The periparturient period is critical according to health, productivity and profitability. As this period is fundamental for the success of the lactation period, the interest in improving periparturient health by dietary supplements increased in recent years. The present study investigated the effects of feeding nicotinic acid (NA) combined with varying dietary energy densities on immunological, hematological and biochemical parameters of periparturient cows differing in parity. Thirty-six multiparous and 20 primiparous dairy cows were enrolled in the study 42 days before expected parturition date until 100 days postpartum with the half of the cows being supplemented with 24 g of NA/d. After parturition a diet with 30% concentrate was fed to all cows which was followed by different concentrate escalation strategies. Dietary NA supplementation was ceased on day 24 postpartum. Dietary NA increased (P = 0.010) serum nicotinamide concentrations (mean of 3.35± 1.65 μg/mL), whereas NA could not be detected. Present data emphasize that periparturient cows are faced with major physiological challenges and that both parity-groups have different prerequisites to adapt to those changes irrespective of NA supplementation. Simple Summary: l-carnitine might limit the mitochondrial energy generation from fatty acids, particularly in periods of enhanced energy requirement such as during lipopolysaccharide (LPS) mediated innate immune responses. This study examined the influence of a dietary l-carnitine supplementation on dairy cows in mid-lactation which were challenged by an injection with LPS. Lipopolysaccharides (LPS) are often used in animal models to study intervention measures during innate immune responses such as APR. Thus, the aim of the study was to investigate the effects of dietary l-carnitine supplementation during an LPS-induced APR in mid-lactating German Holstein cows. Animals were assigned to a control (CON, n = 26) or l-carnitine group (CAR, n = 27, 25 g rumen-protected l-carnitine/cow/d) and received an intravenous injection of LPS (0.5 μg/kg body weight) at day 111 post-partum. Blood samples were collected from day 1 pre-injection until day 14 post-injection (pi). From 0.5 h pi until 72 h pi blood samplings and clinical examinations were performed in short intervals. Clinical signs of the APR were not altered in group CAR except rumen motility which increased at a lower level compared to the CON group after a period of atonia. Simple Summary: Phosphorus and calcium are essential nutrients for body functions including the immune system and are generally supplemented to poultry diets. Phosphorus is also present in plant feedstuffs, bound as phytate, which can be used by enzymatic hydrolyzation in the chicken. A reduction of dietary mineral phosphorus might consequently be conceivable, without negatively influencing the immune system. The high concentration of calcium in diets for laying hens that is needed for eggshell formation may inhibit phytate degrading enzymes, and thus, decrease phosphorus availability for the hen. Both phytate degradation and several immune parameters are known to be strain-specific, making an interaction of the genetic background and the dietary phosphorus and calcium supply with the immune system likely. The aim of the study was to evaluate the impact of reduced concentrations of dietary phosphorus and calcium on the peripheral and gut-associated immune system in two laying hen strains. Reduced mineral phosphorus enhanced, while reduced calcium reduced several immune parameters. The two strains showed differences in many immune parameters, but only the impact of dietary phosphorus was influenced by the genetic background. Reduced calcium levels resulted in lower numbers of T cells in blood and cecal tonsils in both strains, suggesting negative effects on adaptive immunity. Simple Summary: Phosphorus is stored as phytate in plant seeds, which are the main components of poultry feed. Poultry can utilize phytate phosphorus after its cleavage catalyzed by enzymes. These enzymes are inhibited by high concentrations of calcium and phosphorus in diets. As laying hens require a high calcium concentration for eggshell production, the inhibition of enzymes might be high. Therefore, the objective of this study was to characterize the degradation of phytate and the utilization of phosphorus and calcium by two laying hen strains supplied with standard or reduced levels of dietary calcium and phosphorus at the egg production peak. The calcium level had a clear effect on phytate degradation products and mineral utilization. The phosphorus level had no effect on these traits, suggesting that actual recommendations for phosphorus supply of laying hens are too high. Differences were noted between the two hen strains and among individual hens regarding calcium and phosphorus metabolism. This is a first step in identifying individual birds that are more capable than others in using phytate phosphorus under challenging conditions. Thus, the two strains may use different mechanisms to meet their respective P demand, i.e., via effective phytate degradation and transcellular transport. Simple Summary: Dairy cows develop metabolic diseases especially in the transition period due to high energy requirements for the process of calving, beginning milk production and, simultaneously, restricted feed intake capacity. L-carnitine is endogenously synthesised as an obligatory, quaternary amine for the initial step of ß-oxidation, but with the onset of lactation it is also excreted with milk, whereby its availability for other metabolic pathways might be limited. Supplemental L-carnitine might be able to fill in this apparent gap and to enhance the efficiency of ß-oxidation, whereby the magnitude of negative energy balance would be decreased. The present experiment mainly focused on the energy-consuming process of calving itself and on the energy metabolism during the first weeks of lactation. Dairy cows are metabolically challenged during the transition period. Furthermore, the process of parturition represents an energy-consuming process. The degree of negative energy balance and recovery from calving also depends on the efficiency of mitochondrial energy generation. At this point, L-carnitine plays an important role for the transfer of fatty acids to the site of their mitochondrial utilisation. Blood and clinical parameters were recorded in high resolution from 0.5 h to 72 h pp. L-carnitine-supplemented cows had higher amounts of milk fat in early lactation and higher triacylglyceride concentrations in plasma ap, indicating increased efficiency of fat oxidation. Simple Summary: Weaning calves from liquid to solid feed can be a stressful event in their life and can affect growth, development and welfare. It is commonly done at the age of 7 to 8 weeks on dairy farms, but weaning at a greater age could potentially reduce the associated stress. Therefore, it might improve growth rates and enable a smooth transition to an adult liver metabolism. To confirm this hypothesis this study evaluated the effect of two different weaning ages (7 vs. 17 weeks of age) on female Holstein calves. Furthermore, the effect of mothers' parity was analyzed (primiparous vs. multiparouos). Primiparous cows were often immature and still developing during their first pregnancy. This can lead to negative intrauterine conditions and result in long-term changes in the calf's metabolism. Late-weaned calves consumed high amounts of concentrate feed before weaning despite their high milk replacer intake, indicating the maturation of their rumen. In addition, they experienced a smooth transition to an adult liver metabolism as reflected by steady plasma glucose and cholesterol concentrations. Later weaning corrected the reduced growth of calves born to primiparous cows as well, indicating that those particularly benefitted from late weaning. All benefits were indicated by slower changes of blood metabolites and higher growth rates, which might lead to better health and productivity in their subsequent lifetime. Development of calves depends on prenatal and postnatal conditions. Primiparous cows were still maturing during pregnancy, which can lead to negative intrauterine conditions and affect the calf's metabolism. It is hypothesized that weaning calves at higher maturity has positive effects due to reduced metabolic stress. We aimed to evaluate effects of mothers' parity and calves' weaning age on growth performance and blood metabolites. Fifty-nine female Holstein calves (38.8 ± 5.3 kg birth weight, about 8 days old) were used in a 2 × 2 factorial experiment with factors weaning age (7 vs. 17 weeks) and parity of mother (primiparous vs. multiparous cows). Calves were randomly assigned one of these four groups. Live weight, live weight gain and morphometry increased over time and were greater in calves weaned later. Metabolic indicators except total protein were interactively affected by time and weaning age. Leptin remained low in early-weaned calves born to primiparous cows, while it increased in the other groups. The results suggest that weaning more mature calves has a positive effect on body growth, and calves born to primiparous cows particularly benefit from this weaning regimen.
