While many watershed models estimate overland flow-related soil loss, streambank erosion is often overlooked even though it can be the dominant source of sediment in a catchment. We used the enhanced generalized watershed loading functions (GWLF-E) model to simulate the water budget, field erosion from the landscape, and streambank erosion along a gradient of agricultural to urban land cover from 1997 to 2015 in the Indian Mill Creek watershed of Michigan, USA. We evaluated discharge simulations using in-streamflow measurements and evaluated streambank erosion simulations using field-collected erosion pin measurements from a previous study. Annual water budget results suggest the creek is primarily groundwater fed, but that a per-subbasin average of 6–15% of precipitation becomes runoff. Field erosion contributed a per-subbasin average of 0.5–2.5 Mg ha−1year−1of sediment, while streambank erosion accounted for 0.2–50.1% of the subbasins’ total sediment yields. Average lateral erosion rate of streambanks in subbasins ranged from 0.04 to 7.37 cm year−1, with four subbasins exceeding 1.0 cm year−1. Evaluation with field-collected discharge data suggest that GWLF-E may have been overestimating discharge in the subbasins. Evaluation with streambank erosion data found that GWLF-E may have difficulty capturing the complexity of erosion rates, including instances where sediment is deposited on streambanks from upstream sources, and that it underestimates streambank erosion in headwater catchments sometimes over an order of magnitude. Our findings are important for watershed managers to evaluate modeling approaches and highlight the importance of modeling both field and streambank erosion.

The successful use of remote site incubators (RSIs) to rear eggs of Arctic Grayling Thymallus arcticusalong Montana streams has sparked interest in reestablishing the species in Michigan. As a preparatory step, we assessed the efficacy of RSIs by deploying them along three Michigan streams during 2 years using surrogate eggs from Rainbow Trout Oncorhynchus mykiss. Our objectives were to (1) compare hatching success between two different RSI designs (19‐L versus 265‐L RSIs), (2) test whether the removal of dead eggs (“picking”) from 19‐L RSIs affected hatching success, and (3) develop a simple model to predict fry yield and its uncertainty. Overall survival was 41.3% in 2018 and 52.4% in 2019. Differences in survival between unpicked 19‐L and 265‐L RSIs tended to be small, with mean differences from 4.82% (95% CI = –0.60 to +10.25) in 2018 to 0.08% (95% CI = –0.14 to +0.30) in 2019. On average, picked 19‐L RSIs had greater, although not always statistically significant, survival than unpicked 19‐L RSIs during both years (mean difference = 1.6% [2018] and 10.4% [2019]). We documented a significant positive correlation between survival and RSI flow rate. Survival abruptly declined in unpicked 19‐L RSIs when RSI flow rates dropped below ~0.3 L/min, suggesting that removing dead eggs from 19‐L RSIs likely increased survival when RSI flow rates were <0.3 L/min. The most notable result from our fry yield model was that increasing the number of RSIs reduced the coefficient of variation in fry yield following a pattern of diminishing returns, suggesting two or three RSIs usually will be a good choice. We showed that 19‐L and 265‐L RSIs can be used successfully in Michigan streams, with our model providing a tool for managers to explore the relative importance of several properties of RSI design and operation on fry yield and uncertainty.

ABSTRACTA novel lateral flow immunochromatographic device (LFD) was evaluated in several veterinary diagnostic laboratories. It was confirmed to be specific for Mycobacterium bovisand M.capraecells. The performance of the novel LFD was assessed relative to the confirmatory tests routinely applied after culture (spoligotyping or quantitative PCR [qPCR]) in each laboratory; liquid (MGIT or BacT/Alert) and/or solid (Stonebrink, Coletsos, or Lowenstein-Jensen) cultures were tested. In comparison to spoligotyping of acid-fast-positive MGIT cultures, percent agreement between positive LFD and spoligotyping results was excellent in two United Kingdom laboratories (97.7 to 100%) but lower in the Spanish context (76%), where spoligotyping was applied to MGIT cultures previously confirmed to be positive for M. tuberculosiscomplex (MTBC) by qPCR. Certain spoligotypes of M. bovisand M. capraewere not detected by the LFD in Spanish MGIT cultures. Compared to qPCR confirmation, the agreement between positive LFD and qPCR results was 42.3% and 50% for BacT/Alert and MGIT liquid cultures, respectively, and for solid cultures, it ranged from 11.1 to 89.2%, depending on the solid medium employed (Coletsos, 11.1%; Lowenstein-Jensen, 55.6%; Stonebrinks, 89.2%). Correlation between the novel LFD and BD MGIT TBc Identification test results was excellent when 190 MGIT cultures were tested (r= 0.9791; P< 0.0001), with the added benefit that M. boviswas differentiated from another MTBC species in one MGIT culture by the novel LFD. This multilaboratory evaluation demonstrated the novel LFD's potential utility as a rapid test to confirm isolation of M. bovisand M. capraefrom veterinary specimens following culture.

