The wetsalts tiger beetle, Cicindelidia haemorrhagica(LeConte) (Coleoptera: Cicindelidae), is found in several active thermal hot spring areas in Yellowstone National Park (YNP) where substrate surface temperatures can exceed 50 °C. However, relationships between surface temperatures and the time adults spend on them remain poorly understood. Therefore, we characterized thermal profiles of Dragon Spring and Rabbit Creek, 2 thermally active research sites containing C. haemorrhagicain YNP, to quantify the time adults spend at different surface temperatures. We took 58 thermal video recordings of adults over 6 total days of observation ranging from 10 to 15 min for each adult. Thermal video analysis results indicated a positive relationship between the total time adult beetles spent on surface temperatures from Dragon Spring and Rabbit Creek as temperatures increased from 20 °C. Once surface temperatures exceeded 40 °C, the total time spent at those surface temperatures declined. Adults were recorded on substrates exceeding 50 °C at one of the 2 research locations. Rabbit Creek had substantially more instances of adults present with surface temperatures exceeding 40 °C, including one individual on a surface temperature of 61.5 °C. There were 3 instances of beetles spending more than 4 min at a particular surface temperature, all within the preferred range of 30–40 °C. Our thermal profile results and previous behavioral observations suggest that adults may be resistant to the heat produced from the thermal waters that influence the substrate temperatures but may not be subject to high surface temperatures as previously reported.

There is a growing interest in enhancing the resilience of the transportation network in the face of hazards such as storms, flooding, and climate change impacts. Transportation agencies and planning organizations need tools to assess potential resilience-focused infrastructure projects and to support infrastructure project justifications. In support of the U.S. Department of Transportation and the Federal Highway Administration, we developed the open-source, publicly available Resilience and Disaster Recovery (RDR) Tool Suite to evaluate resilient infrastructure return on investment. This tool suite is based on robust decision-making concepts that enable scenario analysis and decision making under uncertainty. The RDR Tool Suite leverages widely used travel demand modeling as well as benefit–cost analysis, breakeven analysis, and benefit–cost analysis under uncertainty to accommodate a range of data availability. The tool suite also includes an Exposure Analysis Tool to assist with assigning hazard severity to the transportation network. This paper provides an overview of the newly released RDR Tool Suite and its utility for transportation planning.

In Australia, there is a growing concern about the well-being of teachers with many expressing their intention to leave the profession or indeed have already left. Various reasons have been suggested for this trend, with burnout being identified as one of the factors. This study investigates burnout in Australian teachers as one of the constructs which make up compassion fatigue (CF), a reduced ability to empathise with others. Moreover, it explores secondary traumatic stress (STS), which also contributes to CF and occurs when a person learns about the traumatic experiences of someone under their care. Both constructs may severely impact the ability of teachers to form close relationships with their students. As part of the present study, 1939 Australian teachers were surveyed about their quality of life, well-being, classroom efficacy, and trauma awareness. Findings demonstrate that teachers with higher levels of well-being and with higher perceived classroom efficacy are less prone to burnout, reducing the risk of emotional exhaustion and disengagement often associated with this phenomenon. Conversely, connections were found between lower well-being of teachers and educators' awareness of trauma and their susceptibility to STS. Recommendations are made for further research exploring the barriers and enablers of compassion fatigue as well as positive teacher well-being, in order to develop targeted initiatives to better prepare and protect teachers to work with a cohort of students who are increasingly demonstrating symptoms of trauma and poor well-being.

Beckwith-Wiedemann syndrome (BWS) is an epigenetic overgrowth disorder and cancer predisposition syndrome caused by imprinting defects of chromosome 11p15.5-11p15.4. BWS should be considered in children with atypical presentations of embryonal tumors regardless of clinical phenotype. Risk of malignancy correlates with specific molecular subgroups of BWS making molecular subclassification important for appropriate cancer screening. We report the first case of concurrent embryonal tumors in a phenotypically normal child, leading to the diagnosis of BWS with paternal uniparental disomy and describe the molecular classification of BWS as it relates to malignancy risk, along with approach to management.

