Increasing evidence suggests that environmental exposures can modify epigenetic marks in the germline, leading to the transmission of abnormal post-fertilization sperm epigenetic indicators and affecting embryonic development. Given the pivotal role of sperm cells in determining embryo quality, there is growing interest in understanding the potential effects of sperm sex sorting on embryo quality. This study aimed to investigate the impact of bovine sperm sexing on in vitro embryo production (IVP) and to associate molecular aspects of embryos analysis. Frozen semen samples from five Nellore bulls were used, with each bull contributing unsexed sperm (conventional semen – CV treatment) and female and male sexed sperm pooled after thawing (SX treatment). First, semen quality was assessed, including motility, morphology, acrosome integrity, and chromatin integrity to denaturation. Then, IVP was carried out, focusing on embryonic production and developmental kinetics. In the third experiment, embryo quality was evaluated by examining the gene expression of key markers (OCT4, NANOG, DNMT3A, TET1 , and Fematrin-1) and the methylation pattern of the Satellite-1 and α-Satellite genes in blastocysts. Differences between CV and SX semen were only observed in motility, which was lower in SX compared with CV (P < 0.05). Although cleavage was similar, the SX groups showed lower blastocyst production than CV (P < 0.05). Of the genes evaluated, only NANOG showed high expression in the CV blastocysts compared with the SX blastocysts, but the methylation pattern revealed no differences. Gonadal soma-derived factor (gsdf) is a teleost- and gonad-specific growth factor involved in early germ cell development. The red spotted grouper, Epinephelus akaara , as a protogynous hermaphrodite, provides a novel model for understanding the mechanisms of sex determination and differentiation in teleosts. In the present study, a 2307-bp long gsdf gene was cloned from E. akaara and there was further analysis of its tissue distribution and gonadal patterns of gene expression during the female phase and sex change developmental stages. The cellular localization of gsdf at the late transitional developmental stage was also analyzed. In addition, the concentrations of serum sex steroid hormones ( E 2 , 11-KT and DHP) were determined. The gsdf transcripts were exclusively localized in the gonad. During the female phase at an early developmental stage, when the ovotestis contained mainly oogonia and primary growth oocytes, the gsdf mRNA was relatively more abundant. The relative abundance of gsdf decreased, however, and the lesser amount was sustained with the advancement of oocyte development. During the transitional phase, the relative abundance of gsdf mRNA increased slightly at the early developmental stage and there were further increases in relative abundance in the late developmental stage, and the gsdf transcripts were observed in the Sertoli cells surrounding early developing spermatogonia. Among the sex steroids, 11-KT concentrations were positively correlated with amount of gsdf mRNA during sex change. Twenty bitches were seen in consultation at the Department of Reproduction at ONIRIS (College of Veterinary Medicine, Food Science and Engineering, Loire Atlantique, Nantes, France) between 25 and 50 days of gestation for early sex determination of the canine foetus using ultrasound. The genital tubercle is not visible before 26 days; between 26 and 30 days, it is visible between the pelvic limbs; between 33 and 50 days, the position of the genital tubercle enables sex determination as it migrates caudally in the female and cranially in the male. Good statistical concordance between sexing via ultrasound and sexing at birth has been established (kappa coefficient of 0.8). Macroscopic, microscopic, and histological examinations of the external genital organs were also performed on 10 foetuses at 35 days of gestation; a cartilaginous structure was visualized in the genital apparatus of the male but also in half of the females. Finally, the development of a PCR technique on the SRY gene using formaldehyde-preserved tissues has been described for the first time in this study. Abstract: In the equine species, sex determination of the conceptus is of growing interest for the breeding industry. In horses, the sex ratio of the offspring depends on changes in body condition of the mother at conception and under natural conditions