The objective of this study was to compare 2 laying hen strains in 5 production periods regarding phytase activity, phytate (InsP6) degradation, and myo-inositol (MI) release in the digestive tract and phosphorus (P) and calcium (Ca) utilization. One offspring of 10 nonrelated roosters per strain (Lohmann Brown-classic (LB) or Lohmann LSL-classic (LSL)) was placed in one of 20 metabolic units in a completely randomized block design in week 8, 14, 22, 28, and 58 of life. All hens were fed the same corn and soybean meal–based diet at one time, but the diet composition was adjusted to the requirements in the respective period. For 4 consecutive days, excreta were collected quantitatively at 24-hour intervals. In week 10, 16, 24, 30, and 60, the blood plasma, digesta of crop, gizzard, jejunum, ileum, and ceca, and mucosa of the jejunum was collected. The concentration of inorganic P in the blood plasma was higher in LB than in LSL hens (P = 0.026). Plasma Ca concentrations increased with each period (P < 0.001) in both strains. In jejunum digesta, the MI concentration did not differ between strains, but InsP6concentration was higher in LB than in LSL hens (P = 0.002) and the highest in week 30 and 60. Total phosphatase and phytase activities were higher in LB than in LSL hens (P ≤ 0.009). Period effects were also significant for these enzymes. Concentrations of some constituents of the cecal content were different between the strains. The MI concentration in the egg albumen and yolk was higher in LB than in LSL hens. Differences in InsP6- and MI-related metabolism of the 2 hen strains existed. These differences were partly dependent of the period. Especially, week 24 was a period of remarkable change of metabolism. Great differences also existed among individuals, making it worth to have a closer look at the metabolism of individuals in addition to evaluating treatment means. Further studies on metabolic, genetic, and microbiome level may help explain these differences.

As a constituent of animal cells, myo-inositol (MI) has been hypothesized to be crucial in several metabolic and regulatory pathways. Recently, it was shown that dietary phytase contributes to release of MI from phytate in the poultry digestive tract, increasing its systemic concentrations. This study investigated the activities of phosphatases in the jejunum and systemic plasma MI concentration in broilers not supplemented or supplemented with phytase through analyses based on modifications from commercial enzyme activity kits. Three hundred sixty male Ross 308 broilers were randomly allocated to 24 pens (15 birds per pen) in 4 dietary groups. The positive control group was fed with an adequate basal diet. The negative control group (NC) was fed with a reduced level of P and Ca. Groups Phy1500 and Phy3000 were fed with the NC diet plus 1,500 or 3,000 FTU of phytase per kilogram of feed, respectively. One bird per pen was selected for the measurement of jejunal phosphatase activity; MI concentration in plasma, the liver, and the kidney; and key MI enzyme concentrations (liver inositol monophosphatase 1 [IMPase 1] and kidney myo-inositol oxygenase [MIOX]). Endogenous phytase and alkaline phosphatase activity as well as IMPase 1 and MIOX expression were not statistically different among the dietary groups. The supplementation of 1500 FTU of phytase per kilogram of feed resulted in increase of plasma (P < 0.001) and kidney (P < 0.05) but not liver MI concentrations. The results indicated that systemic MI might reflect MI released from dietary sources; however, it did not appear to change expression of enzymes related to endogenous MI synthesis in the liver and catabolism in the kidney. New and larger studies are necessary to reach stronger evidence on the effects of dietary phytase on intestinal and systemic MI concentrations in broilers.

Myo-inositol (MI) has gained relevance in physiology research during the last decade. As a constituent of animal cells, MI was proven to be crucial in several metabolic and regulatory processes. Myo-inositol is involved in lipid signaling, osmolarity, glucose, and insulin metabolism. In humans and rodents, dietary MI was assessed to be important for health so that MI supplementation appeared to be a valuable alternative for treatment of several diseases as well as for improvements in metabolic performance. In poultry, there is a lack of evidence not only related to specific species-linked metabolic processes but also about the effects of dietary MI on performance and health. This review intends to provide information about the meaning of dietary MI in animal metabolism as well as to discuss potential implications of dietary MI in poultry health and performance with the aim to identify open questions in poultry research.

