When oil is spilled into the environment its toxicity is affected by abiotic conditions. The cumulative and interactive stressors of chemical contaminants and environmental factors are especially relevant in estuaries where tidal fluctuations cause wide variability in salinity, temperature, and ultraviolet (UV) light penetration, which is an important modifying factor for polycyclic aromatic hydrocarbon (PAH) toxicity. Characterizing the interactions of multiple stressors on oil toxicity will improve prediction of environmental impacts under various spill scenarios. This study examined changes in crude oil toxicity with temperature, salinity, and UV light. Oil exposures included high-energy, water-accommodated fractions (HEWAFs) and thin oil sheens. Larval (24-48 h post hatch) estuarine species representing different trophic levels and habitats were evaluated. Mean 96 h LC 50 values for oil prepared as a HEWAF and tested under standard conditions (20 ppt, 25 °C, No-UV) were 62.5 µg/L tPAH 50 (mud snails), 198.5 µg/L (grass shrimp), and 774.5 µg/L (sheepshead minnows). Thin oil sheen 96 h LC 50 values were 5.3 µg/L tPAH 50 (mud snails), 14.7 µg/L (grass shrimp), and 22.0 µg/L (sheepshead minnows) under standard conditions. UV light significantly increased the toxicity of oil in all species tested. Oil toxicity also was greater under elevated temperature and lower salinity. Multi-stressor (oil combined with either increased temperature, decreased salinity, or both) LC 50 values were reduced to 3 µg/L tPAH 50 for HEWAFs and < 1.0 µg/L tPAH 50 for thin oil sheens. Environmental conditions at the time of an oil spill will significantly influence oil toxicity and organismal response and should be taken into consideration in toxicity testing and oil spill damage assessments.

Evaluation of effects on fish reproduction and development during chemical exposures lasting for multiple generations is sometimes limited by variable reproductive responses and the time required for the exposure. Established testing methods and the short life cycle of the sheepshead minnow, Cyprinodon variegatus, make this species particularly suitable for use in identifying potential impacts of contaminants in estuarine and marine environments. This study describes the refinement of life-cycle exposure methods that increased the reliability of reproduction in sheepshead minnows and reduced the time to maturation for larvae and juvenile fishes. A test of three spawning chamber designs, three sex ratios, and two photoperiods identified conditions that reduced the coefficient of variation in egg production from >100% to as little as 32%. The most reliable results were produced with groups of three female and two male fishes (all of similar size) when they were placed in a rectangular chamber and acclimated for 12 days. A test water temperature of 26.5 +/- 2 degrees C and a 14L:10D photoperiod resulted in fish producing a mean of 74 embryos per female per day, with a coefficient of variation of 31.8%. Egg fertility exceeded 90%, with a hatch rate of 95% for normal embryos (>or=80% yolk) and a hatch rate of or=2.7 cm standard length) was critical for spawning readiness. Adult fish were prepared for the spawning assessment by adding frozen brine shrimp to their diet. Results of these experiments provide methods that are of particular interest in assessment of endocrine-disrupting chemicals that are known to affect reproduction.

We used three populations of mummichogs (Fundulus heteroclitus), one from a polluted site (Piles Creek [PC], New Jersey) and two from cleaner sites (Tuckerton [TK], New Jersey, and East Hampton [EH], New York), to study (1) whether embryonic, embryonic plus larval, or larval exposure to methylmercury (MeHg) altered larval prey capture ability and growth; and (2) whether there were differences in tolerance to MeHg-induced behavioral changes among the three populations. Eggs and sperm were obtained from mummichogs captured in the field, and their embryos and larvae were kept in clean sea water or MeHg solution (5, 10 microg/L). Larvae were then tested regularly for prey capture rates and prey capture efficiencies, and their lengths were measured. Embryonic exposure to MeHg induced transitory and recoverable impairments in larval prey capture ability, whereas larval exposure alone was relatively ineffective. When both embryos and larvae were treated, larval prey capture ability was affected at a lower concentration and a wider range of larval ages. In terms of growth and prey capture ability, response of larvae to embryonic or larval or exposure to both stages to MeHg varied with populations. TK fish were the most tolerant with respect to behavioral changes but were the most sensitive to MeHg in reduction of growth. EH fish were the most sensitive whenever embryos were treated, and PC fish were the most vulnerable after larval exposure. The population differences in response to MeHg intoxication may be due to pollution related factors or differences in behavioral-related genetic factors.

