Peroxisome proliferator-activated receptor γ (PPARγ) belongs to the nuclear receptor superfamily and is involved in the inflammatory process. Previously, we synthesized the ligands of PPARγ that possess the hybrid structure of a food-derived cinnamic acid derivative (CA) and GW9662, an irreversible PPARγ antagonist. These ligands activate the transcription of PPARγ through the covalent bond formation with the Cys285 residue of PPARγ, whereas their anti-inflammatory effect has not been examined yet. Here, we show the anti-inflammatory effect of the covalent PPARγ ligands in RAW264 cells, murine macrophage-like cells. GW9662 suppressed the production of nitric oxide (NO) stimulated by lipopolysaccharide and exerted a synergistic effect in combination with CA. The compounds bearing their hybrid structure dramatically inhibited NO production and transcription of proinflammatory cytokines. A comparison study suggested that the 2-chloro-5-nitrobenzoyl group of the ligands is important for anti-inflammation. Furthermore, we synthesized an alkyne-tagged analogue that becomes an activity-based probe for future mechanistic study.Graphical AbstractAbility of covalent bond formation and attachment of cinnamic acid unit to GW9662 are important for suppression of inflammatory biomarker production in RAW264 cells.

The chemogenetic control of cellular protein stability using degron tags is a powerful experimental strategy in biomedical research. However, this technique requires permanent fusion of the degron to a target protein, which may interfere with the proper function of the protein. Here, we report a peptide fragment from the carboxyl terminus of ubiquitin as a cleavable linker that exhibits the slow but efficient cleavage of a degron tag via cellular deubiquitinating enzymes (DUBs). We designed a fusion protein consisting of a cleavable linker and a destabilizing domain (DD), which conditionally controls the expression and release of a target protein in a ligand-induced state, allowing the free unmodified protein to perform its function. Insertion of an AGIA epitope at the carboxyl terminus of the linker made space for the DUBs to access the site to assist the cleavage reaction when the amino terminus of the target protein caused steric hindrance. The developed system, termed a cleavable degron using ubiquitin-derived linkers (c-DUB), provides robust and tunable regulation of target proteins in their native forms. The c-DUB system is a useful tool for the regulation of proteins that have terminal sites that are essential for the proper localization and function. In addition, a mechanistic investigation using proximity labeling showed that DUBs associate with the refolded DD to reverse ubiquitination, suggesting a cellular surveillance system for distinguishing the refolded DD from misfolded proteins. The c-DUB method may benefit from this machinery so that DUBs subsequently cleave the neighboring linker.

O-Methylated and glucuronosylated flavonoids were isolated from Tamarix gallicaas α-glucosidase inhibitors. Structure–activity relationship of these flavonoids suggests that catechol moiety and glucuronic acid at C-3 are factors in the increase in α-glucosidase inhibitory activity. Furthermore, rhamnetin, tamarixetin, rhamnazin, KGlcA, KGlcA-Me, QGlcA, and QGlcA-Me exhibit synergistic potential when applied with a very low concentration of acarbose to α-glucosidase from rat intestine.

In an aging society, the prevalence of Alzheimer disease (AD) and type 2 diabetes (T2D) has increased. It is currently hypothesized that these diseases are caused by the aggregation of amyloid β (Aβ) in the brain and human islet amyloid polypeptide (hIAPP) in the islets of Langerhans, respectively. Therefore, the disaggregation of these existing amyloid aggregates is a promising approach to the prevention and treatment of both diseases. In our previous studies, we found a remarkable Aβ and hIAPP aggregation inhibitory activity of polyphenolic compounds containing catechol moieties. Compared to previous reports on their aggregation inhibitory activity, there are few on the disaggregation activity of polyphenolic compounds. Additionally, there are few findings on the disaggregation activity of polyphenolic compounds on hIAPP. In this study, we investigated the Aβ and hIAPP disaggregation activity of scirpusin B, a polyphenolic compound found in passion fruit seeds, and related compounds. Thioflavin T (Th-T) assays and transmission electron microscopy (TEM) were performed on these compounds to evaluate their Aβ42 and hIAPP disaggregation activities. The results showed that scirpusin B and its related compounds showed remarkable disaggregation activity. The structure–activity relationship of these compounds revealed that the presence of catechol moieties is important for this activity. This study also showed that polyphenols from passion fruit seeds have significant disaggregation activity against amyloid polypeptide aggregation.

