Simple Summary: The leishmaniases are vector-borne, neglected diseases caused by parasites of the genus Leishmania, widely diffused around the globe. Clinical manifestations range from localized cutaneous and mucocutaneous lesions to the visceral form, with potentially fatal outcomes. Associated with malnutrition, a weak immune system, and no access to health-care facilities, these conditions affect the poorest populations worldwide. In addition, dogs develop a multisystemic and fatal disease, and act as the main parasite reservoir for some Leishmania species. Currently, human, and canine leishmaniases share the same treatment, which includes very few, difficult to administer, expensive, and toxic drugs. Moreover, drug resistance is increasingly spreading, and there is no human vaccine. Therefore, improved and safer treatments are a high priority. Here, monovalent ionophores (salinomycin, monensin, and nigericin) were repurposed in vitro for their leishmanicidal abilities against both insect-stage parasites (promastigotes) and intracellular forms (amastigotes) within human and primary canine macrophages. These compounds showed similar antiparasitic effects against distinct Leishmania spp. promastigotes. Interestingly, a differential and host-specific anti-amastigote efficacy was observed, being these compounds more active against human- than canine-infected macrophages. Altogether, these data indicate a potential application of ionophores against Leishmania infections and challenge the concept that the same compounds may be equally effective against both human and canine leishmaniases. Disease control is mainly based on chemotherapy, which, at present, shows serious drawbacks both in humans and dogs. Therefore, the discovery or repurposing of new treatments is mandatory. Here, three monovalent ionophores (salinomycin, monensin, nigericin) were tested against promastigotes of Leishmania (L.) infantum, Leishmania tropica, and Leishmania braziliensis, and against amastigotes of L. infantum within human and, for the first time, canine macrophages. All three drugs were leishmanicidal against all Leishmania spp. promastigotes with IC50 values between 7.98 and 0.23 µM. Monensin and nigericin showed IC50 values < 1 µM, whereas salinomycin was the least active compound (IC50 > 4 µM). Notably, the ionophores killed L. infantum amastigotes within human THP-1 cells with IC50 values ranging from 1.67 to 1.93 µM, but they only reduced by 27–37% the parasite burden in L. infantum-infected canine macrophages, showing a host-specific efficacy. Moreover, a selective higher toxicity against canine macrophages was observed. Overall, repurposed ionophores have the potential to be further investigated as anti-Leishmania agents, but different drug options may be required to tackle human or canine leishmaniases. Simple Summary: The tiger (Panthera tigris) is the largest of the wild cats and is a pivotal member of many ecosystems and cultures. Despite a long history of concern for wild tigers, both their range and total number have collapsed: fewer than 3500 animals now live in the wild. Furthermore, an increased demand for tiger parts and consequent poaching as well as hunting of their prey has decimated populations across Asia, both in and out of reserves. All subspecies of tigers are globally endangered. Given the precarious situation of these wild felid, the health of each individual animal is of utmost importance so thorough health monitoring, including laboratory blood testing and physical evaluation, is vital. In human and veterinary medicine, hematological parameters and blood chemistry analysis are used in the diagnostic process, to establish disease processes and to monitor the progression of disease. Additionally, blood testing in wild animals provides baseline values for future studies and may help to support conservation strategies. It is extremely difficult to obtain biological samples from free-living Panthera tigris, therefore the values obtained from captive tigers provide very useful data. This study reports hematological and serum biochemical data in captive tigers. These results will be useful in the evaluation of physiological and pathological variations in wild and captive tiger populations. The aim of this study was to provide additional information for the values of hematological and serum biochemical parameters in healthy captive tigers. Blood samples were collected from 22 clinically healthy tigers (Panthera tigris). The following parameters were analyzed: glucose, urea, creatinine, alanine aminotransferase (ALT), alkaline phosphatase (ALP), total protein (TP) and red blood cells (RBCs), hemoglobin (Hb), hematocrit (Hct) and red cell indices; such as mean cell volume (MCV), mean cell Hb (MCH), mean cell Hb concentration (MCHC), platelet (PLT) and white blood cells (WBCs). The mean hematological values in our tiger population were not significantly different when compared with the same parameters in the previously studied tiger population. The mean values of RBCs and PLT were statistically significantly