Aims: Good sleep is crucial for healthcare professionals, especially nurses, to prevent errors and injuries that may occur during caregiving. This study investigated the sleep quality and coping styles of nurses during the post-pandemic period, as well as related factors. Subject and methods: An online observational cross-sectional study involving 1022 nursing staff was conducted. Participants completed questionnaires on job characteristics, the Pittsburgh Sleep Quality Index (PSQI) and Simplified Coping Style Questionnaire (SCSQ). Pearson’s correlation and multiple logistic regression analyses confirmed interactions between these factors. Results: A total of 1022 nurses completed the survey, of whom 90.90% (n= 929) were female, and 87.38% were over 25 years old. The PSQI score was 10.51(2.23), and 91.1% (n= 931) of the nurses had sleep disturbances. In this study, we found that PSQI was associated with SCSQ. Multivariate analysis showed that age (36–40 years), frequency of night shifts (9–11 times/month, ≥ 12 times/month), and less humanistic care were risk factors for sleep disorders among night shift nurses (P< 0.05), whereas frequency of exercise (1 time/week), non-attendance of training and learning after night shifts, and SCSQ positivity were protective factors for sleep disorders (P< 0.05). Conclusions: Sleep quality of medical staff, especially nurses, was affected by shift work during the post-pandemic era. Designing the best strategy for employees who are more likely to have sleep disorders, such as experienced nurses, will support them through sleep hygiene education and strengthen the humanistic solicitude of nurses.

With the recent advancement of novel biomedical technologies such as high-throughput sequencing and wearable devices, multi-modal biomedical data ranging from multi-omics molecular data to real-time continuous bio-signals are generated at an unprecedented speed and scale every day. For the first time, these multi-modal biomedical data are able to make precision medicine close to a reality. However, due to data volume and the complexity, making good use of these multi-modal biomedical data requires major effort. Researchers and clinicians are actively developing artificial intelligence (AI) approaches for data-driven knowledge discovery and causal inference using a variety of biomedical data modalities. These AI-based approaches have demonstrated promising results in various biomedical and healthcare applications. In this review paper, we summarize the state-of-the-art AI models for integrating multi-omics data and electronic health records (EHRs) for precision medicine. We discuss the challenges and opportunities in integrating multi-omics data with EHRs and future directions. We hope this review can inspire future research and developing in integrating multi-omics data with EHRs for precision medicine.

Inflammatory pain results from the heightened sensitivity and reduced threshold of nociceptor sensory neurons due to exposure to inflammatory mediators. However, the cellular and transcriptional diversity of immune cell and sensory neuron types makes it challenging to decipher the immune mechanisms underlying pain. Here we used single-cell transcriptomics to determine the immune gene signatures associated with pain development in three skin inflammatory pain models in mice: zymosan injection, skin incision and ultraviolet burn. We found that macrophage and neutrophil recruitment closely mirrored the kinetics of pain development and identified cell-type-specific transcriptional programs associated with pain and its resolution. Using a comprehensive list of potential interactions mediated by receptors, ligands, ion channels and metabolites to generate injury-specific neuroimmune interactomes, we also uncovered that thrombospondin-1 upregulated by immune cells upon injury inhibited nociceptor sensitization. This study lays the groundwork for identifying the neuroimmune axes that modulate pain in diverse disease contexts.

To enhance the stability of nano-selenium, polysaccharide was employed as template to prepare polysaccharide nano-selenium, and its biological activity was studied. The Fagopyrum tataricumpolysaccharide (FTP1, FTP2, FTP3, FTP4) was extracted using a water extraction-alcohol precipitation method, and Fagopyrum tataricumpolysaccharide nano-selenium (FTP3-SeNPs) was synthesized via by HNO3-Na2SeO4method, and its structure was characterized using ultraviolet–Vis (UV–Vis), fourier-transform infrared (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). Then, the antioxidant and hypoglycemic activities of FTP3-SeNPs in vitro were studied. According to the results of UV–Vis, FT-IR, XRD, and SEM, FTP3-SeNPs formed granular surface structure, altering the surface morphology of the polysaccharide while preserving its fundamental structure intact; the UV-Vis spectrum exhibited a novel absorption peak at approximately 290 nm, and an evident dispersion peak emerged within the XRD pattern at 20°~30°, indicating a pronounced interaction between FTP3 and SeNPs, leading to the formation of the FTP3-SeNPs complex. The average particle size of FTP3-SeNPs was 101.76 nm, significantly smaller than FTP3. The stability of FTP3-SeNPs was higher than FTP3, which was because the Zeta potential absolute value of FTP3-SeNPs was greater than FTP3. The highest scavenging activities of FTP3-SeNPs on DPPH, ·OH, and ABTS were 60.97%, 82.53%, and 79.11%, significantly higher than FTP3. The highest inhibitory activities of FTP3-SeNPs on α-glucosidase and α-amylase were 93.17 and 83.26%, significantly higher than FTP3. In conclusion, selenization modification could increase the antioxidant and hypoglycemic activity of polysaccharide from Fagopyrum tataricum.