Concentrations of retinoids, derivatives of vitamin A, were measured in populations of the nonmigratory estuarine fish Fundulus heteroclitus, indigenous to a reference site and a site highly contaminated with polychlorinated biphenyls (PCBs) to address the hypothesis that contaminant exposure causes depletion of stored retinoids in native fish populations. To assess differences related to chemical exposure, as well as season and diet, hepatic retinoid analyses were conducted using fish collected early and late in the natural spawning season and after laboratory holding. Though hepatic retinoid composition was generally similar among groups, hepatic retinoid concentrations differed. Laboratory-held fish had higher hepatic retinyl ester concentrations than field-collected fish. Among field-collected fish, those collected early in the spawning season had higher hepatic retinyl ester concentrations than those collected later in the spawning season. Although there was no evidence of the dramatic retinoid depletions that have been reported in highly exposed populations of other fish species, hepatic retinoid stores were consistently lower in F. heteroclitus indigenous to the highly PCB-contaminated site. These results are consistent with prior findings that fish from this contaminated site are relatively insensitive to some of the toxic effects of PCB exposures, including retinoid depletion.

Many contaminated sites contain a variety of toxicants. Risk assessment and the development of water quality criteria therefore require information on the interactive effects among toxicants in such mixtures. Interactions between metals are relatively well studied, but little is known about interactions between metals and hydrocarbons. This study investigated the interaction between phenanthrene and zinc in the sheepshead minnow Cyprinodon variegatus. Interaction studies were performed with 7-day-old minnows in 96-h bioassays with zinc and phenanthrene at a fixed ratio and with varying proportions of zinc and phenanthrene. Mixture toxicity was quantified with the toxic unit, additive index, and excess function methods. All three methods generally indicated an antagonistic interaction between phenanthrene and zinc, though the results also provide some evidence for a synergistic interaction at low toxicant levels or at specific phenanthrene-to-zinc ratios. Short-term uptake experiments were conducted to determine if the strong antagonistic interaction observed when zinc and phenanthrene were present at 50% of their LC50 values was due to effects of zinc and phenanthrene on each other's uptake. Significantly less 65Zn uptake occurred in the presence of phenanthrene than in its absence. The effects of tributyltin (TBT) to the life cycle of the estuarine fish Cyprinodon variegatus were examined in a 180-day flow-through exposure. The study was initiated with embryos less than 24 h postfertilization and monitored through hatch, maturation, growth, and reproduction under continuous exposure to mean measured TBT concentrations of 5.4, 3.2, 1.3, 0.66, and 0.41 microg TBT/L. Progeny isolated at the onset of reproduction by the parental generation (F0) were monitored for survival as embryos, survival as fry/juveniles, and growth 30 days postisolation. TBT, at a concentration of 5.4 microg/L, significantly reduced embryo survival of the F0 generation sheepshead minnows. By day 145 of the exposure, significant effects to the survival of this generation increased and included all test concentrations >/=0.66 microg/L. Survival of 0.66 microg/L was reduced 59% relative to control survival by termination of the F0 generation. Growth of F0 generation organisms as measured by standard length was significantly reduced only on day 90 at 3.2 microg/L, however no significant reductions of wet or dry weight related to treatment concentration were detected. Due to complete mortality of organisms exposed to 5.4 microg/L by study day 7, effects to fecundity and progeny were monitored at measured concentrations of 3.2, 1.3, 0.66, and 0.41 microg TBT/L. Fecundity, as measured by the production of viable eggs produced per female per day, was unaffected by any of the test treatments. All F1 generation embryos isolated from treatment chambers into 3.2 microg/L died. Survival, standard length, wet and dry weight of the F1 generation at the remaining treatment concentrations were unaffected. The results of this study indicate that exposure to TBT reduced survival of the F0 generation sheepshead minnow and establishes the lowest observed effect concentration (LOEC) as 0.66 microg TBT/L, and the no observed effect concentration (NOEC) as 0.41 microg TBT/L for this species.http://link.springer-ny. com/link/service/journals/00244/bibs/37n2p258.html