higher and the mean values of Hb, PCV, MCV, MCH, MCHC, and WBC were lower than the mean values obtained in previous studies on the Amur tiger. The aim of this study determined platelet, erythrocyte, and leukocyte counts in canine leukocyte- and platelet rich plasma (L-PRP) produced using a commercial semi-automated closed system (CPUNT 20, Eltek group, Casale Monferrato, Alessandria, Italy). Whole blood (WB) from 30 healthy dogs was used. In 10 L-PRP bovine thrombin activated samples platelet-derived growth factor isoform BB (PDGF-BB) concentration was also measured. The CPUNT 20 produced clinically useful quantities of sterile canine L-PRP containing a high concentration of platelets (767,633 ± 291,001 μL, p < 0.001), with a 4.4 fold increase in platelet count, lower concentration of erythrocytes (528,600 ± 222,773 μL, p < 0.001) and similar concentration of leukocytes (8422 ± 6346 μL, p = 0.9918) compared with WB. Neutrophils, lymphocytes and monocytes average percent content in L-PRP was 14.8 ± 13.2, 71.7 ± 18.5 and 10.7 ± 6.4, respectively. Activated L-PRP has an average of 3442 ± 2061 pg/mL of PDGF-BB. Erythrocyte, leukocyte, and platelet counts were determined in both whole blood (WB) and resultant L-PRP. The PDGF-BB concentration was evaluated after bovine thrombin activation of 10 L-PRP samples. Results: This commercial system produced on average 2.3 ± 0.7 mL of L-PRP containing a high concentration of platelets (767,633 ± 291,001 μL, p < 0.001), with a 4.4 fold increase in platelet count, lower concentration of erythrocytes (528,600 ± 222,773 μL, p < 0.001) and similar concentration of leukocytes (8422 ± 6346 μL, p = 0.9918) compared with WB. Simple Summary: Indications for whole blood transfusion in equine critical care include severe anemia from surgical blood loss or acute hemorrhage, hemolysis and neonatal isoerythrolysis. In horses, as in other animals, transfusions are associated with a number of inherent risks such as transfusion reactions. Pretransfusion screening and blood typing are indicated to minimize the risk of incompatible red cell transfusions. Equine blood types include seven systems, namely A, C, D, K, P, Q, and U. The major RBC antigens that warrant identification before packed RBC or whole blood transfusions in horses are Ca and Aa. The frequencies of blood groups can vary from one population to another and from one breed to another. In some situations where testing compatibility is not possible, such as in rural practice, the knowledge of the breed blood type frequencies may help selecting the best donor candidate. The aims of this study were to: estimate the prevalence of Ca blood type in horses from northern Italy; estimate the association between Ca blood type sex and breed of horse; estimate the prevalence of anti-Ca alloantibodies in Ca− horses. The prevalence of the Ca+ blood type was 79.1%. No significant association was found between blood type Ca and sex. The total number of Ca− samples with detectable anti-Ca alloantibodies was 7/23 (30.4%). A knowledge of the blood groups and alloantibodies present is essential for the safe transfusion of blood products in horses. Ca blood type was determined on 110 whole blood samples. The prevalence of the Ca+ blood type was 79.1%. This study also provides data about the prevalence of Ca+ blood group in Italian Saddle Horses (77,3%) and Dutch Warmblood (58,3%). No significant association was found between Ca blood type and sex with 79.5% and 78.8% of females and males testing Ca+, respectively. Simple Summary: The most important blood group system in cats is the AB, in which cats are classified as type-A, B or AB. Cats have antibodies against the blood type they do not possess, called alloantibodies. The aims of this study were to update blood type prevalence in cats from Northern Italy and study for the first time the blood type in cats from an insular region of Southern Italy, Sicily; to detect alloantibodies in these feline populations; to compare results with previous studies performed in Italy and between regions in Northern and Southern Italy. Cats from Southern Italy had the highest prevalence of type-B and type-AB, and the lowest prevalence of type-A blood in Italy. In particular, type-AB prevalence was higher than all previous reports in non-pedigree cats in Europe and the Italian prevalence of anti-type-B alloantibodies in type-A cats was the lowest reported worldwide. These results highlight the usefulness of regional studies to report different prevalences in feline blood types. Compatibility tests such as blood typing and cross matching must be considered fundamental in cats of any origin to ensure safe and efficient blood transfusion and to prevent neonatal isoerythrolysis. The aims of this study were to determine the prevalence of A, B and AB blood types and alloantibodies in non-pedigree cats from two regions, one in Northern and one in Southern Italy (Lombardy and Sicily, respectively). A total of 448 samples (52.0% from Northern and 48.0% from Southern Italy) were blood typed. The prevalence of A, B and