The roles of growth factors such as angiopoietin (Ang) and vascular endothelial growth factor (VEGF) in angiogenesis have been known for some time, yet we have just an incipient appreciation for the contribution of Wnts to this process. Cellular proliferation and polarity, apoptosis, branching morphogenesis, inductive processes, and the maintenance of stem cells in an undifferentiated, proliferative state are all regulated by Wnt signaling. The development and maintenance of vascular structures are dependent on all these processes, and their orchestration has, to some extent, been revealed in studies of VEGF and Ang receptors. Recent evidence links the Wnt/Frizzled signaling pathway to proper vascular growth in mammals but our knowledge of Wnt function in the vasculature is rudimentary. Further insight into vascular development and the process of angiogenesis depends on evaluating the function of novel endothelial regulatory pathways such as Wnt/Frizzled signaling.

We put forward an algorithm describing the three principal interconnected sub-processes that influence tumor and vasculature dynamics: (i) tumor cell proliferation (ii) angiogenesis, that is, the formation and regression of immature vessels (IV), and (iii) maturation, i.e., the formation and destabilization of mature vessels (MV). This algorithm takes account of the crucial quantitative interactions of these sub-processes, occurring across the molecular, cellular and organ levels. Implementing this complex algorithm in a computer model, one can evaluate the correlations between various factors influencing angiogenesis and their influence on tumor progression at any given moment. Moreover, the computer simulations enable analysis of the versatile effects of drugs on the growth and decay of both the tumor and the immature and mature blood vessels, as well as on the induction of an array of relevant growth factors such as angiopoietin-1 (Ang1), angiopoietin-2 (Ang2), vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF). Simulation results suggest that vessel maturation and destabilization of MV drive the otherwise non-linearly growing system into a very dynamic region, having irregular, scale-invariant, fluctuations, around certain asymptotic values of all the involved quantities. Destabilization itself adequately explains the experimentally observed eventual decrease of tumor growth, with no need to implicate additional assumptions, such as a new tumor growth inhibitory, or anti-angiogenic, factors. Our results further suggest that mono-therapy alone can slow tumor growth, but is not capable of eliminating it altogether. In contrast, the combined treatment of anti-angiogenic and anti-maturation drugs causes prolonged suppression of tumor growth and a significant linear decrease in average tumor size. Laboratory experiments are warranted for validating our predictions and for providing in vivo evaluated parameters.

The transmembrane tyrosine kinase TIE-2, the receptor for the angiopoietins-1 and -2, has been shown to be involved in angiogenic processes. Investigating the regulation of TIE-2 expression on endothelial cells, we found that stimulators such as PMA induce a decrease of TIE-2 protein from the cell surface without affecting TIE-2 mRNA. In conditioned media of PMA stimulated endothelial cells, a soluble form of this receptor comprising parts of the extracellular domain can be detected. Using a sandwich ELISA, we were able to detect and quantify TIE-2 receptors in cell lysates (representing the whole transmembrane receptor) and in cell culture supernatants (representing a soluble form of this receptor, sTIE-2). Several factors influencing this shedding process e.g. basic FGF could be identified. Finally, the soluble form of TIE-2 could also be detected in human biological fluids such as sera and plasma from healthy controls.

The Tie2 receptor and its known ligands, the angiopoietins, play a critical role in endothelial cell differentiation during the process of angiogenesis. Recent experimental observations indicate that the agonistic ligand, angiopoietin-1, can stimulate endothelial cell sprouting and act as a chemo-attractant in vitro and induce increased and enhanced angiogenesis both alone and in conjunction with vascular endothelial growth factor (VEGF) in vivo. Here, we present a monoclonal antibody (MAb), which binds to the extracellular portion of the Tie2 receptor and elicits similar agonist effects. Upon MAb binding to the native Tie2 receptor of cultured human umblical vein endothelial cells (HUVEC), there is a rapid increase in receptor autophosphorylation with a concomitant enhancement in the recruitment and association of the signalling intermediates Grb2 and SH-PTP2. The antibody further demonstrates functional activity in vascular tissues. In vitro, the antibody promotes the survival of cultured HUVEC and elicits a dose dependent outgrowth and branching of microvessels from cultured explants of rat aorta. When administered in vivo, the antibody enhances the vascularization of subcutaneous Matrigel implants in mice. Together these data suggest that the antibody is capable of acting as a surrogate ligand for Tie2 and further confirms the role of Tie2 in the differentiation of endothelial cells during angiogenesis.

