2 mg of 2-Hydroxystilbamidine isethionate when dissolved in 50 ml 0.1 M citric acid produced nuclear fluorescence in paraffin sections. Pre-hydrolysis in 5N HCl at room temperature increased selectivity of nuclear fluorescence. The addition of 100–200 mg sodium metabisulphite to the fluorochrome solution and preoxidation in periodic acid produced selective fluorescence of mucosubstances. Pre-oxidation with potassium permanganate induced selective fluorescence of elastic fibres. Yellow nuclear fluorescence contrasted clearly with blue/white fluorescence of mucosubstances and elastic fibres when excited with UV light. Unwanted nuclear fluorescence was quenched with 5% iron alum solution. Mast cells selectively fluoresced in acid alcoholic solutions of the fluorochrome. The procedures described were simple and rapid and produced permanent fluorescent preparations. The metachromatic fluorescence of nuclei in contrast to that of mucosubstances and elastic fibres eliminated the need for counterstaining.

The results of a survey of the major pathological conditions encountered in an established breeding colony of common cotton-eared marmosets (Callithrix jacchus) is presented. 265 home-bred and 70 imported wild-caught marmosets were examined. A Heinz body haemolytic anaemia and skeletal muscle myopathy were the most common pathological findings and were considered to be a result of a complex nutritional deficiency involving vitamin E, selenium and protein. Inflammatory disease of the intestinal tract was also a major feature. Chronic colitis was particularly common in older marmosets.Pneumonia, otitis media, meningitis and brain abscesses were important pathological findings in home-bred marmosets and were commonly associated with bacterial infections, particularly Bordetella bronchisepticaand Klebsiella species. Trichospirura leptostomawithin pancreatic ducts of wild-caught marmosets was the only significant parasitic disease encountered. Mycotic infections of the upper alimentary tract with Candidaspecies were occasional findings in debilitated animals. No pathological features suggesting viral diseases were found.

The compositions of fifty-nine common histological dyes, as well as duplicate samples of several dyes from different suppliers, have been studied by agar gel electrophoresis, agarose gel electrophoresis, paper electrophoresis, paper chromatography and thin layer chromatography. Tables are presented to show the number of components present in each dye as disclosed by the different methods; the cases where duplicate samples were available are summarised in a separate table.

Specific staining of glycogen in rat liver fixed in chilled 80% alcohol, chilled formol alcohol or 10% neutral formalin has been accomplished with acid alizarin blue SWR, alizarin brilliant blue BS, alizarin red S, gallein, haematein, and haematoxylin solutions. TO prepare a staining solution, 1 gm dye, 1 gm K2CO3 and 5 gm KCl were dissolved by heating in 60 ml of water. Concentrated NH4OH (0.880 sp.gr.), 15 ml, followed by 15 ml of dry methanol were added to 20 ml of the cooled solution. Paraffi sections were stained for 5 min, rinsed in dry methanol, cleared in xylene, and mounted in D.P.X. The high specificity obviated the need for counterstaining: nuclei and cytoplasm were unstained. Precipitation of stain onto the slide was rare. As all the dyes carried, like carminic acid, numerous groups capable of forming hydrogen bonds, it is suggested that the staining mechanism involved hydrogen bonding.

A detailed comparison of fixatives used for the demonstration of glycogen has been based on chemical assay and microspectrophotometry. Rat liver containing known amounts of glycogen was fixed in formol alcohol, Rossman's fluid, 10% neutral formalin, Bouin, Helly, SUSA, and Zenker's fluid at 4 C and 18 C. Chemical assay was carried out before and after fixation and paraffin sections were prepared from the fixed material. Sections were stained with PAS and the silver methenamine method. Visual examination was carried out with a comparison microscope and quantitative estimations on PAS-stained sections were performed by scanning microspectrophotometry. The histochemical methods were compared with the chemical results obtained from the same tissue and a reasonable degree of correlation between the sets of results was observed. Cold formol alcohol and cold Rossman's fluid preserved the most glycogen and Zenker and SUSA fixation preserved the least. Cold formol alcohol was the only fixative that preserved threshold values of glycogen i.e. 0.3% and the silver methenamine method is recommended for the demonstration of these small amounts.

