With the European Space Agency (ESA) and NASA working to return humans to the moon and onwards to Mars, it has never been more important to study the impact of altered gravity conditions on biological organisms. These include astronauts but also useful micro-organisms they may bring with them to produce food, medicine, and other useful compounds by synthetic biology. Parabolic flights are one of the most accessible microgravity research platforms but present their own challenges: relatively short periods of altered gravity (~20s) and aircraft vibration. Live-imaging is necessary in these altered-gravity conditions to readout any real-time phenotypes. Here we present Flight-Scope, a new microscopy and microfluidics platform to study dynamic cellular processes during the short, altered gravity periods on parabolic flights. We demonstrated Flight-Scopes capability by performing live and dynamic imaging of fluorescent glucose uptake by yeast, S. cerevisiae, on board an ESA parabolic flight. Tuberculosis (TB) is the world's deadliest infectious disease, with over 1.5 million deaths annually and 10 million new cases reported each year. The causative organism, Mycobacterium tuberculosis (Mtb) can take nearly 40 days to culture, a required step to determine the pathogen's antibiotic susceptibility. Both rapid identification of Mtb and rapid antibiotic susceptibility testing (AST) are essential for effective patient treatment and combating antimicrobial resistance. Here, we demonstrate a rapid, culture-free, and antibiotic incubation-free drug susceptibility test for TB using Raman spectroscopy and machine learning. We collect few-to-single-cell Raman spectra from over 25,000 cells of the MtB complex strain Bacillus Calmette Guerin (BCG) resistant to one of the four mainstay anti-TB drugs, isoniazid, rifampicin, moxifloxacin and amikacin, as well as a pan susceptible wildtype strain. By training a neural network on this data, we classify the antibiotic resistance profile of each strain, both on dried samples and in patient sputum samples. On dried samples, we achieve >98% resistant versus susceptible classification accuracy across all 5 BCG strains. In patient sputum samples, we achieve ~79% average classification accuracy. We develop a feature recognition algorithm in order to verify that our machine learning model is using biologically relevant spectral features to assess the resistance profiles of our mycobacterial strains. Finally, we demonstrate how this approach can be deployed in resource-limited settings by developing a low-cost, portable Raman microscope that costs <$5000. We show how this instrument and our machine learning model enables combined microscopy and spectroscopy for accurate few-to-single-cell drug susceptibility testing of BCG.

Microscopy plays a crucial role in hematology and diagnosis of infectious diseases worldwide. For malaria alone, more than 200 million slides are read by manual microscopists every year. High quality thin blood smears are essential for subsequent microscopy examinations including malaria microscopy, but are hard to make in field settings. Existing devices for assisting in making thin smears are available but are limited by cost or complexity for wider use. Here we present Inkwell, a portable mechanical device capable of making high quality thin blood smears in field settings. Inkwell is simple, low-cost, does not use electricity, and requires minimal training prior to use. By utilizing passive dissipative dynamics of a spiral spring coupled to an air dashpot with a tunable valve - we demonstrate a highly tunable mechanism for constant velocity smears at prescribed angle. Inkwell is capable of producing high quality blood smears of tunable cell density with more than 12 million individually distinguishable red blood cells on a single slide. The current design, which exploits precision manufacturing of a 17 cents plastic syringe and a spring, can be printed on a standard 3D printer with overall unit cost of less than a few dollars in large quantities. We further present usability tests to confirm performance over 10,000 unit cycle operations with no degradation in quality of the smear and demonstrate ease of use with minimal training. Inkwell enhances the broader toolbox of open innovations in diagnostics for providing high quality medical care in low and medium resource settings. Combined with rise of 3D printing, Inkwell presents an alternative to traditional centralized manufacturing and opens up distributed manufacturing of medical diagnostics in global context.

Fish swimming style and schooling behaviour are two important aspects of underwater locomotion. How swimming style affects fish schooling is investigated by a representative problem setup consisting of two tethered NACA0012 hydrofoils undulating in a free stream flow at various wavelengths $ \lambda^* = 0.8 - 8 $, front-back distance $ D = 0,\ 0.25,\ 0.5,\ 0.75 $, phase difference $ \phi/\pi = 0,\ 0.5,\ 1,\ 1.5 $, and lateral gap distance $ G = 0.25,\ 0.3,\ 0.35 $ with fixed Reynolds number $ Re = 5000 $, Strouhal number $ St = 0.4 $, and maximum amplitude $ A_{max} = 0.1 $. In total, 336 cases were simulated by open-source software IBAMR based on immersed boundary method. The increase in wavelength generally leads to higher thrust and lateral force, consistent with a previous study regarding a single swimmer. The highest Froude efficiency is obtained at intermediate wavelength $ \lambda^* = 2 $ for either individual or two foils as a group. At side-by-side arrangement $ D = 0 $, the thrust force upon the two foils can be equivalent, indicating a stable formation. At staggered arrangement, $ D > 0 $, the follower can take significant advantage of the leader in locomotion performance by tuning phase difference. The follower's benefit decreases with relative distance. Various combinations of wavelength and relative distance can lead to distinct flow structures, indicating a tunable stealth capacity of the schooling swimmers., Comment: 53 pages, 21 figures

In this paper, we propose a novel design of dielectric laser-driven accelerator (DLA) utilizing evanescent electric field of racetrack ring resonator structures. Driven by laser light with the correctly designed optical phase window, sustained acceleration of electrons with controlled deflection is shown. Based on this design, we calculate an acceleration from 30 keV to 148.312 keV in 104.655 {\mu}m using a cascaded 11-stage racetrack ring resonators. This new idea poses a solution for on-chip integration of many DLA stages, while maintains high average accelerating gradients, providing a potential practical realization for "accelerator on a chip"., Comment: 5 pages, 3 figures, 1 table

Surface enhanced Raman scattering (SERS) in nanoscale hotspots has been placed great hopes upon for identification of minimum chemical traces and in-situ investigation of single molecule structures and dynamics. However, previous work consists of either irreproducible enhancement factors (EF) from random aggregates, or moderate EFs despite better reproducibility. Consequently, systematic study of SERS at the single and few molecules level is still very limited, and the promised applications are far from being realized. Here we report EFs as high as the most intense hotspots in previous work yet achieved in a reproducible and well controlled manner, that is, electromagnetic EFs (EMEF) of 10^9~10 with an error down to 10^+/-0.08 from gold nanospheres on atomically flat gold planes under radially polarized (RP) laser excitation.