Lake Reserve Manage. 32:366–372.Invasive plants are a major concern for environmental managers around the world. Cryptic invasive taxa present additional challenges because of their potential to respond differently to management efforts while being difficult to distinguish morphologically. Here, we consider the potential for different responses of Eurasian watermilfoil (Myriophyllum spicatum) and hybrid watermilfoil (Myriophyllum spicatumx Myriophyllum sibiricum), which cannot be reliably distinguished based on morphological characters. Laboratory studies show that hybrid watermilfoil genotypes can grow faster, branch more, and be less responsive to herbicide control methods designed for Eurasian watermilfoil. To date, however, there have been no statistical comparisons of the responses of hybrid versus Eurasian watermilfoil under operational management in the field. In this study, we use genetic methods to distinguish morphologically cryptic Eurasian and hybrid watermilfoil in Houghton Lake, Michigan, and test whether they exhibit divergent responses for distribution and abundance following treatment with 2,4-dichlorophenoxyacetic acid-amine and triclopyr. Our results suggest that treatment reduced the frequency of occurrence and abundance of pure Eurasian watermilfoil more than it did for hybrids in Houghton Lake. These findings highlight the importance of distinguishing these cryptic taxa during management evaluations.

Most studies that investigate biases in stream fish abundance estimators focus on salmonines, yet nongame species comprise a major portion of fish assemblages. We evaluated mark–recapture (the Lincoln–Petersen estimator with Chapman correction) and removal (constant capture probability and generalized removal estimators) methods for estimating abundance of Mottled Sculpin Cottus bairdii, and we tested whether assumptions of the abundance estimators (i.e., the population is closed during sampling; and marks are detected) were reasonable. Over 2-d periods, fish in eight streams were sampled by using backpack electrofishing in 90-m reaches (each divided into three 30-m subreaches) to assess movement. Removal abundance estimates were significantly lower than mark–recapture estimates. Moreover, removal estimates were 52% lower than the known number of marked individuals, likely because capture probability (q) was low and declined with subsequent sampling passes. Survival of fish held in cages was 100%; there were no undetected marks among fish held overnight. For all streams combined, 11% of recaptured fish that were marked in the core subreach were recaptured on day 2 in an adjoining subreach. In five of the eight study streams, movement of marked fish from the core subreach was not detected, suggesting that in most streams the population was closed to movement. Using computer simulations, we found that low numbers of recaptures (mean < 6) negatively biased the mark–recapture estimator; however, the estimator was positively biased when the study population was open to movement—meaning that marked and unmarked fish were lost from the study reach as new, unmarked fish entered. Our results indicate that the removal estimator was negatively biased. We recommend using the mark–recapture method to estimate abundance when qis low (e.g., 0.21 in our study), with the caveat that study reach length must be sufficient to minimize fish movement across reach boundaries when block nets cannot be maintained between sampling events. The free-water dissolved-oxygen (FWDO) method employs high-frequency time series of dissolved oxygen (DO) and several other variables to estimate gross primary production (GPP), total respiration (R), and net production (NP) of a lake. It requires a process-based model of DO dynamics to represent the roles of oxygenic photosynthesis, aerobic respiration, and atmospheric exchange in determining observed DO concentrations, as well as a procedure for estimating parameter values. We review approaches that have been used in conjunction with the FWDO method for modeling DO dynamics in lakes and estimating model parameters. Most studies have used mixed-layer models of DO dynamics that assume stratified lakes have a well-mixed upper layer (mixed layer) whose DO concentration is not affected by deeper water, which therefore is excluded from the models. We have found that mixed-layer models often cannot adequately account for observed diel dynamics of DO in our study system (Muskegon Lake, USA), indicating that model improvement is necessary. We outline various alternative forms of mixed-layer DO models that are compatible with the FWDO method (including several new ones), review methods for estimating GPP, R, and NP with them, and illustrate their performance in Muskegon Lake. None of the alternative forms resolve the problem of frequent poor model fits. It therefore appears that an important process is missing from mixed-layer DO models. We suggest the main problem is that the mixed-layer assumption is too simple to adequately represent the key physical transport and mixing processes in Muskegon Lake on most days, and we argue that this problem is likely to arise in other lakes, as well. We conclude by discussing several alternatives to mixed-layer models that address the entire water column, are straightforward to implement, and retain compatibility with the FWDO method.