may thus markedly deviate from an expected 1:1 ratio. Insemination with sex-sorted spermatozoa allows a pronounced shift of the sex ratio but at present pregnancy rates are low and vary considerably under field conditions. In equine embryo transfer programmes, sex determination in embryos before transfer via genetic methods is a promising approach with high reliability. In ongoing pregnancies, fetal sex can be determined in utero by transrectal or transabdominal ultrasound between days 57 and 220 after ovulation, but experience is required to achieve satisfying accuracy. Recently, genetic sexing via identification of circulating cell-free fetal DNA in the maternal circulation has been successfully performed in the last three months of pregnancy. Development of this technique may also allow fetal sex determination at earlier stages of pregnancy. Further research is required to allow for techniques that enable sex determination in equine embryos as well as in ongoing pregnancies under field conditions. [Copyright &y& Elsevier]

Flow cytometry is a widely used application for validating the accuracy of sperm sexing. However, this method is relatively expensive and requires considerable technical support. An alternative method employing simpler technology at low cost could be suitable for the evaluation of bovine semen in laboratories with low budgets. We used a SYBR Green Real-Time PCR assay to determinate sex ratio in bovine semen. The PLP and SRY genes were amplified to isolate the specific fragments of X- and Y-chromosome sequences, respectively. Two certified standard curves were obtained using two plasmids containing PLP and SRY amplicons. Our results show no significant difference in semen sex ratio in unsorted semen (54.7±0.52% X and 47.6±0.60% Y). However, significant difference was observed in X/Y-sorted semen (93.3±0.08% X and 91.4±0.06% Y-sperm), as compared to the expected ratio in unsorted semen or the post-sorting reanalysis data. The evolution of X-chromosome bearing sperm content in unsorted samples showed an average of 52.6 for ejaculates and 51.8 for the commercial semen. In order to confirm our results, the accuracy, repeatability and reproducibility of the method were tested resulting in 98.2% accuracy, repeatability of CV=5.59% and reproducibility of CV=5.40%. Abstract: The aim has been to set up a rapid and accurate microarray assay using sandwich mode for sex determination of bovine preimplantation embryos. Twelve sequence-specific oligonucleotide capture probes used to discriminate 12 samples were spotted onto the aldehyde-modified glass slides by Arrayer. The 2 recognition probes used to identify coding regions of the sex-determining region of the Y chromosome gene (SRY) and β-casein (CSN2) reference gene were coupled with biotin. The assay was optimized by using genomic DNA extracted from blood samples of known sex individuals. Polymerase chain reaction (PCR) was used to amplify the fragments in the HMG box region of SRY gene and CSN2 gene with sequence-specific primers. The sex of samples was identified by detecting both the SRY and CSN2 genes simultaneously in 2 reaction cells of microarrays, with the male having SRY and CSN2 signals and the female only CSN2. The sex of 20 bovine preimplantation embryos was determined by oligonucleotide microarray. The protocol was run with a blind test that showed a 100% (82/82) specificity and accuracy in sexing of leukocytes. The bovine embryos were transferred into 20 bovine recipients, with a pregnant rate of 40% (8/20). Three calves were born at term, and 5 fetuses were miscarried. Their sexes were fully in accordance with the embryonic sex predetermination predicted by oligonucleotide microarray. This suggests that the oligonucleotide microarray method of SRY gene analysis can be used in early sex prediction of bovine embryos in breeding programs. [Copyright &y& Elsevier]

