The peptidase system in Drosophila melanogaster (dipeptidase-A, -B, and -C and leucine aminopeptidases G and P) was used as a model to study the effects of modifier genes on activity of enzymes with similar functions. A screen of X, second, and third chromosome substitution isogenic lines revealed the presence of activity modifiers for peptidases on all three chromosomes. Correlation analyses indicated that covariation between some of the peptidase activities is independent of genetic background, while others are associated with variable second chromosomes. Chromosome-specific effects on Km, Vmax, and specific activity of partially purified peptidases were also detected. Moreover, a repeatable technique using anion-exchange column chromatography allowed the characterization of possibly two putative peptidic enzymes, glycyl-L-isoleucine-ase and L-leucyl-L-proline-ase, whose kinetic properties differ from the dipeptidases and the leucine aminopeptidases. These findings confirm the existence of activity modifiers for peptidases, much like other enzymes in Drosophila melanogaster.

Dip-A, Dip-B, and Dip-C constitute structural genes for three peptidic enzymes in Drosophila melanogaster distinct from the leucine aminopeptidases. Their ontogenetic and tissue distributions of activities suggest the involvement of these enzymes in a general metabolic role, such as the regulation of amino acid and oligopeptide pools to make amino acids available for protein synthesis. Screening of chromosome substitution isogenic lines for DIP-C activity indicated that, like DIP-A and DIP-B, unlinked activity modifiers exist for Dip-C. The developmental profiles of dipeptidase activities are very similar, except in the pupal stage, during which DIP-C activity is markedly low compared to the other two enzymes. Intercorrelations of dipeptidase activities vary ontogenetically, which is consistent with the need for coordinate expression of these enzymes during certain developmental stages. Tissue-specific expression of dipeptidases in larvae and adults are also similar, although the relative levels of DIP-A activity differ from those of DIP-B and DIP-C in certain organs and body parts. Some of the differences among chromosome substitution lines for dipeptidase activities appear to be systemic, while others are developmental stage-specific and tissue-specific. Second- and third-chromosome variants for DIP-C activity differed in their tissue distribution. This is consistent with the presence of temporal and spatial variants in natural populations for other Drosophila enzymes.

Genetic polymorphism of peptidases A, B, C, and D in the wild rabbit (Oryctolagus cuniculus) was assessed by horizontal starch gel electrophoresis, in a total of 1003 individuals comprising 16 Iberian and 2 French populations and 1 domestic stock. Twenty-five different alleles were detected: 6 in PEPA, 4 in PEPB, 8 in PEPC, and 7 in PEPD. The highest values of observed heterozygosity averaged over the four loci were obtained for the southwestern Iberian populations and a clinal loss of variability in a northeastern direction was detected. A clear separation between the two putative subspecies O. c. cuniculus and O. c. algirus was not obtained.

Genetic variation in the golden-striped salamander (Chioglossa lusitanica) was assessed in 231 individuals from four Portuguese populations by means of horizontal starch gel electrophoresis and isoelectric focusing. Three of 19 enzyme systems, representing 21 presumptive loci, were found to be polymorphic: phosphoglucomutase 1 (PGM1), peptidase B (PEPB), and peptidase D (PEPD). The observed average heterozygosity in Chioglossa lusitanica (0.027) is significantly lower than that observed for other amphibians, either urodeles or salamandrids. Differences in allele frequencies and the presence of private alleles are indicative of a high degree of population differentiation. PEPD, in particular, seems to be a diagnostic locus separating the southernmost population studied from the others.

An inherited electrophoretic variant of prolidase (EC 3.4.13.9), also called peptidase 4 (PEP-4), has been discovered among inbred strains of mice. Analysis of progeny from reciprocal backcrosses established that the electrophoretic forms are expressed codominantly and that Pep-4 is located between the genes for glucosephosphate isomerase (Gpi-1) and pink-eyed dilution (p) on chromosome 7. These data define a region of conserved gene linkage between mouse chromosome 7 and human chromosome 19, as originally indicated by somatic cell hybrid studies, and imply that human prolidase (PEPD) is located in the region of human chromosome 19 pter greater than q13.

Carnosinase is a peptidase which cleaves B-alanyl-l-histidine (carnosine) and closely related dipeptides. Its activity in kidney cytosol of various mouse strains varies more than 50-fold. The highest activity occurs in random-bred CD-1 and inbred NZB/BINJ mice, while it is barely detectable in BALB/cJ, C57BL/6J, and AU/SsJ among others. Carnosinase is immunologically and enzymologically identical in all high-activity strains. This is the first report of quantitative interstrain differences in carnosinase activity. No other peptidase activity has been reported which exhibits the same strain distribution shown here. In matings and backcrosses between the NZB/BINJ and the BALB/cJ strains, the levels of kidney carnosinase activity in the progeny behave as a classical Mendelian trait.