Natural rates of straying are difficult to quantify over large spatial scales using direct observations, particularly for long-lived fish species characterized by delayed sexual maturity and long interspawning intervals. Using multilocus microsatellite genotypes and likelihood-based statistical methods, we quantified rates of immigration and emigration for six genetically differentiated (mean FST= 0.041) lake sturgeon Acipenser fulvescenspopulations in Lake Michigan based on adults (n= 437) captured in tributaries during the spawning season. Estimated rates of straying were high (mean = 0.105), asymmetrical, and highly variable across populations. We found no significant association between the total length (a surrogate measure of age) of individuals that strayed and those that did not. Linear distance between streams was more predictive of straying rates and FSTthan least-cost distances estimated based on lakescape features (bathymetry and lake current patterns). Historical rates of gene flow estimated using coalescent analysis indicated a fully parameterized model with variable evolutionarily effective population sizes (? range, 0.684–0.989), and variable and nonsymmetrical migration rates best explained the genetic data. Comparatively high estimates of relative historical gene flow from several numerically depressed populations suggest that these populations were once larger contributors to basinwide gene flow than indicated by estimates of contemporary straying rates. High rates of interpopulation straying contrast with high FST, suggesting that straying rates are poor indicators of successful reproduction following dispersal.

ABSTRACTThis study describes the development and optimization of an immunomagnetic separation (IMS) method to isolate Mycobacterium boviscells from lymph node tissues. Gamma-irradiated whole M. bovisAF2122/97 cells and ethanol-extracted surface antigens of such cells were used to produce M. bovis-specific polyclonal and monoclonal antibodies in rabbits and mice. They were also used to generate M. bovis-specific peptide ligands by phage display biopanning. The various antibodies and peptide ligands obtained were used to coat MyOne tosyl-activated Dynabeads (Life Technologies), singly or in combination, and evaluated for IMS. Initially, M. boviscapture from Middlebrook 7H9 broth suspensions (concentration range, 10 to 105CFU/ml) was evaluated by IMS combined with an M. bovis-specific touchdown PCR. IMS-PCR results and, subsequently, IMS-culture results indicated that the beads with greatest immunocapture capability for M. bovisin broth were those coated simultaneously with a monoclonal antibody and a biotinylated 12-mer peptide. These dually coated beads exhibited minimal capture (mean of 0.36% recovery) of 12 other Mycobacteriumspp. occasionally encountered in veterinary tuberculosis (TB) diagnostic laboratories. When the optimized IMS method was applied to various M. bovis-spiked lymph node matrices, it demonstrated excellent detection sensitivities (50% limits of detection of 3.16 and 57.7 CFU/ml of lymph node tissue homogenate for IMS-PCR and IMS-culture, respectively). The optimized IMS method therefore has the potential to improve isolation of M. bovisfrom lymph nodes and hence the diagnosis of bovine tuberculosis.

ABSTRACTThe tuberculin skin test has been used for the diagnosis of bovine and human tuberculosis (TB) for over a hundred years. However, the specificity of the test is compromised by vaccination with the Mycobacterium bovis-derived vaccine strain bacille Calmette-Gue´rin (BCG). Since current promising vaccines against bovine TB are based on heterologous prime-boost combinations that include BCG, there is a need for diagnostic tests for differentiating infected from vaccinated animals (DIVA). The application of antigens such as ESAT-6 and CFP-10 for DIVA has so far been realized largely through their application in the blood-based gamma interferon release assay. In the current study, we have reassessed the potential of such antigens as skin test reagents for DIVA in cattle. A cocktail of the Mycobacterium tuberculosiscomplex recombinant protein antigens ESAT-6, CFP-10, MPB70, and MPB83 elicited delayed-type hypersensitivity (DTH) skin test responses in 78% of naturally infected tuberculin-positive cattle. Importantly, this cocktail induced no skin responses in BCG-vaccinated cattle despite them being sensitized for strong tuberculin responses. Further optimization of skin test antigen combinations identified that the inclusion of Rv3615c (Mb3645c) enhanced skin test sensitivity in naturally infected cattle without compromising specificity. In addition, we demonstrate for the first time the utility of synthetic peptides as promising skin test antigens for bovine TB for DIVA. Our data provide a promising basis for the future development of skin tests for DIVA with practical relevance for TB diagnosis in both veterinary and clinical settings.
