Abstract: Microquasars are laboratories for the study of jets of relativistic particles produced by accretion onto a spinning black hole. Microquasars are near enough to allow detailed imaging of spatial features across the multiwavelength spectrum. The recent extension measurement of the spatial morphology of a microquasar, SS 433, to TeV gamma rays1localizes the acceleration of electrons at shocks in the jet far from the black hole2. V4641 Sagittarii (V4641 Sgr) is a similar binary system with a black hole and B-type main-sequence companion star and has an orbit period of 2.8 days (refs. 3,4). It stands out for its super-Eddington accretion5and for its radio jet, which is one of the fastest superluminal jets in the Milky Way. Previous observations of V4641 Sgr did not report gamma-ray emission6. Here we report TeV gamma-ray emission from V4641 Sgr that reveals particle acceleration at similar distances from the black hole as SS 433. Furthermore, the gamma-ray spectrum of V4641 Sgr is among the hardest TeV spectra observed from any known gamma-ray source and is detected above 200 TeV. Gamma rays are produced by particles, either electrons or protons, of higher energies. Because energetic electrons lose energy more quickly the higher their energy, such a spectrum either very strongly constrains the electron-production mechanism or points to the acceleration of high-energy protons. This suggests that large-scale jets from microquasars could be more common than previously expected and that they could be a notable source of galactic cosmic rays7–9.

Abstract: SS 433 is a binary system containing a supergiant star that is overflowing its Roche lobe with matter accreting onto a compact object (either a black hole or neutron star)1–3. Two jets of ionized matter with a bulk velocity of approximately 0.26c(where cis the speed of light in vacuum) extend from the binary, perpendicular to the line of sight, and terminate inside W50, a supernova remnant that is being distorted by the jets2,4–8. SS 433 differs from other microquasars (small-scale versions of quasars that are present within our own Galaxy) in that the accretion is believed to be super-Eddington9–11, and the luminosity of the system is about 1040ergs per second2,9,12,13. The lobes of W50 in which the jets terminate, about 40 parsecs from the central source, are expected to accelerate charged particles, and indeed radio and X-ray emission consistent with electron synchrotron emission in a magnetic field have been observed14–16. At higher energies (greater than 100 gigaelectronvolts), the particle fluxes of ?-rays from X-ray hotspots around SS 433 have been reported as flux upper limits6,17–20. In this energy regime, it has been unclear whether the emission is dominated by electrons that are interacting with photons from the cosmic microwave background through inverse-Compton scattering or by protons that are interacting with the ambient gas. Here we report teraelectronvolt ?-ray observations of the SS 433/W50 system that spatially resolve the lobes. The teraelectronvolt emission is localized to structures in the lobes, far from the centre of the system where the jets are formed. We have measured photon energies of at least 25 teraelectronvolts, and these are certainly not Doppler-boosted, because of the viewing geometry. We conclude that the emission—from radio to teraelectronvolt energies—is consistent with a single population of electrons with energies extending to at least hundreds of teraelectronvolts in a magnetic field of about 16 microgauss.

Abstract: e-ASTROGAM (‘enhanced ASTROGAM’) is a breakthrough Observatory space mission, with a detector composed by a Silicon tracker, a calorimeter, and an anticoincidence system, dedicated to the study of the non-thermal Universe in the photon energy range from 0.3 MeV to 3 GeV – the lower energy limit can be pushed to energies as low as 150 keV for the tracker, and to 30 keV for calorimetric detection. The mission is based on an advanced space-proven detector technology, with unprecedented sensitivity, angular and energy resolution, combined with polarimetric capability. Thanks to its performance in the MeV–GeV domain, substantially improving its predecessors, e-ASTROGAM will open a new window on the non-thermal Universe, making pioneering observations of the most powerful Galactic and extragalactic sources, elucidating the nature of their relativistic outflows and their effects on the surroundings. With a line sensitivity in the MeV energy range one to two orders of magnitude better than previous generation instruments, e-ASTROGAM will determine the origin of key isotopes fundamental for the understanding of supernova explosion and the chemical evolution of our Galaxy. The mission will provide unique data of significant interest to a broad astronomical community, complementary to powerful observatories such as LIGO-Virgo-GEO600-KAGRA, SKA, ALMA, E-ELT, TMT, LSST, JWST, Athena, CTA, IceCube, KM3NeT, and LISA.

