The application of nitrogen fertilizer is crucial to the growth and biological nitrogen fixation of peanut, especially in the seedling stage where nodules have not yet formed. However, it is still uncertain how much initial nitrogen fertilizer should be applied to promote peanut root growth, nodule formation, and biological nitrogen fixation (BNF). There, a 2-year pot experiment was conducted using Huayu 22 (HY22, large-grain cultivar) and Huayu 39 (HY39, small-grain cultivar) as experimental materials to research the effects of different initial nitrogen fertilizer application rates on peanut root growth (root weight, root length, root mean diameter, root activity) and biological nitrogen fixation capacity (nodule number, nodule weight, biological nitrogen fixation, and nitrogen fixation potential per plant). N0, as control, four initial nitrogen fertilizer application rates were established: 15 kg·hm−2 (N15), 30 kg·hm−2 (N30), 45 kg·hm−2 (N45), and 60 kg·hm−2 (N60). The present results showed that the nodule number, nodule dry weight, nitrogenase activity, and biological nitrogen fixation of the HY22 cultivar under the N15 treatment were higher compared to those under other treatments over the two growing seasons. In addition, the cultivar of HY39 treated with the N15 treatment also increased the nitrogen fixation potential per plant and BNF relative to other treatments. Although the application of 60 kg·hm−2 nitrogen increased the root surface area and root volume, it decreased the nitrogenase activity, nodule dry weight, and nitrogen fixation potential per plant of HY22 and HY39 varieties in both growing seasons. Above all, an initial nitrogen application of 15 kg·hm−2 may be the optimal treatment for promoting peanut nodule formation and biological nitrogen fixation. [ABSTRACT FROM AUTHOR]