We examined the effects of acteoside (1a), which was isolated from Orobanche minor, and its derivatives on the aggregation of a 42-mer amyloid β protein (Aβ42) in our search for anti-amyloidogenic compounds for Alzheimer's disease (AD) therapy. Acteoside (1a) strongly inhibited the aggregation of Aβ42 in a dose-dependent manner. The structure-activity relationship for acteoside (1a) and related compounds suggests the catechol moiety of phenylethanoid glycosides to be essential for this inhibitory activity.

Caffeoylquinic acid (CQA) derivatives are polyphenolic compounds found in wide variety of plants. Previously, we have demonstrated that di-CQA and tri-CQA may be neuroprotective through their ability to promote intracellular adenosine-5′-triphosphate (ATP) generation and by up-regulation of glycolytic enzyme expression. In the present study we have investigated the effect of di-CQA and tri-CQA on energy metabolism in SH-SY5Y cells using a metabolomic approach. Results indicate that di-CQA treatment of SH-SY5Y cells significantly increases the production of an array of glycolysis metabolites and tricarboxylic acid (TCA) cycle metabolites, including acetyl-CoA, succinic acid, fumaric acid, and malic acid. Tri-CQA treatment was also observed to increase the levels of all glycolysis and TCA cycle metabolites and evoked a stronger effect than that of di-CQA. In addition to their effects on glycolytic metabolites, di-CQA and tri-CQA exposure also induced a significant increase in the production of ATP, ADP, GTP, and GDP. Our results suggest that CQA-induction of intracellular ATP synthesis is mediated by the activation of central metabolic pathways including the activation of glycolysis and the TCA cycle.

The physicochemical and biological properties of fulvic acid extracted and purified from excess sludge and solubilized excess sludge were studied. Solubilization was introduced to improve the recovery rate of fulvic acid from the sludge. The structural features of fulvic acid from excess sludge and solubilized excess sludge were characterized by using an elemental analysis, Fourier transform infrared spectroscopy and 1H-nuclear magnetic resonance spectroscopy, and were compared with fulvic acid extracted from peat which had an inhibitory effect on the type I allergy in our previous study. The results show that they had a higher aliphatic characteristic with lower oxygen group content than fulvic acid from peat, and that the aliphatic characteristic was further strengthened by the use of solubilization. The biological properties of fulvic acid from excess sludge and solubilized excess sludge showed an inhibitory effect on β-hexosaminidase release at the antigen-antibody binding stage and antigen-receptor binding stage by using rat basophilic leukemia cells.

Two new daphnane diterpenoids, hirseins A (1) and B (2), were isolated from the aerial parts of Thymelaea hirsuta, and their structures were elucidated on the basis of spectroscopic data interpretation. Hirsein B (2) is an unusual daphnane in possessing a coumaroyl moiety. NOESY correlations of 2implied that isomerization of the coumaroyl group in 2was caused by equilibrium between the E(2e) and Z(2z) forms. Compounds 1and 2were found to inhibit melanogenesis in B16 murine melanoma cells.

Four new oxindole alkaloids, paratunamides A−D (1−4), containing a secologanin unit, were isolated from the bark of Cinnamodendron axillare, and their structures and relative configurations were elucidated by spectroscopic data. The absolute configuration at C-7 in 1−4was assigned as S, S, R, and S, respectively, on the basis of the CD spectra.

The structure-activity relationship of 12 isolated acetylenes from galls of Hedera rhombea (Araliaceae) induced by Asphondylia sp. (Cecidomyiidae) and their derivatives has been studied for the inhibition of the shoot and root growth of rice, perennial ryegrass, cockscomb, lettuce, and cress. Almost all acetylenes generally showed growth inhibitory activity. The diacetylenes exhibited higher activity than the monoacetylenes, suggesting that a conjugated diyne segment is essential for the activity. On the other hand, the acetylenes with a nonoxidated methylene group at C-8 showed stronger activity comparing with those possessing hydroxy and acetoxy groups at C-8. Furthermore, it has been demonstrated that the acetylenes bearing a terminal olefinic group at C-16,C-17 enhanced the activity. It is thus clarified that important sites for the activity of the acetylenes from galls of H. rhombea are a conjugated diyne and a terminal olefinic group connecting to the aliphatic chain and that less oxidated compounds show more activity.