4. Ancient Pulsar Wind Nebulae as a natural explanation for unidentified gamma-ray sources

Abstract: A large part of the Galactic sources emitting very high energy (VHE; > 1011eV) gamma-rays are currently still unidentified. The evolution of Pulsar Wind Nebulae (PWNe) plays a crucial role in interpreting these sources. The time-dependent modeling of PWNe has been tested on a sample of well-known young and intermediate age PWNe; and it is currently applied to the full-sample of unidentified VHE Galactic sources. The consequences of this interpretation go far beyond the interpretation of “dark sources” (i.e. VHE gamma-ray sources without lower energies, radio or X-ray, counterparts): e.g. there could be strong implication in the origin of cosmic rays and (when considering a leptonic origin of the gamma-ray signal) they can be important for reinterpreting the detection of starburst galaxies in the TeV gamma-ray band. Moreover, the number of Galactic VHE sources is currently increasing with further observation by Imaging Atmospheric Cherenkov Telescopes (IACTs) and by the advent of more sensitive water Cherenkov telescopes such as HAWC (High-Altitude Water Cherenkov Observatory); therefore the physical interpretation of unidentified sources becomes more and more crucial.

5. The efficacy of a discontinuous graded exercise test in measuring peak oxygen uptake in children aged 8 to 10 years.

Abstract: As children's natural activity patterns are highly intermittent in nature, and characterised by rapid changes from rest to vigorous physical activity, discontinuous exercise tests may be considered ecologically valid for this population group. This study compared the peak physiological responses from a discontinuous and continuous graded exercise test (GXT_D, GXT_C, respectively) during treadmill exercise in children. Twentyone healthy children (9.6 ± 0.6 y) completed GXT_D and GXT_C in a randomised order, separated by 72-hours. Following each GXT, and after a 15-minute recovery, participants completed a verification test at 105% of the velocity attained at peak oxygen consumption (VO2peak). There were no differences in VO2peak (55.3 ± 8.2 cf. 54.4 ± 7.6 mL⋅kg-1#8901;min-1) or maximal heart rate (202 ± 10 cf. 204 ± 8 b#8901;min-1) between GXT_C and GXT_D, respectively (P>.05). Peak running speed (10.7 ± 0.9 cf. 12.1 ± 1.3 km#8901;h-1) and respiratory exchange ratio (1.04 ± 0.05 cf. 0.92 ± 0.05) were however different between tests (P<.001). Although similar peak physiological values were revealed between GXT_C and the corresponding verification test (P>.05), VO2peak (53.3 ± 7.3 mL#8901;kg-1#8901;min-1) and heart rate (197 ± 13 b#8901;min-1) were significantly lower in the GXT_D verification test (P<.05). In conclusion, a discontinuous GXT is an accurate measure of VO2peak in children aged 8 to 10 years and may be a valid alternative to a continuous GXT, despite its longer duration. 6. Histone deacetylase inhibitors interrupt HSP90•RASGRP1 and HSP90•CRAF interactions to upregulate BIM and circumvent drug resistance in lymphoma cells

Abstract: Histone deacetylase (HDAC) inhibitors, which are approved for the treatment of cutaneous T-cell lymphoma and multiple myeloma, are undergoing evaluation in other lymphoid neoplasms. How they kill susceptible cells is incompletely understood. Here, we show that trichostatin A, romidepsin and panobinostat induce apoptosis across a panel of malignant B cell lines, including lines that are intrinsically resistant to bortezomib, etoposide, cytarabine and BH3 mimetics. Further analysis traces the pro-apoptotic effects of HDAC inhibitors to increased acetylation of the chaperone heat shock protein 90 (HSP90), causing release and degradation of the HSP90 client proteins RASGRP1 and CRAF, which in turn leads to downregulation of mitogen-activated protein kinase pathway signaling and upregulation of the pro-apoptotic BCL2 family member BIM in vitroand in vivo. Importantly, these pro-apoptotic effects are mimicked by RASGRP1 small interfering RNA (siRNA) or HSP90 inhibition and reversed by overexpression of constitutively active MEK1 or siRNA-mediated downregulation of BIM. Collectively, these observations not only identify a new HSP90 client protein, RASGRP1, but also delineate a complete signaling pathway from HSP90 acetylation through RASGRP1 and CRAF degradation to BIM upregulation that contributes to selective cytotoxicity of HDAC inhibitors in lymphoid malignancies.

