Two methods for quantifying elemental impurities in a highly potent analgesic activity agent (2R,4aR,7R,8aR)-4,7-dimethyl-2-(thiophen-2-yl)octahydro-2H-chromen-4-ol have been developed using acid decomposition assisted by microwave digestion system or heating in metallic block followed by analyte determination using microwave-induced plasma optical emission spectrometry (MIP OES). Both methods have been validated. The fast linear interference correction (FLIC) and inter-element correction (IEC) methods have been used to take into account possible spectral interferences. Arsenic and mercury have been determined using the multimode sample introduction system (MSIS). The detection limits have been calculated (µg L−1) – 5.4 for Cd, 5.1 for Pb, 11 for As, 1.3 for Hg, 10 for Co, 2.4 for V, 1.4 for Ni. The accuracy of the results was established using an independent inductively coupled plasma mass spectrometry (ICP-MS) method. The relative standard deviation (RSD) (n ≥ 4) was ≤ 12% for Cd, 3.8% for Pb, 6.2% for As, 11% for Hg, 3.0% for V (sample decomposition assisted by heating on metallic block), and 10% for Ni (sample decomposition assisted by microwave radiation). It was shown that the concentration of Cd, As, Hg, Co, V, Ni, Pd, Ru, Ag, Pt, and Mo as impurities in (2R,4aR,7R,8aR)-4,7-dimethyl-2-(thiophen-2-yl)octahydro-2H-chromen-4-ol was lower than the upper limit concentration.

Condensation of (−)-isopulegol with acetone was investigated at 25–40 °C over clay based materials (K10, cloisite Na+and halloysite nanotubes) modified with sulfonic acid groups by one-pot sulfonation with chlorosulfonic acid or via organosilylation with 2-(4-chlrosulfonylphenyl)-ethyltrimethoxysilane. The target product was the Rdiastereomer of dimethyl-substituted octahydro-2H-chromen-4-ol with exhibits antiviral activity. The catalysts were characterized by a range of physico-chemical methods. The effect of the initial isopulegol concentrations on the yield of ketone-derived chromenols and the R/Sisomers ratio was studied for the first time. The best catalyst was a highly acidic, large pore K10-clay modified by sulfonic acid. Varying the initial concentration of the reagents allows to find a balance between dehydration by-products and a side etherification reaction. A high yield (73%) of the desired chromenols was reached using 0.52 mol/l initial (−)-isopulegol concentration after 240 min at 87% substrate conversion and reaction temperature 25 °C. The yield exceeded that of other previously reported in the open literature catalysts.

Currently, the spectrum of agents against orthopoxviruses, in particular smallpox, is very narrow. Despite the fact that smallpox is well controlled, there is, for many reasons, a real threat of epidemics associated with this or a similar virus. In order to search for new low molecular weight orthopoxvirus inhibitors, a series of amides combining adamantane and monoterpene moieties were synthesized using 1- and 2-adamantanecarboxylic acids as well as myrtenic, citronellic and camphorsulfonic acids as acid components. The produced compounds exhibited high activity against the vaccinia virus (an enveloped virus belonging to the poxvirus family), which was combined with low cytotoxicity. Some compounds had a selectivity index higher than that of the reference drug cidofovir; the highest SI = 1123 was exhibited by 1-adamantanecarboxylic acid amide containing the (−)-10-amino-2-pinene moiety. The produced compounds demonstrated inhibitory activity against other orthopoxviruses: cowpox virus (SI = 30–406) and ectromelia virus (mousepox virus, SI = 39–707).

Background: Despite a variety of drugs used to stop acute pain, problems related to their insufficient efficacy and undesirable side effects have remained unresolved. Therefore, the search for analgesics of new structural types, which combine high activity with low toxicity, is a topical issue. It is known that a number of compounds with a hydrogenated 2H-chromene skeleton exhibit significant analgesic activity in in vivo tests. Methods: New hydro-2H-chromenols containing monoterpenoid moieties were obtained via one-pot synthesis by the interaction between para-menthane alcohols and commercially available monoterpene aldehydes: Citral, hydroxycitronellal, myrtenal, and perillaldehyde. The analgesic activity of these compounds wаs studied in the acetic acid-induced writhing test and hot plate test. Results: The target compounds were characterized using NMR and HR-MS. Most of them exhibited pronounced analgesic activity. Conclusion: Due to high analgesic activity, (2S,4aR,8R,8aR)-2-((E)-2,6-dimethylhepta-1,5-dien-1- yl)-4,7-dimethyl-3,4,4a,5,8,8a-hexahydro-2H-chromene-4,8-diol is considered as candidate compound to participate in further research.