Three dipeptidases in Drosophila melanogaster are under independent genetic control and their structural genes have been localized, Dip-A to 2R and Dip-B and Dip-C to 3R (Voelker and Langley, 1978; Ohnishi and Voelker, 1981). These enzymes were characterized with respect to their substrate specificities, genetic variability (electrophoretic mobility and quantitative activity level), ontogeny (activity and isozyme pattern), and tissue localization. The dipeptide substrate specificities of DIP-A and DIP-B overlap each other considerably, but do not overlap with DIP-C. In natural populations, DIP-B and DIP-C are essentially monomorphic electrophoretically whereas DIP-A is polymorphic for three allozymes. Both DIP-A and DIP-B show quantitative genetic variation of activity level within an allozyme class. All three enzymes are expressed at all stages in the life cycle, but DIP-A and DIP-B activities vary considerably according to developmental stage and sex of adult. The tissue localizations of DIP-A and DIP-B activities show similar patterns and a nearly ubiquitous occurrence of both enzymes, but with particularly high values in larval and adult midguts and in the adult female reproductive system. These results suggest a general metabolic role for the enzymes, such as regulation of the concentrated pools of amino acids and oligopeptides found in Drosophila tissues.

Evidence is presented for the assignment of the gene for dipeptidase 2 to Mus musculus chromosome 18 by synteny testing and karyotypic analysis of Chinese hamster × mouse somatic cell hybrid clones. DIP-2 and chromosome 18 were expressed concordantly in 24/24 clones examined (ten primary clones and 14 secondary clones). Synteny testing indicated that DIP-2 was not expressed concordantly with the expression of any marker enzymes.

The cytosolic di- and tripeptidases of fish were studied in an electrophoretic phylogenetic survey that included elasmobranchs, a holostean, and teleosts. Antisera against four of the peptidases from tuna were raised in rabbits and used to establish homologies between the peptidases of tuna and other fish and between piscine PEP A, B, and S and corresponding enzymes of the higher vertebrates. Substrate specificities, tissue distributions, and electrophoretic mobilities were conserved during the evolution of the fish. The nomenclature for mammalian peptidases was extended to the piscine enzymes, but with reservations in the case of PEP C and E. Using this nomenclature, the six major, genetically independent peptidases are PEP A, B, C, D, E, and S. Within the fish substrate specificity was a reliable indicator of identity. The peptidases of vertebrates thus consist of a widely distributed group of enzymes with constant characteristics. Much of the confusion in the field is probably due to variable and poorly defined species-specific enzymes.

An examination of Drosophila melanogaster from natural populations revealed genetic variation for dipeptidase-A (DIP-A) and dipeptidase-B (DIP-B) activities within sets of lines that differed from one another only in the second or the third chromosome. Analyses of diallel crosses indicate that both activities are inherited additively, and coordinate control of expression is suggested by the significant positive correlation between the two activities. Electrophoresis and thermal denaturation studies failed to detect structural differences among lines with different levels of DIP-A activity. No characteristic level of activity could be associated with any DIP-A allozyme. Mapping experiments revealed the presence of activity modifiers that are in tight linkage with the structural gene, as well as those that manifest their effects from a distance. The maximum genetic distance between a high-activity effect on DIP-A and the structural gene was determined to be 0.029 map unit. These results are in accordance with the prevalence of activity modifiers for various enzymes in Drosophila melanogaster.

Four major peptidases of Drosophila melanogaster have been described and distinguished by their electrophoretic mobilities, molecular weights, net electrical charges, and substrate specificities. The previously described leucine aminopeptidase, LAP D, consists of at least two isozymes, designated here LAP P and LAP G. In pupae most LAP activity results from LAP P (pupal); in larvae and adults, in contrast, most LAP activity results from LAP G (gut). These two LAPs may be separated by electrophoresis in the presence of the nonionic detergent Triton X-100. A specific assay for LAP P, which exploits the large difference between the net electrical charge of LAP P and that of LAP G, is described. The activity levels of two dipeptidases, Dip A and Dip B, were high in all the postembryonic stages examined. Specific assays for Dip A and Dip B were used to show that for each of these isozymes, the activity in an adult is proportional to gene dosage.