7. Depletion of ozone and reservoir species of chlorine and nitrogen oxide in the lower Antarctic polar vortex measured from aircraft

Abstract: Novel airborne in situ measurements of inorganic chlorine, nitrogen oxide species, and ozone were performed inside the lower Antarctic polar vortex and at its edge in September 2012. We focus on one flight during the Transport and Composition of the LMS/Earth System Model Validation (TACTS/ESMVal) campaign with the German research aircraft HALO (High‐Altitude LOng range research aircraft), reaching latitudes of 65°S and potential temperatures up to 405 K. Using the early winter correlations of reactive trace gases with N2O from the Atmospheric Chemistry Experiment‐Fourier Transform Spectrometer (ACE‐FTS), we find high depletion of chlorine reservoir gases up to ∼40% (0.8 ppbv) at 12 km to 14 km altitude in the vortex and 0.4 ppbv at the edge in subsided stratospheric air with mean ages up to 4.5 years. We observe denitrification of up to 4 ppbv, while ozone was depleted by 1.2 ppmv at potential temperatures as low as 380 K. The advanced instrumentation aboard HALO enables high‐resolution measurements with implications for the oxidation capacity of the lowermost stratosphere. Chemistry climate models reveal large uncertainties in the future ozone projection until the end of the century in the lower polar and midlatitude stratosphere. One process that impacts the ozone lifetime during the polar winter is the formation of active chlorine from chlorine reservoir species. Here we present high‐resolution measurements performed aboard the new German research aircraft HALO (High‐Altitude LOng range research aircraft) in the lower Antarctic vortex in winter 2012. We find significant amounts of active chlorine in the lower vortex that has been transported from higher altitudes and latitudes to the flight altitude of HALO (∼14 km). This enhanced activated chlorine content has implications on the ozone lifetime in this region. Our measurements complement satellite observations but feature higher‐altitude resolution and extend to lower altitudes. With our case study we investigate the intersection of midlatitude and polar air as well as the effects of transport from the upper to the lower polar vortex. These process studies help to improve chemistry climate models. Here we present an overview over the AGN research and results from an ongoing multi-band campaign on the active galaxy NGC1052. Beyond these studies, we address the latest technical developments and its impact in the AGN field: the Square Kilometre Array, a new radio interferometer planned for the next decade, and the oncoming x-ray and gamma ray missions. Abstract: The measurement of an excess in the cosmic-ray electron spectrum between 300 and 800 GeV by the ATIC experiment has – together with the PAMELA detection of a rise in the positron fraction up to ≈100 GeV – motivated many interpretations in terms of dark matter scenarios; alternative explanations assume a nearby electron source like a pulsar or supernova remnant. Here we present a measurement of the cosmic-ray electron spectrum with H.E.S.S. starting at 340 GeV. While the overall electron flux measured by H.E.S.S. is consistent with the ATIC data within statistical and systematic errors, the H.E.S.S. data exclude a pronounced peak in the electron spectrum as suggested for interpretation by ATIC. The H.E.S.S. data follow a power-law spectrum with spectral index of 3.0±0.1(stat.)±0.3(syst.), which steepens at about 1 TeV.

Abstract: Aims. This article presents very-high-energy (VHE; E> 100 GeV) data from the γ-ray binary PSR B1259-63 as taken during the years 2005, 2006 and before as well as shortly after the 2007 periastron passage. These data extend the knowledge of the lightcurve of this object to all phases of the 3.4 year binary orbit. The lightcurve constrains physical mechanisms present in this TeV source.

Abstract: Observations of the globular cluster 47 Tucanae (NGC 104), which contains at least 23 ms pulsars, were performed with the HESS telescope system. The observations lead to an upper limit of $F(E>800\ {\rm GeV}) < 6.7 \times 10^{-13}~{\rm cm^{-2}\,s^{-1}}$on the integral γ-ray photon flux from 47 Tucanae. Considering millisecond pulsars as the unique potential source of γ-rays in the globular cluster, constraints based on emission models are derived: on the magnetic field in the average pulsar nebula and on the conversion efficiency of spin-down power to γ-ray photons or to relativistic leptons.

Abstract: We discovered the >100 GeV γ-ray source, HESS J1713-381, apparently associated with the shell-type supernova remnant (SNR) CTB 37B, using HESS in 2006. In 2007 we performed X-ray follow-up observations with Chandrawith the aim of identifying a synchrotron counterpart to the TeV source and/or thermal emission from the SNR shell. These new Chandradata, together with additional TeV data, allow us to investigate the nature of this object in much greater detail than was previously possible. The new X-ray data reveal thermal emission from a ~4'region in close proximity to the radio shell of CTB 37B. The temperature of this emission implies an age for the remnant of ~5000 years and an ambient gas density of ~0.5 cm-3. Both these estimates are considerably uncertain due to the asymmetry of the SNR and possible modifications of the kinematics due to efficient cosmic ray (CR) acceleration. A bright (≈7 $\times$10-13erg cm-2s-1) and unresolved (<1″) source (CXOU J171405.7-381031), with a soft (Γ≈3.3) non-thermal spectrum is also detected in coincidence with the radio shell. Absorption indicates a column density consistent with the thermal emission from the shell, suggesting a genuine association rather than a chance alignment. The observed TeV morphology is consistent with an origin in the complete shell of CTB 37B. The lack of diffuse non-thermal X-ray emission suggests an origin of the γ-ray emission via the decay of neutral pions produced in interactions of protons and nuclei, rather than inverse Compton (IC) emission from relativistic electrons.