Rapid testing devices with cellulose membrane-based lateral flow assay (LFA) could play a significant role due to their immense advantageous features to prevent and control infectious diseases like COVID-19. Compared to lab-based testing, it still lags behind due to its lower sensitivity. Herein, to increase the LFA sensitivity, barriers were designed and developed in such a way that there is a gradual increase and decrease of sample flow along with conjugated gold nanoparticles and the flow rate distribution is controlled across the thickness of the cellulose membrane-based LFA device. Finally, the barriers enhance reaction time, resulting in improved sensitivity. The two barriers, one between the conjugate pad and test zone and the other between the test and control zones, improve the detection limit from 2.0 (control or unmodified LFA device) to 0.5 nM, indicating 4-fold higher sensitivity. No additional chemicals or materials are added. ORF1ab nucleic acid of COVID-19 was used as the model target to demonstrate the concept. The stability analysis also supports the potential use of the same concept in other cellulose membrane-based platforms including point-of-care, do-it-yourself-based microfluidics paper-based analytical devices and environment monitoring purposes.

Ultra-precision diamond milling (UPDM) provides a promising generation of high-quality surface up to sub-micrometric form accuracy and nanometric surface roughness. Prior to its machining process, the investigation on surface generation would be beneficial to achieve desired quality with reduced cost and boosted productivity. However, few studies discussed this issue so far. In this study, a theoretical and experimental investigation into surface generation was carried out in UPDM. Firstly, a geometric model was built up considering the material removal process between the milling tool and the workpiece. Secondly, surface generation process was elaborated according to the developed model. Finally, UPDM experiments were performed on a typical brass material at different cutting conditions and surface texture parameters were characterized according to ISO 25178-2. Autocorrelation function (ACF) analysis was employed for a detailed evaluation of surface generation. The results showed that the static factors (cutting parameters) as well as the dynamic factors (material effects and spindle vibration), which were associated with the cutting conditions, yielded a great influence on surface generation. Surface texture characterization indicated that there was a good consistence between simulation and experiment, demonstrating the feasibility of the proposed method. ACF analysis indicated that, with the increased feedrate/step ratio, surface generation approximately achieved a transition from the periodical surface (dominated by cutting residuals) to the random surface (determined by multiple components). Significantly, this study draws up a fundamental understanding toward surface generation in UPDM.

Real-time functional MRI neurofeedback (rtfMRI-NF) is a noninvasive technique that extracts concurrent brain states and provides feedback to subjects in an online method. Our study aims to investigate the effect of rtfMRI-NF on amygdala-based emotion self-regulation by analyzing resting-state functional connectivity. We conducted a task experiment to train subjects in self-regulating amygdala activity in response to emotional stimuli. Twenty subjects were divided into two groups. The up-regulate group (URG) viewed positive stimulus, while the down-regulate group (DRG) viewed negative stimulus. The rtfMRI-NF experiment paradigm consisted of three conditions. The URG’s percent amplitude fluctuation (PerAF) scores are significant, indicating that positive emotions may be a partial side effect, with increased activity in the left hemisphere. Resting-state functional connectivity was analyzed via a paired-sample t-test before and after neurofeedback training. Brain network properties and functional connectivity analysis showed a significant difference between the default mode network (DMN) and the brain region associated with the limbic system. These results reveal to some extent the mechanism of neurofeedback training to improve individuals’ emotional regulate regulation ability. Our study has shown that rtfMRI-neurofeedback training can effectively enhance the ability to voluntarily control brain responses. Furthermore, the results of the functional analysis have revealed distinct changes in the amygdala functional connectivity circuits following rtfMRI-neurofeedback training. These findings may suggest the potential clinical applications of rtfMRI-neurofeedback as a new therapy for emotionally related mental disorders.