Inoculants containing strains of bacteria that fix atmospheric N that are tolerant or resistant to chemical products commonly used in seed treatment are classified as long-life inoculants, which contribute to improving the efficiency of nodulating bacteria in soybean crops. The objective of this work was to evaluate the effects of applying commonly used pesticides in seed treatment and a long-life inoculant, alone or in combination, on the nodulation process and seed quality of soybeans during storage. The first experiment was carried out in a greenhouse using a completely randomized design, in an 8 × 2 factorial arrangement, with three replications. The treatments consisted of applications of industrial seed treatments: control, MaximAdvanced, Fortenza, long-life inoculant, MaximAdvanced + Fortenza, MaximAdvanced + long-life inoculant, Fortenza + long-life inoculant, and MaximAdvanced + Fortenza + long-life inoculant. The seeds were sown in pots containing soils from two crop areas. The second experiment was carried out in a laboratory, using a completely randomized design in an 8 × 7 factorial arrangement, with four replications. The treatments consisted of the same eight treatments used in the greenhouse experiment, which were applied before packaging and storing the seeds for two months. The industrial seed treatment with the mixture of fungicide, insecticide, and inoculants improved the soybean photosynthesis and nodulation processes, regardless of the history of the area. Soybean seeds can be stored for commercial purposes for up to 40 days, regardless of the seed treatment with fungicide, insecticide, and long-life inoculant applied alone or in combination. Sustainable corn production requires a dramatic shift toward natural soil fertility rather than relying solely on synthetic fertilizers. Cover crops play an important role in improving the productivity of subsequent row crops through improving soil properties. The main goal of this study was to investigate if increasing cover crop biomass through applying a higher density can enhance soil characteristics in the short term and contribute more nitrogen to succeeding corn silage. In a two-year field study (2018–2019), the influence of faba bean (Vicia faba L.) as a cover crop on soil characteristics and corn silage (Zea mays L.) production was evaluated. Treatments consisted of five levels of faba bean density (0, 25, 35, 40, and 80 plants m−2) and four application rates of urea-based nitrogen fertilizer (0, 100, 200, and 300 kg ha−1) in a no-till system. The measured soil characteristics were not significantly affected through increasing cover crop density to 80 plants m−2. The faba bean roots comprised 33% of total biomass in densities ranging from 25–40 plants m−2. The highest total N yield (root + shoot) was 133 kg N ha−1, obtained from 40 faba bean plants m−2. The faba bean root decomposed faster than the shoot, and the addition of N to the corn accelerated 50% N release from the roots but had no significant effect on shoot decomposition. Corn planted after 40 plants m−2 faba bean yielded 28% more than the corn with no faba bean. Corn yielded less in no-cover-crop fields even when it received the highest synthetic N rate (300 kg N ha−1), indicating the value of including faba bean in rotation with corn. This study evaluated the combined effect of foliar spray inoculation with plant growth-promoting bacteria (PGPB) and nitrogen doses on the yield, development, and nutritive value of Megathyrsus maximus cv. BRS Zuri. The experimental design was randomized blocks with four replications, with repeated measures in time. Foliar inoculation of two bacteria (Azospirillum brasilense Ab-V5 (CNPSo 2083) and Ab-V6 (CNPSo 2084) and Pseudomonas fluorescens (CNPSo 2799)) and 40 and 80 kg ha−1 N (urea) rates, in addition to the control (without inoculation and N fertilization), were applied. In the rainy season, at the level of 40 kg ha−1 of N, inoculation of both A. brasilense and P. fluorescens increased, respectively, the tiller number by 33% and 25% (22 February), and the N accumulated in tissues by 42% and 25% (22 January), while in the previous year (21 February) the beneficial effects of both bacteria were observed in the percentage of leaf blade and in the true digestibility in vitro. When the foliar spray was inoculated with A. brasilense Ab-V5 and Ab-V6 and P. fluorescens CNPSo 2799, with 80 kg N ha−1, the root system of Zuri grass increased by 61% and 30%, respectively. Summer legume cover crops (CC) such as Crotalaria juncea, Crotalaria spectabilis, Crotalaria ochroleuca, and Cajanus cajan could offer diverse advantages for the environment and productive cropping systems. A low transpiration efficiency (TE) of CC can induce soil water content to levels that present a challenge for the subsequent crop. In a 75-day growth chamber experiment, using the natural abundance of 13C, 18O, and 15N we evaluated the TE and BNF under two soil water conditions. Our results showed that the four species tested are good candidates for their use as CC because they showed good results in terms of productivity parameters, TE, and BFN. Cajanus cajan had the highest TE, a high shoot dry matter production, and accumulated more N from BFN in the shoot than C. spectabilis, C. juncea, and C. ochroleuca. ∆18O increased under moderate water deficit and showed an inversely proportional relationship with the amount of transpired water, supporting the use of this isotopic indicator as a proxy for transpiration and stomatal conductance. For the isotopic parameters no interaction between the factors water regimen and species were found. We propose the mass ratio of nitrogen fixed by the volume of transpired water and the isotopic discrimination of 13C as useful indicators of drought fixing legumes tolerance.

The field experiment was carried out in 2013 and 2014 as part of a long-term experiment to test the influence of different soil tillage systems (conventional, reduced, direct sowing) on (a) the yield of summer pea (Pisum sativum L.) and (b) the amount of biologically reduced atmospheric nitrogen (N), which was determined using the 15N dilution method (ID15N). Spring barley was used as a reference plant. Climatic conditions did not have a significant influence on the yield of pea seeds (mean value 4.56 t ha−1), post-harvest residues (3.76 t ha−1) and total biomass (8.33 t ha−1). Soil tillage system was found to have a significant impact on all components of the pea yield in years of experiment, with the highest average seed yield values observed with the conventional system (5.19 t ha−1) and significantly lower values observed with reduced tillage (4.34 t ha−1) and direct sowing (4.17 t ha−1). The content of total N was greatest in the pea biomass that was harvested in the conventional system (202 kg ha−1) and lowest with direct sowing (155 kg ha−1). Nitrogen accumulated in the pea seeds from three sources: atmosphere (mean value 35.2%), fertilizers (6.8%) and soil (57.9%), equating to 48.6, 9.9, and 85.4 kg ha−1, respectively. Soil tillage system was found to have a significant impact on the amount of N that was fixed from the atmosphere by the peas: 17.7% in conventional tillage, 37.9 in reduced system and 50.2% in direct sowing, which equates to a harvested seed yield of 28.9, 52.0 and 64.4 kg ha−1, respectively. In the post-harvest residues, the amount of N fixed from the atmosphere by the pea crop was also modified by the soil tillage system in range: 20.2% in conventional tillage and 32.6% in direct sowing (which equates to 7.4 and 8.5 kg N ha−1, respectively), but the difference was not significant.