Abstract: Aims. The BL Lac object RGB J0152+017($z=0.080$) was predicted to be a very high-energy (VHE; >100 GeV) γ-ray source, due to its high X-ray and radio fluxes. Our aim is to understand the radiative processes by investigating the observed emission and its production mechanism using the High Energy Stereoscopic System (HESS) experiment.

Abstract: Acquired thrombotic thrombocytopenic purpura (TTP) is caused by an autoantibody‐mediated deficiency of the von Willebrand factor‐cleaving protease ADAMTS‐13. Acute episodes of the disease are treated with a combination of immunosuppression and repeated cycles of plasma exchange to remove anti‐ADAMTS‐13 autoantibodies and, at the same time, replenish functional ADAMTS‐13. Although this is often effective, the mortality rate has remained between 10% and 20%, highlighting the need for safer treatment options.

Abstract: With the aid of stimulus-responsive hydrogel substrates composed of ABA triblock copolymer micelles, we monitored the morphological dynamics of myoblast (C2C12) cells in response to an abrupt change in the substrate elasticity by live cell imaging. The remodeling of actin cytoskeletons could be monitored by means of transient transfection with LifeAct-GFP. Dynamic changes in the orientational order of actin filaments were characterized by an order parameter, which enables one to generalize the mechanically induced actin cytoskeletons as a break of symmetry. The critical role that acto-myosin complexes play in the morphological transition was verified by the treatment of cells with myosin II inhibitor (blebbistatin) and the fluorescence localization of focal adhesion contacts. Such dynamically tunable hydrogels can be utilized as in vitrocellular micro-environments that can exert time-dependent stimuli to mechanically regulate target cells.

23. Effect of a synthetic GnRF vaccine (Improvac®) on daily weight gain and carcass quality of boars. A field trial in Bavaria.

Abstract: Objective: The effects of vaccination against gonadotropin releasing factor (GnRF) with Improvac® (Pfizer Animal Health) were compared with surgical castration in fattening pigs. Material and methods: A total of 205 pigs were surgically castrated (group K) and 191 were vaccinated twice (group V) using the boar taint vaccine (Improvac®; 2ml s.c.). The first dose was administered atthe age of 12 weeks when the animals were moved into the fattening unit and the second dose in week 18, 4–6 weeks before the planned slaughter date. Live weights were recorded in weeks 1, 4, 12, 18, and 22. In weeks 18 and 20, length and width of the testicles of 171 animals of group V were measured. After slaughtering cold carcass weight, back fat depth, muscle thickness, percent lean meat, and fat and muscle areas of the carcasses were determined. A piece of the neck muscle from each pig was used to conduct a cooking and melting sensory test. Results: While no significant weight difference was evident in week 22 (K=89.4kg; V=88.6kg), cold carcass weight, and back fat and muscle thickness were lower for vaccinates. Vaccinates had higher average daily weight gains (ADW) after the second injection from week 18 up to the cut-off weighing in week 22 (V=1121g; K=1007g; p<0.001) in contrast to average daily weight gains between weeks 12 and 18 (K=740g; V=668g; p<0.001). After the second injection, testicle size of vaccinated pigs decreased significantly. All animals were negative for boar taint by both cooking and melting tests. Conclusion: Boars vaccinated against boar taint had lower ADW before the second vaccination, but compensated the weight difference after complete vaccination. The significant reduction in the testicle size after the second injection indicates a vaccination success. After vaccination no boar taint was detected in carcasses. Clinical relevance: Vaccination as well as surgical castration reliably prevents the incidence of boar taint. The late rise in daily gain can be beneficial if management is aligned.

Abstract: Aims. This article presents very-high-energy (VHE; E > 100?GeV) data from the ?-ray binary PSR?B1259-63 as taken during the years 2005, 2006 and before as well as shortly after the 2007 periastron passage. These data extend the knowledge of the lightcurve of this object to all phases of the 3.4?year binary orbit. The lightcurve constrains physical mechanisms present in this TeV source.Methods. Observations of VHE ?-rays with the HESS telescope array using the Imaging Atmospheric Cherenkov Technique were performed. The HESS?instrument features an angular resolution of < 0.1? and an energy resolution of < 20%. Gamma-ray events in an energy range of 0.5-70?TeV were recorded. From these data, energy spectra and lightcurve with a monthly time sampling were extracted.Results. VHE ?-ray emission from PSR?B1259-63?was detected with an overall significance of 9.5?standard deviations using 55h of exposure, obtained from April to August 2007. The monthly flux of ?-rays during the observation period was measured, yielding VHE lightcurve data for the early pre-periastron phase of the system for the first time. No spectral variability was found on timescales of months. The spectrum is described by a power law with a photon index of ? = 2.8 ?0.2stat?0.2sysand flux normalisation ?0= (1.1 ?0.1stat?0.2sys) ? 10-12?TeV-1?cm-2?s-1. PSR?B1259-63?was also monitored in 2005 and 2006, far from periastron passage, comprising 8.9?h and 7.5?h of exposure, respectively. No significant excess of ?-rays is seen in those observations.Conclusions. PSR B1259-63? has been re-confirmed as a variable TeV ?-ray emitter. The firm detection of VHE photons emitted at a true anomaly ??-0.35 of the pulsar orbit, i.e.?already ~50?days prior to the periastron passage, disfavors the stellar disc target scenario as a primary emission mechanism, based on current knowledge about the companion star's disc inclination, extension, and density profile.