Abstract: The objective of this study was to evaluate the use of polymerase chain reaction analysis (PCR) of fetal cells/DNA in the maternal plasma of pregnant cows to determine the sex of the fetus. Plasma was harvested from 35 cows of mixed genotype at different stages of pregnancy ranging from 5 to 35 weeks. A male calf and a heifer calf provided the control samples. Fetal sex was determined by amplification of Y-specific sequences. For the 35 cows, the fetal sex predicted by this technique was in accordance with the sex of the calf at birth in 88.6% of cases. The agreement between predicted and observed fetal sex was less for cows with a gestational length of 35–48 days (63.6%). Regression analysis showed that there was a strong relationship between the probability of correctly predicting fetal sex and the stage of gestation. It was estimated that the test performed at 43.8 days post fertilization would have 95% accuracy, increasing to 99% accuracy for testing at 48.4 days and 99.9% accuracy for tests at 55.0 days or later. It was concluded that PCR analysis of fetal cells in maternal plasma can be used to predict successfully the sex of the fetus in cattle. [Copyright &y& Elsevier]

Abstract: The aim of this study was to evaluate embryo production in superovulated wapiti hinds inseminated with either Y-sorted or unsorted semen. Eighteen hinds were allocated to three treatment groups: AI following multiple ovulation (CIDR/FSH) with 10×106 Y-sorted frozen–thawed semen (Y group, n =6), or 10×106 and 100×106 unsorted frozen–thawed semen for the unsorted (n =6) and the control group (n =6). The embryos from the sixth day following insemination were collected and classified. Fifteen embryos from the unsorted or the control group, and four embryos from the Y group were sex determinated based on DNA analysis of the amelogenin gene. Twenty-one embryos from the Y group and 42 embryos from the unsorted or the control group were transferred into 21 and 42 synchronized recipients via standard procedures on 6th day post estrus, respectively. There were no significant differences in the number of recovered eggs, transferable embryos, degenerated embryos or unfertilized oocytes per hind among the three groups of the control (9.2±3.6, 4.7±1.9, 3.0±2.0, 1.5±1.4), the unsorted (8.2±1.9, 4.8±0.7, 1.7±1.0, 1.7±1.0) and the Y group (8.8±4.2, 4.2±1.8, 2.2±1.2, 2.5±2.1), respectively (P >0.05). The sex ratio of embryos from the Y group (4M/0F) was significantly (P <0.05) distinct from that of the unsorted and control group (8M/7F). The sex ratio of the offspring from sexed embryos (8M/0F) was deviated significantly (P <0.05) from that of the non-sexed embryos (11M/9F). In conclusion, the results suggested that the male embryos of predicted sex can be achieved with AI of sex-sorted cryopreserved sperm. PCR amplification using the amelogenin gene primers can be applied to DNA analysis of micro samples from wapiti embryo biopsies for sex identification. Abstract: Despite considerable cryobiology research there is no industry standard for the concentration to which ram spermatozoa should be diluted before freezing. Ram semen is highly concentrated and often frozen at a high sperm concentration, necessitating the use of small laparoscopic insemination doses. The aim of this paper was to ascertain the effect of dilution on the integrity of frozen-thawed ram spermatozoa. In the first experiment, spermatozoa were extended with a Tris-buffered diluent before freezing or after thawing to yield a final sperm concentration of 20×106/ml, or were not diluted. Motility characteristics, viability and acrosome integrity of spermatozoa were analysed over a 6h incubation period at 37°C. In the second experiment, spermatozoa were either diluted before freezing, subjected to sex-sorting or not diluted before freezing. Thawed spermatozoa were separated into sub-populations using centrifugal counter-current distribution (CCCD) and the profile of partition and functional integrity (viability, chlortetracycline status and Annexin-V binding) in the sub-populations assessed. Dilution before freezing significantly improved post-thaw viability, acrosome integrity and total motility whereas dilution post-thaw decreased viability and motility of spermatozoa. Sperm heterogeneity, as assessed by CCCD profile, was not different for control, diluted and sex-sorted spermatozoa. Analysis of CCCD sub-populations showed the proportion of functional cells (displaying the F-Pattern or no PS translocation) was similar for all sperm types. The results show that ram spermatozoa retain normal function at higher pre-freeze dilution rates than are commonly used in the sheep industry. Abstract: This study aimed to detect the sex of bovine embryo by adding ethidium bromide (EB) or 5μl of CuSO4 at different concentrations (0.5M, 1M, 1.5M, 2M, 3M and 5M) to the product of loop-mediated isothermal amplification (LAMP) reaction. The result of these additions was a colour change and a precipitate. This allows detection