System-on-chip (SoC) architectures are a heterogeneous mix of microprocessors, custom accelerators, memories, interfaces, peripherals, and other resources. These resources communicate using complex on-chip interconnect networks that attempt to quickly and efficiently arbitrate memory transactions whose behaviors can vary drastically depending on the current mode of operation and system operating state. Security- and safety-critical applications require access control policies that define how these resources interact to ensure that malicious and unsafe behaviors do not occur. Aker is a design and verification framework for on-chip access control. The core of Aker is the access control wrapper (ACW)–a high-performance yet efficient hardware module that dynamically arbitrates on-chip communications. Aker distributes ACWs across the SoC and programs them to perform local access control. Aker provides a firmware generation tool and a property-driven security verification methodology to ensure that the ACWs are properly integrated and configured. Aker security verification confirms that the ACW behaves properly at IP level. It verifies the hardware root of trust firmware configures the ACW correctly. And it evaluates system-level security threats due to interactions between shared resources. Aker is experimentally validated on a Xilinx UltraScale+ programmable SoC. Additionally, an Aker access control system is integrated into the OpenPULP multicore archtiecture that uses OpenTitan hardware root-of-trust for firmware configuration.

IMPORTANCE: Pregnancies resulting from assisted reproductive technology are associated with an increased risk of adverse perinatal outcomes compared with those following natural conception. Previous studies have shown an association of pregnancies resulting from transfer of multiple embryos with these negative findings. OBJECTIVE: To determine the risk for adverse outcomes in singletons conceived through assisted reproduction using double-embryo transfer (DET) vs single-embryo transfer (SET). DESIGN, SETTING, AND PARTICIPANTS: This cohort study used data from women who achieved singleton deliveries after SET or DET in Sweden between 2007 and 2017 as recorded in the National Quality Registry for Assisted Reproduction. All embryo transfers, at cleavage or blastocyst stage, replaced in fresh or frozen treatment cycles were included. Data on obstetric and neonatal outcomes were retrieved by linkage to the National Medical Birth Register. Naturally conceived singletons were included as a reference group. Data were analyzed between September 2021 and August 2022. EXPOSURES: Double-embryo transfer leading to singleton birth. MAIN OUTCOMES AND MEASURES: Relative risk ratios or odds ratios (ORs) and absolute risk differences (ARDs) in percentage points with 95% CIs were calculated for obstetric and perinatal outcomes in singleton births conceived using DET vs SET. RESULTS: Among 1 115 863 singleton births, 30 713 singletons were born after SET and 5123 after DET. A higher risk of neonatal death was found in singletons after DET vs SET (OR, 2.67 [95% CI, 1.28-5.55]; ARD, 0.2 percentage points [95% CI, 0.0-0.4 percentage points]). In frozen embryo transfers, DET was associated with a higher risk of low birth weight (OR, 1.64 [95% CI, 1.19-2.25]; ARD, 2.0 percentage points [95% CI, 0.5-3.5 percentage points]). Among blastocyst transfers, DET was associated with very preterm birth (relative risk ratio, 2.64 [95% CI, 1.50-4.63]; ARD, 1.8 percentage points [95% CI, 0.3-3.4 percentage points]) and low birth weight (OR, 1.83 [95% CI, 1.29-2.60]; ARD, 3.2 percentage points [95% CI, 0.9-5.5 percentage points]). CONCLUSIONS AND RELEVANCE: These results indicate a higher risk of adverse outcomes following DET, even when the result is a singleton birth, vs singletons born after SET. Adverse outcomes were mainly observed in singletons following DET using frozen embryos and blastocysts.

We describe the security verification of OpenTitan. We illustrate how information flow tracking turns human knowledge of assets and security requirements into formal security properties verified using Cycuity’s Radix. The verification uncovered weaknesses and helped produce hardware fixes to eliminate vulnerabilities.

Aberrant CD4+T cell reactivity against intestinal microorganisms is considered to drive mucosal inflammation in inflammatory bowel diseases. The disease-relevant microbial species and the corresponding microorganism-specific, pathogenic T cell phenotypes remain largely unknown. In the present study, we identified common gut commensal and food-derived yeasts, as direct activators of altered CD4+T cell reactions in patients with Crohn’s disease (CD). Yeast-responsive CD4+T cells in CD display a cytotoxic T helper cell (TH1 cell) phenotype and show selective expansion of T cell clones that are highly cross-reactive to several commensal, as well as food-derived, fungal species. This indicates cross-reactive T cell selection by repeated encounter with conserved fungal antigens in the context of chronic intestinal disease. Our results highlighted a role of yeasts as drivers of aberrant CD4+T cell reactivity in patients with CD and suggest that both gut-resident fungal commensals and daily dietary intake of yeasts might contribute to chronic activation of inflammatory CD4+T cell responses in patients with CD.