Starch gel electrophoresis and histochemical staining with l-leucyl-l-tyrosine have revealed genetic variation for dipeptidase in Rattus norvegicus. The tissue distribution, substrate specificity, and heterozygous expression as a monmeric protein suggest homology of the variant peptidase to human PEP-C and mouse Pep-3 (Dip-1). We propose Peptidase-3 (Pep-3) as a name for this autosomal locus in the rat. The allele responsible for slower (less anodal) electrophoretic migration is designated Pep-3 a and is characteristic of strain ACI/Pit. A faster (more anodal) electrophoretic mobility is the product of the Pep-3 b allele in strain F344/Pit. Twenty-five additional inbred strains carry Pep-3 a and 16 others carry Pep-3 b . Wild rats trapped in Pittsburgh were polymorphic for this locus. Alleles at Pep-3 segregated independently of c (linkage group I), a (linkage group IV), RT2 and Es-1 (linkage group V), h (linkage group VI), and RTI (linkage group VIII).

Genetic variation at three dipeptidase loci (Dip-A, Dip-B, and Dip-C) in Drosophila simulans was analyzed by starch gel electrophoresis. Dip-A was found to be polymorphic in four populations, while Dip-B and Dip-C were found to be polymorphic in one. The numbers of different alleles found at each respective locus were: Dip-A, two; Dip-B, two; and Dip-C, three. Dip-A was genetically mapped at 57.9 on the second chromosome, and Dip-B and Dip-C at 80.9 and 87.9 on the third chromosome, respectively. Neither Dip-B nor Dip-C has been mapped in D. melanogaster because both loci are apparently monomorphic. Their map positions in D. simulans with respect to flanking markers whose homologous genes have been cytogenetically localized in D. melanogaster suggested that they might be mapped cytogenetically by using available deficiencies in D. melanogaster. Accordingly, by the construction of interspecific hybrids which carried deficiencies of melanogaster and an allele of simulans with a mobility different from that of the fixed melanogaster allele, Dip-B and Dip-C were localized between 87 F12-14 and 88 C1-3 and between 87 B5-6 and 87 B8-10, respectively, in the salivary gland chromosomes of D. melanogaster. The similarity between these two species is discussed on the basis of these findings.

Starch gel electrophoresis in conjunction with a specific staining method revealed the occurrence of five distinct peptidases in mouse red blood cells. These enzymes can be distinguished on the basis of substrate specificity and electrophoretic mobility. They have been designated peptidases A, B, C, D, and E to correspond with the nomenclature adopted for human peptidases with which the mouse enzymes appear to be homologous. Genetically determined variants of peptidase C are described. The phenotype Pep C1 occurs in C57BL/Gr mice and the phenotype Pep C2 in CBA/Gr and Strong A/Gr mice. These phenotypes and the presumed heterozygote, Pep C2-1, appear to be due to the occurrence of codominant autosomal alleles which have been designated Pep-C1 and Pep-C2. F1 and F2 crosses show segregation in the expected Mendelian ratios. F2 embryos and their placentae show the same electrophoretic pattern for peptidase C. The occurrence of a separate locus controlling the structure of each distinct peptidase is postulated.

The linkages of the isozyme genes Mod-2, Pgm-2, and Dip-1 have been determined in tests with established linkage group markers among inbred strains of mice. Unique alleles for both Mod-2 and Pgm-2 have been observed in the strain of SM/J. Linkage was determined from backcross progeny of the matings C57BL/6J×(SM/J×C57BL/6J)F1, (SM/J×SWR/J)F1×SM/J, and (SM/J×SWR/J)F1×SJL/J. The gene Mod-2 is on linkage group 1. In a three-point cross of the loci Gpi-1, c, and Mod-2, the c locus was determined to be the middle gene. No double crossovers were observed. Our combined data show the following linkages: Gpi-1 to c, 28.3±3.2%; Gpi-1 to Mod-2, 33.3±3.0%; and c to Mod-2, 4.1±2.8%. The proposed gene order for four markers on LG I is Gpi-1-p-c-Mod-2. The gene Pgm-2 was linked to Gpd-1 (27.0±4.2%) on LGVIII. Two backcrosses segregating for Pgm-2 and b, (SM/J×DBA/2J) F1×DBA/2J and (SM/J×DBA/2J)F1×C57BR/cdJ, showed 9.1±4.3% recombination. The proposed gene order on LG VIII is b-Pgm-2-Gpd-1. The genes Pgm-1 and Pgm-2 are not linked (53.4±4.4%). Linkage of the isozyme genes Dip-1 and Id-1 on LG XIII was observed in backcross progeny of the crosses (SJL/J×C57BL/6J)F1×SJL/J and C57BL/6J×(SM/J×C57BL/6J)F1. The combined recombination was 23.8±2.8%. Two cases are established where genes whose enzyme products share substrate affinities (Pgm-1 and Pgm-2; Mod-1 and Mod-2) are not linked. Our data generally support the conclusion that functionally or metabolically related isozyme genes are not contiguous on mouse linkage groups.