Reactive oxygen species are generated during the processes of photosynthesis and nitrate reduction, which can compromise the integrity of biomolecules and membranes. During the vegetative phase of Fabaceae species, around half of translocated carbohydrate is used for nodule growth, while the other half returns to the aerial part with nitrogen incorporated. These sugars may be yet involved with membrane stabilization, signaling, and activation of important genetic pathways for plant development. Thus, the aim was to study the adjustments of the photosynthetic and antioxidant systems and the accumulation of carbohydrates and biomass in Glycine–Bradyrhizobium cultivated with nitrate (NO3−). Four treatments were evaluated in completely randomized blocks. Glycine–Bradyrhizobium was grown with 1.7 mM of NO3− (GB: 1.7 mM NO3−) and without NO3− (GB: 0 mM NO3−), and Glycine was grown with 1.7 mM of NO3− (G: 1.7 mM NO3−) and without NO3− (G: 0 mM NO3−). Glycine–Bradyrhizobium symbiosis contributes to photosynthetic metabolism and total sugars, reduces the action of antioxidant enzymes, and minimizes the use of nitrate in soybean cultivation.; Glycine–Bradyrhizobium with nitrate provided greater plant dry mass in the vegetative phase, along with increased enzymatic activity and reduced nodule mass.

Inoculants or biofertilizers aiming to partially or fully replace chemical fertilizers are becoming increasingly important in agriculture, as there is a global perception of the need to increase sustainability. In this review, we discuss some important results of inoculation of a variety of crops with rhizobia and other plant growth-promoting bacteria (PGPB). Important improvements in the quality of the inoculants and on the release of new strains and formulations have been achieved. However, agriculture will continue to demand chemical pesticides, and their low compatibility with inoculants, especially when applied to seeds, represents a major limitation to the success of inoculation. The differences in the compatibility between pesticides and inoculants depend on their active principle, formulation, time of application, and period of contact with living microorganisms; however, in general they have a high impact on cell survival and metabolism, affecting the microbial contribution to plant growth. New strategies to solve the incompatibility between pesticides and inoculants are needed, as those that have been proposed to date are still very modest in terms of demand.

Low soil phosphorus levels in savanna soils of Ghana limit cowpea response to inoculation. A two-year experiment was carried out on 2 soil types of the Guinea and Sudan savanna zones of Ghana based on the hypothesis that Bradyrhizobia inoculant (BR3267) in combination with phosphorus and potassium fertilizer will significantly increase cowpea root nodulation, growth and yield. The study aimed to determine the effect of phosphorus and potassium fertilizer on cowpea response to Bradyrhizobia inoculant. The treatments were laid out in Randomized Complete Block Design, replicated four times. The plot size was 8 × 3 m, with the sowing distance of 60 × 20 cm. The treatment comprises of commercial Bradyrhizobia inoculant, phosphorus (0, 30, 40 kg P2O5 ha−1) and potassium (0, 10, 20, 30 K2O ha−1). Application of Bradyrhizobia inoculant with 30 kg P2O5 ha−1 and 20 kg K2O ha−1 gave the highest grain yield (1.68 and 1.86 tons ha−1) at both soils which did not differ from the yield obtained from BR-40-30 kg ha−1 P2O5 and K2O ha−1 on the Ferric Lixisol and BR-40-20 kg ha−1 P2O5 and K2O on the Ferric Luvisol. The same treatment also gave the highest nodule number and nodule dry weight. The results of this study have shown that the application of Bradyrhizobia inoculant followed by P and K fertilizer was effective for cowpea growth in field conditions.

