Background: Gold nanoclusters (AuNCs) obtained by electroreduction have excellent electrochemiluminescence (ECL) properties, and its ECL intensity is regulated by the valence state. In addition, their ECL signals can be rapidly quenched by reactive oxygen species (ROS). Based on this observation, a sensitive ROS biosensor was designed based on valence regulation of AuNCs. Excessive ROS in seminal plasma can lead to male infertility, and the short half-life and instability of ROS pose a challenge for their detection. Since valence regulation can be done quickly and is very sensitive, this ECL biosensor holds promise to address this issue., Results: The ECL mechanism of AuNCs and the quenching mechanism of AuNCs by ROS were explored, mainly because ROS change the valence state of AuNCs. The ECL signals of the biosensor have a linear relationship with logarithm of the target concentration in the range of 1.0 × 10 -8 to 1.0 × 10 -3  M and 1.0 × 10 -3 to 1.0 × 10 -1  M, with a detection limit of 0.75 × 10 -10  M (S/N = 3). The biosensor enables rapid one-step detection of ROS and has the advantage of being stable and reusable. More notably, the results of 57 real samples showed that the biosensor can be used to accurately assess the concentration of seminal plasma ROS, guiding the monitoring of sperm quality and the diagnosis of male infertility., Significance: Compared with the traditional strategy of applying AuNCs only as a luminescent body, this strategy of regulating the valence state of AuNCs to achieve sensitive and rapid detection broadens the application of AuNCs in the field of analysis. In this study, preparation of novel pH-sensitive N-doped carbon dots (NCDs) using glucose and urea is reported. The prepared NCDs present strong excitation-dependent fluorescence changes towards the pH that is a new behavior from these nanomaterials. By taking advantage of this unique behavior, two separated ratiometric pH sensors using emission spectra of the NCDs for both acidic (pH 2.0 to 8.0) and basic (pH 7.0 to 14.0) ranges of pH are constructed. Additionally, by considering the entire Excitation-Emission Matrix (EEM) of NCDs as analytical signal and using a suitable multivariate calibration method, a broad range of pH from 2.0 to 14.0 was well calibrated. The multivariate calibration method was independent from the concentration of NCDs and resulted in a very low average prediction error of 0.067 pH units. No changes in the predicted pH under UV irradiation (for 3 h) and at high ionic strength (up to 2 M NaCl) indicated the high stability of this pH nanosensor. In this paper, the comparison of two kinds of electrochemically reduced graphene oxide (ERGNO) and zirconia composites, obtained by one-step (ZrO2-ERGNO) and stepwise (ZrO2/ERGNO) electrodeposition for DNA sensing, is systematically studied. The resulting composites were characterized by scanning electron microscopy, cyclic voltammetry, and differential pulse voltammetry. The results indicated that the ZrO2-ERGNO presented fine globular nanostructure. However, ZrO2/ERGNO presented agglomerate massive microstructure due to the absence of the oxygen-containing groups of graphene oxide, confirming the oxygen-containing groups provided a better affinity for the deposition of ZrO2. Due to the strong binding of the phosphate groups of DNA with the zirconia film, DNA probes were attached on the ZrO2-based composites. ZrO2-ERGNO/Au owning fine nanostructure presented larger surface area than microstructured ZrO2/ERGNO/Au. Moreover, compared with microstructured ZrO2/ERGNO, the nanostructured ZrO2-ERGNO provided more accessible space for immobilized DNA probe hybridization with target sequence, which consequently resulted in higher hybridization efficiency. In this communication, the first nonelectrocatalysis-type hydrogen peroxide electrochemical sensor is reported. The electroactive iron(III) diethylenetriaminepentaacetic acid (DTPA-Fe(III)) complex is immobilized on the cysteamine (cys) modified nanoporous gold (NPG) films by covalent method. The immobilized DTPA-Fe(III) complex quickly communicates an electron with the electrode. Upon addition of hydrogen peroxide, however, hydrogen peroxide inhibits the direct electron transfer of the DTPA-Fe(III) complex due to the generation