. Aktuelle Herausforderungen betreffen die Weiterentwicklung von Richtlinien, insbesondere zur Sammlung von Webseiten und zur Langzeitarchivierung. Ziel der SLB ist es, eine repräsentative Pflichtexemplarsammlung aufzubauen, die die brandenburgische Verlags- und Publikationsgeschichte umfassend dokumentiert.

Magnetism of oxide antiferromagnets (AFMs) has been studied in single crystals and extended thin films. The properties of AFM nanostructures still remain underexplored. Here, we report on the fabrication and magnetic imaging of granular 100 nm-thick magnetoelectric Cr2O3films patterned in circular bits with diameters ranging from 500 down to 100 nm. With the change of the lateral size, the domain structure evolves from a multidomain state for larger bits to a single domain state for the smallest bits. Based on spin–lattice simulations, we show that the physics of the domain pattern formation in granular AFM bits is primarily determined by the energy dissipation upon cooling, which results in motion and expelling of AFM domain walls of the bit. Our results provide a way toward the fabrication of single domain AFM-bit-patterned memory devices and the exploration of the interplay between AFM nanostructures and their geometric shape.

In systemic lupus erythematosus, loss of immune tolerance, autoantibody production and immune complex deposition are required but not sufficient for organ damage1. How inflammatory signals are initiated and amplified in the setting of autoimmunity remains elusive. Here we set out to dissect layers and hierarchies of autoimmune kidney inflammation to identify tissue-specific cellular hubs that amplify autoinflammatory responses. Using high-resolution single-cell profiling of kidney immune and parenchymal cells, in combination with antibody blockade and genetic deficiency, we show that tissue-resident NKp46+innate lymphoid cells (ILCs) are crucial signal amplifiers of disease-associated macrophage expansion and epithelial cell injury in lupus nephritis, downstream of autoantibody production. NKp46 signalling in a distinct subset of group 1 ILCs (ILC1s) instructed an unconventional immune-regulatory transcriptional program, which included the expression of the myeloid cell growth factor CSF2. CSF2 production by NKp46+ILCs promoted the population expansion of monocyte-derived macrophages. Blockade of the NKp46 receptor (using the antibody clone mNCR1.15; ref. 2) or genetic deficiency of NKp46 abrogated epithelial cell injury. The same cellular and molecular patterns were operative in human lupus nephritis. Our data provide support for the idea that NKp46+ILC1s promote parenchymal cell injury by granting monocyte-derived macrophages access to epithelial cell niches. NKp46 activation in ILC1s therefore constitutes a previously unrecognized, crucial tissue rheostat that amplifies organ damage in autoimmune hosts, with broad implications for inflammatory pathologies and therapies.

In this work, we have synthesized a set of peptoid-based histone deacetylase inhibitors (HDACi) with a substituted hydrazide moiety as zinc-binding group. Subsequently, all compounds were evaluated in biochemical HDAC inhibition assays and for their antiproliferative activity against native and cisplatin-resistant cancer cell lines. The hydrazide derivatives with a propyl or butyl substituent (compounds 5and 6) emerged as the most potent class I HDAC selective inhibitors (HDAC1–3). Further, compounds 5and 6outperformed entinostat in cytotoxicity assays and were able to reverse chemoresistance in cisplatin-resistant A2780 (ovarian) and Cal27 (head-neck) cancer cell lines. Moreover, the hydrazide derivatives 5and 6showed strong synergism with cisplatin (combination indices <0.2), again outperforming entinostat, and increased DNA damage, p21, and pro-apoptotic BIM expression, leading to caspase-mediated apoptosis and cell death. Thus, compounds 5and 6represent promising lead structures for developing new HDACi capable of reversing chemoresistance in cisplatin resistant cancer cells.

Im ersten Teil des Beitrags (FR 2024, 836) wurde die Verfassungsmäßigkeit der Besteuerung der Energiepreispauschalen I und II (EPP I und II) untersucht. Im Rahmen des zweiten Teils des Beitrags sollen nunmehr Überlegungen zu den dem Gesetzgeber zur Verfügung stehenden Gestaltungsmöglichkeiten, die auch in Zukunft für (steuerliche) Transferzahlungen eingesetzt werden könn(t)en, aufgezeigt werden. Diese Überlegungen sollen insbesondere deshalb angestellt werden, um Aussagen dazu treffen zu können, ob eine entsprechende (alternative) Umsetzung bereits im Rahmen der Umsetzung der EPP I und der EPP II hätte in Erwägung gezogen werden können bzw. sollen, da die bestehenden Gestaltungsmöglichkeiten den verfolgten Zielen zielführender bzw. einfacher Rechnung getragen hätten als die gewählten Ausgestaltungen.