Background: Striatal-enriched Tyrosine Phosphatase (STEP) plays a key role in the mechanisms of neuronal signaling and is a potential molecular target for new generation of psychotropic drugs. STEP inhibitor, 8-(trifluoromethyl-1,2,3,4,5-benzopentathiepin-6-amine hydrochloride (TC-2153), shows anxiolytic effect on mice. Zebrafish (Danio rerio) is a suitable model for the study of anxiety pharmacology. Objective: The objective of this study is to investigate the effects of acute and chronic TC-2153 treatment on zebrafish anxiety-related behavior. Methods: The effects of acute (0.125 and 0.25 mg/l, 3 h) and chronic (0.125 mg/l, 14 days) administration of TC-2153 on locomotion and anxiety-related behavior (time spent near the bottom and mean distance from the bottom) of adult zebrafish in the Novel Tank (NT) test were compared with those of the same doses of fluoxetine chosen as a positive control. Results: Acute treatment with 0.125 mg/l and 0.25 mg/l of TC-2153 or fluoxetine decreased time spent near the bottom, increased time spent near the surface and increased mean distance from the bottom of tank. Chronic treatment with 0.125 mg/l of TC-2153 reduced only time spent near the tank bottom without any effect on time spent near the surface and mean distance from the bottom, while chronic administration of 0.125 mg/l of fluoxetine altered these three indices of anxiety. Conclusion: Both acute and chronic TC-2153 produces anxiety-like effect indicating STEP involved in the mechanism of anxiety-related behavior in zebrafish. At the same time, chronic treatment with TC-2153 reduced locomotor activity. Zebrafish is a promising laboratory object to study the role of STEP in the nervous system.

Background: Inhibition of the DNA repair enzyme, tyrosyl-DNA phosphodiesterase 1 (TDP1), may increase the efficacy of cancer drugs that cause damage to tumor cell DNA. Among the known TDP1 inhibitors, there are compounds containing moieties of natural substances, e.g., monoterpenoids. In this work, we synthesized several compounds containing aromatic/ heteroaromatic amines and monoterpenoid groups and assessed their TDP1 inhibition potential. Methods: Structures of all the synthesized compounds were confirmed by 1H and 13C NMR as well as HRMS. The TDP1 inhibitory activity of the amines was determined by real-time fluorescence oligonucleotide biosensor. Results: The synthesized secondary amines had TDP1 inhibitory activity IC50 in the range of 0.79-9.2 μM. The highest activity was found for (–)-myrtenal derivatives containing p-bromoaniline or m-(trifluoromethyl)aniline residue. Conclusion: We synthesized 22 secondary amines; of these, 17 amines are novel chemical structures. Many of the amines inhibit TDP1 activity in the low micromolar range. Therefore, these compounds are promising for further study of their antiproliferative activity in conjunction with DNA damaging drugs.

Background: Parkinson's disease (PD) is the second most common neurodegenerative disorder worldwide, the lifetime risk of developing this disease is 1.5%. Motor diagnostic symptoms of PD are caused by degeneration of nigrostriatal dopamine neurons. There is no cure for PD and current therapy is limited to supportive care that partially alleviates disease signs and symptoms. As diagnostic symptoms of PD result from progressive degeneration of dopamine neurons, drugs restoring these neurons may significantly improve treatment of PD. Method: A literature search was performed using the PubMed, Web of Science and Scopus databases to discuss the progress achieved in the development of neuroregenerative agents for PD. Papers published before early 2018 were taken into account. Results: Here, we review several groups of potential agents capable of protecting and restoring dopamine neurons in cultures or animal models of PD including neurotrophic factors and small molecular weight compounds. Conclusion: Despite the promising results of in vitro and in vivo experiments, none of the found agents have yet shown conclusive neurorestorative properties in PD patients. Meanwhile, a few promising biologicals and small molecules have been identified. Their further clinical development can eventually give rise to disease-modifying drugs for PD. Thus, intensive research in the field is justified.

