Simple Summary: Procambarus clarkii is typically viewed as a predator rather than prey, due to its strong anti-predatory capabilities. However, during its juvenile stage, it remains vulnerable to predators. Nonetheless, complex outdoor habitats can offer some protection. By examining the effects of three different types of shelters with various coverages and combinations on the predation rhythm, behavior, and efficiency of Silurus asotus on juvenile Procambarus clarkii, we found that the presence and complexity of shelters in the environment have a limited impact on the predatory activities of Silurus asotus. Among the shelter types, PVC pipes offered the best protection, followed by stone, with water grass providing the least protection. Furthermore, the protective effect of shelters increased with coverage rate. Therefore, we suggest that the type and abundance of shelters significantly influence the predation rhythm and behavior of Silurus asotus on Procambarus clarkii in outdoor environments. Procambarus clarkii is adept at using natural shelters and caves to evade attacks from predators. However, the concealment abilities and mechanisms of P. clarkii for different types of shelters under predation pressure have not yet been reported. In this study, laboratory experiments were carried out to determine the effects of different coverages (25%, 50%, and 75%) and different combinations (I–VII) of three types of shelters (PVC pipes, water grass, and stone) on the predation rhythm, behavior, and abilities of Silurus asotus on P. clarkii. The results indicated that the predation of S. asotus on P. clarkii exhibited significant rhythmicity under shelter conditions, excluding PVC pipes, 75% stone, and combination VI. Among the three types of shelters, PVC pipes provided the strongest concealment, followed by stone and water grass. With the increase in shelter coverage, the anti-predation ability of P. clarkii continued to increase, and the optimal shade rate for water grass was 50%. In the different shelter combinations, the environmental complexity had little effect on the predation activity of S. asotus on P. clarkii. Simple Summary: The study described the effects of Myostatin gene editing on cattle to improve the growth and muscle development of hybrid cattle offspring. We focused on the Myostatin gene, which plays a crucial role in regulating muscle development. Gene editing could be able to enhance the size and performance of the crossbred cattle. The results showed that the Myostatin gene-edited cattle offspring displayed improved body size parameters, particularly body weight, cross height, and hip height, indicating the significant development of the hindquarters. The edited cattle have "double-muscled" forequarters and hindquarters with clear muscle masses, and their coat color reverted back to the original yellow. The study also found differences in blood glucose, calcium, and low-density lipoprotein levels, as well as a decrease in serum insulin levels in the edited cattle. Overall, the findings suggest that breeding with these Myostatin gene-edited bulls improve growth and performance in crossbred cattle, benefiting both farmers and consumers. The loss-of-function mutation of MSTN gene can induce the muscle hypertrophic phenotype. (2) Methods: Growth indexes and blood parameters of the cattle of different months were analyzed via multiple linear regression. (3) Results: Compared with the control group, the body shape parameters of F2 cattle were improved, especially the body weight, cross height, and hip height, representing significant development of hindquarters, and the coat color of the F2 generation returned to the yellow of Luxi cattle. As adults, MSTN gene-edited bulls have a tall, wide acromion and a deep, wide chest. Both the forequarters and hindquarters are double-muscled with clear muscle masses. The multiple linear regression demonstrates that MSTN gene-edited hybrid beef cattle gained weight due to the higher height of the hindquarters. Significant differences in blood glucose, calcium, and low-density lipoprotein. Serum insulin levels decreased significantly at 24 months of age. MSTN gene editing improves the adaptability of cattle. Simple Summary: Salinity is an important environmental factor which can influence the behavior of Eriocheir sinensis. In this study, female crabs were more active in a saline environment, especially low salinity stress, and the changes of antennae were obviously different under salinity shifts. Interestingly, E. sinensis had obvious behavioral differences in the high and low salinity stress, suggesting E. sinensis has different behaviors to adapt to the change of water salinity. The effects of salinity on behavior are far-reaching, and Eriocheir sinensis showed disparate behaviors under different salinity conditions. Female crabs were more active in saline water, especially low salinity stress, which is beneficial for female crabs to escape from the low-salinity environment quickly. Then, antennal movement indicated that antennae might be the main osmoreceptors in E. sinensis, and 65 min might be a good starting time for salinity stress to analyze osmoregulation in crabs. Interestingly, E. sinensis had obvious