Availability of nitrogen (N) limits pasture production. Inclusion of legumes into grass pastures can provide an alternative N source through biological N2 fixation (BNF), and enhance retention and cycling of soil C and N. Despite the use of alfalfa (Medicago sativa L.), cicer milkvetch (Astragalus cicer L.) and sainfoin (Onobrychis viciifolia Scop.) in grass-legume pastures to improve forage quality, relative BNF potentials and resource partitioning are unknown. We quantified BNF using 15N isotope dilution and estimated resource partitioning in alfalfa, two cultivars of cicer milkvetch and two cultivars of sainfoin under controlled conditions. Percentage of nitrogen derived from atmosphere followed the order alfalfa (92%) > cicer milkvetch (87%) > sainfoin (81%); corresponding to estimated N contributions of 200, 128 and 65 kg N ha−1 yr−1, respectively, based on total herbage. Root dry matter was 24% to 36% greater than shoot dry matter in all of the legumes, providing substantial below-ground C and N. Cultivars of the same species did not differ in any measured parameter (p > 0.05). Despite the lower BNF in cicer milkvetch and sainfoin compared to alfalfa, their use may not negatively affect stand productivity and C storage.

The common bean (Phaseolous vulgaris L.) is a grain legume functionally characterized by its capacity for symbiotic of biological nitrogen fixation. As such, it does not demand the application of synthetic nitrogenous fertilizer and can offer environmental benefits as part of holistic cropping systems. While common bean commodities are highly nutritious, commercial cultivation of this crop is declining in already-industrialized countries. However, recent interest of consumers towards diets that benefit environmental and personal health has rekindled commercial interest in legumes, including the common bean. The aim of this protocol is to identify agronomic practices that are capable of increasing the yield and quality of the common bean for use as food. To address this research question, published literature will be screened for inclusion on the basis of defined eligibility criteria to ensure data sources are selected in an objective and consistent manner. Consistency checks will be carried out for the title, abstract and full texts of the literature collated. The output is expected to be a summary of the knowledge available to maximize the productivity and quality of the common bean as food. This anticipated synthesis will be of utility for a wide range of value-chain stakeholders from farmers and consumers, to research scientists and policy makers.

The introduction of nitrogen fixing bacteria (NFB) and arbuscular mycorrhizal fungi (AMF) into the soil is an advisable agricultural practice for the crop, since it enhances nutrient and water uptake and tolerance to biotic and abiotic stresses. The aim of this work was to study plant nutrition, biological nitrogen fixation (BNF) and crop yield and quality, after inoculating seeds with NFBs ((Rhizobium leguminosarum, Burkholderia cenocepacia, Burkholderia vietnamiensis)) and/or AMFs (Rhizophagus irregularis, Claroideoglomus etunicatum, Claroideoglomus claroideum and Funneliformis mosseae) in a fava bean crop in two seasons. The composition of the nodule bacterial community was evaluated by the high-throughput sequencing analysis of bacterial 16 S rRNA genes. It was found that microbial inoculation accompanied by a 20% decrease in mineral fertilization had no significant effect on crop yield or the nutritional characteristics compared with a non-inoculated crop, except for an increase in the grain protein content in inoculated plants. None of the inoculation treatments increased biological nitrogen fixation over a non-inoculated level. The bacterial rRNA analysis demonstrated that the genus Rhizobium predominated in all nodules, both in inoculated and non-inoculated treatments, suggesting the previous presence of these bacteria in the soil. In our study, inoculation with Rhizobium leguminosarum was the most effective treatment for increasing protein content in seeds, while Burkholderia sp. was not able to colonise the plant nodules. Inoculation techniques used in fava beans can be considered an environmentally friendly alternative, reducing the input of fertilizers, while maintaining crop yield and quality, with the additional benefit of increasing the grain protein content. However, further research is required on the selection and detection of efficient rhizobial strains under local field conditions, above all those related to pH and soil type, in order to achieve superior nitrogen-fixing bacteria.

Nitrogen (N) in soybean (Glycine max L.) plants derived from biological nitrogen fixation was shown to be a sustainable N resource to substitute for N fertilizer. However, the limited water supply in sandy soil is a critical factor for soybean nodulation and crop growth. This study investigated the potential mechanism of the effect of biochar and irrigation on the soybean-Rhizobium symbiotic performance and soil biological activity in a field trial. In the absence of N fertilizer, 10 t ha−1 of black cherry wood-derived biochar were applied under irrigated and rainfed conditions on an experimental, sandy field site. The plant biomass, plant nutrient concentrations, nodule number, nodule leghemoglobin content, soil enzyme activities, and soil-available nutrients were examined. Our results show that biochar application caused a significant increase in the nodule number by 35% in the irrigated condition. Shoot biomass and soil fluorescein diacetate hydrolytic activity were significantly increased by irrigation in comparison to the rainfed condition. The activity of soil protease reduced significantly, by 8%, with the biochar application in the irrigated condition. Further, a linear correlation analysis and redundancy analysis performed on the plant, nodule, and soil variables suggested that the biochar application may affect soybean N uptake in the sandy field. Nodulation was enhanced with biochar addition, however, the plant N concentration and nodule Lb content remained unaffected.