of nonelectroactive DTPA-Fe(III)-H2O2 complex. Based on quenching mechanism, the first hydrogen peroxide electrochemical sensor based on a nonelectrocatalytic mechanism is developed. The novel hydrogen peroxide electrochemical sensor has the ultralow detection limit (1.0×10(-14) M) and wide linear range (1.0×10(-13) to 1.0×10(-8) M) with excellent reproducibility and stability., (Copyright © 2013 Elsevier B.V. A highly selective fluorescent probe (OHBT) was designed and synthesized by linking the ESIPT fluorophore N-(3-(benzo[d]thiazol-2-yl)-4-(hydroxyphenyl) benzamide) (HBTBC) to the palladium specificity response group, allyl group, for the detection of palladium species in aqueous solution. The allyl group can be hydrolyzed by Pd(0) species through the Pd(0)-catalyzed Tsuji-Trost reaction and thus release the fluorophore HBTBC, which shows two emission bands. The maximum emission spectra originated from the enol and keto forms at 415 and 555 nm respectively and with no overlap, which implies the high resolution of the palladium detection. The palladium species can also be detected by paper strip because of the solid-state fluorescence of probe HOBT catalyzed by palladium. In this paper, a structure-switching aptamer assay based on a fluorescence polarization (FP) signal transduction approach and dedicated to the L-tyrosinamide sensing was described and optimized. A fluorescently labelled complementary strand (CS) of the aptamer central region was used as a probe. The effects of critical parameters such as buffer composition and pH, temperature, aptamer:CS stoichiometry, nature of the dye (Fluorescein (F) or Texas Red (TR)) and length of the CS (15-, 12-, 9- and 6-mer) on the assay analytical performances were evaluated. Under optimized experimental conditions (10 mM Tris-HCl, 5 mM MgCl(2) and 25 mM NaCl, pH 7.5 temperature of 22°C and stoichiometry 1:1), the results showed that, for a 12-mer CS, the F dye moderately increased the method sensitivity in comparison to the TR label. The F labelled 9-mer CS, however, did not allow the hybrid formation with the functional nucleic acid, thus emphasizing the importance of the nature of the fluorophore. In contrast, the same 9-mer CS labelled with the TR dye was able to effectively associate with the aptamer and was easily displaced upon target binding as demonstrated by a significant improvement of the sensitivity and a detection limit of 250 nM, comparable to those reported with direct aptasensing methods. The present study demonstrates that not only the CS length but also the nature of the dye played a preponderant role in the performance of the structure-switching aptamer assay, highlighting the importance of interdependently controlling these two factors for an optimal FP-based sensing platform., (Copyright © 2011 Elsevier B.V. A new dual-amplification strategy of electrochemical signal based on the catalytic recycling of the product was developed for the antigen-antibody interaction by glucose oxidase (GOD)- conjugated gold-silver hollow microspheres (AuAgHSs) coupled with an artificial catalase, Prussian blue nanoparticles (PB), on a graphene-based immunosensing platform. The first signal amplification introduced in this study was based on the labeled GOD on the AuAgHSs toward the catalytic oxidation of glucose. The generated H(2)O(2) was catalytically reduced by the immobilized PB on the graphene nanosheets with the second amplification. With a sandwich-type immunoassay format, carcinoembryonic antigen (CEA) was monitored as a model analyte by using the synthesized AuAgHSs as labels in pH 6.0 phosphate buffer containing 10mM glucose. Under optimal conditions, the electrochemical immunosensor exhibited a wide dynamic range of 0.005-50 ng mL(-1) with a low detection limit (LOD) of 1.0 pg mL(-1) CEA (at 3σ). Both the intra- and inter-assay coefficients of variation (CVs) were lower than 10%. The specificity and stability of the immunosensor were acceptable. In addition, the assay was evaluated for clinical serum specimens, and received a good correlation with those obtained by the referenced electrochemiluminescent (ECL)., (Copyright © 2011 Elsevier B.V. A new sandwich-type electrochemical immunosensor with enhanced sensitivity was developed for detection of alpha-fetoprotein (AFP, as a model analyte) in biological fluids by using nanogold-enclosed titania nanoparticle (AuTi)-labeled secondary antibody on a gold-silver-graphene hybrid nanosheet (AuAgGP)-functionalized glassy carbon electrode (GCE). The presence of the AuAgGP nanosheets not only enhanced the immobilized amount of biomolecules, but also improved the electrochemical properties of the immunosensor. With the aid of AuTi nanolabels, the electrochemical signal was greatly amplified in comparison with pure nanogold or titania-based labels. Under optimal conditions, the sensitivity and dynamic range of the immunosensor were evaluated by using the labeled horseradish peroxidase on the AuTi as trace and H(2)O(2) as enzyme substrate, and exhibited a wide dynamic range of 0.001-200 ng mL(-1) with a low detection limit (LOD) of 0.5 pg mL(-1) AFP (at 3σ). Both the intra- and inter-assay coefficients of variation were less than 10%. The current of the immunosensor at 13th day was as much as 90% of the initial current. In addition, the methodology was evaluated for 8 positive serum specimens obtained from hepatocarcinoma patients and 19 negative sera, and validated with the commercially available Roche 2010 Electrochemiluminescent (ECL) Automatic Analyzer. Uricase has been covalently immobilized using glutaraldehyde as cross-linker onto electrochemically synthesized polyaniline (PANI) films. These PANI-uricase electrodes have been characterized using spectroscopic, cyclic voltammetry and impedance measurements. The morphology and covalent linkage of uricase lead to high enzyme loading and better shelf life. The value of the Michaelis-Menton constant obtained as 5.1x10(-3) mM L(-1) for the immobilized uricase compared to 3.4x10(-1) mM L(-1) for the free uricase enzyme, suggests enhancement in affinity and/or activity of uricase attached to PANI. The influence of pH, temperature and concentration on electrode activity were studied. The enzyme electrodes were found to retain 95% of activity after 17-18 weeks when stored at 4 degrees C. These electrodes have a response time of about 60 s and have been used to measure uric acid concentration in serum. These PANI-uricase electrodes can be used for about 30 times for electrochemical measurements while retaining about 90% of its activity indicating improved performance.

This paper presents different experimental results of the influence of an immersion angle (theta, the angle between the surface of a quartz crystal resonator and the horizon) on the resonant frequency of a quartz crystal microbalance (QCM) sensor exposed one side of its sensing surfaces to liquid. The experimental results show that the immersion angle is an added factor that may influence the frequency of the QCM sensor. This type of influence is caused by variation of the reflection conditions of the longitudinal wave between the QCM sensor and the walls of the detection cell. The frequency shifts, measured by varying theta, are related to the QCM sensor used. When a QCM sensor with a weak longitudinal wave is used, its resonant frequency is nearly independent of theta. But, if a QCM sensor with a strong longitudinal wave is employed, the immersion angle is a potential error source for the measurements performed on the QCM sensor. When the reflection conditions of the longitudinal wave are reduced, the influence of theta on the resonant frequency of the QCM sensor is negligible. The slope of the plot of frequency shifts (deltaF) versus (rho eta)(1/2), the square root of the product of solution density (rho) and viscosity (eta), may be influenced by theta in a single experiment for the QCM sensor with a strong longitudinal wave in low viscous liquids, which can however, be effectively weakened by using the averaged values of reduplicated experiments. In solutions with a large (rho eta)(1/2) region (0-55 wt% sucrose solution as an example, with rho value from 1.00 to 1.26 g cm(-3) and eta value from 0.01 to 0.22 g cm(-1) s(-1), respectively), the slope of the plot of deltaF versus (rho eta)(1/2) is independent of theta even for the QCM sensor with a strong longitudinal wave in a single experiment. The influence of theta on the resonant frequency of the QCM sensor should be taken into consideration in its applications in liquid phase.
