l-(+)-Carbafuranomycin is a novel analogue of l-(+)-furanomycin, an unusual antibiotic α-amino acid that attracted great interest due to its activity as an isoleucine antagonist. We present here a concise and efficient asymmetric synthesis of this carba-analogue starting with the 1,3-dipolar cycloaddition of a chiral nitrile oxide with cyclopentadiene. Notably, the methyl group was introduced by an SN2‘ cuprate substitution with high stereo- and regioselectivity.

Animal cells can be grown in different reactors. An Eppstein-Barr-Virus (EBV) transformed human B-cell line was cultivated for the antibody production in two different reactor systems: First, in a well mixed reactor equipped with a double membrane stirrer and an ultrafiltration module for antibody retention and second, in a hollow fibre cartridge reactor. Their performance in respect to antibody production was compared according to the antibody yield per liter medium consumed. The best yield was obtained in the well mixed reactor, where the micro-invironment of the cells can be considered as uniform for all cells.