with the naked eye without the use of electrophoresis or a turbidity meter. The in vitro produced bovine embryos were divided into one to eight pieces using a microblade attached to a micromanipulator. The cell number in each piece was counted before sexing. Sexing of DNA samples extracted from one to five biopsies cells was performed by LAMP. After biopsy, the remaining part of the embryos was used to confirm the sex by polymerase chain reaction (PCR). The time for the whole procedure was about 45min. LAMP is a novel DNA amplification method which amplifies a target sequence specifically under isothermal condition. We used this method to examine 58 bovine embryos and the accuracy of sex prediction was 100% when the blastomeres dissociated from a morula exceeds three. This study showed that the present method can be applied in bovine breeding programs to facilitate manipulation of the sex ratio of offspring. The aim of this study is to develop an improved embryo sexing technique based on the LAMP reaction. Abstract: This study was conducted to use B-mode trans-rectal (TR) and trans-abdominal (TA) ultrasonography to determine early pregnancy and fetometry as crown-rump-length (CRL) and bi-parietal-diameter (BPD). A total of 110 does aged between 8 and 36 months were used. The accuracy for detecting early pregnancy (fetal fluids and heartbeats) and fetal number (single or twins) was measured. The relationship between gestation age and CRL or BPD was determined from day 40 to 109 of gestation. The accuracy of fetal sexing was determined by differentiation of genital tubercle (GT) at different stages of gestation (from day 40 to 109 of gestation). The examination revealed 95.5% of examined does were pregnant, with accuracy 100% in detecting pregnancy for positive cases. The fetal number was 45.7% and 54.3% for single and twins+triplets, respectively. The TR probe enabled the reliable and earlier recognition of fetal fluid and heartbeats (indicating pregnancy) than TA probe. For maximum reliability, the TR observation of heartbeats is recommended as conclusive evidence of the presence of a live fetus. The TA convex probe was used from day 40 to 89 for measuring CRL and from day 40 to 109 for measuring the BPD. Gestation equations were: CRL=0.464x −17.767 and BPD=0.055x −1.431 (x =gestational age in days). The relation between gestational age and CRL or BPD was highly (p <0.0001) significant. The fetal sexing was found in 100, 83.3 and 64.3% of single pregnancies and in 85.7, 80 and 52.3% of twins+triplets pregnancies during 40–60 days, 61–70 days and 71–109 days of gestation, respectively. The accuracy of sex identification among the 3 groups was not significantly (p >0.05) higher in single than twins+triplets pregnancies. However, identification of GT in male fetus was possible from day 40 onward. From a total of 105 scanned does, 80 (76.25) were sexed. In conclusion, B-mode real-time ultrasonography is recommended as a reliable mean for early detection of gestation as early as 19–27 days after mating, for CRL or BPD measuring as well as fetal sex determination from day 40 of gestation onwards under field conditions. Abstract: Currently, there are few inexpensive, reliable, effective methods for commercially separating X- and Y-chromosome bearing fresh and frozen bovine sperm. The objective of these experiments was to determine the efficacy of a commercially available post-thaw bovine semen sexing kit, HeiferPlus™ (HP) which claims to alter the sex ratio in favor of female calves following artificial insemination. Three trials included the insemination of hyperstimulated cows with Control or HP-treated semen, non-surgical embryo collection on Day 7, and a combined PCR/dot blot assay to determine embryo sex. Chi-square analysis showed that the Control group produced a greater proportion (p <0.0005) of female embryos than the HP group. There were no differences in the proportions of transferable compared with degenerate embryos or in number of ovulations, embryos, and unfertilized ova collected from Control compared with HP groups. When treatments were combined, one of the two bulls used in the hyperstimulation studies produced an overall greater proportion of females (p <0.05), suggesting a bull effect. Another trial involved the insemination of cows synchronized via OvSynch® with fetal sexing via ultrasonography. Results of these studies indicated that HP semen sexing kit did not alter the sex ratio in favor of females in either hyperstimulated or single-ovulating cows; however, potential bull effects may be further evaluated to understand the capacity of HP with semen from specific bulls. Additionally, perhaps the sex of the surviving embryo can be manipulated by the maternal side, through ovarian, hormonal, oviductal, or uterine influences. [Copyright &y& Elsevier]