behavioral differences in the high and low salinity stress, and behaviors were more intense in a salinity dip from salinity 18 to salinity 0. This study analyzed the osmoregulatory process of catadromous E. sinensis in different salinity from the point of osmoregulatory organ and behavioral response. These results will provide a scientific basis for the osmoregulatory mechanism of E. sinensis, which are conducive to evaluating and analyzing the impact of saltwater intrusion in the Yangtze River estuary on resource fluctuation. Simple Summary: DNA methylation is an important epigenetic modification and plays an important role in the regulation of gene expression. The study of DNA methylation will help to explore the effects of epigenetic modifications, other than DNA sequence variation, on biological phenotypes and physiological functions, promoting the revolution of livestock selection and breeding practices. DairyMeade sheep (thin-tailed) and Mongolian sheep (fat-tailed) have large differences in their dairy and meat production performances, as well as their tail phenotype, thus providing us with good materials for genomic DNA methylation studies. The below results provided a genome-wide DNA methylation landscape of muscle and tail-fat tissues between DairyMeade sheep and Mongolian sheep and a series of differentiated methylation regions (DMRs) in which CAMK2D may play a crucial role in fat metabolism and meat quality traits. These results may help us to understand how DNA methylation affects the economic traits of domestic animals like sheep. This study aimed to explore the genome-wide DNA methylation differences between muscle and tail-fat tissues of DairyMeade sheep (thin-tailed, lean carcass) and Mongolian sheep (fat-tailed, fat-deposited carcass). Whole-genome bisulfite sequencing (WGBS) was conducted and the global DNA methylation dynamics were mapped. Generally, CGs had a higher DNA methylation level than CHHs and CHGs, and tail-fat tissues had higher CG methylation levels than muscle tissues. For DNA repeat elements, SINE had the highest methylation level, while Simple had the lowest. When dividing the gene promoter region into small bins (200 bp per bin), the bins near the transcription start site (±200 bp) had the highest CG count per bin but the lowest DNA methylation levels. A series of DMRs were identified in muscle and tail-fat tissues between the two breeds. Among them, the introns of gene CAMK2D (calcium/calmodulin-dependent protein kinase II δ) demonstrated significant DNA methylation level differences between the two breeds in both muscle and tail-fat tissues, and it may play a crucial role in fat metabolism and meat quality traits. Simple Summary: Myostatin (MSTN) gene knockout can increase lean muscle mass and has been widely used in livestock breeding. MSTN deficiency also regulates various metabolic processes. However, the effect of MSTN knockout on the liver, the largest metabolic organ, has not been reported. In this study, physiological and biochemical parameters of serum, untargeted and targeted metabolomics of MSTN+/− and WT cattle were studied, and we found that the knockout of MSTN could regulate liver metabolism and promote bile acid metabolism. This may be due to the enhanced expression of bile acid synthesis genes in MSTN+/− bovine livers. In conclusion, MSTN knockout regulated bile acid metabolism via enhanced bile acid synthesis. Myostatin (MSTN) is a major negative regulator of skeletal muscle mass and causes a variety of metabolic changes. However, the effect of MSTN knockout on bile acid metabolism has rarely been reported. In this study, the physiological and biochemical alterations of serum in MSTN+/− and wild type (WT) cattle were investigated. There were no significant changes in liver and kidney biochemical indexes. However, compared with the WT cattle, lactate dehydrogenase, total bile acid (TBA), cholesterol, and high-density lipoprotein (HDL) in the MSTN+/− cattle were significantly increased, and glucose, low-density lipoprotein (LDL), and triglycerides (TG) were significantly decreased, indicating that MSTN knockout affected glucose and lipid metabolism and total bile acids content. Targeted metabolomic analysis of the bile acids and their derivatives was performed on serum samples and found that bile acids were significantly increased in the MSTN+/− cattle compared with the WT cattle. As the only bile acid synthesis organ in the body, we performed metabolomic analysis on the liver to study the effect of MSTN knockout on hepatic metabolism. Metabolic pathway enrichment analysis of differential metabolites showed significant enrichment of the primary bile acid biosynthesis and bile secretion pathway in the MSTN+/− cattle. Targeted metabolomics data further showed that MSTN knockout significantly increased bile acid content in the liver, which may have resulted from enhanced bile acid synthesis due to the expression of bile acid synthesis genes, cholesterol 7 alpha-hydroxylase (CYP7A1) and sterol 27-hydroxylase (CYP27A1), and upregulation in the liver of the MSTN+/− cattle. These results indicate that MSTN knockout does not adversely affect bovine fitness but regulates bile acid metabolism via enhanced bile acid synthesis.