The Trail Historical Society, established in 1953, is responsible for preserving the history of the City of Trail in British Columbia. The society owns and manages collections, exhibits, and programming in the Trail Museum & Archives. One significant aspect of Trail's history is the Doukhobor community, which settled in the city around 1910. The Doukhobors cultivated gardens and became known for their fresh produce, particularly tomatoes. The Doukhobor communal house, built by Peter Verigin, still stands today but is currently vacant as the city works on a project to convert the surrounding area into a park. Twin Rivers Elementary School in Castlegar, British Columbia, is a trilingual school that focuses on English, French, and Russian languages. The school's administrators recognized the unique need of their community, which has a large population of children with Doukhobor heritage, and incorporated this heritage into the curriculum. The program began in 1983 and has been successful in improving academic performance and engagement among students. The Russian community in Castlegar has been supportive of the program, and the school continues to celebrate Russian language and culture. However, the program faces challenges as the fluency of Russian-speaking grandparents decreases with each generation. Professional women engage with individual and social issues of ageing, appearance and identity throughout their lives. The challenges professional women have faced since the 1970s have changed as to how they view themselves in new and varied contemporary careers and their perceptions of how others view them. Eleven career women in the Twin Cities, MN, were interviewed about how they managed their appearances throughout their careers. Responses were organized into groups built around the women’s early career narratives – 20s–30s, mid-career narratives – 40s–60s, late and post-career narratives – 70s–90s. We found that these women consciously thought about the meaning and impact of work and their work wardrobe as they passed through each career stage, especially in terms of impression and identity management. We learned through the stories they told how the art, practice and meaning around dressing for work changed over time – with both continuity and change in their expressed outcomes.

The radical SAM superfamily (RSS), arguably the most functionally diverse enzyme superfamily, is also one of the largest with ∼700 K members currently in the UniProt database. The vast majority of the members have uncharacterized enzymatic activities and metabolic functions. In this Perspective, we describe RadicalSAM.org, a new web-based resource that enables a user-friendly genomic enzymology strategy to explore sequence-function space in the RSS. The resource attempts to enable identification of isofunctional groups of radical SAM enzymes using sequence similarity networks (SSNs) and the genome context of the bacterial, archaeal, and fungal members provided by genome neighborhood diagrams (GNDs). Enzymatic activities and in vivofunctions frequently can be inferred from genome context given the tendency for genes of related function to be clustered. We invite the scientific community to use RadicalSAM.org to (i) guide their experimental studies to discover new enzymatic activities and metabolic functions, (ii) contribute experimentally verified annotations to RadicalSAM.org to enhance the ability to predict novel activities and functions, and (iii) provide suggestions for improving this resource.

Dopamine (DA) and glutamate neurotransmission are strongly implicated in schizophrenia pathophysiology. While most studies focus on contributions of neurons that release only DA or glutamate, neither DA nor glutamate models alone recapitulate the full spectrum of schizophrenia pathophysiology. Similarly, therapeutic strategies limited to either system cannot effectively treat all three major symptom domains of schizophrenia: positive, negative, and cognitive symptoms. Increasing evidence suggests extensive interactions between the DA and glutamate systems and more effective treatments may therefore require the targeting of both DA and glutamate signaling. This offers the possibility that disrupting DA-glutamate circuitry between these two systems, particularly in the striatum and forebrain, culminate in schizophrenia pathophysiology. Yet, the mechanisms behind these interactions and their contributions to schizophrenia remain unclear. In addition to circuit- or system-level interactions between neurons that solely release either DA or glutamate, here we posit that functional alterations involving a subpopulation of neurons that co-release both DA and glutamate provide a novel point of integration between DA and glutamate systems, offering a key missing link in our understanding of schizophrenia pathophysiology. Better understanding of mechanisms underlying DA/glutamate co-release from these neurons may therefore shed new light on schizophrenia pathophysiology and lead to more effective therapeutics.