Background and Objective: The DNA repair enzyme tyrosyl-DNA-phosphodiesterase 1 (TDP1) is a current inhibition target to improve the efficacy of cancer chemotherapy. Previous studies showed that compounds combining adamantane and monoterpenoid fragments are active against TDP1 enzyme. This investigation is focused on the synthesis of monoterpenoid derived esters of 1-adamantane carboxylic acid as TDP1 inhibitors. Methods: New esters were synthesized by the interaction between 1-adamantane carboxylic acid chloride and monoterpenoid alcohols. The esters were tested against TDP1 and its binding to the enzyme was modeling. Results: 13 Novel ester-based TDP1 inhibitors were synthesized with yields of 21–94%; of these, nine esters had not been previously described. A number of the esters were found to inhibit TDP1, with IC50 values ranging from 0.86–4.08 μM. Molecular modelling against the TDP1 crystal structure showed a good fit of the active esters in the catalytic pocket, explaining their potency. A non-toxic dose of ester, containing a 3,7- dimethyloctanol fragment, was found to enhance the cytotoxic effect of topotecan, a clinically used anti-cancer drug, against the human lung adenocarcinoma cell line A549. Conclusion: The esters synthesized were found to be active against TDP1 in the lower micromolar concentration range, with these findings being corroborated by molecular modeling. Simultaneous action of the ester synthesized from 3,7-dimethyloctanol-1 and topotecan revealed a synergistic effect.

Chiral oxygen-containing heterocyclic compounds are of great interest for the development of pharmaceuticals. Monoterpenes and their derivatives are naturally abundant precursors of novel synthetic chiral oxygen-containing heterocyclic compounds. In this study, acid catalyzed reactions of salicylic aldehydes with (−)-8-acetoxy-6-hydroxymethyllimonene, readily accessible from α-pinene, leads to the formation of chiral polycyclic products of various structural types. Three of the six isolated chiral heterocyclic products obtained from salicylic aldehyde contain previously unknown polycyclic ring types. Having carried out the reaction in the presence of Brønsted or Lewis acids (Amberlyst 15, trifluoromethanesulfonic acid, trifluoroacetic acid and boron trifluoride etherate) or aluminosilicates (montmorillonite K10, halloysite nanotubes), we found that the nature of products depends on the catalyst as well as the reaction conditions (reaction time, reactant ratio, presence or absence of solvent). Detailed mechanistic insight on the complex cascade reactions for product formation is provided with extensive experimental and quantum mechanical computational studies.

Neurodegenerative disorders (NDDs) like Alzheimer's disease, Parkinson’s disease and Huntington’s disease are a heterogeneous group of disorders with the progressive and severe loss of neurons. There are no full proof cures for these diseases, and only medicines are available that can alleviate some of the symptoms. Developing effective treatments for the NDDs is a difficult but necessary task. Hence, the investigation of monoterpenoids which modulate targets applicable to many NDDs is highly relevant. Many monoterpenoids have demonstrated promising neuroprotective activity mediated by various systems. It can form the basis for elaboration of agents which will be useful both for the alleviation of symptoms of NDDs and for the treatment of diseases progression and also for prevention of neurodegeneration. The further developments including detections of monoterpenoids and their derivatives with high neuroprotective or neurotrophic activity as well as the results of qualified clinical trials are needed to draw solid conclusions regarding the efficacy of these agents.

The review integrates data on the synthesis of chiral oxygen-containing heterocycles of various structural types by reactions of monoterpenoids with carbonyl compounds. Depending on the structure of the monoterpene and carbonyl components and the catalyst used, these reactions lead to mono-, bi-, tri- or tetracyclic products. The possible reaction mechanisms are discussed and data on the biological activities of the products are given.The bibliography includes 125 references.

