The experiment was conducted to evaluate alternative protein ingredients in a low-fish meal (FM) diet for red seabream (Pagrus major). Twelve experimental diets were formulated. Control diet (CON) was designed to contain 60% FM. Other experimental diets were formulated by replacing 50% of FM from the CON with soy protein concentrate (SPC), corn gluten (CG), meat meal (MM), and/or chicken byproduct meal (CBM). Four diets were designed including one of SPC, CG, MM, or CBM as FM replacer and designated as SPC, CG, MM, and CBM. Six other diets were formulated by adding two ingredients as SPC and CG, SPC and MM, SPC and CBM, CG and MM, CG and CBM, or MM and CBM, and designated as SCG, SMM, SCM, CMM, CCM, and MCM, respectively. The 12th diet (MIX) was formulated by including SPC, CGM, MM, and CBM. Triplicate fish groups (50.2 ± 0.1 g) were hand-fed for 12 weeks. Weight gain (WG) of fish was significantly improved by MM and MCM diets compared to CG, SCG, CMM, and CCM diets. WG of CON, SPC, CM, SMM, SCM, and MIX groups were comparable with MM and MCM groups. The lowest WG was observed in CG and CMM groups. Feed efficiency (FE) was significantly higher in MM group compared to SPC, CG, SGC, and CMC groups. FE of MCM group was significantly higher than CG and SCG groups. Fillet linolenic acid (C18:2n–6) level in CG group was significantly higher than CON, MM, CM, SCM, CCM, and MCM groups. Serum lysozyme activity was significantly higher in MCM and MIX groups. Therefore, a high level of dietary CG reduces the growth performance and feed utilization of red seabream. A 50-day feeding trial was conducted to evaluate the efficiency of single and mixed strains of probiotic bacteria supplements on juvenile red sea bream (Pagrus major). The study investigated the growth, digestibility, hemato-biochemistry, antioxidant, immune, immune, and growth gene expression and stress responses of P. major. Three hundred juvenile P. major (21.56 ± 0.09 g) were randomly distributed into fifteen 200-L polyethylene tanks (20 fish per tank) in triplicate of 5 treatments designated as D1 = the basal diet; Streptococcus faecalis (SF) T − 110 5 × 106 cfu/g diet and Bacillus amyloliquefaciens (BA) TOA 5001 5 × 105 cfu/g diet in the second group (D2) at 0.2%; mix SF T − 110 1 × 106 cfu/g diet, Lactobacillus plantarum (LP) TO − A 4 × 105 cfu/g diet, Bacillus mesentericus (BM) TO − A 1 × 102 cfu/g diet, BA TOA 5001 5 × 105 cfu/g diet in the third group (D3) at 1%; mix SF T − 110 1 × 106 cfu/g diet, LP TO − A 4 × 105 cfu/g diet, and BM TO − A 1 × 102 cfu/g diet in fourth group (D4) at 0.5%; and single strain BA TOA5001 5×105 cfu/g diet in the fifth group (D5) at 0.5% of dietary proportion. Results showed that D2 and D3 fish groups exhibit better performance, followed by D4, D5, and control group D1. This finding demonstrated that the use of BA in mix strain probiotic bacteria diet (D2, D3) had improved immune response, antioxidant enzymes, immunity (TNF-a, IL-1b), and growth-related (IGF-1, IGF-2) mRNA expression of juvenile P. major compared to the mix strain D4, single strain D5, and the control D1. Furthermore, single strain D5 and mix strain D4 also exhibit relatively better immune responses in P. major than in control D1. Considering the overall fish performances, mix strain SF+BA (D2) and mix strain SF+BM+LP+BA (D3) were the recommended potential mix strain probiotic bacteria supplement for P. major and may be useful also for other related aquatic species. The optimum level of fishmeal (FM) protein replacement by bacterial protein meal (BPM) was determined in the diet of juvenile red sea bream, Pagrus major. Six isoproteic and isolipidic diets were formulated to replace 0 (control diet), 190 , 280, 370, 470 and 560 g of the FM protein kg‐1 diet with BPM protein, and referred to as C, BP19, BP28, BP37, BP47 and BP56, respectively. Each diet was fed to triplicate groups of twenty fish (mean weight ~43 g) twice per day to apparent satiation. Fish fed diets C and BP19 did not show significant differences (p >.05) in growth, apparent digestibility or protein and lipid retention efficiency, but the daily feeding rate (DFR) was significantly lower under BP19 than under C. However, almost all growth parameters were significantly reduced when FM protein replacement with BPM was further increased from 280 to 