AB blood types in northern and southern cats were 91.0%, 5.2%, 3.8%, and 77.2%, 12.1% and 10.7%, respectively. The prevalence of type-A blood in southern cats was significantly lower (p = 0.0001) than in northern cats, while type-B and AB blood were significantly higher (p = 0.0085 and p = 0.0051, respectively) in Southern compared to Northern Italian cats. Alloantibodies against type-A blood were found in 94.1% of type-B cats, 11.2% of type-A cats had alloantibodies against type-B blood, while no type-AB cats had alloantibodies with no significant difference between the two Italian populations. Type-AB prevalence in non-pedigree cats in Southern Italy was the highest reported in Europe. Italian type-A cats had the lowest worldwide prevalence of alloantibodies against type-B blood. These results highlight the usefulness of regional studies to report different prevalences in feline blood types and reinforce the importance of blood typing cats before transfusions and mating. Simple Summary: Leishmaniosis caused by Leishmania infantum is a parasitic disease of people and animals transmitted by sand fly vectors. Although dogs in south and central Italy are most affected, in recent decades feline leishmaniosis (FeL) has become an emerging disease. This study aimed to update information on the presence of FeL in stray cats in Milan, in northern Italy; to compare these results with previous studies performed in the same area; and to report aspects of Leishmania infection in cats. A total of 117 stray cats were included and 10 (8.6%) had L. infantum infection. The parasite was identified in popliteal lymph nodes in five cats and five had antibodies against L. infantum. Most infected cats were from a specific area of Milan. Increased gammaglobulins were seen in infected cats, which also had antibodies against the pathogens Anaplasma phagocytophilum, Chlamydophila felis, and Toxoplasma gondii. There was a high prevalence of FeL in the surveyed area of northern Italy. Further studies are needed to understand if these infected cats are being imported from other areas, or if there is a real Leishmania focus in Milan. Investigation of the presence of sand fly vectors in Milan would be useful. Feline leishmaniosis (FeL) is an emerging vector-borne feline disease, with increasing numbers of cases reported and studies performed internationally. This study aimed to update the epidemiological status for FeL in stray cats in Milan, northern Italy; compare these results with previous studies in Northern Italy; and report clinicopathologic findings and coinfections in cats infected with Leishmania spp. A total of 117 cats were tested for L. infantum and retrovirus infection, hematological, and biochemical parameters. Demographic and clinical data were collected and FeL affected cats screened for selected coinfections. Overall, 10/117 (8.6%) cats tested positive for L. infantum: in five cats L. infantum DNA was found in popliteal lymph nodes and five were IFAT seropositive at titers from 1:80 to 1:160. Infected cats were concentrated in a specific area of Milan (p = 0.0154). No specific clinicopathologic abnormalities or retroviral infections were significantly linked to the infection, other than hypergammaglobulinemia (p = 0.0127). Seroreactivity to Anaplasma phagocytophilum, Chlamydophila felis, and Toxoplasma gondii was found in some infected cats. A high prevalence of FeL was found in a non-endemic area of northern Italy and future studies should continually monitor this data to understand whether these cases are imported or if Leishmania vectors are present in this area. Due to pressures from poaching and retaliatory killings, this carnivore had lost an estimated 93% of its historic range. Given the critical situation of these wild felines, the health of each individual is of prime importance and laboratory blood testing, as well as evaluation of their physical condition, is important in their health assessment. Protein concentrations in the blood can be altered by malnutrition and dehydration as well as by disease. Serum electrophoresis allows the identification of the different protein fractions present in the blood and represents a useful tool in the diagnosis and monitoring of a number of diseases. Serum protein electrophoresis is a useful tool in the diagnosis and monitoring of a number of diseases. In this study, we evaluated agarose gel serum protein electrophoresis on samples from 11 healthy captive tigers. Serum electrophoresis on all 11 tiger samples successfully separated proteins into albumin, α1, α2, β1, β2 and γ globulin fractions as in other mammals. Electrophoretic patterns were comparable in all tigers. Mean± standard deviation or median and range values obtained for each protein fraction in healthy tigers were, respectively: 3.6 ± 0.2, 0.21 (0.2–0.23), 1.2 ± 0.2, 10.7 ± 0.2, 0.4 (0.3–0.6), 1.2 (1–1.8) gr/dL. The results of this preliminary study provide the first data on serum electrophoretic patterns in tigers and may be a useful diagnostic tool in the health assessment of this endangered species.