The determination of ligand–receptor binding affinities plays a key role in the development process of pharmaceuticals. While the classical radiochemical binding assay uses radioligands, fluorescence-based binding assays require fluorescent probes. Usually, radio- and fluorescence-labeled ligands are dissimilar in terms of structure and bioactivity, and can be used in either radiochemical or fluorescence-based assays. Aiming for a close comparison of both assay types, we synthesized tritiated fluorescent neurotensin receptor ligands ([3H]13, [3H]18) and their nontritiated analogues (13, 18). The labeled probes were studied in radiochemical and fluorescence-based (high-content imaging, flow cytometry, fluorescence anisotropy) binding assays. Equilibrium saturation binding yielded well-comparable ligand–receptor affinities, indicating that all these setups can be used for the screening of new drugs. In contrast, discrepancies were found in the kinetic behavior of the probes, which can be attributed to technical differences of the methods and require further studies with respect to the elucidation of the underlying mechanisms.

Herein, we describe the design, synthesis, and biological evaluation of 15 Contilisant+Tubastatin Ahybrids. These ligands are polyfunctionalized indole derivatives developed by juxtaposing selected pharmacophoric moieties of Contilisantand Tubastatin Ato act as multifunctional ligands. Compounds 3and 4were identified as potent HDAC6 inhibitors (IC50= 0.012 μM and 0.035 μM, respectively), so they were further evaluated in Drosophilaand human cell models of Parkinson’s disease (PD). Both compounds attenuated PD-like phenotypes, such as motor defects, oxidative stress, and mitochondrial dysfunction in PD model flies. Ligands 3and 4were also studied in the transgenic Caenorhabditis elegansCL2006 model of Alzheimer’s disease (AD). Both compounds were nontoxic, did not induce undesirable animal functional changes, inhibited age-related paralysis, and improved cognition in the thrashing assay. These results highlight 3and 4as novel multifunctional ligands that improve the features of PD and AD hallmarks in the respective animal models.

The compound Na2B6Si2was synthesized under high-pressure, high-temperature conditions at pressures ranging from 6 to 9.5 GPa and temperatures from 1070 to 1270 K before quenching to room temperature followed by slow decompression. The crystal structure was determined from microcrystals using precession-assisted electron diffraction tomography, validated by dynamical refinement and full-profile refinements using optimized coordinates from quantum chemical calculations (space group R3̅m, Pearson symbol hR30, a= 5.0735(1) Å and c= 16.0004(7) Å). The atomic arrangement consists of a unique framework formed by electron-precise octahedral closo (B6)2–clusters connected via ethane-like (Si2)0dumbbells. The Na+cations occupy cavities in the hierarchical variation of a Heusler-type framework. The balance (Na+)2([B6]2–)(Si0)2reveals an electron precise Zintl-Wade phase, which is in line with electronic band structure calculations predicting semiconducting behavior.

Nachdem im ersten Teil des Beitrags (Ubg 2024, 430) zunächst die Problematik von Mitarbeiteroptionen im Kontext der Wegzugsbesteuerung aufgearbeitet worden ist und zwei konkrete Praxisfälle unter Berücksichtigung des neugefassten AEAStG kritisch analysiert wurden, werden im zweiten Teil des Beitrags zwei weitere Praxisfälle genauer analysiert, bevor abschließend der aktuelle Stand der (noch immer nicht beendeten) Diskussion um die Gewährung einer dauerhaften zinslosen Stundung bei einem Wegzug in die Schweiz aufgearbeitet und erörtert wird.

Im vorliegenden ersten Teil des Beitrags untersuchen die Verfasser die Frage der Verfassungsmäßigkeit der Besteuerung der Energiepreispauschalen I und II. Im anschließenden zweiten Teil des Beitrags werden dann Überlegungen zu Gestaltungsmöglichkeiten zukünftiger (steuerlicher) Transferzahlungen angestellt, um prüfen zu können, ob durch diese den mit beiden Energiepreispauschalen verfolgten gesetzgeberischen Zielen besser hätte Rechnung getragen werden können als durch die gewählten Ausgestaltungen.