When seedlings of lettuce, cress, rice and wheat were incubated with the leachate of wheat straw, the roots growth of lettuce and garden cress were particularly inhibited. The leachate of wheat straw (100 g eq./l) showed 80.5 and 79.4% inhibition for lettuce and cress roots, respectively. The inhibitory activity was stronger as the concentration of wheat straw leachate was greater. This result indicates that allelochemical(s) inhibiting the roots growth of lettuce and cress are leached from the wheat straw into the water. Two potent compounds were isolated from the leachate of the wheat straw and identified as syringoylglycerol 9-O-β-d-glucopyranoside and l-tryptophan by spectral analyses. Syringoylglycerol 9-O-β-d-glucopyranoside inhibited the roots growth of lettuce and cress at concentrations greater than 0.1 and 10.0 μM, respectively. On the other hand, l-tryptophan inhibited the roots growth of lettuce and cress at concentrations greater than 0.1 and 1.0 μM, respectively. The content of syringoylglycerol 9-O-β-d-glucopyranoside and l-tryptophan in the leachate of wheat straw (100 g eq./l) was 18.4 ± 0.7 and 6.2 ± 0.6 μM, respectively. Syringoylglycerol 9-O-β-d-glucopyranoside (18.4 μM) showed 21.5 and 13.5% inhibition in the lettuce and cress roots assay, respectively. On the other hand, 6.2 μM of l-tryptophan showed 47.5 and 35.0% inhibition in the lettuce and cress roots assay, respectively. The structure–activity relationships of alkaloids (1–5) from mesquite were subjected to assessment of growth inhibition against the shoot and root growth of monocotyledonous plants, barnyard grass, rice and timothy, and dicotyledonous ones, amaranth, lettuce and cress. All alkaloids tested generally showed growth inhibitory against both monocotyledonous and dicotyledonous plants. Furthermore, these alkaloids exhibited higher activity against the growth of root than that of shoot of all plant species used, except that juliprosopine (5) showed higher activity against the shoot growth than the root growth of rice seedling. Among these alkaloids, the highest active compound appeared to be juliprosine (4), followed by a (1:1) mixture of 3-oxo- and 3'-oxo-juliprosine (3a and 3b), and juliprosopine (5). The activity of juliprosine (4) containing 2-methyl piperidine bearing hydroxyl groups at C-3 and C-3' was higher than that of 3-oxo- and 3'-oxo-juliprosine (3a and 3b) containing 3-oxo- and 3'-oxo-2-methylpiperidine. Compound 3 and 4 containing dihydroindolizinium ring showed higher activity than compound 5 containing tetrahydroindolizine ring, whereas compound 1 containing tetrahydroindolizinone ring showed weaker activity. The activity of secojuliprosopinal (2) without indolizine ring was very weak. It was thus clarified that the active sites in the chemical structure of alkaloids from mesquite are the functional group at C-3 and C-3' of piperidine and indolizine skeleton.

Phototropic stimulation of dark-grown hypocotyls of Arabidopsis thalianaincreased a growth inhibitor in the wild-type but not in the non-phototropic nph3-101mutant. From light-grown wild-type shoots the inhibitor was isolated and identified as indole-3-acetonitrile (IAN) from its 1H NMR spectrum. The content of endogenous IAN in the hypocotyls of wild-type and mutant unilaterally exposed to blue light was determined using a physicochemical assay. The IAN concentration (28 μM) in the phototropically stimulated wild-type hypocotyls was about three times larger than in the dark control. However, its content in the mutant hypocotyls did not change. IAN inhibited the hypocotyl growth of the nph3-101to the same extent as in the wild-type at concentrations higher than 10 μM. These results suggest that IAN plays a role in the phototropism of Arabidopsis thalianahypocotyls.

The allelopathic potential of -tryptophan in the leachate from the foliage of mesquite (Prosopis juliflora(Sw.) DC.) plants was investigated. Distilled water (500 ml) was sprayed on mesquite plants from above for 50 min and water passing through the foliage was collected. The content of -tryptophan in the sample (30 ml eq.) was analyzed using a physicochemical method and the concentration was shown to be 17.9 µM. The sample (30 ml eq.) caused 69.8% inhibition of the root growth of barnyard grass (Echinochloa crus-galliL.). The concentration of authentic -tryptophan required for 69.8% inhibition of the root growth of barnyard grass was estimated at 20.0 µM from the dose-response curve. Moreover, to establish the origin of -tryptophan in the leachate, the amount of -tryptophan in both the leaves and the leachates was determined. The amount of -tryptophan containing in the foliage significantly decreased by soaking with time, whereas in the leachates the amount increased. After 60 min, its content in the leachates was nearly equivalent to that of the leaves. These results suggest that -tryptophan leached from the foliage may play an important role in the allelopathy imposed by mesquite.