Research on the use of soil enhancer materials such as biochar from soil chemical perspective still provide differing results; therefore, investigations focusing on soil-biochar-plant interactions are still necessary to constrain our understanding of complex biochar effects. The present study investigated the changes in biological nitrogen fixation rates (BNF) and overall nutrient dynamics (NO3−, NH4+, total N, K2O, and P2O5) during the growth of Capsicum annuum (pepper) in pot experiments amended with biochar made of paper fiber sludge and grain husk. Four treatments were studied with 0, 0.5%, 2.5%, and 5.0% (by weight) added biochar (BC) amount to temperate silt loam soil. Peppers were planted at 2–4 leave stages and grown for the duration of 12.5 weeks. Our results showed that total nitrogen had relatively small changes in all treatments over time compared to the dynamic changes observed in the case of inorganic nutrients. NO3−-N and NH4+-N abundances presented a continuous decrease during the course of the study after an initial increase. The pepper plant facilitated the BNF rates to triple in the control soils, while plants were in the growing phase (weeks 1–6), which further increased an additional 61% by harvesting (week 12). A high amount of biochar addition suppressed potential BNF rates of the investigated soil, indicating its potentially negative effects on soil indigenous microbial communities if added in excess. We also found a plateau in plant biomass production that after reaching an optimal (2.5%) biochar amendment in the soils, and excess biochar addition did not result in significant changes in the soils’ pH to achieve better nutrient (potassium, nitrogen, phosphorous) use or crop growth.

