TH17 differentiation is critically controlled by “signal 3” of cytokines (IL-6/IL-23) through STAT3. However, cytokines alone induced only a moderate level of STAT3 phosphorylation. Surprisingly, TCR stimulation alone induced STAT3 phosphorylation through Lck/Fyn, and synergistically with IL-6/IL-23 induced robust and optimal STAT3 phosphorylation at Y705. Inhibition of Lck/Fyn kinase activity by Srci1 or disrupting the interaction between Lck/Fyn and STAT3 by disease-causing STAT3 mutations selectively impaired TCR stimulation, but not cytokine-induced STAT3 phosphorylation, which consequently abolished TH17 differentiation and converted them to FOXP3+ Treg cells. Srci1 administration or disrupting the interaction between Lck/Fyn and STAT3 significantly ameliorated TH17 cell–mediated EAE disease. These findings uncover an unexpected deterministic role of TCR signaling in fate determination between TH17 and Treg cells through Lck/Fyn-dependent phosphorylation of STAT3, which can be exploited to develop therapeutics selectively against TH17-related autoimmune diseases. Our study thus provides insight into how TCR signaling could integrate with cytokine signal to direct T cell differentiation.

A novel self-assembling peptide-functionalized core–shell mesoporous silica nanoparticle was developed as a drug carrier. Superparamagnetic manganese- and cobalt-doped iron oxide nanoparticles formed the core for the mesoporous silica shell coating. On the silica outer shell, the peptide Boc–Phe–Phe–Gly–Gly–COOH was covalently conjugated by 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride and N-hydroxysulfosuccinimide sodium salt coupling. The self-assembling property of the peptide at physiological temperature was utilized to block the pore openings, while the disassembly at elevated local particle temperature released cargo molecules without bulk heating that would cause cell damage. Both conventional heating and heating in an alternating magnetic field were tested for the release of fluorescein and daunorubicin. In vitro experiments showed high cytotoxicity on pancreatic carcinoma cells (PANC-1) when this delivery system was activated by an alternating magnetic field, while control particles without drugs showed no obvious cytotoxicity.

Graphical abstract Abstract Although nano "green" coatings with excellent corrosion resistance have attracted great attention, the inhibition efficiency is still limited due to the lack of knowledge about the correlation between molecular structure and anticorrosion performance. Here, we fabricated a series of 3,4-dihydroxy- l -phenylalanine adlayers on self-assembled monolayers (SAMs) with varying end groups. We found that both NH 2 and CF 3 SAMs were more conducive to the adsorption of DOPA and a flat adsorption conformation was preferentially adopted, with the plane of the phenylene ring parallel to the surface via cation-π interactions or hydrophobic interactions, leading to a compact and dense adlayer. Such DOPA-SAM multilayers can effectively protect the substrate from corrosion by suppressing the diffusion of aggressive water and acid molecules as well as the electrodissolution of metals. The lowest corrosion current of adlayers reaches 6.96 μA cm−2 which is much lower than that of bare substrate and other anticorrosion surfaces reported previously. Motor vehicle ammonia (NH3) emissions have attracted increasing attention for their potential to form secondary aerosols in urban atmospheres. However, vehicle NH3emission factors (EFs) remain largely unknown due to a lack of measurements. Thus, we conducted detailed measurements of NH3emissions from 18 Euro 2 to Euro 5 light-duty gasoline vehicles (LDGVs) in Shanghai, China. The distance- and fuel-based NH3EFs average 29.2 ± 24.1 mg·km–1and 0.49 ± 0.41 g·kg–1, respectively. The average NH3-to-CO2ratio is 0.41 ± 0.34 ppbv·ppmv–1. The measurements reveal that NH3emissions from LDGVs are strongly correlated with both vehicle specific power (VSP) and the modified combustion efficiency (MCE); these relationships were used to predict LDGV NH3EFs via a newly developed model. The predicted LDGV NH3EFs under urban and highway driving cycles are 23.3 mg·km–1and 84.5 mg·km–1, respectively, which are consistent with field measurements. The NH3EF has decreased by 32% in average since the implementation of vehicle emission control policies in China five years ago. The model presented herein more accurately predicts LDGV NH3emissions, contributing substantially to the compilation of NH3emission inventories and prediction of future motor vehicle emissions in China.

ABSTRACTMycobacterium tuberculosis(M.tb), which causes tuberculosis, is a host-adapted intracellular pathogen that can live within macrophages owning to its ability to arrest phagolysosome biogenesis. The guanine nucleotide exchange factor H1 (GEF-H1) may contribute to the phagocytosis of bacteria by macrophages through mediating the crosstalk between microtubules and the actin cytoskeleton. Its role in Shigellainfection has been determined but little is known about the role of GEF-H1 in mycobacterial infection. In the present study, we demonstrated that GEF-H1 functioned as a key regulator of the macrophage-mediated anti-mycobacterial response. We found that both mRNA and protein expression levels of GEF-H1 were significantly upregulated in macrophage during mycobacterial infection. Moreover, silencing of GEF-H1 with specific siRNAs reduced the phosphorylation of p38 mitogen-activated protein kinase and TANK binding kinase 1 as well as the expression of interleukin-1β (IL-1β), IL-6, and interferon-β (IFN-β), without affecting nitric oxide production or autophagy. Importantly, GEF-H1 depletion attenuated macrophages-mediated mycobacterial phagocytosis and elimination. Taken together, our data supported that GEF-H1 was a novel regulator of inflammatory cytokine production and mycobacterial elimination, and may serve as a novel potential target for clinical treatment of tuberculosis.

