Quantitative analysis of microRNAs (miRNAs) in a noninvasive manner is of vital importance for disease diagnosis and prognosis evaluation. However, conventional strategies for realizing accurate, simple, and sensitive detection of target molecules are still a challenge, especially for miRNAs due to their low abundance and susceptibility in the complex biological environment. Here, a novel surface-enhanced Raman scattering (SERS) strategy was established for quantitative detection and monitoring of miRNA-21-5p (miR-21-5p) in living cells and in vivo cerebrospinal fluid (CSF) by applying hairpin DNA (hpDNA)-conjugated gold nanostars (GNSs) SERS probes combined with acupuncture-based technology. This strategy enabled ultrasensitive exploration toward miR-21-5p in a wide range from 1 fM to 100 pM in cell lysates. Moreover, SERS analysis facilitated the detection and long-term monitoring for in vivo miR-21-5p noninvasively. This developed strategy promises to offer a powerful method for the analysis of multiple biomolecules in single cells and living bodies.

Diabetic wounds severely influence life, facing grand challenges in clinical treatments. The demand for better treatment is growing dramatically. Diabetic wound healing is challenging because of inflammation, angiogenesis disruptions, and tissue remodeling. Based on sequencing results of diabetic patients' skins and artificial intelligence (AI)-assisted bioinformatics, we excavate a potential therapeutic agent Trichostatin A (TSA) and a potential target histone deacetylase 4 (HDAC4) for diabetic wound healing. The molecular docking simulation reveals the favorable interaction between TSA and HDAC4. Taking advantage of the microneedle (MN) minimally invasive way to pierce the skin barrier for drug administration, we develop a swelling modified MN-mediated patch loaded with TSA to reduce the probability of injection-caused iatrogenic secondary damage. The MN-mediated TSA patch has been demonstrated to reduce inflammation, promote tissue regeneration, and inhibit HDAC4, which provides superior results in diabetic wound healing. We envisage that our explored specific drug TSA and the related MN-mediated drug delivery system can provide an innovative approach for diabetic wound treatment with simple, effective, and safe features and find a broad spectrum of applications in related biomedical fields.

Magneto-sensitive soft materials that can accomplish fast, remote, and reversible shape morphing are highly desirable for practical applications including biomedical devices, soft robotics, and flexible electronics. In conventional magneto-sensitive elastomers (MSEs), there is a tradeoff between employing hard magnetic particles with costly magnetic programming and utilizing soft magnetic particle chains causing tedious and small deformation. Here, inspired by the shape and movement of mimosa, a novel soft magnetic particle doped shape material bianisotropic magneto-sensitive elastomer (SM bianisotropic MSE) with multimodal transformation and superior deformability is developed. The high-aspect-ratio shape anisotropy and the material anisotropy in which the magnetic particles are arranged in a chainlike structure together impart magnetic anisotropy to the SM bianisotropic MSE. A magneto-elastic analysis model is proposed, and it is elucidated that magnetic anisotropy leads to peculiar field-direction-dependent multimodal transformation. More importantly, a quadrilateral assembly and a regular hexagon assembly based on this SM bianisotropic MSE are designed, and they exhibit 2.4 and 1.7 times the deformation capacity of shape anisotropic samples, respectively. By exploiting the multidegree of freedom and excellent deformability of the SM bianisotropic MSE, flexible logic switches and ultrasoft magnetic manipulators are further demonstrated, which prove its potential applications in future intelligent flexible electronics and autonomous soft robotics.