A series of new taxoids, named taxuspines A–H and J–Z (1–25) and taxezopidines A–H and J–L (26–36), have been isolated together with 37 known taxoids (37–73) including paclitaxel (53) from the Japanese yew, Taxus cuspidataSieb. et Zucc. (Taxaceae). These new taxoids possess various skeletons containing 5/7/6, 6/10/6, 6/5/5/6, 6/8/6, or 6/12‐membered ring systems. Among the new taxoids, some non‐taxol‐type compounds remarkably reduced CaCl2‐induced depolymerization of microtubules, or increased cellular accumulation of vincristine in multidrug‐resistant tumor cells as potent as verapamil. On the other hand, chemical derivatization of taxinine (37), one of major taxoids obtained from this yew, led to the discovery of unusual reactions of taxinine derivatives. Here we describe our recent results on the isolation, structure elucidation, and bioactivity of these new and known taxoids and the formation of unexpected products of the unusual reactions of taxinine. © 2002 Wiley Periodicals, Inc. Some non‐taxol‐type taxoids having neither an oxetane ring at C‐4 and C‐5 nor an N‐acylphenylisoserine group at C‐13, such as taxuspine C, 2′‐desacetoxyaustrospicatine, and 2‐desacetoxytaxinine J, which were isolated from the Japanese yew Taxus cuspidata, increased cellular accumulation of vincristine (VCR) in multidrug‐resistant 2780AD cells as potently as verapamil, and efficiently inhibited [3H]azidopine photolabeling of P‐glycoprotein (P‐gp). Taxuspine C, 2′‐desacetoxyaustrospicatine, and 2‐desacetoxytaxinine J at 10μMcompletely reversed the resistance to colchicine, VCR, and taxol in KB‐C2 cells, which overexpress P‐gp, while taxinine and taxinine M showed no effect. Taxuspine C, 2′‐desacetoxyaustrospicatine, and 2‐desacetoxytaxinine J may be candidate pharmaceuticals for reversing multidrug resistance (MDR) and also may be good modifiers of MDR in cancer chemotherapy.

Leaf-movement in nyctinastic plants has long been believed to be controlled by plant hormones that are common among all nyctinastic plants. We have identified several bioactive substances for nyctinasty, whose bioactivities were highly specific to the original plant, based on the bioassay using the original plant leaf, and have shown that nyctinastic leaf-movement is not regulated by plant hormones. Our present results are in accordance with Umrath et al. physiologically significant opinion.

Boron trifluoride-catalyzed reaction of β-4(20)-epoxy-5- O-triethylsilyltaxinine A ( 1) gave the 3,5-diene ( 3) and the 3,8-cyclopropane ( 4) derivatives, while the similar reaction of the corresponding α-4(20)-epoxide ( 2) afforded the ring contracted derivative ( 5) and its hemiacetal dimer ( 6). Plausible mechanisms of these reactions containing 1,2-hydride shift ( 3and 4from 1) or pinacol-type rearrangement ( 5and 6from 2) are proposed.

Nakijiquinones C (1) and D (2), new sesquiterpenoid quinones with a serine or a threonine residue, respectively, have been isolated from an Okinawan marine sponge of the family Spongiidae and the structures were determined by spectroscopic data and chemical correlations with a known compound, isospongiaquinone (5). Nakijiquinones and synthetic related compounds exhibited inhibitory activity against c-erbB-2 kinase.

Oxidation of taxinine A ( 1) with tetrapropylammonium perruthenate afforded 5-oxotaxinine A ( 2) which subsequently gave a new dimeric compound ( 3) through regio- and stereo-specific Diels-Alder cycloaddition. The relative stereostructure of 3was established by spectral data and X-ray analysis.

Four new metabolites (citreobiphenyl, precitreobiphenyl, citreoviranol, and demethylcitreoviranol) have been isolated from the mycelium of Penicillium citreo-virideB. (IFO 4692) incubated on the polished rice together with sodium bromide, and their structures have also been elucidated on the basis of their spectral data.

Two new cyclic peptides, keramamides K (1) and L (2), with an unusual tryptophan residue have been isolated from an Okinawan marine sponge Theonellasp., and the structures including absolute stereochemistry were elucidated on the basis of 2D NMR and FABMS/MS data and degradation experiments.

Crystal and solution state conformations of two taxoids, taxinine and taxinine B, inhibiting the drug transport by P-glycoprotein in multidrug-resistant cells, were analyzed by X-ray crystallographic analysis and ROE experiments. This study demonstrated that in solid state, both taxinine and taxinine B took similar cage conformation in which A-ring and the cinnamoyl side chain at C-ring were specially very close to each other. These conformations were also observed in their solution conformations deduced by ROE correlations in CDCl3, Monte Carlo simulation using MM2∗force field, and semiempirical molecular orbital calculation using PM3 method.