•A generalised approach to calculate the hydraulic head difference at two sides of each suspended waterproof curtain during multi-grade dewatering is proposed.•A comprehensive parametric analysis is carried out to investigate the influence of the relative inserted depth of curtain into confined aquifer, the distance between two curtains, and the anisotropy of aquifer hydraulic conductivity on the magnitudes of flow rate of the seepage near the curtain.•Based on the results of the parametric study, an equation is established to calculate the hydraulic head between the two sides of each curtain.

Promoting the electron occupancy of active sites to unity is an effective method to enhance the oxygen evolution reaction (OER) performance of spinel oxides, but it remains a great challenge. Here, an in situapproach is developed to modify the valence state of octahedral Ni cations in NiFe2O4inverse spinel via surface sulfates (SO42–). Different from previous studies, SO42–is directly anchored on the spinel surface instead of forming from uncontrolled conversion or surface reconstruction. Experiment and theoretical calculations reveal the precise adsorption sites and spatial arrangement for SO42–species. As a main promoting factor, surface SO42–effectively converts the crystal field stable Ni state (t2g6eg2) to the near-unity egelectron state (t2g6eg1). Moreover, the inevitable oxygen vacancies (Vo) further optimize the energy barrier of the potential-determining step (from OH* to O*). This co-modification strategy enhances turnover frequency-based electrocatalytic activity about two orders higher than the control sample without surface sulfates. This work may provide insight into the OER activity enhancement mechanism by the oxyanion groups.

Despite the myriad peer-reviewed papers demonstrating novel Artificial Intelligence (AI)-based solutions to COVID-19 challenges during the pandemic, few have made a significant clinical impact, especially in diagnosis and disease precision staging. One major cause for such low impact is the lack of model transparency, significantly limiting the AI adoption in real clinical practice. To solve this problem, AI models need to be explained to users. Thus, we have conducted a comprehensive study of Explainable Artificial Intelligence (XAI) using PRISMA technology. Our findings suggest that XAI can improve model performance, instill trust in the users, and assist users in decision-making. In this systematic review, we introduce common XAI techniques and their utility with specific examples of their application. We discuss the evaluation of XAI results because it is an important step for maximizing the value of AI-based clinical decision support systems. Additionally, we present the traditional, modern, and advanced XAI models to demonstrate the evolution of novel techniques. Finally, we provide a best practice guideline that developers can refer to during the model experimentation. We also offer potential solutions with specific examples for common challenges in AI model experimentation. This comprehensive review, hopefully, can promote AI adoption in biomedicine and healthcare.

Purpose: The dietary inflammatory index (DII) is a scoring system to quantify the inflammatory effects of nutrients and foods. Inflammation may affect bone health. The purpose of this study was to explore the relationships of DII with bone mineral density (BMD) and osteoporosis. Methods: This study involved 1023 women and 1080 men (age ≥ 50) in the US National Health and Nutrition Survey (NHANES), 2017–2018. Multivariable linear regression models were used to estimate the associations between DII and BMD. Association between DII and osteoporosis was tested with multivariable logistic regression models. Results: In women, DII was negatively associated with total hip and femoral neck BMD after adjusting for covariates (P< 0.05). In men, DII was negatively associated with lumbar spine BMD (P< 0.05). DII was positively associated with osteoporosis in women (P< 0.05). The odds ratios (ORs) (95% CI) for osteoporosis associated with DII quartiles 2, 3 and 4 vs. quartile 1 were 2.95 (1.08, 8.09), 5.63 (2.87, 11.04), and 6.14(2.55, 14.78), respectively. No significant association was observed in men. Conclusions: Higher DII scores were associated with increase osteoporosis risk in women, while no association was found in men. Greater pro-inflammatory diets might be associated with lower BMD in both women and men.

Circulating tumor DNA (ctDNA) sequencing guides therapy decisions but has been studied mostly in small cohorts without sufficient follow-up to determine its influence on overall survival. We prospectively followed an international cohort of 1,127 patients with non-small-cell lung cancer and ctDNA-guided therapy. ctDNA detection was associated with shorter survival (hazard ratio (HR), 2.05; 95% confidence interval (CI), 1.74–2.42; P< 0.001) independently of clinicopathologic features and metabolic tumor volume. Among the 722 (64%) patients with detectable ctDNA, 255 (23%) matched to targeted therapy by ctDNA sequencing had longer survival than those not treated with targeted therapy (HR, 0.63; 95% CI, 0.52–0.76; P< 0.001). Genomic alterations in ctDNA not detected by time-matched tissue sequencing were found in 25% of the patients. These ctDNA-only alterations disproportionately featured subclonal drivers of resistance, including RICTORand PIK3CAalterations, and were associated with short survival. Minimally invasive ctDNA profiling can identify heterogeneous drivers not captured in tissue sequencing and expand community access to life-prolonging therapy.