Abstract: Changes in the expression of repellent factors, i.e., Netrins and their receptors, may be responsible for the invasive behavior of the synovial tissue cells in patients with rheumatoid arthritis (RA) and osteoarthritis (OA). This study was carried out to analyze the expression of Netrins and their receptors in synovial cells of patients with RA, OA, and control subjects without synovial inflammation. Quantitative RT-PCR was performed to measure the expression of Netrin-1, −3, −4, Neogenin, DCC, UNC5A-D. The influence of Netrin-1 on synovial fibroblasts (SF) was analyzed by determining proliferation, migration, and their ability to organize collagen. SF expressed all repellent factors of the Netrin family. When comparing SF of healthy donors to patients with RA and OA, a stronger expression of UNC5B (4 fold) and UNC5C (769 fold) in RA and OA was found, whereas expression of the other molecules revealed no significant differences. Treating the SF-cells with recombinant Netrin-1 resulted in inhibition of migration of RA- and OA-SFs whereas control cells were not affected. The stronger expression of UNC5B and UNC5C receptors might contribute to the disordered phenotype of RA- and OA-SFs. Addition of Netrin-1 reduces the migratory ability of SFs, potentially by repulsion, as seen in neuronal cells in embryonic development. Due to its function, Netrin-1 may constitute a novel target in the treatment of OA and RA.

Abstract: Aims. Very-high-energy (VHE; ?100?GeV) ?-rays are expected from ?-ray bursts (GRBs) in some scenarios. Exploring this photon energy regime is necessary for understanding the energetics and properties of GRBs. Methods. GRBs have been one of the prime targets for the HESS experiment, which makes use of four Imaging Atmospheric Cherenkov Telescopes (IACTs) to detect VHE ?-rays. Dedicated observations of 32?GRB positions were made in the years 2003?2007 and a search for VHE ?-ray counterparts of these GRBs was made. Depending on the visibility and observing conditions, the observations mostly start minutes to hours after the burst and typically last two hours.Results. Results from observations of 22?GRB positions are presented. Evidence of a VHE signal was found neither in observations of any individual GRBs nor from stacking data from subsets of GRBs with higher expected VHE flux according to a model-independent ranking scheme. Upper limits for the VHE??-ray flux from the GRB positions were derived. For those GRBs with measured redshifts, differential upper limits at the energy threshold after correcting for absorption due to extra-galactic background light are also presented.

Abstract: Aims. A detailed study of the spectrum and variability of the source HESS J1745-290 in the Galactic Center (GC) region using new data from the H.E.S.S. array of Cherenkov telescopes is presented. Flaring activity and quasi periodic oscillations (QPO) of HESS J1745-290 are investigated.Methods. The image analysis is performed with a combination of a semi-analytical shower model and the statistical moment-based Hillas technique. The spectrum and lightcurves of HESS J1745-290 are derived with a likelihood method based on a spectral shape hypothesis. Rayleigh tests and Fourier analysis of the H.E.S.S. GC signal are used to study the periodicity of the source.Results. With a three-fold increase in statistics compared to previous work, a deviation from a simple power law spectrum is detected for the first time. The measured energy spectrum over the three years 2004, 2005 and 2006 of data taking is compatible with both a power law spectrum with an exponential cut-off and a broken power law spectrum. The curvature of the energy spectrum is likely to be intrinsic to the photon source, as opposed to effects of interstellar absorption. The power law spectrum with an exponential cut-off is characterized by a photon index of 2.10 ? 0.04stat? 0.10systand a cut-off energy at 15.7 ? 3.4stat? 2.5systTeV. The broken power law spectrum exhibits spectral indices of 2.02 ? 0.08stat? 0.10systand 2.63 ? 0.14stat? 0.10systwith a break energy at 2.57 ? 0.19stat? 0.44systTeV. No significant flux variation is found. Increases in the ?-ray flux of HESS J1745-290 by at least a factor of two would be required for a 3? detection of a flare with time scales of an hour. Investigation of possible QPO activity at periods claimed to be detected in X-rays does not show any periodicities in the H.E.S.S. signal.