Heusler-type Co–Ni–Ga shape memory alloys attracted significant attention due to their excellent functional properties in single-crystalline state under both compressive and tensile loading. The present study investigates the superelastic deformation behavior under torsion. Using a newly installed torsion testing setup, in situ synchrotron diffraction was carried out on single-crystalline material in order to investigate the martensitic phase transformation. Incremental deformation experiments reveal a fully reversible martensitic transformation under torsional loading at room temperature, leading to excellent strain recovery after deformation to 6.5% shear strain. Furthermore, relevant aspects towards the analysis of powder diffraction data obtained for single-crystalline material in transmission mode under torsional loading are presented and critically discussed.

The delta opioid receptor (δOR or DOR) is a G protein-coupled receptor (GPCR) showing a promising profile as a drug target for nociception and analgesia. Herein, we design and synthesize new fluorescent antagonist probes with high δOR selectivity that are ideally suited for single-molecule microscopy (SMM) applications in unmodified, untagged receptors. Using our new probes, we investigated wild-type δOR localization and mobility at low physiological receptor densities for the first time. Furthermore, we investigate the potential formation of δOR homodimers, as such a receptor organization might exhibit distinct pharmacological activity, potentially paving the way for innovative pharmacological therapies. Our findings indicate that the majority of δORs labeled with these probes exist as freely diffusing monomers on the cell surface in a simple cell model. This discovery advances our understanding of OR behavior and offers potential implications for future therapeutic research.

For healthspan and lifespan, ERK, AMPK and mTORC1 represent critical pathways and inflammation is a centrally important hallmark1–7. Here we examined whether IL-11, a pro-inflammatory cytokine of the IL-6 family, has a negative effect on age-associated disease and lifespan. As mice age, IL-11 is upregulated across cell types and tissues to regulate an ERK–AMPK–mTORC1 axis to modulate cellular, tissue- and organismal-level ageing pathologies. Deletion of Il11or Il11ra1protects against metabolic decline, multi-morbidity and frailty in old age. Administration of anti-IL-11 to 75-week-old mice for 25 weeks improves metabolism and muscle function, and reduces ageing biomarkers and frailty across sexes. In lifespan studies, genetic deletion of Il11extended the lives of mice of both sexes, by 24.9% on average. Treatment with anti-IL-11 from 75 weeks of age until death extends the median lifespan of male mice by 22.5% and of female mice by 25%. Together, these results demonstrate a role for the pro-inflammatory factor IL-11 in mammalian healthspan and lifespan. We suggest that anti-IL-11 therapy, which is currently in early-stage clinical trials for fibrotic lung disease, may provide a translational opportunity to determine the effects of IL-11 inhibition on ageing pathologies in older people.

Introduction of Th into synthetic disordered fluorite-type Gd2Zr2O7induces a transition to an ordered pyrochlore-type phase at a Th concentration of 10 % at the Gd site (Gd1.8Th0.2Zr2O7composition). The degree of order of the fluorite-type phase reaches 50 % for a Th concentration of 25 % (Gd1.5Th0.5Zr2O7composition). Upon application of high pressure, the Gd2Zr2O7phase retains the fluorite-type structure until 33 GPa (K0= 167(1) GPa), where it undergoes reversible amorphization. The Gd1.7Th0.3Zr2O7phase was found to be stable up to at least a pressure of 25 GPa (K0= 169(3) GPa). Upon heating to Tmaxof 1135 K, the Gd2Zr2O7phase retains its disordered fluorite-type structural arrangement (α= 3.03 × 10−5K−1). The excellent stability of the Gd2−xThxZr2O7phases under extreme conditions of temperature and pressure makes Gd2Zr2O7a promising candidate as a host matrix for radioactive elements for safe long-term underground storage of nuclear waste.

Anthropogenic activities have fundamentally changed the chemistry of the Baltic Sea. According to results reported in this study, not even the thallium (Tl) isotope cycle is immune to these activities. In the anoxic and sulfidic (“euxinic”) East Gotland Basin today, Tl and its two stable isotopes are cycled between waters and sediments as predicted based on studies of other redox-stratified basins (e.g., the Black Sea and Cariaco Trench). The Baltic seawater Tl isotope composition (ε205Tl) is, however, higher than predicted based on the results of conservative mixing calculations. Data from a short sediment core from East Gotland Basin demonstrates that this high seawater ε205Tl value originated sometime between about 1940 and 1947 CE, around the same time other prominent anthropogenic signatures begin to appear in the same core. This juxtaposition is unlikely to be coincidental and suggests that human activities in the surrounding area have altered the seawater Tl isotope mass-balance of the Baltic Sea.