Deoxysilydianin methyl ether has been readily synthesized by means of anodic oxidation of the corresponding phenol as a key step, indicating a general method for synthesis of 9-oxaisotwist-8-en-2-ones in high yields.

The first total synthesis of 4-methylthio-3-butenyl glucosinolate (MTBG), a natural bioactive compound and a precursor of radish phototropism-regulating substances, was achieved from commercially available 1,4-butanediol. The glucosinolate framework was prepared by coupling of an oximyl chloride derivative and tetraacetyl thioglucose. A methylthio group was introduced to the framework by a Wittig reaction between triphenylphosphonium thiomethylmethylide and an aldehyde intermediate of glucosinolate. The synthetic route should facilitate preparation of various derivatives needed for probe synthesis based on MTBG.

Nowadays, the incidence of Alzheimer’s disease (AD) and type 2 diabetes (T2D) is increasing at an alarming rate. More and more studies have been investigating the relationship between these two diseases and are trying to find an effective treatment. According to amyloid hypothesis, it is very necessary to find phenolic compounds with catechol moieties, which would inhibit the aggregation of amyloid β (Aβ) and human islet amyloid polypeptide (hIAPP), while also exhibiting antioxidant activity and protective effect. We isolated rosmarinic acid (RA) from the plant Isodon japonicus(Burm.f.) H. Hara. Thioflavin T assay and transmission electron microscopy observation were carried out to evaluate the inhibitory effect of RA, caffeic acid, and 3,4-dihydroxyphenyllactic acid, which are the substructures of RA, on both Aβ and hIAPP fibrillization. 2,2-Diphenyl-1-picrylhydrazyl assay was applied to test the antioxidant activity. RA showed inhibitory effect on both peptides and strong antioxidant activity. These results suggest that the existence of catechol units plays an important role on the inhibitory activity. Therefore, RA will be a promising strategy to prevent AD and T2D.

The new chlorinated cyclopentenols, palmaenol A (1) and palmaetriol (3), were isolated from the culture broth of the discomycete Lachnum palmae(NBRC-106495). In addition, a new chlorinated cyclopentenol, palmaenol B (2), a geometric isomer of 1, was also obtained from the culture broth containing KCl. The structures of 1–3were elucidated from spectroscopic data. Compounds 1-3are cyclopentenols containing three or two chlorines; 1and 2are also reduced forms of palmaenones A (4) and B (5), which have been isolated from the same discomycete, respectively. Compounds 4and 5had potent antimicrobial activity, while compounds 1–3showed no such activity. Thus, it is suggested that the unsaturated carbonyl group at C-6 has an important role for the antimicrobial activity.

A new iridoid glucoside, sesinoside (1), was isolated from the seedlings of Sesamum indicum. The structure of 1was elucidated by spectroscopic analyses and by methanolysis of 1, which produced the known compounds, phlorigidosides C (2) and (6Z)-foliamenthic acid methyl ester (3). This is the first report of an iridoid glucoside with 3.

The diterpenoid moiety of brasilicardin A ( 1), isolated from the actinomycete Nocardia brasiliensisIFM 0406, was shown to be biosynthesized from d-glucose viathe non-mevalonate pathway.

A novel taxoid, taxezopidine A ( 1), has been isolated from seeds of Japanese yew Taxus cuspidataSieb. et Zucc. and the structure elucidated on the basis of spectroscopic data. Taxezopidine A ( 1) is the first taxoid with a hemiketal ring at C-11 ∼ C-13, C-15, and C-17. © 1997 Elsevier Science Ltd.

Three polyacetylenes, 8-(β-D-glucopyranosyloxy)-3-hydroxy-1,9, 14-pentadecatriene-4,6-diyne, termed “helian”(1), 8-acetoxy-3-hydroxy-1,9, 14-pentadecatriene-4,6-diyne (2), and 3,8-dihydroxy-1,9, 14-pentadecatriene-4,6-diyne (3) were isolated from seedlings of sunflower, Helianthus annuusL. cv Russia. Compounds 1and 2, having a β-glucose and an acetoxy group at C-8, respectively, showed a weak effect on the growth of roots and shoots of rice (Oryza sativaL.) and cress (Lepidium sativumL.), while compound 3, having a free hydroxyl group at C-8, exhibited a growth promoting effect on the roots and shoots of rice and cress.
