Time-resolved measurements of changes in the size and shape of nanobiological objects and layers are crucial to understand their properties and optimize their performance. Optical sensing is particularly attractive with high throughput and sensitivity, and label-free operation. However, most state-of-the-art solutions require intricate modeling or multiparameter measurements to disentangle conformational or thickness changes of biomolecular layers from complex interfacial refractive index variations. Here, we present a dual-band nanoplasmonic ruler comprising mixed arrays of plasmonic nanoparticles with spectrally separated resonance peaks. As electrodynamic simulations and model experiments show, the ruler enables real-time simultaneous measurements of thickness and refractive index variations in uniform and heterogeneous layers with sub-nanometer resolution. Additionally, nanostructure shape changes can be tracked, as demonstrated by quantifying the degree of lipid vesicle deformation at the critical coverage prior to rupture and supported lipid bilayer formation. In a broader context, the presented nanofabrication approach constitutes a generic route for multimodal nanoplasmonic optical sensing.

The challenge of inducing and controlling localized fluid flows for generic force actuation and for achieving efficient mass transport in microfluidics is key to the development of next-generation miniaturized systems for chemistry and life sciences. Here we demonstrate a methodology for the robust generation and precise quantification of extremely strong flow transients driven by vapor bubble nucleation on spatially isolated plasmonic nanoantennas excited by light. The system is capable of producing peak flow speeds of the order mm/s at modulation rates up to ∼100 Hz in water, thus allowing for a variety of high-throughput applications. Analysis of flow dynamics and fluid viscosity dependence indicates that the transient originates in the rapid bubble expansion that follows nucleation rather than being strictly thermocapillary in nature.

The highly localized sensitivity of metallic nanoparticles sustaining localized surface plasmon resonance (LSPR) enables detection of minute events occurring close to the particle surface and forms the basis for nanoplasmonic sensing. To date, nanoplasmonic sensors typically consist of two-dimensional (2D) nanoparticle arrays and can therefore only probe processes that occur within the array plane, leaving unaddressed the potential of sensing in three dimensions (3D). Here, we present a plasmonic metasurface comprising arrays of stacked Ag nanodisks separated by a thick SiO 2 dielectric layer, which, through rational design, exhibit two distinct and spectrally separated LSPR sensing peaks and corresponding spatially separated sensing locations in the axial direction. This arrangement thus enables real-time plasmonic sensing in 3D. As a proof-of-principle, we successfully determine in a single experiment the layer-specific glass transition temperatures of a bilayer polymer thin film of poly(methyl methacrylate), PMMA, and poly(methyl methacrylate)/poly(methacrylic acid), P(MMA-MAA). Our work thus demonstrates a strategy for nanoplasmonic sensor design and utilization to simultaneously probe local chemical or physical processes at spatially different locations. In a wider perspective, it stimulates further development of sensors that employ multiple detection elements to generate distinct and spectrally individually addressable LSPR modes.

Antibody-antigen interactions are complex events central to immune response, in vivo and in vitro diagnostics, and development of therapeutic substances. We developed an ultrastable single-molecule localized surface plasmon resonance (LSPR) sensing platform optimized for studying antibody-antigen interaction kinetics over very long time scales. The setup allowed us to perform equilibrium fluctuations analysis of the PEG/anti-PEG interaction. By time and frequency domain analysis, we demonstrate that reversible adsorption of monovalently bound anti-PEG antibodies is the dominant factor affecting the LSPR fluctuations. The results suggest that equilibrium fluctuation analysis can be an alternative to established methods for determination of interaction rates. In particular, the methodology is suited to analyze molecular systems whose properties change during the initial interaction phases, for example, due to mass transport limitations or, as demonstrated here, because the effective association rate constant varies with surface concentration of adsorbed molecules.

Quasicrystals are structures that possess long-range order without being periodic. We investigate the unique characteristics of a photonic quasicrystal that consists of plasmonic Ag nanodisks arranged in a Penrose pattern. The quasicrystal scatters light in a complex but spectacular diffraction pattern that can be directly imaged in the back focal plane of an optical microscope, allowing us to assess the excitation efficiency of the various diffraction modes. Furthermore, surface plasmon polaritons can be launched almost isotropically through near-field grating coupling when the quasicrystal is positioned close to a homogeneous silver surface. We characterize the dispersion relation of the different excited plasmon modes by reflection measurements and simulations. It is demonstrated that the quasicrystal in-coupling efficiency is strongly enhanced compared to a nanoparticle array with the same particle density but only short-range lateral order. We envision that the system can be useful for a number of advanced light harvesting and optoelectronic applications.

Ultrasensitive biosensing is one of the main driving forces behind the dynamic research field of plasmonics. We have previously demonstrated that the sensitivity of single nanoparticle plasmon spectroscopy can be greatly enhanced by enzymatic amplification of the refractive index footprint of individual protein molecules, so-called plasmon-enhanced enzyme-linked immunosorbent assay (ELISA). The technique, which is based on generation of an optically dense precipitate catalyzed by horseradish peroxidase at the metal surface, allowed for colorimetric analysis of ultralow molecular surface coverages with a limit of detection approaching the single molecule limit. However, the plasmonic response induced by a single enzyme can be expected to vary for a number of reasons, including inhomogeneous broadening of the sensing properties of individual particles, variation in electric field enhancement over the surface of a single particle and variation in size and morphology of the enzymatic precipitate. In this report, we discuss how such inhomogeneities affect the possibility to quantify the number of molecules bound to a single nanoparticle. The discussion is based on simulations and measurements of large arrays of well-separated gold nanoparticles fabricated by electron beam lithography (EBL). The new data confirms the intrinsic single-molecule sensitivity of the technique but we were not able to clearly resolve the exact number of adsorbed molecules per single particle. The results indicate that the main sources of uncertainty come from variations in sensitivity across the surface of individual particles and between different particles. There is also a considerable uncertainty in the actual precipitate morphology produced by individual enzyme molecules. Possible routes toward further improvements of the methodology are discussed.