Inclusion of legumes into grass pastures can provide an alternative N source through biological N2 fixation (BNF), and enhance retention and cycling of soil C and N. Despite the use of alfalfa (Medicago sativa L.), cicer milkvetch (Astragalus cicer L.) and sainfoin (Onobrychis viciifolia Scop.) in grass-legume pastures to improve forage quality, relative BNF potentials and resource partitioning are unknown. We quantified BNF using 15N isotope dilution and estimated resource partitioning in alfalfa, two cultivars of cicer milkvetch and two cultivars of sainfoin under controlled conditions. Percentage of nitrogen derived from atmosphere followed the order alfalfa (92%) > cicer milkvetch (87%) > sainfoin (81%); corresponding to estimated N contributions of 200, 128 and 65 kg N ha−1 yr−1, respectively, based on total herbage. Root dry matter was 24% to 36% greater than shoot dry matter in all of the legumes, providing substantial below-ground C and N. Cultivars of the same species did not differ in any measured parameter (p > 0.05). The common bean (Phaseolous vulgaris L.) is a grain legume functionally characterized by its capacity for symbiotic of biological nitrogen fixation. As such, it does not demand the application of synthetic nitrogenous fertilizer and can offer environmental benefits as part of holistic cropping systems. While common bean commodities are highly nutritious, commercial cultivation of this crop is declining in already-industrialized countries. However, recent interest of consumers towards diets that benefit environmental and personal health has rekindled commercial interest in legumes, including the common bean. The aim of this protocol is to identify agronomic practices that are capable of increasing the yield and quality of the common bean for use as food. To address this research question, published literature will be screened for inclusion on the basis of defined eligibility criteria to ensure data sources are selected in an objective and consistent manner. Consistency checks will be carried out for the title, abstract and full texts of the literature collated. The output is expected to be a summary of the knowledge available to maximize the productivity and quality of the common bean as food. The introduction of nitrogen fixing bacteria (NFB) and arbuscular mycorrhizal fungi (AMF) into the soil is an advisable agricultural practice for the crop, since it enhances nutrient and water uptake and tolerance to biotic and abiotic stresses. The aim of this work was to study plant nutrition, biological nitrogen fixation (BNF) and crop yield and quality, after inoculating seeds with NFBs ((Rhizobium leguminosarum, Burkholderia cenocepacia, Burkholderia vietnamiensis)) and/or AMFs (Rhizophagus irregularis, Claroideoglomus etunicatum, Claroideoglomus claroideum and Funneliformis mosseae) in a fava bean crop in two seasons. The composition of the nodule bacterial community was evaluated by the high-throughput sequencing analysis of bacterial 16 S rRNA genes. It was found that microbial inoculation accompanied by a 20% decrease in mineral fertilization had no significant effect on crop yield or the nutritional characteristics compared with a non-inoculated crop, except for an increase in the grain protein content in inoculated plants. None of the inoculation treatments increased biological nitrogen fixation over a non-inoculated level. The bacterial rRNA analysis demonstrated that the genus Rhizobium predominated in all nodules, both in inoculated and non-inoculated treatments, suggesting the previous presence of these bacteria in the soil. In our study, inoculation with Rhizobium leguminosarum was the most effective treatment for increasing protein content in seeds, while Burkholderia sp. was not able to colonise the plant nodules. Inoculation techniques used in fava beans can be considered an environmentally friendly alternative, reducing the input of fertilizers, while maintaining crop yield and quality, with the additional benefit of increasing the grain protein content. However, further research is required on the selection and detection of efficient rhizobial strains under local field conditions, above all those related to pH and soil type, in order to achieve superior nitrogen-fixing bacteria. Nitrogen (N) in soybean (Glycine max L.) plants derived from biological nitrogen fixation was shown to be a sustainable N resource to substitute for N fertilizer. However, the limited water supply in sandy soil is a critical factor for soybean nodulation and crop growth. This study investigated the potential mechanism of the effect of biochar and irrigation on the soybean-Rhizobium symbiotic performance and soil biological activity in a field trial. In the absence of N fertilizer, 10 t ha−1 of black cherry wood-derived biochar were applied under irrigated and rainfed conditions on an experimental, sandy field site. The plant biomass, plant nutrient concentrations, nodule number, nodule leghemoglobin content, soil enzyme activities, and soil-available nutrients were examined. Our results show that biochar application caused a significant increase in the nodule number by 35% in the irrigated condition. Shoot biomass and soil fluorescein diacetate hydrolytic activity were significantly increased by irrigation in comparison to the rainfed condition. The activity of soil protease reduced significantly, by 8%, with the biochar application in the irrigated condition. Further, a linear correlation analysis and redundancy analysis performed on the plant, nodule, and soil variables suggested that the biochar application may affect soybean N uptake in the sandy field. Research on the use of soil enhancer materials such as biochar from soil chemical perspective still provide differing results; therefore, investigations focusing on soil-biochar-plant interactions are still necessary to constrain our understanding of complex biochar effects. The present study investigated the changes in biological nitrogen fixation rates (BNF) and overall nutrient dynamics (NO3−, NH4+, total N, K2O, and P2O5) during the growth of Capsicum annuum (pepper) in pot experiments amended with biochar made of paper fiber sludge and grain husk. Four treatments were studied with 0, 0.5%, 2.5%, and 5.0% (by weight) added biochar (BC) amount to temperate silt loam soil. Peppers were planted at 2–4 leave stages and grown for the duration of 12.5 weeks. Our results showed that total nitrogen had relatively small changes in all treatments over time compared to the dynamic changes observed in the case of inorganic nutrients. NO3−-N and NH4+-N abundances presented a continuous decrease during the course of the study after an initial increase. The pepper plant facilitated the BNF rates to triple in the control soils, while plants were in the growing phase (weeks 1–6), which further increased an additional 61% by harvesting (week 12). A high amount of biochar addition suppressed potential BNF rates of the investigated soil, indicating its potentially negative effects on soil indigenous microbial communities if added in excess. We also found a plateau in plant biomass production that after reaching an optimal (2.5%) biochar amendment in the soils, and excess biochar addition did not result in significant changes in the soils' pH to achieve better nutrient (potassium, nitrogen, phosphorous) use or crop growth.
