1. Epidermal growth factor-induced ANGPTL4 enhances anoikis resistance and tumour metastasis in head and neck squamous cell carcinoma

Abstract

Epidermal growth factor (EGF) is important for cancer cell proliferation, angiogenesis and metastasis in many types of cancer. However, the mechanisms involved in EGF-induced head and neck squamous cell carcinoma (HNSCC) metastasis remain largely unknown. In this study, we reveal that angiopoietin-like 4 (ANGPTL4) plays an important role in the regulation of EGF-induced cancer metastasis. We showed that EGF-induced ANGPTL4 expression promoted anoikis resistance and cancer cell migration and invasion in HNSCC. In addition, depletion of ANGPTL4 inhibited EGF-induced cancer cell invasion. Autocrine production of EGF-induced ANGPTL4 regulated the expression of matrix metalloproteinases (MMPs). The induction of MMP-1 gene expression by ANGPTL4-activated integrin β1 signalling occurred through the AP-1 binding site in the MMP-1 gene promoter. Furthermore, down-regulation of MMP-1 impeded EGF- and recombinant ANGPTL4-enhanced HNSCC cell migration and invasion. Depletion of ANGPTL4 significantly blocked EGF-primed extravasation and metastatic seeding of tumour cells and MMP-1 expression in lungs. However, no effect of ANGPTL4 on tumour growth was observed. These results suggest that EGF-induced expression and autocrine production of ANGPTL4 enhances HNSCC metastasis via the up-regulation of MMP-1 expression. Inhibition of ANGPTL4 expression may be a potential strategy for the treatment of EGFR-mediated HNSCC metastasis.

2. Characterisation of bio-oil from fast pyrolysis of rice husk in a fluidised bed reactor.

Abstract

Fast pyrolysis of rice husk was conducted to produce bio-oil. The physical-chemical properties of the bio-oil were analysed. The results show that bio-oil has relatively high water content (47·05%), high ash content (0·29%), high carbon residue (5·84%) and low higher heating value (16·42 MJ kg<superscript>-1</superscript>). The density, kinetic viscosity at room temperature and pH value of the bio-oil were 1100 kg m<superscript>-3</superscript>, 3·41 mm<superscript>2</superscript> s<superscript>-1</superscript> and 3·23 respectively. Simultaneously, the kinetic viscosity of the bio-oil decreases significantly with increasing measurement temperature from 25 to 100°C, and its prominent viscosity-temperature characteristics were proved. Analysis of chemical functional groups and chemical structure of the bio-oil through Fourier transformation infrared spectrometer and Fourier transformation nuclear magnetic resonance technologies demonstrated the complexity of chemical compositions of the rice husk bio-oil. The current study investigated the involvement of Th17 cells in multiple myeloma. METHODS: Flow cytometry was used to investigate the frequencies of Th17 cells in peripheral blood mononuclear cells from 30 patients with multiple myeloma and from 14 healthy control subjects. The concentrations of Th17-associated cytokines (interleukin [IL]-6, IL-17, IL-1β and IL-23) were measured by enzyme- linked immunosorbent assay. RESULTS: There was a significantly increased proportion of Th17 cells, and increased plasma concentrations of Th17-associated cytokines, in patients with multiple myeloma compared with healthy controls. There was a significant relationship between the proportion of Th17 cells and clinical tumour stage, serum lactate dehydrogenase concentration and serum creatinine concentration. Prospective areas for extension of the Tan sheep industry were also investigated. Two sheep groups were sampled; one consisted of artificial insemination flocks (1241 lambs sampled from 25 flocks) and the other of natural mating flocks (1009 lambs from 38 flocks). Six phenotypical traits were measured for each lamb and six hydrothermal factors were collected from the meteorological stations located in the sampling areas. A significant correlation was found between wool characteristics of the lambskin and hydrothermal conditions. Data further indicated that among all the hydrothermal factors measured, those causing the geographical differentiation of soil and vegetation were the major factors responsible for the corresponding differentiation of the lambskin and wool quality of Tan sheep. Thermal factors were mostly positively correlated with wool quality, while the influence of moisture was negative. Three eco-geographical regions were defined based on a combination of the hydrothermal conditions and corresponding wool characters: (i) typical region or super-suitable region, (ii) sub-typical region or suitable region and (iii) transitional region. The delimitation could be used as a basis for the extension of the Tan sheep industry. By Far-Western blotting and yeast two-hybrid assay, we demonstrate that p45, the large subunit of NF-E2, is capable of binding to a specific set of WW domain-containing proteins, including the ubiquitin ligase hRPF1. This binding is mediated through the interaction between the WW domains and a PY motif located within the amino-terminal region of p45. Interestingly, the carboxyl-terminal domain of mammalian RNA polymerase II binds a similar set of WW domains to which p45 interacts with.