560 g kg‐1 diet (p <.05). The results of polynomial regression analysis (second‐order) revealed significant negative correlations between dietary copper levels and final mean weight (R2 =.913), specific growth rate (R2 =.913) and DFR (R2 =.956). The results indicated that BPM could replace approximately 190 g of FM protein kg‐1 diet in the juvenile red sea bream diet without compromising growth performance or feed efficiency. Purine nucleotides regulate the cellular functions in the animal body. The current study evaluates the comparative efficacy of dietary purine nucleotides, that is, adenosine monophosphate (AMP), guanosine monophosphate (GMP) and inosine monophosphate (IMP) in red sea bream Pagrus major. Semi‐purified basal diet was formulated (Control, D1) containing 550 g/kg protein, supplemented with purine nucleotides AMP, GMP and IMP at their optimum supplementation level (2, 4 and 4 g/kg) to formulate the experimental diet groups D2, D3 and D4, respectively. Initial weight of 3.5 ± 0.01 g fish was randomly fed test diets in triplicate. After 56 days, % weight gain (p =.003), specific growth rate (p =.003) and apparent lipid digestibility (p =.04) were significantly higher in fish fed diet group D4 followed by D3 and D2. Supplemented groups showed significantly higher feed intake in comparison to control (p =.001). Supplemented groups showed significantly increased and decreased NBT (p =.003) and CAT (p =.003) activity, respectively. Fish fed IMP supplemented diets had significantly lower blood urea nitrogen (p =.04), glutamyl oxaloacetic transaminase (p =.04) and glutamic‐pyruvate transaminase (p =.001) followed by other supplemented groups. Supplemented diet groups showed enhanced stress resistance. Interestingly, D2 and D4 groups showed best oxidative stress status of fish. The effects of dietary protein and lipid sources on growth and survival of the red sea bream Pagrus major (30-days old) and Japanese flounder Paralichthys olivaceus (15-days old) were examined using four zein-microbound diets (MBD) containing the following nitrogen and lipid sources: MBD-1, SBP-1000 (soybean peptides with molecular weights of about 1000) + fish oil calcium soap (FOCS); MBD-2, SBP-3000 (molecular weights of about 3000) + FOCS; MBD-3, soybean protein isolate (SPI) + FOCS; MBD-4, SBP-1000 + stearoyl calcium lactate (SCL). Six groups of the test fish were maintained on the following diets for 30 days; group 1, live food; group 2, 1/2 live food + 1/2 MBD-1; group 3, MBD-1; group 4, MBD-2; group 5, MBD-3; group 6, MBD-4. In the red sea bream, high performance in terms of total length (TL) and body weight gain (BWG) was obtained in groups 1, 2, 3 and 4. However, the fish receiving MBD-3 (group 5) and MBD-4 (group 6) had shorter TL and lower BWG than those receiving the other diets. Regarding the Japanese flounder, high performance in terms of TL, BWG, and survival rate (SR) was obtained in groups 1 and 2. Group 3 receiving MBD-1 gave the BWG and SR close to those of group 2 but lower than group 1. In contrast, groups 5 and 6 receiving MBD-3 or MBD-4 alone had significantly ( P < 0.05) shorter TL and lower BWG than groups 1 and 2. The present study thus indicated that soybean peptides with molecular weights of 1000–3000 and FOCS are superior to SPI and SCL as nitrogen and lipid sources of MBD for the red sea bream and Japanese flounder at larval and early juvenile stage. The nutritional value of the kernel meals of three species of lupin ( Lupinus albus, L. angustifolius and L. luteus) was compared against each other and solvent extracted soya bean meal and wheat gluten, when fed to either rainbow trout or red seabream. The digestible nutrient and energy values were determined for each ingredient, based on the diet substitution digestibility method. Each test ingredient was included in test diets at 300 g kg−1. As different faecal collection methods were used on each species, only ingredient comparisons within each fish species study are valid. Protein digestibility for all lupin kernel meals was significantly better than for soya bean meal when fed to red seabream, although only numerically so when fed to rainbow trout. The highest protein digestibility in both fish species was that from the wheat gluten (100.0% for both rainbow trout and red seabream). Energy digestibility