Abstract: The rapidly varying (~10?min timescale) non-thermal X-ray emission observed from Sgr A*implies that particle acceleration is occuring close to the event horizon of the supermassive black hole. The TeV ?-ray source HESS?J1745-290 is coincident with Sgr?A*and may be closely related to its X-ray emission. Simultaneous X-ray and TeV observations are required to elucidate the relationship between these objects. We report on joint HESS/Chandra observations performed in July?2005, during which an X-ray flare was detected. Despite a factor of ?9 increase in the X-ray flux of Sgr?A*, no evidence is found for an increase in the TeV ?-ray flux from this region. We find that an increase in the ?-ray flux of a factor of?2 or greater can be excluded at a confidence level of?99%. This finding disfavours scenarios in which the keV and TeV emission are associated with the same population of accelerated particles and in which the bulk of the ?-ray emission is produced within ~1014?cm (~100RS) of the supermassive black hole.

Abstract: Aims. The supernova remnant (SNR) complex CTB?37 is an interesting candidate for observations with very high energy (VHE) ?-ray telescopes such as HESS. In this region, three SNRs are seen. One of them is potentially associated with several molecular clouds, a circumstance that can be used to probe the acceleration of hadronic cosmic rays.Methods. This region was observed with the HESS Cherenkov telescopes and the data were analyzed with standard HESS procedures. Recent X-ray observations with Chandra and XMM-Newton were used to search for X-ray counterparts.Results. The discovery of a new VHE ?-ray source HESS?J1714-385 coincident with the remnant CTB?37A is reported. The energy spectrum is well described by a power-law with a photon index of ? = 2.30 ? 0.13 and a differential flux at 1?TeV of ?0= (8.7 ?1.0stat?1.8sys) ?10-13cm-2?s-1?TeV-1. The integrated flux above 1?TeV is equivalent to?3% of the flux of the Crab nebula above the same energy. This VHE ?-ray source is a counterpart candidate for the unidentified EGRET source 3EG?J1714-3857. The observed VHE?emission is consistent with the molecular gas distribution around CTB?37A; a close match is expected in a hadronic scenario for ?-ray production. The X-ray observations reveal the presence of thermal X-rays from the NE part of the?SNR. In the NW?part of the remnant, an extended non-thermal X-ray source, CXOU?J171419.8-383023, is discovered as well. Possible connections of the X-ray emission to the newly found VHE?source are discussed.

Abstract: Aims. The BL?Lac object RGB?J0152+017?(z=0.080) was predicted to be a very high-energy (VHE; >100?GeV) ?-ray source, due to its high X-ray and radio fluxes. Our aim is to understand the radiative processes by investigating the observed emission and its production mechanism using the High Energy Stereoscopic System (HESS) experiment. Methods. We report recent observations of the BL?Lac source RGB?J0152+017?made in late October and November 2007 with the HESS array consisting of four imaging atmospheric Cherenkov telescopes. Contemporaneous observations were made in X-rays by the Swift and RXTE satellites, in the optical band with the ATOM telescope, and in the radio band with the Nan?ay Radio Telescope. Results. A signal of 173 ?-ray photons corresponding to a statistical significance of 6.6? was found in the data. The energy spectrum of the source can be described by a powerlaw with a spectral index of ?=2.95?0.36stat?0.20syst. The integral flux above 300?GeV corresponds to ~2% of the flux of the Crab nebula. The source spectral energy distribution (SED) can be described using a two-component non-thermal synchrotron self-Compton (SSC) leptonic model, except in the optical band, which is dominated by a thermal host galaxy component. The parameters that are found are very close to those found in similar SSC studies in TeV blazars. Conclusions. RGB?J0152+017?is discovered as a source of VHE ?-rays by HESS The location of its synchrotron peak, as derived from the SED in Swift data, allows clear classification as a high-frequency-peaked BL?Lac (HBL).