Background: It was found earlier that compounds combining diazaadamantane and monoterpene moieties possessed promising analgesic activity along with low acute toxicity and the lack of ulcerogenic activity. Objective: In this paper, new structural analogues of the most active compounds were synthesized and evaluated for their analgesic activity. Methods: Their structures were confirmed by various analytical methods, such as 1H and 13C NMR, HRMS. All of them were evaluated for analgesic activity at a dose of 20 mg/kg or less using acetic acid-induced writhing test and hot plate test. Results: Some compounds showed analgesic activity in acetic acid-induced writhing test. One of the synthesized compounds demonstrated high analgesic activity in both tests with 46% effect in acetic acid-induced writhing test and 89% effect in hot plate test. Both structural fragments of this compound did not possess any analgesic effect at a dose of 20 mg/kg. Conclusion: Structure-activity relationships indicated that the most active compound combines fragments of (–)-myrtenal and 6-amino-5,7-dimethyl-1,3-diazaadamantane. Both parts of the molecule are important for demonstrating analgesic activity.

Background: Depressive disorders are among four leading causes of burden among the most disabling medical illnesses and antidepressant drugs (AD) occupying leading positions in the worldwide pharmaceutical market. However, typical AD show prolonged delay, refractory and adverse side effects. Methodology: The review analyzes experimental data and theoretical models of possible antidepressant effect of benzopentathiepine, 8-(trifluoromethyl)-1,2,3,4,5 - benzopentathiepin-6- amine hydrochloride (TC-2153). The antidepressant effect of TC-2153 is based on its ability to inhibit striatal enriched protein tyrosine phosphatase (STEP)- an enzyme that plays a key role in the intraneuronal transduction of signals from receptors of classic neurotransmitters and the brain derived neurotrophic factor (BDNF). The mechanism underlying antidepressant effect of TC-2153 includes increase in production as well as intracellular effect of BDNF on neuronal survival and neurogenesis. Results and Conclusion: There is no visible mechanisms of delay, refractory and adverse side effects of TC-2153. Thus, TC-2153 is a promising drug of new generation of atypical AD.

Background: The positional effects of the methoxy- and hydroxyl substituents in the phenyl ring were examined in vivo for distinct receptor classes in order to gain an insight into the mechanism by which isomeric compounds (2S,4R,4aR,8R,8aR)-2-(3(4)-hydroxy-4(3)-methoxyphenyl)-4,7-dimethyl- 3,4,4a,5,8, 8a-hexahydro-2H-chromene-4,8-diols 1 and 2 exhibit their pharmacological activity. Conclusion: Our findings suggest a strong structure-function relationship between the substitution pattern and the mechanism of biological activity of compound. The methoxy substituent at C4 and the hydroxyl substituent at C3 (compound 1) seem to employ the cannabinoid and adrenergic systems, whereas compound 2 with the methoxy substituent at C3 and the hydroxyl substituent at C4 possibly targets the opioid and dopaminergic mechanisms.

The brain serotonin (5-HT) and dopamine (DA) systems are targets for many clinically effective antidepressants. Here, we studied the effects of acute administration of a new potential antidepressant drug, 8-(trifluoromethyl)-1,2,3,4,5 - benzopentathiepin-6-amine hydrochloride (TC-2153), on the concentration of 5-HT and DA as well as their main metabolites, 5-hydroxyindoleacetic (5-HIAA) and homovanillic acids (HVA), in the brain. TC-2153 at the doses of 10, 20 or 40 mg/kg was administered by ip injection to adult C57BL/6 mouse males. The levels of 5-HT, 5-HIAA, DA and HVA in the hypothalamus, hippocampus, striatum, midbrain were assayed an hour after treatment using high performance liquid chromatography with electrochemical detection. It was found that 20 and 40 mg/kg of TC-2153 significantly increased 5-HT, 5-HIAA, DA and HVA levels as well as the HVA/DA ratio in the hypothalamus. At the same time, the drug produced no effect on 5-HT, 5-HIAA, DA and HVA levels in the hippocampus, striatum, midbrain or substantia nigra. Therefore, the hypothalamic 5-HT and DA systems are targets for acute TC-2153 injection.