for each of the lupin kernel meals (range 62.4%–64.9% in rainbow trout and 60.9%–69.5% in red seabream) was less than that obtained from soya bean meal (75.1% and 81.0% for rainbow trout and red seabream, respectively). However, the higher gross energy content of the lupin kernel meals still resulted in both L. albus and L. luteus providing equivalent levels of digestible energy as that of soya bean meal. Organic matter digestibility was generally poorer for each of the lupin kernel meals relative to that for the soya bean meal and particularly compared with the wheat gluten. Phosphorus digestibility was significantly better in all lupin kernel meals than that from both the soya bean meal and wheat gluten. Highest phosphorus digestibility was that obtained from L. albus kernel meal (100.0% for both rainbow trout and red seabream) although notably the phosphorus digestibility of all lupin kernel meals when fed to rainbow trout was determined at 100%. Similarly the digestibility of phosphorus from lupin kernel meals fed to red seabream was also very high. This study supported the good nutritional value to both rainbow trout and red seabream of kernel meals of all three species of lupin, although the digestible nutrient attributes of kernel meal from L. luteus were particularly favourable. Due to its traditionally good availability, digestibility and high content of n − 3 HUFA, fish oil is the main lipid source in fish feeds. However, world demand for this product has grown significantly in recent years, whereas its production, based on fisheries landings, is static. The purpose of the present study was to assess the effect of partial replacement of fish oil in compound diets for gilthead seabream and seabass, by several vegetable oil sources, on growth, dietary fatty acid utilization and flesh quality. Five iso-energetic and isoproteic experimental diets were formulated (25% lipid content). Fish oil was the only added lipid source in the control (FO) diet, and it was included in the other experimental diets at a level high enough (40% of FO diet) to keep the n − 3 HUFA levels well over 3% in order to cover the essential fatty acid requirements of these species. Fish oil was replaced by soyabean oil (SO), rapeseed oil (RO) and linseed oil (LO) or a mixture (Mix) of them. Feed intake in all dietary groups was in the range of results obtained for commercial diets in both species, and growth and feed utilization were very good. The results show that, providing a minimum content of essential fatty acids in the diet, it is possible to replace up to 60% of the fish oil by SO, LO and RO or a mixture of them in diets for seabream and seabass, without compromising fish growth. Fatty acid composition of liver and muscle reflected that of the diet, but utilization of dietary lipids differed between these two tissues and was also different for the different fatty acids. Despite reduction in dietary saturated fatty acids by the inclusion of vegetable oils, their levels in fish liver were as high as in fish fed the fish oil diet, whereas, in muscle, levels were reduced according to that in the diet. Linoleic and linolenic acids were accumulated in the liver proportionally to their levels in the diet, suggesting a lower oxidation of these fatty acids in comparison to other 18C fatty acids. Regarding eicosapentaenoic acid (20 : 5 n − 3; EPA), docosahexaenoic acid (22 : 6 n − 3; DHA) and arachidonic acid (20 : 4 n − 6; ARA), these essential fatty acids were reduced in the liver at a similar rate, whereas DHA was preferentially retained in the muscle in comparison with the other fatty acids, denoting a higher oxidation particularly of EPA, in the muscle. Some other PUFA increased despite their low dietary levels in seabream fed LO diets and in seabass fed SO diet, suggesting the stimulation of delta-6 and delta-5 desaturase activity in marine fish. This study evaluated the potential for manipulating the fatty acid composition of juvenile red seabream, Pagrus auratus. Prior to the start of the study, three groups of fish had been reared for 3 months on a fish oil based diet or diets where the added fish oil had been replaced with either canola or soybean oil. In the present study, fish that had previously been fed either the canola or soybean oil diets were fed a fish oil based