Abstract

Results demonstrate that histone deacetylase inhibitors induce H4 acetylation, suggesting that this enzyme maintains low-level H4 acetylation at the Ey promoter site. Furthermore, histone deacetylase-dependent control mechanism counteracts constitutive histone acetyltransferase leading to a low-level H4 acetylation maintenance.

Abstract

The yeast transcriptional coactivator protein GCN5 was previously found to be a nuclear histone acetyltransferase (HAT). Conditions for chromatographic fractionation of HATs and adapter protein AD2 from human K562 erythroleukemia cells were determined. Two populations of GCN5 with Stokes' radii of 67 and 33 angstroms were observed. The HAT complexes were resistant to 0.3 M NaCl and DNase I. Results showed that human GCN5 and ADA2 are not stably associated with each other.

Abstract

The erythroid-specific transcription factor NF-E2 is present as distinct DNA binding forms, capable and incapable. The conforms have similar subunits but, different charge properties. Vanadate (V) increases DNA binding by increasing the number of active DNA binding molecules. The increase is not due to the suppression of the activity of a phosphate. V interacts with one of the conforms and activates it by stabilizing the molecule or mimicking a bound phosphate molecule necessary for the active state.