Abstract

One’s response is not arbitrary when confronted with the coronavirus disease 2019 (COVID-19). Previous research has found that individuals with coronavirus stress still be more prone to overeat. We also know from other research that anxiety is the most salient aspect of overeating. However, no research has investigated whether the coronavirus stress of college students is significantly associated with overeating and examine the potential indirect pathway and moderating mechanisms in this association. With the aid of 2926 participants, we found that, coronavirus stress was linked to college students’ overeating. We also found that this relationship was partially explained by anxiety. In addition, the association between anxiety and overeating was stronger for those with higher COVID-19 burnout. This study is an important step in unpacking how coronavirus stress relates to overeating of Chinese college students. However, they are limited by the cross-sectional nature of the study, meaning we cannot imply causality. We recommend that further research replicate our findings in people with diagnosed feeding and eating disorders using a longitudinal design.

The ecological engineering pond aquaculture system was promoted in China in recent years as it can save water and reduce pollution. In the present study, an ecological engineering pond aquaculture system was established, consisting of a drainage ditch, a settling pond, an aeration pond, a filter dam, eco-ponds, and fish ponds for tilapia farming. Water from different ponds were sampled and sequenced by Illumina sequencing, and then the microbial communities were analyzed. The metabolic activity of bacteria in the water samples was tested using Biolog EcoPlates. The concentrations of total nitrogen (TN), ammonia-nitrogen (NH4-N), nitrate-nitrogen (NO3-N), nitrite-nitrogen (NO2-N), and total phosphorus (TP) in water after treatment by the system were 0.71 ± 0.19, 0.48 ± 0.15, 0.51 ± 0.21, 0.03 ± 0.01, and 0.04 ± 0.01 mg/L, respectively. All the units of the system could remove TN in water. Proteobacteria, Actinobacteria, Cyanobacteria, Bacteroidetes, and Planctomycetes were the dominant phyla in the water of the system. The genera Candidatus Aquilunaand Dechloromonaspositively correlated with the microbial metabolic activity of organic nitrogen and NH4-N concentrations in water, which could be the functional bacteria for nitrogen removal. Several genera showed positive correlation with the TN and NH4-N concentrations of water and were probably involved in nitrogen cycling of the system. Our data indicated that the ecological engineering pond aquaculture system notably improves water quality and alters the microbial populations. The results supply substantial information on the functional microbiota for nitrogen removal from freshwater aquaculture ecosystems.

The growth of GeTe thin films on a Si(111)-(√3 × √3)R30°-Sb surface is reported. At growth onset, the rapid formation of fully relaxed crystalline GeTe(0001)-(1 × 1) is observed. During growth, a GeTe(0001)-(√3 × √3)R30° surface reconstruction is also detected. Indeed, density functional theory (DFT) simulations indicate that the reconstructed GeTe(0001)-(√3 × √3)R30° structure is energetically competing with the GeTe(0001)-(1 × 1) reconstruction. The out-of-plane α-GeTe<0001>||Si<111> and in-plane α-GeTe<−1010>||Si<−211> epitaxial relationships are confirmed by X-ray diffraction (XRD). Suppression of rotational twist and reduction of twinned domains are achieved. The formation of rotational domains in GeTe grown on Si(111)-(7 × 7) is explained by domain matched coincidence lattice formation with the Si(111)-(1 × 1) surface. Atomic force microscopy (AFM) images show the coalescence of well-oriented islands with subnanometer roughness on their top part. van der Pauw measurements are performed to verify the electric properties of the films. The quality of epitaxial GeTe thin film is discussed and related to the crystalline structure of GeTe and its rhombohedrally distorted resonant bonds.

A highly unconventional growth scenario is reported upon deposition of GeTe films on the hydrogen passivated Si(111) surface. Initially, an amorphous film forms for growth parameters that should yield a crystalline material. The entire amorphous film then crystallizes once a critical thickness of four GeTe bilayers is reached, subsequently following the GeTe(111) || Si(111): GeTe[−110] || Si[−110] epitaxial relationship rigorously. Hence, in striking contrast to conventional lattice-matched epitaxial systems, a drastic improvement in atomic order is observed above a critical film thickness. Raman spectra show a remarkable change of vibrational modes above the critical thickness that is attributed to a change in the nature of the bonds: While ordinary covalent bonding is found in ultrathin films, resonant bonding can prevail only once a critical thickness is reached. This scenario is further supported by density functional theory calculations showing that ultrathin films do not utilize resonant bonding in contrast to the bulk phase. These findings are important not only for ultrathin films of phase-change materials such as GeTe and GeSbTe, which are employed in phase-change memories, but also for thermoelectrics and topological insulators such as Bi2Te3and Sb2Te3, where resonant bonding might also have a significant role.
