Nucleus movement, positioning, and orientation is precisely specified and actively regulated within cells, and it plays a critical role in many cellular and developmental processes. Mutation of proteins that regulate the nucleus anchoring and movement lead to diverse pathologies, laminopathies in particular, suggesting that the nucleus correct positioning and movement is essential for proper cellular function. In motile cells that polarize toward the direction of migration, the nucleus undergoes controlled rotation promoting the alignment of the nucleus with the axis of migration. Such spatial organization of the cell appears to be optimal for the cell migration. Nuclear reorientation requires the cytoskeleton to be anchored to the nuclear envelope, which exerts pulling or pushing torque on the nucleus. Here we discuss the possible molecular mechanisms regulating the nuclear rotation and reorientation and the significance of this type of nuclear movement for cell migration.

Abstract: Markers of apoptosis were followed in batch hybridoma cultures carried out in protein-free medium. Samples were collected on day 0, representing early exponential phase (viability 91%), and on day 8, corresponding to late stationary phase (viability 8%). The apoptotic index reflecting the relative number of bodies insoluble in 6 M guanidinium hydrochloride in the culture of day 8 (30%) exceeded markedly the index in the culture of day 0 (2.5%). A gel chromatography on Sepharose 2B was developed for quantitative evaluation of fragmented cellular DNA. This analysis, including a correction for nonspecific fragmentation, showed that on day 8 more than 30% of cellular DNA was fragmented, whereas on day 0 it was less than 5%. Control necrotic cells prepared by rapid killing in 1% sodium azide displayed a low apoptotic index (2.4%) and low DNA fragmentation. Electrophoretic patterns in agarose gel showed a typical “ladder” of fragments in the DNA sample of day 8. The demonstration of fragmented cellular DNA and of the high incidence of apoptotic bodies at late stationary phase adds substantial weight to the view that in hybridoma cultures apoptosis represents the prevalent mode of cell death.

Expression of the dnaKand groELgenes during sporulation was assayed by determination of their mRNA levels by Northern blotting and compared with the relative level and rate of synthesis of the corresponding proteins. The ability of sporulating cells to respond to a heat shock by an increase in dnaKand groELexpression was determined at the same time. Synthesis of DnaK and GroEL encoding mRNAs during sporulation in non‐shocked cells was low suggesting that this kind of cytodifferentiation was not accompanied by enhanced synthesis of these chaperones. Also the ability of sporulating cells to respond to a heat shock by stimulating their synthesis substantially decreased during the reversible and dropped to negligible values during the irreversible sporulation phase. Nevertheless, some dependence of the heat shock response on sporulation exists because sporulation suppression by mutation or by netropsin treatment further decreased the cells' capacity to respond to a heat shock.