Abstract: Abstract:: In cell culture, liquid crystal analogues are formed by elongated, migrating, and interacting amoeboid cells. An apolar nematic liquid crystal analogue is formed by different cell types like human melanocytes (=pigment cells of the skin), human fibroblasts (=connective tissue cells), human osteoblasts (=bone cells), human adipocytes (=fat cells), etc. The nematic analogue is quite well described by i) a stochastic machine equation responsible for cell orientation and ii) a self-organized extracellular guiding signal, E2, which is proportional to the orientational order parameter as well as to the cell density. The investigations were mainly made with melanocytes. The transition to an isotropic state analogue can be accomplished either by changing the strength of interaction (e.g. variation of the cell density) or by influencing the cellular machinery by an externally applied signal: i) An isotropic gaseous state analogue is observed at low cell density (melanocytes/mm^2) and a nematic liquid crystal state analogue at higher cell density. ii) The nematic state analogue disappears if the bipolar shaped melanocytes are forced to become a star-like shape (induced by colchicine or staurosporine). The analogy between nematic liquid crystal state analogue formed by elongated, migrating and interacting cells and the nematic liquid crystal phase formed by interacting elongated molecules is discussed.

Abstract: Numbers of gamma interferon (IFN-gamma)-producing cells reactive to ESAT-6 antigen were increased in recent converters to purified protein derivative positivity and in tuberculosis patients but not in unvaccinated or Mycobacterium bovis BCG-vaccinated healthy donors. ESAT-6-reactive IFN-gamma-producing cells in recent converters and tuberculosis patients recognized similar synthetic peptides. Thus, ESAT-6 is a potential candidate for use in detection of early, as well as active, tuberculosis and for control of the disease.

38. Retention of the human Rad9 checkpoint complex in extraction-resistant nuclear complexes after DNA damage.

Abstract: Studies in yeasts and mammals have identified many genes important for DNA damage-induced checkpoint activation, including Rad9, Hus1, and Rad1; however, the functions of these gene products are unknown. In this study we show by immunolocalization that human Rad9 (hRad9) is localized exclusively in the nucleus. However, hRad9 was readily released from the nucleus into the soluble extract upon biochemical fractionation of un-irradiated cells. In contrast, DNA damage promptly converted hRad9 to an extraction-resistant form that was retained at discrete sites within the nucleus. Conversion of hRad9 to the extraction-resistant nuclear form occurred in response to diverse DNA-damaging agents and the replication inhibitor hydroxyurea but not other cytotoxic stimuli. Additionally, extraction-resistant hRad9 interacted with its binding partners, hHus1 and an inducibly phosphorylated form of hRad1. Thus, these studies demonstrate that hRad9 is a nuclear protein that becomes more firmly anchored to nuclear components after DNA damage, consistent with a proximal function in DNA damage-activated checkpoint signaling pathways.

39. Effect of 6-aminonicotinamide and other protein synthesis inhibitors on formation of platinum-DNA adducts and cisplatin sensitivity.

Abstract: The present study was undertaken to examine the mechanistic basis for the recent observation that the pyridine nucleotide derivative 6-aminonicotinamide (6AN, NSC 21206) enhances the accumulation and resulting cytotoxicity of cisplatin in a variety of tumor cell lines. When A549 lung cancer cells or K562 leukemia cells were treated with 62.5 microM 6AN for 21 h and then pulse-labeled with [(35)S]methionine for 1 h, increased labeling of five polypeptides, one of which corresponded to a M(r) approximately 78,000 glucose-regulated protein (GRP78), was observed. Two subsequent observations, however, suggested that up-regulation of these polypeptides was unlikely to explain the interaction between 6AN and cisplatin: 1) the concentration of 6AN required to induce GRP78 was 4-fold higher than the dose required to sensitize cells to cisplatin; and 2) simultaneous treatment of cells with 6AN and cycloheximide prevented the increase in GRP78 but not the sensitizing effect of 6AN. On the contrary, treatment with the protein synthesis inhibitors cycloheximide, anisomycin, or puromycin as well as prolonged exposure to the RNA synthesis inhibitor actinomycin D mimicked the biochemical modulating effects of 6AN on cisplatin action. Conversely, 6AN inhibited protein synthesis, whereas 18 6AN analogs that failed to enhance Pt-DNA adducts and cisplatin cytotoxicity failed to inhibit protein synthesis. These observations are consistent with a model in which 6AN and other inhibitors of protein synthesis act as modulating agents by increasing cisplatin accumulation, thereby enhancing the formation of Pt-DNA adducts and subsequent cisplatin-induced cell death.

40. Successful Virtual Screening of a Chemical Database for Farnesyltransferase Inhibitor Leads

Abstract: Virtual screening of chemical databases is an emerging approach in drug discovery that uses computers to dock chemicals into the active site of a drug target to identify leads through evaluation of binding affinities of the chemicals. However, there are concerns about the validity and scope of the reported virtual screens due to lack of studies to show that randomly selected chemicals are not equally active and due to the fact that metalloproteins were rarely used as drug targets. We have performed a virtual screening of a chemical database to identify prototypic inhibitors of farnesyltransferase (FT) with zinc present in the active site. Among the 21 compounds identified by computers, four inhibited FT in vitro with IC50 values in the range from 25 to 100 μM. The most potent inhibitor also inhibited FT in human lung cancer cells. In contrast, none of 21 randomly selected compounds have an IC50 lower than 100 μM. The results demonstrate the validity of virtual screening and the feasibility of applications of this approach to metalloprotein drug targets, such as matrix metalloproteinases, farnesyltransferase, and HIV-1 integrase, for the treatments of cardiovascular diseases, cancers, and AIDS.