Isomerization of verbenol oxide to (1R, 2R, 6S)-3-methyl-6-(prop-1-en-2-yl)cyclohex-3-ene-1,2-diol, which exhibits anti-Parkinson activity, has been investigated with large and medium pore zeolites, namely H-USY and H-ZSM-5 types of catalysts with different SiO2to Al2O3ratio in dimethylacetamide as a solvent. The catalysts were characterized with nitrogen adsorption, 27Al MAS NMR, pyridine desorption, XRD, SEM and TEM techniques. The main reaction product was the target diol with the highest selectivity of 52 % achieved with H-ZSM-5-23 (the last number corresponds to SiO2/Al2O3ratio) at 49 % conversion of verbenol oxide. The catalyst deactivation was, however, extensive with medium pore zeolites, compared to large pore zeolites. The second highest selectivities to diol with H-USY-80 and H-ZSM-5-80 were about 47–50 mol% at 100 and 78 % of conversion, respectively, achieved at 140 °C within 3 and 5 h. The other main products were the corresponding cyclopentylhydroxyketone and oxetane. A reaction mechanism was proposed.

A series of secondary amines combining monoterpenoid and aminoadamantane moieties have been synthesized. Their cytotoxic activity against human cancer cells CEM-13, MT-4, and U-937 has been studied for the first time. Most of the obtained compounds exhibited a significant cytotoxic activity with the median cytotoxic dose (CTD50) ranging from 6 to 84 μM. The most promising results were obtained for compound 2b which was synthesized from 1-aminoadamantane and (-)-myrtenal and revealed a high activity against all tumor lines used (CTD50 = 12÷21 μM) along with low toxicity with respect to MDCK cells (CTD50 = 1500 μM). The synthesized amines do not exert the genotoxic effect on cells of the biosensor strain based on recombinant E. coli cells bearing the pRAC-gfp plasmid.

Reduction of several polynitroarenes with isopropanol in a flow-type reactor in the presence of Al2O3catalyst was studied. Temperature-controlled highly selective reduction of one, two, or three nitro groups in all the studied polynitroarenes proved to be feasible. N-alkylation of the reaction products was the predominant side reaction at high temperatures, except for the reduction of 2,7-dinitronaphthalene: in this case, a considerable amount of alkoxylated product was obtained. Benzo[c]cinnoline was the main product of reduction of 2,2′-dinitrobiphenyl.

Studies of the neuroprotective activity of a series of monoterpene-derived compounds on the MPTP mice model showed that 4-[(3aR,7aS)-1,3,3a,4,5,7a-hexahydro-3,3,6-trimethylisobenzofuran-1-yl]-2-methoxyphenol 15 possessed a considerable neuroprotective activity. Compound 15 was synthesized by one-stage reaction of (+)-2-carene with vanillin. It has low acute toxicity and significantly decreased the development of catalepsy evoked by haloperidol in rats.

Creation of effective antidepressant drugs is the key problem of the modern neuropharmacology. Here, the effect of acute administration of a new potential psychotropic drug, 8-(trifluoromethyl)-1,2,3,4,5-benzopentathiepin-6- amine hydrochloride (TC-2153) (10 and 20 mg/kg), on depressive-like behavior in the forced swim test (FST), anxietylike behavior and locomotion in the open field (OFT) and in the elevated plus-maze (PMT) tests was compared with those of classic antidepressant drugs, fluoxetine (20 mg/kg) and imipramine (20 mg/kg). Experiments were performed on adult male mice of AKR.CBA-D13Mit76 recombinant strain. TC-2153 at the doses of 10 or 20 mg/kg as well as fluoxetine and imipramine significantly decreased immobility time in the FST. Both doses of TC-2153 did not alter mouse behavior in the OFT. At the same time, imipramine significantly reduced the distance travelled and the number of rearing bouts, while fluoxetine decreased the number of rearing bouts in the OFT. All drugs produced no effect on the time spent in the center of the OFT and in the open arms in the PMT. These results suggest that TC-2153 is a promising psychotropic drug with the antidepressant-like activity similar to imipramine or fluoxetine, but lacking any visible negative side effect (reduction of locomotion or exploratory behavior).

High selectivity (82 %) to 3-methyl-6-(prop-1-en-2-yl)cyclohex-3-ene-1,2-diol, a potent anti-Parkinson drug, was achieved in the isomerization of verbenol oxide at the conversion level of 96 % in N,N-dimethylacetamide at 140 °C over a metal modified zeolite, Fe-Beta-300 with SiO2to Al2O3ratio of 300. This catalyst exhibits both weak and medium strong Brønsted and Lewis acid sites.