IMPORTANCE: Induction chemotherapy added to concurrent chemoradiotherapy significantly improves survival for patients with locoregionally advanced nasopharyngeal carcinoma, but the optimal induction regimen remains unclear. OBJECTIVE: To determine whether induction chemotherapy with paclitaxel, cisplatin, and capecitabine (TPC) improves survival vs cisplatin and fluorouracil (PF) prior to chemoradiotherapy for patients with stage IVA to IVB nasopharyngeal carcinoma. DESIGN, SETTING, AND PARTICIPANTS: This randomized, open-label, phase 3 clinical trial recruited 238 patients at 4 hospitals in China from October 20, 2016, to August 29, 2019. Patients were 18 to 65 years of age with treatment-naive, nonkeratinizing stage IVA to IVB nasopharyngeal carcinoma and an Eastern Cooperative Oncology Group performance status of 0 to 1. INTERVENTIONS: Patients were randomly assigned (1:1) to receive induction chemotherapy with two 21-day cycles of TPC (intravenous paclitaxel [150 mg/m2, day 1], intravenous cisplatin [60 mg/m2, day 1], and oral capecitabine [1000 mg/m2 orally twice daily, days 1-14]) or PF (intravenous cisplatin [100 mg/m2, day 1] and fluorouracil [800 mg/m2 daily, days 1-5]), followed by chemoradiotherapy. MAIN OUTCOMES AND MEASURES: The primary end point was failure-free survival in the intention-to-treat population. Secondary end points included distant metastasis–free survival, locoregional relapse–free survival, overall survival, tumor response, and safety. RESULTS: Overall, 238 eligible patients (187 men [78.6%]; median age, 45 years [range, 18-65 years]) were randomly assigned to receive TPC (n = 118) or PF (n = 120). The median follow-up duration was 48.4 months (IQR, 39.6-53.3 months). Failure-free survival at 3 years was 83.5% (95% CI, 77.0%-90.6%) in the TPC group and 68.9% (95% CI, 61.1%-77.8%) in the PF group (stratified hazard ratio [HR] for recurrence or death, 0.47; 95% CI, 0.28-0.79; P = .004). Induction with the TPC regimen resulted in a significant reduction in the risk of distant metastases (stratified HR, 0.49 [95% CI, 0.24-0.98]; P = .04) and locoregional recurrence (stratified HR, 0.40 [95% CI, 0.18-0.93]; P = .03) compared with the PF regimen. However, there was no effect on early overall survival (stratified HR, 0.45 [95% CI, 0.17-1.18]; P = .10). The incidences of grade 3 to 4 acute adverse events and late-onset toxicities were 57.6% (n = 68) and 13.6% (16 of 118), respectively, in the TPC group and 65.8% (n = 79) and 17.9% (21 of 117), respectively, in the PF group. One treatment-related death occurred in the PF group. CONCLUSIONS AND RELEVANCE: This randomized clinical trial found that induction chemotherapy with 2 cycles of TPC for patients with stage IVA to IVB nasopharyngeal carcinoma improved failure-free survival compared with 2 cycles of PF, with no increase in the toxicity profile. Recently, nanozymes have been widely applied for the enhancement of reactive oxygen species (ROS) and hypoxia, which is a major obstacle to the quick repairing of skin wound. However, their intrinsic low bioavailability and biosafety is still a great challenge for the broad introduction in the wound healing. Meanwhile, the fabrication of hydrogel wound dressing mostly accompany the complicated polymer modification, synthetic polymer usage and following pollution and health risk. Nanocellulose (CNC) has been remarked as a good candidate for functional hydrogel dressing, however, its role is generally confined to a simple mechanical reinforce or Schiff base participant in the oxidized form. With this regards, in the present paper, we first developed a ROS-scavenging, MnO2-curcumin nanocomplex stabilized by bovine serum albumin (BSA) (called BMC NPs). Dopamine was oxidative polymerized on the nanocellulose chain to form the CNC-polydopamine (CNC-PDA) in a facile way, and cross-linked quickly with thiol-fortified gelatin (Gel-SH) via the catechol-thiol addition to produce the adhesive, self-healing and hemostatic hydrogel. The CNC-PDA + Gel-SH hydrogel loading BMC NPs showed an excellent anti-oxidant, and anti-bacterial capacity. In vivofull-thickness wound experiment also confirmed that CNC-PDA + Gel-SH + BMC NPs hydrogel has a good re-epithelization, collagen synthesis, ROS clearance and anti-inflammation with the NIR treatment. Our result demonstrates the promising effect of nanocomposite -biomolecule conjugate in the wound healing as well as the great potential of PDA-coating chemistry in the hydrogel preparation.