48. Comparison of paclitaxel-, 5-fluoro-2'-deoxyuridine-, and epidermal growth factor (EGF)-induced apoptosis. Evidence for EGF-induced anoikis.

Abstract: Epidermal growth factor (EGF), a hormone that stimulates proliferation of many cell types, induces apoptosis in some cell lines that overexpress the EGF receptor. To evaluate the mechanism of EGF-induced apoptosis, MDA-MB-468 breast cancer cells were examined by microscopy, flow cytometry, immunoblotting, enzyme assays, and affinity labeling after treatment with EGF, paclitaxel, or 5-fluoro-2'-deoxyuridine (5FUdR). Apoptosis induced by all three agents was accompanied by activation of caspases-3, -6, and -7, as indicated by disappearance of the corresponding zymogens from immunoblots, cleavage of substrate polypeptides in situ, and detection of active forms of these caspases in cytosol and nuclei using fluorogenic assays and affinity labeling. Further analysis indicated involvement of the cytochrome c/Apaf-1/caspase-9 pathway of caspase activation, but not the Fas/Fas ligand pathway. Interestingly, caspase activation was consistently lower after EGF treatment than after paclitaxel or 5FUdR treatment. Additional experiments revealed that the majority of cells detaching from the substratum after EGF (but not paclitaxel or 5FUdR) were morphologically normal and retained the capacity to readhere, suggesting that EGF-induced apoptosis involves cell detachment followed by anoikis. These observations not only indicate that EGF- and chemotherapy-induced apoptosis in this cell line involve the same downstream pathways but also suggest that detachment-induced apoptosis is responsible for the paradoxical antiproliferative effects of EGF.

49. Characterization of caspase processing and activation in HL-60 cell cytosol under cell-free conditions. Nucleotide requirement and inhibitor profile.

Abstract: The present studies compared caspase activation under cell-free conditions in vitro and in etoposide-treated HL-60 leukemia cells in situ. Immunoblotting revealed that incubation of HL-60 cytosol at 30 degrees C in the presence of cytochrome c and ATP (or dATP) resulted in activation of procaspases-3, -6, and -7 but not -2 and -8. Although similar selectivity was observed in intact cells, affinity labeling revealed that the active caspase species generated in vitro and in situ differed in charge and abundance. ATP and dATP levels in intact HL-60 cells were higher than required for caspase activation in vitro and did not change before caspase activation in situ. Replacement of ATP with the poorly hydrolyzable analogs 5'-adenylyl methylenediphosphate, 5'-adenylyl imidodiphosphate, or 5'-adenylyl-O-(3-thiotriphos-phate) slowed caspase activation in vitro, suggesting that ATP hydrolysis is required. Caspase activation in vitro was insensitive to phosphatase and kinase inhibitors (okadaic acid, staurosporine, and genistein) but was inhibited by Zn(2+), aurintricarboxylic acid, and various protease inhibitors, including 3,4-dichloroisocoumarin, N(alpha)-p-tosyl-L-phenylalanine chloromethyl ketone, N(alpha)-p-tosyl-L-lysine chloromethyl ketone, and N-(N(alpha)-benzyloxycarbonylphenylalanyl)alanine fluoromethyl ketone, each of which inhibited recombinant caspases-3, -6, -7, and -9. Experiments with anti-neoepitope antiserum confirmed that these agents inhibited caspase-9 activation. Collectively, these results suggest that caspase-9 activation requires nucleotide hydrolysis and is inhibited by agents previously thought to affect apoptosis by other means.

50. Cell-mediated immunity to mycobacteria: a double-sided sword?

Abstract: Mycobacteria are intracellular pathogens capable of replicating in resting macrophages. Specific helper T lymphocytes which activate antimycobacterial capacities in infected macrophages represent an important constituent of acquired resistance. In addition, cytolytic T lymphocytes may contribute to resistance. On the other hand, lysis of infected host cells may also comprise autoaggressive consequences. Recent evidence suggest that T cells with specificity for mycobacterial heat shock proteins are involved in the antimycobacterial immune response. Heat shock proteins are evolutionarily highly conserved and cross-reactivity between microbial and mammalian molecules may occur on the B-cell and T-cell level. Thus, T cells directed against shared epitopes of mycobacterial and autologous origin could initiate autoimmune reactions.