Compounds with different sets of three of the four functional groups of (1R,2R,6S)-3-methyl-6-(prop-1-en-2- yl)cyclohex-3-ene-1,2-diol 1 possessing high antiparkinsonian activity were synthesized. The synthesized compounds were tested for the antiparkinsonian activity in vivo on a mouse model with MPTP neurotoxin. A pronounced antiparkinsonian effect of 1 can only be achieved if it contains all the four functional groups (two hydroxy groups and two double bonds). The 2-hydroxy group or the 3,4-double bond is not required for stimulating the exploratory activity of the animals.

A set of new nitrogen-containing compounds with adamantane and monoterpene frameworks was synthesized. The antiviral activity of the amine products against the influenza virus A/California/07/09 (H1N1)pdm09 was studied. The introduction of a monoterpenoid fragment led to increase of the antiviral activity of adamantylamine derivatives against adamantylamine resistant virus: the selectivity index of the majority of the synthesized amines was higher than that of rimantadine and amantadine.

8-(Trifluoromethyl)-1,2,3,4,5-benzopentathiepine-6-amine 1a and its hydrochloride revealed significant analgesic activity in the acetic acid-induced writhing test, and ED50 of the compound 1a was 0.66 mg/kg. In the hot plate pain test, the reliable analgesic activity was found in hydrochloride 1aHCl, whereas the free base 1a proved to be inactive. Based on the results of the naloxone test, the analgesic effect of the compound 1a*HCl in both tests is mediated with the opioidergic system. Both the free base 1a and its hydrochloride did not show the anti-inflammatory activity. Replacement of the CF3 group for the fluorine atom resulted in complete loss of analgesic activity in the acetic acidinduced writhing test.

A number of new derivatives of monoterpenoid with a para-menthane framework 3-methyl-6-(prop-1-en-2- yl)cyclohex-3-ene-1,2-diol (-)-3 were synthesized. The antiviral activity of (-)-3 and its derivatives against the pandemic influenza virus A/California/07/09 (H1N1)v was studied in vitro. Compound (-)-3 was found to be active against this virus (selectivity index 7.5); for mononicotinate ()-6 the selectivity index was 17. The absolute configuration of compound 6 was shown to be critical for its antiviral activity.

The reactions of benzodithiol-2-ones containing a 4-nitro group with NaHS led to mixtures of 6- aminobenzopentathiepines and 4-nitrobenzotrithiols. The product ratio and yields depend on the substituent in the aromatic ring. Based on the yields of benzopentathiepines, the following series of substituent efficiency can be inferred: CF3 F, Cl CN. Aminobenzopentathiepines are probably formed via the intermediate benzotrithiols; the transformation apparently starts with the reduction of the nitro group.

High anticonvulsive and anxiolytic activities of 8-trifluoromethyl-6-amino-benzopentathiepin 5, which can be synthesized by new simple and effective method, have been found. The transformation of the amino group in compound 5 to the amide group drastically changed the biological activity of the compounds.

Published data on the asymmetric synthesis of chiral biologically active sulfoxides by oxidation of appropriate sulfides catalyzed by titanium and vanadium complexes are generalized. Bibliography -- 87 references.

The reactivity of verbenone epoxide (5), a terpenoid from the pinane series, towards different aliphatic and aromatic aldehydes in the presence of natural montmorillonite (askanite–bentonite) clay has been studied. A series of mechanistically different transformations afforded a number of new, optically active, polyfunctional compounds (7, 8, 10–14, 16). These products are potentially interesting synthons in asymmetric synthesis.

The reaction of salicylaldehyde with some dienes and hydroxyolefins of natural origin on the askanite-bentonite clay under mild conditions was shown to lead to a double heterocyclization forming compounds with the xanthene framework, condensed with tetrahydropyran or tetrahydrofuran rings.

The benzene ring substitution pattern of benzodithiol (I) strongly influences the reaction outcome.

Published data on the asymmetric synthesis of chiral biologically active sulfoxides by oxidation of appropriate sulfides catalyzed by titanium and vanadium complexes are generalized. Bibliography — 87 references.

The results of studies of some terpenoids, alkaloids and phenolic derivatives isolated from Siberian plants are summarised.