The presence of sodium gluconate in Cheddar cheese, which can be added as a processing aid during manufacture, has been shown as a contributing factor to gas production and gas defects in aged cheese. A survey of gluconate content in retail Cheddar cheese was performed. Fifty-three retail packs of cheddar cheese were purchased from stores in Utah as either blocks, shreds, sliced cheeses or individual-sized cuts and sticks. This included mild, medium and sharp cheeses. Analysis of gluconate content was performed using an enzyme assay. Levels of gluconate ≥ 0.1% were found in 7 samples with the highest level being 0.45% (wt/wt).

An item of headwear, the pussy hat, made news in 2017 when women gathered through social media and local events to create the iconic hat. The hat became a part of the Women’s March that took place in January in Washington DC as well as in cities such as St. Paul, Minnesota. This research involves participant observation of the hat-making process and the hat wearing during marches in Washington DC and Minnesota. Findings revealed the effectiveness of the pussy hat in calling attention to the events that took place prior to and during the marches in January 2017. However, examination of the pussy hat included many variations in form, i.e. colour, texture and shape and the meaning represented some but not all voices of the march.

Christianity arrived on the island of Shikoku, Japan, from the neighboring island of Kyushu in the mid-sixteenth century, an event commemorated by a signboard and gravesite where some of the early converts to the faith were buried. The sancti"ed area exhibits what might be expected of Hidden Christian spatiality: a quasi-Buddhist nature, syncretistic Shinto elements, and o#ertory tools; each of which would be quite out of place in any other “Christian” context. What may the sacrality of this ground have entailed? What signi"cance did its objects contain for those who created them and visited them? Moreover, how “ecumenical” could worship there have been if one half (the Christian) was for political reasons forcibly kept hidden while the other half (Buddhist/Shinto) was open? These are the questions we pursue, although our conclusions can perhaps do no more than indicate a direction.

Intermolecular C–C bond-forming reactions are underdeveloped transformations in the field of biocatalysis. Here we report a photoenzymatic intermolecular hydroalkylation of olefins catalyzed by flavin-dependent ‘ene’-reductases. Radical initiation occurs via photoexcitation of a rare high-order enzyme-templated charge-transfer complex that forms between an alkene, α-chloroamide, and flavin hydroquinone. This unique mechanism ensures that radical formation only occurs when both substrates are present within the protein active site. This active site can control the radical terminating hydrogen atom transfer, enabling the synthesis of enantioenriched γ-stereogenic amides. This work highlights the potential for photoenzymatic catalysis to enable new biocatalytic transformations via previously unknown electron transfer mechanisms.

The absence of a reliable, universal biomarker is a significant limitation in neuroendocrine neoplasia (NEN) management. We prospectively evaluated two CgA assays, (NEOLISA, EuroDiagnostica) and (CgA ELISA, Demeditec Diagnostics (DD)) and compared the results to the NETest.NEN cohort (n= 258): pancreatic, n= 67; small intestine, n= 40; appendiceal, n= 10; rectal, n= 45; duodenal, n= 9; gastric, n= 44; lung, n= 43. Image-positive disease (IPD) (n= 123), image & histology- negative (IND) (n= 106), and image-negative and histology positive (n= 29). CgA metrics: NEOLISA, ULN: 108 ng/mL, DD: ULN: 99 ng/mL. Data mean ± s.e.m.NETest: qRT-PCR – multianalyte analyses, ULN: 20. All samples de-identified and assessed blinded. Statistics: Mann–Whitney U-test, Pearson correlation and McNemar-test.CgA positive in 53/258 (NEOLISA), 32 (DD) and NETest-positive in 157/258. In image- positive disease (IPD, n= 123), NEOLISA-positive: 33% and DD: 19%. NETest-positive: 122/123 (99%; McNemar’s Chi2= 79–97, P <0.0001). NEOLISA was more accurate than DD (P = 0.0003). In image- negative disease (IND), CgA was NEOLISA-positive (11%), DD (8%), P= NS, and NETest (33%). CgA assays could not distinguish progressive (PD) from stable disease (SD) or localized from metastatic disease (MD). NETest was significantly higher in PD (47 ± 5) than SD (29 ± 1, P= 0.0009). NETest levels in MD (35 ± 2) were elevated vs localized disease (24 ± 1.3, P= 0.008).NETest, a multigenomic mRNA biomarker, was ~99% accurate in the identification of NEN disease. The CgA assays detected NEN disease in 19–33%. Multigenomic blood analysis using NETest is more accurate than CgA and should be considered the biomarker standard of care.
