Objective: Cell division cyclin 25 homolog C (Cdc25C) is a tumor-associated antigen candidate gene, and this may be used as an effective target in cancer treatment. The present study aims to evaluate the lysis effect of cytotoxic T lymphocytes (CTLs) induced by dendritic cell line DC2.4 overexpressing Cdc25C, and the feasibility of Cdc25C as a component in hepatoma immunotherapy. Methods: The mouse Cdc25C gene was ligated into a lentiviral vector, and transfected into DC2.4 cells. The DC2.4 cell phenotype and cytokine secretion were determined by flow cytometry and ELISA, respectively. CD8+T cells were sorted from the spleens of C57BL/6 mice using a magnetic bead sorting kit obtained from Miltenyi Biotech, Germany, and co-cultured with DC2.4 cells for one week as effector cells. Then, IL-2, granzyme B and perforin were detected in the CTL culture medium by ELISA. Next, time-resolved fluorescence immunoassay was used to detect the immune killing effect of Cdc25C-specific CTLs on target cells. Meanwhile, the effect of blocking MHC-I sites on target cells with a monoclonal anti-MHC-I antibody was evaluated. Results: The results revealed that Cdc25C could be stably overexpressed in DC2.4 cells by LV-Cdc25C infection. DC2.4 cells transfected with LV-Cdc25C secreted more IL-6, IL-12, TNF-α and IFN-γ, and had higher expression levels of CD40, CD86, CCR7 and MHC-II than unaltered DC2.4 cells. The elevated Cdc25C in dendritic cells also further increased the secretion of IL-2, granzyme B and perforin to elicit Cdc25C-specific CTLs, and induced the higher cytotoxicity in Hepa1-6 cell lines (P<0.05), but this had no effect on the target cells when MHC-I monoclonal antibodies were blocked. Conclusion: DC2.4 cells transfected with LV-Cdc25C can induce specific CTLs, and result in a strong cellular immune response. The dendritic cells that overexpress Cdc25C may be useful for hepatoma immunotherapy.

In this work, a modified pretreatment method using magnetic molecularly imprinted polymers (MMIPs) was successfully applied to study the metabolites of an important botanical with ultraperformance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). The MMIPs for glucoside-specific adsorption was used to identify metabolites of Pulsatilla chinensisin rat feces. Polymers were prepared by using Fe3O4nanoparticles as the supporting matrix, d-glucose as fragment template, and dopamine as the functional monomer and cross-linker. Results showed that MMIPs exhibited excellent extraction performance, large adsorption capacity (5.65 mg/g), fast kinetics (60 min), and magnetic separation. Furthermore, the MMIPs coupled with UPLC-Q-TOF-MS were successfully utilized for the identification of 17 compounds including 15 metabolites from the Pulsatillasaponin metabolic pool. This study provides a reliable protocol for the separation and identification of saponin metabolites in a complex biological sample, including those from herbal medicines.

Ginseng is a commonly used herbal medicine in treating various medical conditions. Chronic gut inflammation is a recognized factor for the development of colorectal cancer (CRC). In this project, Asian ginseng berry polysaccharide preparations were used to assess their effects on CRC and related immune regulation mechanisms.

A field microenvironment varies with intercropping ecological planting patterns, especially its light conditions. Due to light reflection and absorption by maize leaves, spectral irradiance, red/far-red (R/FR) ratio, and photosynthetic active radiation (PAR) of the soybean canopy are decreased in a maize–soybean intercropping system as compared to those in sole cropping. In this study, a metabolomics method was applied to analyze fatty acid metabolism of soybeans grown under an intercrop shading condition. The results indicate that the fatty acid contents of the intercropped soybean were significantly higher than those of the sole-cropped soybeans. As the shading effect increased, the soybean fatty acids content showed a rise and fall tendency. Moreover, the partial least-squares discriminant analysis (PLS-DA), Pearson correlation, and hierarchical clustering analysis (HCA) multivariate analyses showed a similar result. The results suggested that shading can manipulate soybean fatty acid metabolism in a maize–soybean strip intercropping system. Further, the metabolite profiling combined with multivariate statistical analysis can be used as a useful tool for identifying the metabolic links between fatty acid metabolites.

Much attention has been paid to utilizing polydopamine (PDA) as the stationary phase in open-tubular capillary electrochromatography (OT-CEC) owing to its diverse properties, such as strong adhesion to various surfaces, latent reactivity toward amine and thiol groups and metal ion chelating/redox activities. In this study, a novel open-tubular capillary column coated with polydopamine/gold nanoparticles/thiols (PDA/Au NPs/thiols) has been fabricated based on the multiple properties of PDA for the first time. The capillary inner surface was firstly functionalized with a layer of PDA/Au NPs using the strong adhesive and metal ion redox properties of PDA. Thiols were then introduced and covalently reacted with the hybrid coating based on the Michael addition reaction of PDA and thiols and also Au–S bonds. Moreover, benefitting from the porosity of PDA, layer-by-layer (LBL) self-assembly was further applied to increase the amounts of stationary phase (Au NPs and thiols), which can significantly enhance the separation effectiveness and stability of the coated column. The formation of the PDA/Au NP/thiol coating in the capillary was confirmed and characterized by scanning electron microscopy (SEM), Energy Dispersive Spectrometry (EDS) and AFM (Atomic Force Microscopy). Then the separation effectiveness of the PDA/Au NP/thiol@capillary was verified by the separation of alkylbenzenes, which can achieve baseline separation easily with high column efficiency. In addition, the column showed long lifetime and good stability. The relative standard deviations (RSDs) for intra-day and inter-day repeatability of the PDA/Au NP/thiol@capillary were lower than 5%. Therefore, the layer-by-layer self-assembly of PDA/Au NPs/thiols on the capillary inner-surface could be an effective capillary modification strategy.

Buyang Huanwu decoction (BYHWD), a traditional Chinese herbal prescription, has been widely used clinically to treat stroke in China for hundreds of years; however, the mechanisms of this drug for stroke treatment are still unclear. This study aims to observe the cerebral angiogenesis effects of BYHWD on chronic brain injury after focal cerebral ischemia in rats and to explore its possible mechanisms. The ischemia was induced by occlusion of the right middle cerebral artery for 90 min. BYHWD (12.5 and 25.0 g/(kg·d), equivalent to the dry weight of the raw materials) was orally administered twice a day beginning 2 h after surgery. BYHWD significantly attenuated the neurological dysfunction, infarct volume, and brain atrophy after ischemia. There was a significant increase in the microvessel density, as assessed by immunofluorescence CD31, and a significant increase in angiopoietin-1 (Ang-1) in the penumbra areas of the rats was shown by immunohistochemical staining and Western blotting. The results indicate that the neurorestorative effects of BYHWD are associated with angiogenesis and the enhancement of the expressions of Ang-1 on chronic brain injury after focal cerebral ischemia.
