Abstract

A novel approach utilizing ATP-agarose affinity chromatography is employed to demonstrate that the intracellular domain of guanylyl cyclase C (GC-C) and the kinase homology domain (KHD) bind ATP. Results shows direct ATP binding to the pseudokinase domain of receptor guanylyl cyclase C, as well as to identify dramatic conformational changes that occur in this domain ATP binding, akin to those in catalytically active protein kinases.

Abstract

The residues that are located at positions corresponding to Leu159 and Ala131 in adenine phosporibosyltransferase (hAPRT) are responsible for the base specificities of type 1 PRTases are proposed. The comparative analysis shown provides structural information for the mechanism by which mutations in the human APRT lead to DHA-urolithiasis.