The purity of 27 commercial pyronin and rhodamine samples was studied by thin-layer chromatography and visible spectroscopy. Seven different red dyes were detected and separated. The chemical identities of 6 of these were established by nuclear magnetic resonance spectroscopy. The identities of samples sold as rhodamine B and rhodamine 6G were as labelled 8 out of 9 times, the pyronin (G)Y samples were as labelled 5 out of 8 times and the 10 samples sold as acridine red, pyronin B, rhodamine 36 and rhodamine S were always incorrectly labelled. The dextrin and salt contents of the dyes were determined by solvent extraction of the dye with dry methanol or ethanol. Amounts of dextrin and salt varied from none to nearly 90%. Practical methods for identification, separation of coloured components and removal of dextrin and salt are given.

The composition of 5 common Gallocyanin-chrome alum (GCA) preparations were studied by thin layer chromatography, electrophoresis and spectrophotometry. The GCA preparations were found to be mixtures of one or more Gallocyanin-chromium co-ordination complexes, chromic ions, and, usually, free Gallocyanin. The differences in composition of the various preparations were due to differences in the preparative boiling times. The differences in histological staining properties depended on the concentrations of free Gallocyanin. The staining action of the GCA mixtures was similar to that of a typical basic dye such as Pyronin Y, both with regard to the materials stained and the effects of varying the pH and salt content of the dyebath. The chemistry of the commonest GCA co-ordination complex was investigated. It was found to have the composition 2∶1 Gallocyanin: chromium (G2Cr). The chromic ion was chelated to the aminocarboxylic acid. The complex carried a net positive charge in the pH range 1–9, and would thus be expected to behave similarly to basic dyes. In fact GCA was no more specific for nucleic acids than other basic dyes.

The selective staining of glycogen by Best's Carmine and the other dyes used in a similar manner (e.g. Alizarin Brilliant Blue BS, Alizarin Red S, Gallein and Haematoxylin) is due to hydrogen bonding between phenolate (−O−) groups of the dye and glycogen hydroxyl groups. The variation in intensity obtained with different dyes may be explained by their stereochemistry: the best stains for glycogen had electron-donating groups (hydroxyl or amine) placed so as to increase the basicity of the phenolate anions and in addition lacked symmetry, so increasing their ability to bind by van der Waals forces.

The effect of meropenem on animal kidneys has been assessed in rats (5 of each sex/group), rabbits (3 of each sex/group) and monkeys (3 of each sex/group) in comparative iv studies with ceftazidime, cefotaxime, cephaloridine and imipenem (without cilastatin). Diarrhoea occurred in rabbits and monkeys dosed with imipenem or meropenem. Emesis occurred only after the administration of imipenem to monkeys. After 14 days administration to rats evidence of nephrotoxicity was seen only in males dosed with cephaloridine (850 mg/kg); no changes were seen with ceftazidime, cefotaxime or meropenem (all at 1000 mg/kg). Four days after a single dose to rabbits renal tubular necrosis was seen in all animals receiving imipenem (150 mg/kg) and cephaloridine (250 mg/kg). Minimal histopathological changes to the kidneys were seen with cefotaxime, ceftazidime and meropenem (all at 400 mg/kg). After seven days administration to cynomolgus monkeys imipenem (180 mg/kg) caused moderate to severe tubular necrosis. No tubular damage was seen with meropenem at 180 mg/kg or with cefotaxime or ceftazidime (both at 500 mg/kg). At 500 mg/kg meropenem caused mild tubular regeneration and/or fat accumulation in 3/6 animals, with mild tubular necrosis in one of these. The data from these three species indicate that meropenem has a low nephrotoxic potential in these animal models.
