Abstract: The delayed reaction paradigm, consisting to discover two different rules consecutively (delayed alternation and non-alternation task) followed by a delayed reversal task, is a specific marker for the functioning of primate prefrontal cortex. Although several works in rodents report the use of operant delayed alternation tasks, in none of the studies mice with lesion of the prefrontal cortex were used in this paradigm. In the current study, mouse experiments were conducted using a new, totally automated device, the olfactory H-maze. Here, we show that unilateral lesion of the dorsomedial prefrontal cortex in mice induced similar deficits to those observed after frontal lesions in monkeys and humans. These pronounced learning deficits seem to come from difficulty elaborating a new rule and the inability to inhibit the previous rule, characterized by perseveration after prefrontal cortex lesion. The present results demonstrate that this very simple experimental paradigm using the olfactory H-maze presents the advantage to be fast (one training session) and well suited to assess the frontal functions in mice. It should be useful for testing pharmacological or stem cell approaches in order to reduce organic damages or gain insight into the cognitive functions of the frontal cortex using transgenic or gene-targeting mice. • Independent MDD and loneliness effects were examined during n-back task performance. • No MDD or loneliness effects were observed on performance measures. • Loneliness increased, while MDD decreased, frontoparietal task-based connectivity. • MDD and loneliness exerted interactive effect on connectivity in the frontal cortex. • The connectivities are implicated in self-referential regulation and action control. Loneliness affects up to 40 % of middle-aged and older adults, and is closely associated with major depressive disorder (MDD). However, the relationship between loneliness and neural network functioning during executive cognitive processes, such as working memory, in MDD is still unclear. To address this gap, our study recruited 21 medicated MDD patients (mean age = 52.0 ± 5 years) and 24 matched healthy controls (HC) (mean age = 48.7 ± 6 years) who completed an n-back fMRI task. For behavioural performance, we observed no significant moderating effect of MDD or loneliness on the task condition effect. However, loneliness was positively associated, and MDD was negatively associated, with the functional connectivity between the inferior parietal cortex and the rostral dorsomedial prefrontal cortex (DMPFC) during task performance. Furthermore, an interactive effect of loneliness and MDD was observed on the functional connectivity between the supplementary motor area and the caudal DMPFC during the n-back task, with loneliness showing a positive relationship in the HC group but a negative relationship in the MDD group with the connectivity. Our results indicated that loneliness may be associated with altered neural regulatory functioning on self-referential processing and action control, which may further depend on the individual's depressive state. These findings can form the theoretical basis for devising intervention programme aimed at improving the mental wellness of the healthy and depressed lonely individuals. [ABSTRACT FROM AUTHOR]

• PPD patients show abnormal resting state interhemispheric functional connectivity in several brain regions. • Decreased voxel-mirrored homotopic connectivity in dorsomedial prefrontal cortex negatively correlated with the depression severity in PPD. • Aberrant interhemispheric connectivity in the brain may involve in the maternal care network in PPD. The aim of this study is to investigate the alterations of interhemispheric functional connectivity in patients with postpartum depression (PPD) during resting state, and their potential correlations with clinical severity. Twenty- eight patients with PPD and twenty-five matched healthy postpartum (HP) women within 4 weeks after delivery were recruited and performed resting-state functional magnetic resonance imaging(fMRI) scans. Voxel-mirrored homotopic connectivity (VMHC), which is useful for exploring interhemispheric functional connectivity, and has been widely utilized to identify abnormal functional connectivity between the symmetrical brain regions in many diseases, was calculated in the present study, and intergroup VMHC differences in the voxel manner were analyzed. Correlations between VMHC values and clinical variables were also analyzed. Compared with HP, patients with PPD exhibited significantly decreased VMHC values in bilateral dorsomedial prefrontal cortex (dmPFC), dorsal anterior cingulate cortex (dACC) and orbitofrontal cortex (OFC). Furthermore, VMHC values within the dmPFC negatively correlated with the Edinburgh postpartum depression scale (EPDS) score. These findings suggested that functional coordination between several homotopic brain regions were impaired in patients with PPD. This study provided evidences of aberrant interhemispheric connectivity within brain regions involved in the maternal care network in PPD, and may contribute to the further understanding of the neural mechanism underlying PPD.