The Angiopoietin/Tie system acts as a vascular specific ligand/receptor system to control endothelial cell survival and vascular maturation. The Angiopoietin family includes four ligands (Angiopoietin-1, Angiopoietin-2 and Angiopoietin-3/4) and two corresponding tyrosine kinase receptors (Tie1 and Tie2). Ang-1 and Ang-2 are specific ligands of Tie2 binding the receptor with similar affinity. Tie2 activation promotes vessel assembly and maturation by mediating survival signals for endothelial cells and regulating the recruitment of mural cells. Ang-1 acts in a paracrine agonistic manner inducing Tie2 phosphorylation and subsequent vessel stabilization. In contrast, Ang-2 is produced by endothelial cells and acts as an autocrine antagonist of Ang-1-mediated Tie2 activation. Ang-2 thereby primes the vascular endothelium to exogenous cytokines and induces vascular destabilization at higher concentrations. Ang-2 is strongly expressed in the vasculature of many tumors and it has been suggested that Ang-2 may act synergistically with other cytokines such as vascular endothelial growth factor to promote tumor-associated angiogenesis and tumor progression. The better mechanistic understanding of the Ang/Tie system is gradually paving the way toward the rationale exploitation of this vascular signaling system as a therapeutic target for neoplastic and non-neoplastic diseases.

Tie2 is a receptor tyrosine kinase that is expressed predominantly in the endothelium and plays key roles in both physiological and pathological angiogenesis. The ligands for Tie2, the angiopoietins (Ang), perform opposing functions in vascular maintenance and angiogenesis; Ang1 regulates vascular quiescence, while Ang2 is thought to promote vascular destabilization and facilitate angiogenesis. However, the mechanisms responsible for these differences are not understood. To begin to elucidate the molecular differences between the angiopoietins, we previously developed a specific RNA aptamer inhibitor of Ang2. Here, we used the same iterative in vitro selection process, termed SELEX (Systematic Evolution of Ligands by EXponential enrichment), to screen a library of 2'-fluoro-modified ribonucleotides for Ang1-binding aptamers. After nine rounds of selection, we identified a single clone, ANG9-4, that bound with high affinity to human Ang1 (K ( d ) 2.8 nM) but not Ang2 (K ( d )1 microM), demonstrating specificity for Ang1. ANG9-4 blocked Ang1-mediated Tie2 phosphorylation and downstream Akt activation. Moreover, ANG9-4 inhibited Ang1-induced endothelial cell survival. Together, these findings demonstrate the feasibility of developing an Ang1-inhibitory aptamer. ANG9-4 and its derivatives may provide useful tools for elucidating the biology of Ang1 and for treating certain angiogenic diseases.

The endothelial cell (EC)-selective receptor tyrosine kinase, Tie2, and its ligands angiopoietin Ang-1 and Ang-2, are essential for blood vessel maintenance and repair. Ang-1 is an agonist of Tie2 receptor activation, whereas Ang-2 is a context-dependent antagonist/agonist. Therefore, we investigated the role of the EC-selective phosphatase, human protein tyrosine phosphatase beta (HPTPbeta), in regulating Tie2 activity.siRNA silencing of HPTPbeta enhanced Ang-1 and Ang-2-induced Tie2 phosphorylation at 10 min (2.5-fold, P0.001; and 1.8-fold, P0.05, respectively). The cell survival response to Ang-1, but not Ang-2, was enhanced by HPTPbeta silencing as measured by flow cytometry (0.85-fold to 0.66-fold, P0.05) and ELISA (0.88-fold to 0.53-fold, P0.01). Hypoxia, which upregulated HPTPbeta expression in endothelial cells, impaired Ang-1-induced Tie2 phosphorylation.These results reveal a novel role for HPTPbeta in modulating Ang-1-Tie2 signaling and endothelial cell survival.
