In vitroantidiabetic efficacy of Cucurbita maximaseed extract (CMSE) has already been studied in our previous findings. Thus, in order to validate these findings in biological system, in vivoantidiabetic activity of aqueous extract was investigated in normal as well as diabetic experimental models.

Although plants have long been a major source of medicine, there is renewed interest in studying the phytochemistry and use of herbal formulations. This paper reports spectroscopic analysis using Laser Induced Breakdown Spectroscopy (LIBS) of a polyherbal formulation, whose antidiabetic activity has also been demonstrated on rat models. LIBS analysis revealed the presence of elements such as Na, K, Mg, Ca, H, O and N. The antidiabetic study showed that amongst the four doses studied (50, 100, 150 and 200 mg/kg bw), the dose of 150 mg/kg bw registered the maximum fall in Blood Glucose Level (BGL) in both normal and diabetic (sub and mild) rats in the Glucose Tolerance Test (GTT) study—normal rats (22 %), sub-diabetic (36.6 %) and mild-diabetic (39 %). The dose of 150 mg/kg also showed the maximum fall of 23.7 % and 22 % in BGL during fasting BGL and GTT studies of normal rats, respectively. The formulation also showed significant antioxidant activity assessed using in vitro assays. The study validates for the first time the therapeutic use of an antidiabetic polyherbal formulation.

Abstract: Antidiabetic efficacy of medicinal plant cannot be ignored in order to develop drugs without toxicity and side effects. Trace elements present in plant material is responsible for their medicinal nature and hence it is necessary to know the major and minor constituents of the plant material. Laser-induced breakdown spectroscopy (LIBS) is a very efficient and powerful analytical tool to monitor the elements present in sample. The present study deals with the LIBS-based validation of glycemic elements present in Withania coagulans and Cajanus cajan medicinal herbs. The decreasing blood glucose level and improving glucose tolerance test significantly showed that the higher content of Mg and Ca in W. coagulans in comparison to C. cajan is responsible for its significant role in diabetes management, whereas the concentration of other elements are nearly the same in both extracts.

Abstract: The present study exploits the information based on concentration of trace elements and minerals in understanding the role/mechanism of action of freeze-dried fruit powder suspended in distilled water of Momordica charantia (family: Cucurbitaceae) in diabetes treatment. Laser-induced break down spectroscopy (LIBS) spectra of plant product was recorded under optimized experimental conditions and analyzed. Several atomic lines such as Na, K, Mg, Ca, Fe, Al, etc. have been observed in the LIBS spectra of the above plant product. The concentrations of these minerals are determined by using calibration-free LIBS method. Correlation between the concentration of these elements/minerals and their defined role in diabetes management was studied in normal as well as diabetic animal models.

Novel lipid (mannito-stearate) antiviral nucleoside and oligonucleotide conjugates were prepared with improved lipophilic and membrane associating properties of durgs. Potential advantages of these liponucleotide produugs are lower toxicity, increased cellular uptake, nuclease resistivity and antiviral activity. Oligonucleotide conjugate complementary to a unique segment of viral genome may selectively disrupt the processed dependent on the segment by hybridisation.

A novel fluorophore viz. 4-dansylamido-1,8-naphthalimido-N-pentanol has been designed, prepared and characterised. The comparative fluorescence has been studied in different solvents, solvent gradient, aqueous solutions of inorganic ions and buffers. This can be used for covalent tagging of oligonucleotides having potential application in Molecular Biology.