Pentagalloylglucose (PGG), a gallotannin polyphenolic compound, has been found to possess a host of beneficial pharmacologic activities, such as anti-inflammatory and antioxidative activities. We previously demonstrated that PGG is capable of binding to the cell membrane of renal mesangial cells, but the pharmacological effect of PGG on diabetic renal injury and the underlying mechanisms are still not yet clear. In this study, the effects of PGG on Nrf2/HO-1 and JAK2/STAT3 signaling were explored in AGE-stimulated mesangial cells. Furthermore, the Nrf2 transcriptional inhibitor ML385 was used to verify the involvement of Nrf2 in the PGG-mediated inhibition of the JAK2/STAT3 cascade. Our results showed that PGG significantly inhibited AGE-induced ROS generation and activated AGE-inhibited Nrf2/HO-1 signaling. Moreover, AGE-induced inflammatory cytokines (IL-1β and TNF-α) and their signaling through JAK2/STAT3 were blocked by PGG. Furthermore, ML385 suppressed Nrf2/HO-1 signaling, elevated ROS and cytokine production, and activated JAK2/STAT3 cascade were reversed by PGG. These findings indicate that PGG inhibits the JAK2/STAT3 cascade by activating Nrf2/HO-1 signaling.

Recently, the g-C3N4-based heterojunctions have been widely investigated for their greatly enhanced photogenerated carrier separation efficiency. However, most studies are based on the study of g-C3N4powders. In this study, a novel TiN/C3N4/CdS nanotube arrays core/shell structure is designed to improve the photoelectrochemical catalytic performance of the g-C3N4-based heterojunctions. Among them, TiN nanotube arrays do not respond to simulated solar light, and thus only serve as an excellently conductive nanotube arrays backbone for supporting g-C3N4/CdS heterojunctions. g-C3N4prepared by simple liquid atomic layer deposition, which possesses appropriate energy band position, mainly acts as the electron acceptor to transport and separate electrons. Deposited CdS quantum dots obtained by successive ionic layer adsorption reaction can effectively absorb visible light and thus act as a light absorber. The TiN/C3N4/CdS nanotube arrays core/shell structure could be verified by X-ray diffractions, Raman spectra, scanning electron microscopy, transmission electron microscopy, energy dispersive spectroscopy elemental mappings and X-ray photoelectron spectroscopy. Compared with TiN/C3N4nanotube arrays, the TiN/C3N4/CdS samples greatly improve the photoelectrochemical performance, which can be evaluated by photoelectrochemical tests and photoelectrochemical catalytic degradation. Especially, the optimized photocurrent density of TiN/C3N4/CdS has almost 120 times improvement on TiN/C3N4at 0 V bias under simulated sunlight, which can be ascribed to the effective expansion of the light absorption range and separation of electron-hole pairs.

Stimulator-of-interferon genes (STING) is vital for sensing cytosolic DNA and initiating innate immune responses against microbial infection and tumors. Redox homeostasis is the balance of oxidative and reducing reactions present in all living systems. Yet, how the intracellular redox state controls STING activation is unclear. Here, we show that cellular redox homeostasis maintained by glutathione peroxidase 4 (GPX4) is required for STING activation. GPX4 deficiency enhanced cellular lipid peroxidation and thus specifically inhibited the cGAS–STING pathway. Concordantly, GPX4 deficiency inhibited herpes simplex virus-1 (HSV-1)-induced innate antiviral immune responses and promoted HSV-1 replication in vivo. Mechanistically, GPX4 inactivation increased production of lipid peroxidation, which led to STING carbonylation at C88 and inhibited its trafficking from the endoplasmic reticulum (ER) to the Golgi complex. Thus, cellular stress–induced lipid peroxidation specifically attenuates the STING DNA-sensing pathway, suggesting that GPX4 facilitates STING activation by maintaining redox homeostasis of lipids.