The objective of this study was to characterize intestinal phytate degradation and mineral utilization by two laying hen strains before and after the onset of egg laying using diets without or with a mineral phosphorus (P) supplement. One offspring of 10 roosters per strain (Lohmann Brown-classic (LB) and Lohmann LSL-classic (LSL)) was sacrificed before (wk 19) and after (wk 24) the onset of egg-laying activity and following 4 wk placement in a metabolic unit. Diets were corn-soybean meal-based and without supplemented P (P-) or with 1 g/kg supplemented P (P+) from monocalcium phosphate. In wk 19 and 24, the blood plasma and digesta of duodenum+jejunum and distal ileum were collected. The concentration of P in blood plasma was higher in hens fed P+ than P- (P< 0.001). In duodenum+jejunum and ileum content, the concentrations of InsP6, Ins(1,2,4,5,6)P5and Ins(1,2,3,4,5)P5were lower in P- than in P+ (P≤ 0.009). In duodenum+jejunum, the concentrations of InsP6, Ins(1,2,4,5,6)P5and Ins(1,2,3,4,5)P5were lower in wk 24 than 19 and lower in LSL than LB hens (P< 0.001). The concentration of myo-inositol (MI) in duodenum+jejunum content was lower in wk 19 than 24 (P< 0.001). Following a 4-d total excreta collection, the retained amount of P was higher in P+ than P- (P< 0.001). Phosphorus retention was lower in LB hens fed P- than in other treatments (P × strain: P = 0.039). In the jejunal tissue, some genes related to intracellular InsP metabolism were higher expressed in LB than LSL hens. The renunciation of mineral P increased endogenous phytate degradation, but more P was retained with supplemented P. Differences in endogenous phytate degradation between the periods before and after the onset of egg laying might be attributed to different Ca concentrations in intestinal digesta caused by different Ca needs in both periods.

Insulin dysregulation (ID) with basal or postprandial hyperinsulinemia is one of the key findings in horses and ponies suffering from the equine metabolic syndrome (EMS). Assessment of ID can easily be performed in clinical settings by the use of oral glucose challenge tests. Oral glucose test (OGT) performed with 1 g/kg bodyweight (BW) glucose administered via naso-gastric tube allows the exact administration of a defined glucose dosage in a short time. However, reliable cut-off values have not been available so far. Therefore, the aim of the study was to describe variations in insulin response to OGT via naso-gastric tubing and to provide a clinical useful cut-off value for ID when using the insulin quantification performed with an equine-optimized insulin enzyme-linked immunosorbent assay. Data visualization revealed no clear separation in the serum insulin concentration of insulin sensitive and insulin dysregulated horses during OGT. Therefore, a model based clustering method was used to circumvent the use of an arbitrary limit for categorization. This method considered all data-points for the classification, taking into account the individual insulin trajectory during the OGT. With this method two clusters were differentiated, one with low and one with high insulin responses during OGT. The cluster of individuals with low insulin response was consistently detected, independently of the initialization parameters of the algorithm. In this cluster the 97.5% quantile of insulin is 110 µLU/mL at 120 min. We suggest using this insulin concentration of 110 µLU/mL as a cut-off value for samples obtained at 120 min in OGT. OGT performed with 1 g/kg BW glucose and administration via naso-gastric tubing can easily be performed under clinical settings. Application of the cut-off value of 110 µLU/mL at 120 min allows assessment of ID in horses.

