BACKGROUND: Preeclampsia is a significant pregnancy disorder with an unknown cause, mainly attributed to impaired spiral arterial remodeling. METHODS: Using RNA sequencing, we identified key genes in placental tissues from healthy individuals and preeclampsia patients. Placenta and plasma samples from pregnant women were collected to detect the expression of TPBG (trophoblast glycoprotein). Pregnant rats were injected with TPBG-carrying adenovirus to detect preeclamptic features. HTR-8/SVneo cells transfected with a TPBG overexpression lentiviral vector were used in cell function experiments. The downstream molecular mechanisms of TPBG were explored using RNA sequencing and single-cell RNA sequencing data. TPBG expression was knocked down in the lipopolysaccharide-induced preeclampsia-like rat model to rescue the preeclampsia features. We also assessed TPBG’s potential as an early preeclampsia predictor using clinical plasma samples. RESULTS: TPBG emerged as a crucial differentially expressed gene, expressed specifically in syncytiotrophoblasts and extravillous trophoblasts. Subsequently, we established a rat model with preeclampsia-like phenotypes by intravenously injecting TPBG-expressing adenoviruses, observing impaired spiral arterial remodeling, thus indicating a causal correlation between TPBG overexpression and preeclampsia. Studies with HTR-8/SVneo cells, chorionic villous explants, and transwell assays showed TPBG overexpression disrupts trophoblast/extravillous trophoblast migration/invasion and chemotaxis. Notably, TPBG knockdown alleviated the lipopolysaccharide-induced preeclampsia-like rat model. We enhanced preeclampsia risk prediction in early gestation by combining TPBG expression with established clinical predictors. CONCLUSIONS: These findings are the first to show that TPBG overexpression contributes to preeclampsia development by affecting uterine spiral artery remodeling. We propose TPBG levels in maternal blood as a predictor of preeclampsia risk. The proposed mechanism by which TPBG overexpression contributes to the occurrence of preeclampsia via its disruptive effect on trophoblast and extravillous trophoblast migration/invasion on uterine spiral artery remodeling, thereby increasing the risk of preeclampsia. Background: Oral antibiotics (OA) combined with mechanical bowel preparation (MBP) significantly decrease the rate of surgical site infections (SSIs). However, the prophylactic effects in region-specific colorectal surgery have not been assessed. Materials and methods: A single-centre, single-blind, randomized controlled trial was conducted from 2019 to 2022. Patients were eligible if they were diagnosed with nonmetastatic colorectal malignancy, and laparoscopic colorectal surgery was indicated. Participants were randomly assigned (1:1) to the experimental (OA+MBP preparation) or control group (MBP preparation). The randomization was further stratified by resected region. The primary outcome was the incidence of SSIs. Patients were followed up for 1 month postoperatively, and all complications were recorded. Result: Between 2019 and 2022, 157 and 152 patients were assigned to the experimental and control groups, respectively, after 51 patients were excluded. The incidence of SSIs in the control group (27/152) was significantly higher than that in the experimental group (13/157; P=0.013), as was the incidence of superficial SSIs (5/157 vs. 14/152, P=0.027) and deep SSIs (7/157 vs. 16/152, P=0.042). After redistribution according to the resected region, the incidence of SSIs was significantly higher in the control group with left-sided colorectal resection (descending, sigmoid colon, and rectum) (9/115 vs. 20/111, P=0.022) but was similar between the groups with right-sided colon resection (ascending colon) (3/37 vs. 7/36, P=0.286). No differences were noted between the groups in terms of other perioperative complications. Conclusion: OA+MBP before colorectal surgery significantly reduced the incidence of SSIs. Such a prophylactic effect was particularly significant for left-sided resection. This preparation mode should be routinely adopted before elective left-region colorectal surgeries. Background and objective Non-small cell lung cancer (NSCLC) is the most common histological type of lung cancer, and one of the malignant tumor with the highest mortality. As the main part of the optical molecular imaging probe, peptide can realize the early screening and diagnosis of tumor and improve the survival rate of patients. The aim of this study was to screen the small-molecule peptide that highly binds to NSCLC NCI-H1299 cells using in vivo phage display technology and to identify their binding specificity by in vitro experiment. Methods To prepare a tumor-bearing nude mouse model of NCI-H1299 cells, after 3 rounds of in vivo screening with Ph.D.