diet. Three additional treatments included fish being maintained on their original diets of fish oil, canola oil or soybean oil. Fish were fed their respective diets twice daily to apparent satiety for 32 days. Samples of fish from each treatment were collected after 0, 1, 2, 4, 8, 16 and 32 days. Composition and growth of the fish were determined at each sample point. Most treatments showed no differences in growth performance, although fish fed a fish oil diet after previously being fed a soybean oil diet showed slightly better growth. No significant differences among treatments were observed in proximate composition of the fish, although there was a significant increase in total fat and individual fatty acid (g kg−1 live-weight) content of the fish from all treatments over the period of the study. No significant changes in the relative fatty acid composition (% of total fatty acids) over time were observed in the three treatments where fish were maintained on their original diets. In contrast, fish that were previously fed either the canola or soybean oil diets and were then fed a fish oil diet had significant changes in both the relative (% of total fatty acids) and absolute (g kg−1 live-weight) fatty acid content. Key changes observed included a decrease in the relative levels of polyunsaturated fatty acids (PUFA) such as 18 : 2 n − 6 and 18 : 3 n − 3. Increases in the relative levels of the long-chain polyunsaturated fatty acids (lcPUFA) 20 : 5 n − 3 and 22 : 6 n − 3 were also observed in both treatments. The rates of absolute (g kg−1 live-weight) change/accumulation of these fatty acids followed an exponential equation that differed for each fatty acid in each treatment. Examination of the retention efficiency of specific fatty acids also showed marked differences between fatty acids within treatments and also differences between treatments. Biologically important fatty acids such as 20 : 5 n − 3 and 22 : 6 n − 3 had only moderate retention efficiencies and these were unaffected by treatment. In contrast, the retention efficiencies of 18 : 2 n − 6 and 18 : 3 n − 3 suggested selective retention of these fatty acids when fed fish oil diets, but moderate catabolism when fed the plant oil diets. There were also high retention efficiencies of most saturated and monounsaturated fatty acids suggestive of active retention and/or active synthesis of these fatty acids by the fish. The results of this study, particularly the increases in lcPUFA, support the usefulness of a fish oil based finisher diet for fish raised predominantly on plant oil based diets. Abstract This study assessed the nutritional and biological value of a noncommercial, transgenic line of the Australian sweet lupin, Lupinus angustifolius , (cv. Warrah), produced to increase the methionine content of the seed. An initial experiment demonstrated that differences in the methionine content of the transgenic and nontransgenic control lupins had no apparent influence on the growth of juvenile red seabream fed practical diets. Re-evaluation of the nutritional characteristics of the lupin meals with subsequent digestibility studies allowed the determination of the digestible value of the protein and energy content of each of the varieties. The digestible protein content of either variety was similar, however significant differences in the digestible energy value of each variety existed (56.3% cf. 64.0%). This re-evaluation of the nutritional value of the genetically manipulated (GM) and non-GM lupin varieties enabled the reformulation of diets on a digestible protein and energy basis. A second growth trial was undertaken using sub-satietal pair-feeding regimes, with the experiment also involving protein-restrictive diets to allow expression of the differences in the methionine content of the transgenic and nontransgenic lupin meals. A significant benefit of the enhanced methionine level in the transgenic lupin was observed. It is argued that in the high-protein fish diets used, the importance of amino acid composition is relatively limited. Economic modelling of the potential value of the increased methionine in the transgenic lupin suggests that this will have limited benefit for aquaculture industries, and that greater value would be attributable to higher protein and energy levels in ingredients.