A comparison between 3-wk-old female turkeys (B.U.T. 6) and broilers (Ross 308) was performed to study the effects of species, dietary P, Ca, and phytase levels on gut mucosal phosphatase activity, myo-inositol hexakisphosphate (InsP6) degradation along the digestive tract, digestibility of P, Ca, and amino acids, and concentrations of myo-inositol in the digesta and blood. The experimental diets were corn-soybean meal-based and identical for both species. Two dietary P and Ca concentrations (CaP–: 4.1 g P/kg, 5.5 g Ca/kg and CaP+: 9.0 g P/kg, 12.0 g Ca/kg) and 2 levels of phytase supplementation (0 and 1,500 FTU/kg) were used in a 2 × 2 factorial design and fed to the animals for 7 d in their third week of age. Each diet was randomly assigned to 6 broiler and 6 turkey pens, with 10 birds each. After slaughter, blood, digesta from the crop, gizzard, duodenum, lower ileum, and mucosa from the jejunum were collected. When fed CaP– without phytase supplementation, there were no differences between species in gut mucosal phosphatase activity, prececal InsP6disappearance, and P and Ca digestibility, indicating a similar intrinsic capacity for phytate degradation in both species. When fed CaP+ without phytase supplementation, turkeys showed higher prececal InsP6disappearance than broilers. Phytase supplementation increased prececal InsP6disappearance and digestibility of P and Ca in both species. However, the phytase-induced increase in prececal InsP6disappearance was more pronounced in broilers than in turkeys, possibly due to more adequate conditions for phytase activity in the broiler crop. In broilers, phytase supplementation increased amino acid digestibility overall, whereas, in turkeys, it increased with CaP+ and decreased with CaP–. In addition, the relationship between myo-inositol concentration in the ileum and blood differed between species, indicating differences in myo-inositol metabolism. It was concluded that 3-week-old turkeys and broilers differ in nutrient digestibility and InsP degradation in some segments of the digestive tract but have similar endogenous InsP6degradation when fed low P and Ca diets.

Aggregation of data, including deep sequencing of mRNA and miRNA data in jejunum mucosa, abundance of immune cells, metabolites, or hormones in blood, composition of microbiota in digesta and duodenal mucosa, and production traits collected along the lifespan, provides a comprehensive picture of lifelong adaptation processes. Here, respective data from two laying hen strains (Lohmann Brown-Classic (LB) and Lohmann LSL-Classic (LSL) collected at 10, 16, 24, 30, and 60 wk of age were analyzed. Data integration revealed strain- and stage-specific biosignatures, including elements indicative of molecular pathways discriminating the strains. Although the strains performed the same, they differed in the activity of immunological and metabolic functions and pathways and showed specific gut-microbiota-interactions in different production periods. The study shows that both strains employ different strategies to acquire and maintain their capabilities under high performance conditions, especially during the transition phase. Furthermore, the study demonstrates the capacity of such integrative analyses to elucidate molecular pathways that reflect functional biodiversity. The bioinformatic reduction of the multidimensional data provides good guidance for further manual review of the data.

Female turkeys (B.U.T. 6) and broilers (Ross 308) were compared at 6 weeks of age to evaluate the effects of species, dietary P, Ca, and phytase levels on myo-inositol hexakisphosphate (InsP6) degradation along the digestive tract, gut mucosal phosphatase activity, P and Ca digestibility, and myo-inositol concentrations in the digesta and blood. The environmental conditions and experimental corn-soybean meal-based diets were the same for both species. Four diets with either combination of 2 levels of P and Ca (CaP–: 4.0 g P/kg, 5.4 g Ca/kg and CaP+: 6.0 g P/kg, 8.0 g Ca/kg) and 2 levels of phytase supplementation (0 and 1,500 FTU/kg) were fed to the animals for 7 days at their 6thweek of age. Each diet was randomly assigned to 6 pens per species, with 10 birds each. After slaughter, blood, digesta from the crop, gizzard, duodenum, lower ileum, and jejunal mucosa were collected. Endogenous mucosal phosphatase activity in the jejunum was higher in turkeys than in broilers. Prececal InsP6disappearance was also higher in turkeys than in broilers when phytase was not supplemented. Phytase supplementation led to a higher prececal InsP6disappearance in broilers than in turkeys, likely due to different crop conditions such as moisture content. However, prececal P digestibility was higher in turkeys than broilers. Different relationships between myo-inositol concentration in the ileum digesta and blood were found, depending on the species. A comparison of the results with those obtained in 3-week-old birds of a companion study showed that in diets with low Ca and P levels, prececal InsP6disappearance increased with age in turkeys, but not in broilers. This coincided with changes in the conditions of the digestive tract, such as the water content in the crop, gizzard pH, and mucosal phosphatase activity. In conclusion, occurrence of differences in phytate degradation between turkeys and broilers, fed the same feed, depended on age and can be explained by different physiological development of the digestive tract.