Schulnoten werden seit jeher kontrovers diskutiert, insbesondere in Bezug auf ihre Bedeutung und Vergleichbarkeit. Dies wird in unterschiedlichen Publikationen deutlich, die zum Teil bis in das 19. Jahrhundert zurückreichen, deren kritische Anmerkungen aber bis heute nicht an Aktualität verloren haben. Auch in jüngster Zeit finden immer wieder bildungspolitische Debatten statt, die im Kern auf Widersprüche zwischen der unklaren Aussagekraft von Schulnoten und ihrer gleichzeitig großen Bedeutung für Selektions- und Allokationsprozesse verweisen. Einen besonderen Schwerpunkt in diesen jüngeren Diskursen bildet die Kritik an der mangelnden Vergleichbarkeit von Schulnoten aus unterschiedlichen Bundesländern, die letztlich, so die vielfach vorgetragene Argumentation, insbesondere durch eine Auflösung föderaler Strukturen und eine nationale Bildungsstrategie überwunden werden könne. Der vorliegende Beitrag nimmt diese Diskussion zum Anlass, die Vergleichbarkeit von Schulnoten genauer zu untersuchen. Hierzu erfolgt eine systematische Aufschlüsselung der Beiträge unterschiedlicher Ebenen (innerhalb von Schulen, zwischen Schulen, zwischen Bundesländern) zur mangelnden Vergleichbarkeit von Schulnoten. Grundlage der Untersuchung bilden repräsentative Daten von N= 55.002 Schülerinnen und Schülern aus den IQB-Bildungstrends 2015 und 2018 (Jahrgangsstufe 9). Die Ergebnisse legen nahe, dass Schülerinnen und Schüler mit gleichen Schulnoten substantiell unterschiedliche Kompetenzen aufweisen und dies auch umgekehrt gilt. Ferner zeigte sich, dass die Unterschiede zu einem erheblichen Maß durch Unterschiede zwischen Schulen innerhalb von Bundesländern erklärt werden konnten und deutlich weniger durch Bundeslandunterschiede. Die Ergebnisse verdeutlichen, dass die Debatte zur eingeschränkten Vergleichbarkeit von Schulnoten ihr Augenmerk nicht nur auf die Ebene der Bundesländer, sondern auch auf die Mechanismen der Notengebung und Ursachen von Schulnotenunterschieden zwischen Schulen innerhalb von Bundesländern richten sollte.

Hintergrund: Mit einer Inzidenz von etwa 1–3 % aller Malignome ist das Syndrom des „cancer of unknown primary“ (CUP-Syndrom) nach wie vor eine relevante Tumorentität. Es wird definiert dadurch, dass bei einer zytologisch oder histologisch bestätigten malignen Erkrankung nach standardisierter und erweiterter Diagnostik der Primärtumor unbekannt bleibt. Ziel: In der vorliegenden Arbeit wird der aktuelle Standard der klinischen Diagnostik beim CUP-Syndrom zusammengefasst, dabei werden verschiedene Lokalisationen und Manifestationen berücksichtigt. Material und Methoden: Für diese Arbeit erfolgte eine fokussierte Literaturrecherche in der Datenbank PubMed sowie die Auswertung aktueller Leitlinien und Empfehlungen wissenschaftlicher Fachgesellschaften. Ergebnis: Die Diagnostik erfolgt bei Patienten mit Metastasen unbekannten Ursprungs zum einen lokalisationsbezogen, zum anderen orientiert an der Histologie inklusive Immunhistologie. Eine allgemeine Basisdiagnostik ist zu ergänzen durch anamnese- und symptombezogene Untersuchungen sowie neuerdings bei definierten Konstellationen durch spezifische ergänzende Algorithmen. Zeigt sich dabei kein Primärtumor, liegt ein CUP-Syndrom vor. Die Behandlung richtet sich danach, ob eine definierte Subgruppe vorliegt – dann erfolgt eine spezifische Therapie – oder nicht. Bei der sog. ungünstigen Gruppe erfolgt eine empirische Therapie. Schlussfolgerung: Durch neue spezifische diagnostische Algorithmen gelingt eine deutlich klarere Definition des CUP-Syndroms als bisher. Dadurch wird die Therapiezuordnung verbessert und das Kollektiv für klinische Studien definiert.