-C7CTM Peptide Library, phage clones were randomly picked, using immunohistochemistry and enzyme-linked immunosorbent assay (ELISA) to identify the affinity of phage clones to NCI-H1299 cells. The positive monoclonal phages DNA was extracted and sequenced to obtain the amino acid sequence of the peptides. The peptides with the highest repetition rate was chemically synthesized and labeled with fluorescein (FITC) to prepare optical molecular probe. We preliminary identified the specificity of the probe binding to lung cancer cells by in vitro experiment. Results After three rounds of in vivo screening, the phages enrichment rate was 341.3 times compared with the first round. Immunohistochemical staining showed that with the increase of screening times, the phages binding to tumor tissues continued to increase, and the binding amount was significantly higher than normal tissues; ELISA results showed that 20 clones among the 30 randomly selected phage clones were positive. After sequencing, the peptide with the highest repetition rate was synthesized and named NSP1; Methyl thiazolyl tetrazolium assay (MTT) and would healing assay showed that NSP1 will not affect cell proliferation and migration. Flow cytometry and immunofluorescence showed specific binding of NSP1 to NCI-H1299 cells. Conclusion We successfully obtained the peptide NSP1 that specifically binds to lung cancer NCI-H1299 cells by in vivo phage display, which provide a theoretical basis for NSCLC early diagnosis and targeted therapy. The paper analysis the dynamic relationship between energy consumption and urbanization using SVAR model with Impulse response functions and variance decomposition based on time series data of 1990-2010 in Shanxi Province. The results show that: urbanization accelerates energy consumption in the short term and decreases energy consumption in the long run, but the contribution is smaller. Therefore, it is an important way for the balance between urbanization and energy consumption to improve energy efficiency, to promote energy-saving technology, to use scientific and technological innovation, and to enhance residents awareness of energy saving. In this paper, authors analyzed the dynamic relationship between energy consumption and urbanization using PVAR model with panel data unit root tests, co-integration tests and panel VAR estimate based on provincial panel data from the year of 1990 to 2010 in China. The results show that: (1) there is a long-run co-integration relationship between Urbanization and energy consumption in the eastern, middle and western regions of China. (2) The impact of urbanization on energy consumption is significantly different in the three regions. The strongest effect is in the eastern area, followed by western area and middle area. In order to promote the harmonious development of Urbanization and energy consumption, China should improve the energy use efficiency and develop energy consumption policies according to the different relationships between urbanization and energy consumption in the eastern, middle and western areas. Several 93W-4.6Ni-1.9Fe-0.5Co alloys have been prepared by liquid phase sintering in vacuum atmosphere using powders with different contents of carbon and oxygen. The microstructures and mechanical properties of sintered samples have been investigated. Results show that the carbon in the starting powders has a stronger deoxidization ability in the high temperature vacuum sintering process due to the carbon-oxygen reaction, which decreases the contents of carbon and oxygen in the sintered samples and prevents the formation of carbide and oxide inclusions, resulting in a strong interfacial boundary between tungsten and bonding phase. The mechanical properties are improved significantly when the oxygen content is controlled to be lower than 500 μg/g. [ABSTRACT FROM AUTHOR]

Abstract: This study investigates the applicability of artificial neural networks (ANNs) to predict the performance of chiller systems. The chiller performance is affected by numerous parameters, and the relationships between them are highly nonlinear. ANNs are appropriate for tackle these complex nonlinear issues due to their learning ability and versatile mapping capabilities. An ANN model for one water chiller is presented in this paper with the objective of requiring only those input parameters that are readily available. For this aim, field investigations on the year-round chiller system have been conducted. With the experimental data, an ANN model for the system was developed based on the standard back propagation algorithm. The model was used for predicting the chiller performance parameters of compressor power input and coefficient of performance (COP). With the satisfactory predicting results, this study shows that ANN can be a promising tool for predicting the chiller''s performance for fault detection and other diagnosis purposes. The article compares the potential of metatropic glutamate receptor 1a and (RS)-1-aminoindan-1, 5-dicarboxylic acid (AIDA) in measuring the in vitro mechanical injury of the cortical neurons in rats. Both inhibitors are examined on the rats through the reverse transcription-polymerase chain reaction, which revealed results at 1 hour after the injury. Results of the study proved that metabotropic gluatamate receptor 1a reaction grew, causing AIDA to protect the cortical neurons after the injury.