During life, the number and function of immune cells change with potential consequences for immunocompetence of an organism. In laying hens, studies have primarily focused on early development of immune competence and only few have investigated systemic and lymphatic distribution of leukocyte subsets during adolescence and the egg-laying period. The present study determined the number of various leukocyte types in blood, spleen, and cecal tonsils of 10 Lohmann Brown-Classic and 10 Lohmann LSL-Classic hens per wk of life 9/10, 15/16, 23/24, 29/30, and 59/60, encompassing important production as well as developmental stages, by flow cytometry. Although immune traits differed between the 2 hen strains, identical patterns of age-related immunological changes were found. The numbers of all investigated lymphocyte types in the spleen as well as the numbers of blood γδ T cells increased from wk 9/10 to 15/16. This suggests an ongoing release of lymphocytes from primary lymphoid tissues and an influx of blood lymphocytes into the spleen due to novel pathogen encounters during adolescence. A strong decrease in the number of CTL and γδ T cells and an increase in innate immune cells within blood and spleen were found between wk of life 15/16 and 23/24, covering the transition phase to egg-laying activity. Numbers of peripheral and splenic lymphocytes remained low during the egg-laying period or even further decreased, for example blood CD4+T cells and splenic γδ T cells. Functional assessments showed that in vitro IFN-γ production of mitogen-stimulated splenocytes was lower in wk 60. Taken together, egg-laying activity seems to alter the immune system toward a more pronounced humoral and innate immune response, with probable consequences for the immunocompetence and thus for productivity, health and welfare of the hens.

1We studied the effects of glycine on myenteric neurones and muscle activity in the colon and stomach of adult guinea‐pigs.2Intracellular recordings revealed that myenteric neurones responded to local microejection of glycine (1 mm) with a fast, transient membrane potential depolarisation (57 % of 191 colonic neurones and 26 % of 50 gastric neurones). Most glycine‐sensitive neurones had ascending projections and were choline acetyltransferase immunoreactive. Glycine preferentially activated neurones with a late afterhyperpolarisation (AH‐neurones) and tonic spiking neurones with fast synaptic inputs (tonic S‐neurones) but less frequently phasic S‐neurones and inexcitable (non‐spiking) neurones. The depolarisation had a reversal potential at −19 ± 13 mV, which was increased by 18 ± 10 % upon lowering extracellular chloride concentration and decreased by 38 ± 14 % in furosemide (frusemide, 2 mm).3Strychnine (300 nm) reversibly abolished the glycine‐induced depolarisation and the Cl−channel blocker picrotoxin (100 μm) reduced the amplitude of the depolarisation by 55 ± 5 %. The glycine effect was a postsynaptic response because it was not changed after nerve blockade with tetrodotoxin (1 μm) or blockade of synaptic transmission in reduced extracellular [Ca2+]. The effect was specific since the response was not changed by the nicotinic antagonists hexamethonium (200 μm) and mecamylamine (100 μm), the GABAAreceptor antagonist bicuculline (10 μm), the NMDA antagonist MK‐801 (20 μm) or the 5‐HT3antagonist ICS 205930 (1 μm).4Glycine (1 mm) induced a tetrodotoxin‐ and strychnine‐sensitive contractile response in the colon; the contractile response in the stomach was tetrodotoxin insensitive.5Glycine activated myenteric neurones in the adult enteric nervous system through strychnine‐sensitive mechanisms. The glycine‐evoked depolarisation was caused by Cl−efflux and the maintenance of relatively high intracellular chloride concentrations involved furosemide‐sensitive cation‐chloride co‐transporters.

Studies on P depleted rats compared with control animals kept under pair fed conditions were carried out. Dietary P depletion led to reduced weight gain and a decreased food conversion ratio in comparison with pair fed control animals. Higher N retention, higher urea concentrations in plasma, liver and kidney tissues and a significant reduction of renal glutamate dehydrogenase ‐ a central enzyme of amino acid degradation in kidney mitochondria ‐ in P depleted animals indicated changes in N utilization and N excretion. While lipid metabolism was not affected by P depletion, carbohydrate metabolism was substantially changed in the kidney: Activity of fructose diphosphatase was significantly reduced, implicating a reduced gluconeogenesis in P depletion. Possible mechanisms of these metabolic effects in P depletion are discussed.