Abstract: In captive Asian elephants, there is a strong need for production of female offspring to enhance reproduction, counter premature aging processes in female animals and reduce challenging management situations derived from husbandry of several bulls in one institution. Artificial insemination of flow cytometrically sex-sorted spermatozoa offers the possibility to predetermine the sex of offspring with high accuracy. The aims of this study were to determine a suitable semen extender and basic parameters for flow cytometrical sex-sorting of Asian elephant spermatozoa. In total 18 semen samples were collected by manual rectal stimulation from one bull. Sperm quality parameters and sex sortability of spermatozoa were evaluated after dilution in three semen extenders (MES-HEPES-skim milk, MES-HEPES, TRIS–citric acid) and DNA staining. MES-HEPES-skim milk was the only semen extender found suitable to sex Asian elephant spermatozoa. From 18 ejaculates collected, 12 were successfully sorted with a purity of 94.5±0.7% at an average sort rate of 1945.5±187.5 spermatozoa per second. Sperm integrity, progressive and total motility were 42.6±3.9%, 48.1±3.3%, 59.4±3.8% after DNA labelling, and 64.8±3.2%, 58.0±5.0%, 70.8±4.4% after sorting, respectively. After liquid storage of sorted spermatozoa for 12h at 4°C, sperm integrity, progressive and total motility were 46.4±5.2%, 32.2±4.2% and 58.2±3.9%, respectively. Abstract: Flow cytometry sorting of X- and Y-chromosome bearing sperm has been emerging as a promising technology to alter the sex ratio in progenies of mammals in the resent years. The objective of this study was to evaluate the efficiency of AI by using the sexed sperm to produce sex-preselected calves in buffalo species. A total of 43 buffalo cows were inseminated with X-sorted sperm, 30 of which were confirmed pregnant 3mo following AI. In terms of conception rate, significant difference was observed between AI with sexed sperm derived from different bulls (P <0.05), but not between sexed and non-sexed sperm (P >0.05), nor between heifers and parous buffalo cows (P >0.05). A total of 29 sex-preselected calves, 24 females and 5 males, developed to term and were viable on delivery. Results of this study indicate the feasibility of the application of the sexing technology to accelerate the genetic improvement in swamp buffalo. [Copyright &y& Elsevier]

• Neotropical otter reproductive season lasts six months, from October to March. • Proportion of estradiol and testosterone metabolites allow sex-id of wild otter feces. • Sex-id of feces was possible only during the reproductive season. • This tool can be used in determination of sex structure of wild populations. Before there was use of ultrasonographic imaging, determination of the ratio of estrogens to androgens in the same individual was a technique used for differentiating the sex of monomorphic animals in captivity, with larger estrogen concentrations in the females. Due to species-specific differences in both concentration and changes throughout the year of these hormones, corroboration of the method is needed in each case. In this study, there was use of a chemo-immuno assay to quantify sex steroids in fecal samples collected from seven (five females and two males) Neotropical otters, Lontra longicaudis. The reproductive season for this species was determined to be between October and March, with increased estradiol in the females and relatively greater concentrations of testosterone in the males as compared with other seasons of the year. Results from utilization of a k-means analysis procedure indicated that the use of steroid ratios in fecal samples to differentiate otter sex is an effective technique when there are evaluations during the breeding season. The estrogen to androgen ratios during this period, however, are the inverse of what was expected, with there being larger testosterone concentrations in the female otters. The ratio of estrogens to androgens in feces of captive otters can be effectively used to determine the sex of otters in the field.