Toxic trace elements (TEs), such as Hg, Cr, As, and Pb, emitted from coal-fired industrial boilers (CFIBs) have gained considerable attention because of their harmful effects on environmental quality and human health. In this study, based on the field test data, Hg, Cr, Pb, and As emissions from six CFIBs in China were investigated. Solid samples including feed coal, bottom ash, fly ash, and gypsum, desulfurization wastewater samples, and stack flue gas samples were simultaneously collected for determining the internal partitioning behavior of TEs from these CFIBs. Results showed that Hg, Cr, Pb, and As concentrations in feed coal varied considerably, which were in the range of 0.02–0.22 mg/kg (Hg), 4.75–19.85 mg/kg (Cr), 4.85–35.00 mg/kg (Pb), and 0.50–9.35 mg/kg (As). The relative enrichment factors (REFs) of TEs in fly ash were all higher than those of the corresponding bottom ash, showing the stronger enrichment capacity of TEs in fly ash. TE concentrations from stack flue gas were in the range of 0.11–0.60 μg/Nm3(Hg), 3.56–23.87 μg/Nm3(Cr), 0.35–4.20 μg/Nm3(Pb), and 0.63–5.17 μg/Nm3(As). Current air pollution control devices (APCDs) have obviously positive effects on TE removal, with the integrated removal efficiencies of APCDs in the range of 80.97–98.86% (Hg), 85.82–99.45% (Cr), 98.13–99.99% (Pb), and 70.47–99.92% (As). TEs were mainly enriched in fly ash in the range of 55.22–96.67% (Hg), 56.97–85.76% (Cr), 41.68–91.99% (Pb), and 52.20–95.29% (As). These results could provide new insights into TE synergistic control in the CFIBs and assist policy making.

A series of La0.5K0.5Mn1−yPdyO3catalysts with different doping amounts of Pd (y= 0, 0.01, 0.02, 0.03, 0.04) were synthesized by the citric acid complexation method and tested for the simultaneous removal of NOxand soot. The X-ray diffraction (XRD), specific surface area and scanning electron microscopy (SEM) results reveal the microscopic morphology and structure of the catalysts, and the catalytic performance of the materials was investigated by temperature-programmed experiments. The noble metal Pd was uniformly dispersed on the surface of the materials, as shown in the SEM images. Among all the catalysts, La0.5K0.5Mn0.97Pd0.03O3exhibited the best NOxremoval rate and the lowest soot ignition temperature. The mechanism of Pd in the simultaneous removal of NOxand soot was characterized by XPS, HRTEM and H2-TPR (the redox properties), and the results illustrate that Pd2+substitution for Mn3+and generation of oxygen vacancies are the key factors in promoting soot combustion. Based upon the NOx-storage test and kinetic calculation, the excellent catalytic performance of a small amount of palladium (0.03 atomic ratio) was confirmed, and this system exhibited the high simultaneous removal rate of NOxand soot under lean conditions.

ABSTRACTRegulatory mechanisms of microRNAs (miRNAs) in the development of intestinal sepsis are unclear. This study investigated the role of rno-miR-146b-5p in sepsis-induced intestinal injury. A rat sepsis model was created using the cecal ligation and puncture method. The expression profiles of miRNA and mRNA in sepsis rats were examined using miRNA and mRNA sequencing; rno-miR-146b was selected for further investigation. The mimics and inhibitors of rno-miR-146b-5p were transfected into IEC-6 cells and then with or without lipopolysaccharide (LPS) treatment, and the expressions of Kruppel-like factor 4 (Klf4) and Cyclin D2 (Ccnd2) were assessed by quantitative real-time transcriptase-polymerase chain reaction (qRT-PCR) and western blotting. Next, cell counting kit-8 assay was used to detect cell viability, and scratch wound healing assay was used to assess cell migration. In sepsis rat model, crypt cell proliferation was inhibited and crypt cell apoptosis was increased. Compared with the sham control, results of miRNA and mRNA sequencing showed that there were 17 miRNAs and 1617 mRNAs that were upregulated and 123 miRNAs and 1917 mRNAs that were downregulated in the sepsis model group. The network diagrams and qRT-PCR validation indicated that rno-miR-146b-5p may inhibit the expression of Klf4. By adjusting the expression of rno-miR-146b-5p in IEC-6 cells with or without LPS treatment, we found that increased expression of rno-miR-146b-5p inhibited cell proliferation and migration and inhibited the expression of Ccnd2. rno-miR-146b-5p may play a vital role in the development of sepsis intestinal injury through targeting Klf4 expression and affecting promoter activity of Ccnd2.
